Influence of high axial compression ratios in RC columns on the seismic response of MRF buildings

  • Poorly designed reinforced concrete (RC) columns of actual moment-resisting frame (MRF) buildings can undergo Axial Compression Ratios (ACR) so high as their demand exceeds their capacity, even for serviceability gravity load combinations, this lack commonly leads to insufficient seismic strength. Nonetheless, many seismic design codes do not specify limits for ACR. The main contribution of this research is to investigate the need to limit the ACR in seismic design. For this purpose, three prototype 6 and 11-story RC MRF buildings are analyzed in this paper, these buildings have columns undergoing excessive ACR, according to the limits prescribed by standards. To better that situation, three types of alterations are performed: retrofitting the abovementioned overloaded columns by steel jacketing, increasing the concrete strength, and reducing the number of stories. Several finite element analyses are conducted using the well-known software SAP2000 and the results are used for further calculations. Code-type and pushover analyses are performed on the original and retrofitted buildings, the suitability of the other modified buildings is checked by code-type analyses only. The obtained results suggest that ACR is a rather reliable indicator of the final building strength, hence, apparently, limiting the ACR in the standards (for early stages of design) might avoid unnecessary verifications.

독활의 복합 유산균 발효 추출액의 마우스에 대한 단회경구투여 독성시험 (Mouse Single Oral Dose Toxicity Test of Lactobacillus-fermented Araliae Continentalis Radix Aqueous Extracts (fACR))

  • 본 실험에서는 유산균발효 독활의 마우스 단회 경구 투여 독성 자료를 얻기 위해 식품의약품안전청 고시 제 2013-121 “의약품 등의 독성시험 기준”에 의거하여, 설치류 투여 한계 용량인 2,000 mg/kg을 최고 투여군을 설정하고 공비 2로 1,000 및 500 mg/kg 투여군을 중간 및 저용량 투여군으로 설정하여 실험을 실시하였으며, 그 결과는 독활 열수 추출물 2,000 mg/kg 암수 투여군 및 암수 매체 대조군과 비교 평가 하였다. 본 실험의 결과, 설치류 투여한계 용량인 2,000 mg/kg 투여군까지, 유산균발효 독활 열수 추출물 투여와 관련된 사망례, 임상증상, 체중, 장기중량, 육안부검 및 조직병리학적 소견이 인정되지 않았다. 따라서 유산균발효 독활 열수 추출물의 마우스에 대한 단회 경구 투여 반수 치사량 및 개략적 치사량은 암수 각각 2,000 mg/kg이상으로 산출되었으며, 특정 임상증상 및 표적 장기 역시 없는 것으로 판단되어, 유산균발효 독활은 매우 안전한 물질로 판단된다. 또한 독활 열수 추출물 2,000 mg/kg 투여와 관련된 사망례, 임상 증상, 체중, 장기중량, 육안 및 조직병리학적 변화 역시 인정되지 않았다. 이러한 결과는 독활의 활용도를 증대시키는 기초 자료가 될 것으로 사료된다.

Applying the ACR Preliminary Diagnostic Criteria in the Diagnosis and Assessment of Fibromyalgia

  • Background: Fibromyalgia (FM) is characterized by chronic widespread pain with a low pain threshold. The aim of this study was to compare two criteria for the diagnosis and assessment of FM and to analyze the correlation and agreement between the 1990 and 2010 American College of Rheumatology (ACR) preliminary diagnostic criteria for FM. Methods: We studied 98 patients who had already been diagnosed as having FM using the 1990 criteria or 2010 preliminary criteria. Tender point examination, FM impact questionnaire (FIQ) and pain visual analog scale (VAS) were obtained. According to the preliminary criteria, FM was quantified as WPI (widespread pain index) and the SS scale (symptom severity) and the two criteria were compared. Results: Among 98 patients, 78.6% of the patients were diagnosed with the 1990 ACR criteria and 93.9% of the patients were diagnosed with the ACR preliminary diagnostic criteria, and there was also significant agreement between the two criteria (P < 0.01). There was a correlation with the WPI and the tender point, with the SS and the FIQ, and with the sum of the WPI and SS and the FIQ. Conclusions: The ACR preliminary diagnostic criteria for FM were in agreement with the 1990 ACR criteria during the disease course. The preliminary criteria were the more sensitive method than the 1990 criteria. In addition, the 2010 criteria might have advantages since it is easy to assess the physical and psychological symptoms and can be quantified. Therefore, the ACR preliminary diagnostic criteria for FM could be used more conveniently for clinical diagnosis and follow up evaluation after starting management of FM.

A Study on a Trend of Human Error Types Observed in a Simulated Computerized Nuclear Power Plant Control Room

  • Objective: The aim of this study is to investigate a trend of human error types observed in a series of verification and validation experiments for an Advanced Control Room(ACR) equipped with Lager Display Panel(LDP), Work Station Flat Panel Display(WS FPD), list type Alarm System(AS), Soft Control(SC) and Computerized Procedure System(CPS). Background: Operator behaviors in a fully computerized control room are quite different from those in a traditional hard-wired control room. Operators in an ACR all together monitor plant status and variables through their own interface system such as LDP and WS FPD, are notified of abnormal plant status through their own list type AS, control the plant through their own SC, and follow the structured procedure through their own CPS whereas operators in a traditional control room only separately do their duty directed by their supervisor. Especially the secondary task such as manipulating the user interface of ACR can be an extra burden to all the operators including the supervisor. Method: The Reason's human error classification method was applied to operators' behavioral data collected from a series of verification and validation experiments where operators showed their plant operational behaviors under a couple of harsh scenarios using the ACR simulator. Results: As operators accustomed to the new ACR system, knowledge or rule based mistakes appearing frequently in the early series of experiments decreased drastically in the latest stage of the series. Slip and lapse types of errors were observed throughout the series of experiments. Conclusion: Education and training can be one of the most important factors for the operators accustomed to the traditional control room to be adapted to the new system and to run the ACR successfully. Application: The results of this study implied that knowledge or rule based mistakes can be reduced by training and education but that lapse type errors might be reduced only through innovative improvement in human-system interface design or teamwork culture design including a new leadership style suitable for ACR.

THP-1 세포에서 융합 발효 독활의 항염증 효과 (Anti-Inflammatory Effects of Fusion-Fermented Aralia continentalis Radix (fACR) on THP-1 cells)

  • 본 연구는 독활의 기능을 부가시키고자 발효과정을 통해 융합-발효 추물물을 얻어 항염증효과를 확인 하고자 한다. 독활을 이용하는데 한계가 있어 유산균주들을 활용한 발효과정을 통해 이용 가능한 균주를 선별하고, 독활의 열수 추출물과 비교하여 총 폴리페놀, 아미노산 총량, 주요 생리활성 미네랄 성분 분석 결과 발효과정을 통해 증가함을 확인하였다. 독활의 발효추출물의 항염증 효과를 확인하고자 관절염 염증반응과 관련된 사이토카인들을 측정하였고, 관절염시 주로 동반되는 연골조직 파괴를 억제 효능을 평가하기 위해 MMP-9 활성도를 확인하였다. 이러한 결과들은 유산균을 이용한 발효과정을 통해 독활의 주요 생리활성이 증가하고 염증반응 억제 효과도 증가하는 것으로 보여진다. 이 연구는 독활의 관절염 예방적 효과를 연구하는데 있어 기초 연구가 될 것으로 기대된다.

Structure of the Tripartite Multidrug Efflux Pump AcrAB-TolC Suggests an Alternative Assembly Mode

  • Escherichia coli AcrAB-TolC is a multidrug efflux pump that expels a wide range of toxic substrates. The dynamic nature of the binding or low affinity between the components has impeded elucidation of how the three components assemble in the functional state. Here, we created fusion proteins composed of AcrB, a transmembrane linker, and two copies of AcrA. The fusion protein exhibited acridine pumping activity, suggesting that the protein reflects the functional structure in vivo. To discern the assembling mode with TolC, the AcrBA fusion protein was incubated with TolC or a chimeric protein containing the TolC aperture tip region. Three-dimensional structures of the complex proteins were determined through transmission electron microscopy. The overall structure exemplifies the adaptor bridging model, wherein the funnel-like AcrA hexamer forms an intermeshing cogwheel interaction with the ${\alpha}$-barrel tip region of TolC, and a direct interaction between AcrB and TolC is not allowed. These observations provide a structural blueprint for understanding multidrug resistance in pathogenic Gram-negative bacteria.

자기공명영상에서 ACR 팬텀을 이용한 해상력 특성 평가 (Evaluation of the Resolution Characteristics by Using American College of Radiology Phantom for Magnetic Resonance Imaging)

  • This study was purpose to quantitative assessment of the resolution characteristics by using American college of radiology(ACR) phantom for magnetic resonance imaging (MRI). The MRI equipment was used (Achiva 3.0T MRI, Philips system, Netherlands) and the head/neck matrix shim SENSE head coil were 32 channels(elements) receive MR coil. And the MRI equipment was used (Discovery MR 750, 3.0T MRI, GE medical system, America) and the head/neck matrix shim MC 3003G-32R 32-CH head coil were receive MR coil. As for the modulation transfer function(MTF) comparison result by using ACR magnetic resonance imaging phantom, the MTF value of the ACR standard T2 image in GE equipment is 0.199 when the frequency is 1.0 mm-1 and the MTF value of the hospital T2 image in Philips equipment is 0.528. It was used efficiently by using a general sequence more than the standard sequence method using the ACR phantom. In addition it is significant that the quantitative quality assurance evaluation method for resolution characteristics was applied mutatis mutandis, and the result values of the physical image characteristics of the 3.0T MRI device were presented.

Evaluation of Albumin Creatinine Ratio as an Early Urinary Biomarker for Chronic Kidney Disease in Dogs

  • Chronic kidney disease (CKD) occurs in more than 15% of the dogs over 10 years of age and causes irreversible renal function deterioration. Therefore, it is important to diagnose CKD early and treat the disease properly. The purpose of this study aimed to to evaluate the clinical utility of urine albumin/creatinine ratio (ACR) using POC (point-of-care) device as an early detection urinary biomarker in CKD dogs and to confirm the correlation between ACR and other known CKD biomarkers. Urine and serum samples were obtained from 50 healthy dogs and 50 dogs with CKD. Serum blood urea nitrogen (BUN), creatinine, and symmetric dimethylarginine (SDMA) concentrations, and urine protein creatinine ratio (UPC) were measured. Urine specific gravity (USG) was evaluated using refractometer, and ACR was measured using an i-SENS A1Care analyzer. The ACR values of dogs with CKD were significantly different from those of healthy dogs (p < 0.001), as with other renal biomarkers. ACR showed significant differences between healthy dogs and dogs with CKD at every IRIS stage (p < 0.005), whereas no significant differences were observed between dogs with CKD IRIS stage I and healthy dogs with UPC. There are significant positive correlation between ACR and BUN (r = 0.611, p < 0.001), creatinine (r = 0.788, p < 0.001), SDMA (r = 0.747, p < 0.001), and UPC (r = 0.784, p < 0.001), and significant negative correlation between ACR and USG (r = -0.700, p < 0.001). In receiver operator characteristic curve analysis, the area under the curve (AUC) was 0.982 (95% CI 0.963-1.000, p < 0.001), with an optimal cut-off value of 64.20 mg/g (94% sensitivity and 94% specificity). Thus, ACR is a useful urinary biomarker for the early diagnosis of proteinuria in CKD and combined use of ACR and other renal biomarkers may be helpful for early diagnosis and prevention of CKD in dogs.

백서의 조건회피반응-유지에 대한 경련성 전기충격의 저해작용에 미치는 CDP-Choline, Aminoguanidine, 및 Difluoromethylornithine의 영향에 관한 연구 : 뇌내 Acetylcholine과 Polyamine 함량-변동에 연관하여 (Effects of CDP-Choline, Aminoguanidine and Difluoromethylornithine on the ECS-induced Impairment of Active Conditioned Response Retention)

  • Two-way shuttle box에서 active conditioned response (ACR)을 10일간 매일 30회 부하하여 10회 이상 반응한 웅성 Wistar 백서를 이용하여, 경련성 전기충격 (ECS: 50 mA, 100 Hz, 1.5 sec)에 의한 ACR-유지의 변동에 미치는 CDP-choline (250 mg/kg/day/CC), aminoguanidine (100 mg/kg/day: AG), ${\alpha}-difluoromethylornithine$ (250 mg/kg/day, DO), 및 spermine (10/mg/kg/day: SM) 각각의 10일간 복강내-주사의 영향을 검토하였다. 그 결과, 10일간 획득된 ACR은 그 다음 10일후에 더욱 증가되었으며, 이는 일일 간격의 5회 ECS (5-ECS) 또는 ACR-유지검사 3시간전 일회 ECS (ECS-3h)로 현저히 억제되나, ACR-유지검사 24시간전의 일회 ECS (ECS-24h)로는 별 영향을 받지 않았다. 아울러, ACR의 자연증가현상은 CC와 SM에 의하여 현저히 저하되었으나 AG와 DFMO에 의하여는 영향을 받지 않았다. 5-ECS에 의한 ACR-유지저해는 AG, SM, 및 CC에 의하여 유의하게 되었으며 DFMO에 의하여는 영향을 받지 않았다. ECS-3h 후의 ACR-유지저해는 SM와 AG에 의하여 다소 더 악화되었고 CC와 DFMO에 의하여는 별 영향을 받지 않았는데, 정상 백서에 비하여 큰 차를 보이지 않은 ECS-24h 후의 ACR은 CC, SM, DO, 및 AG에 의하여도 영향을 받지 않았다. 한편, ECS-3h와 ECS-24는 백서-대뇌 시상하부(HT), 해마(HC), 및 내후피질(EC)의 acetylcholine (ACh) 함량에 별 영향을 미치지 않았으나, $5{\times}ECS$는 다소 증가시켰으며 특히 EC의 ACh증가는 유의하였다. 아울러 CC과 SM도 대뇌-ACh 함량을 유의하게 증가시켰으나, DO와 AG는 별 영향을 미치지 않았다. ECS-3h와 ECS-24h는 대뇌 HT, HC, 및 EC의 polyamine 함량에 영향을 미치지 않았으나, $5{\times}ECS$는 HT와 HC의 putrescine (Pt) 함량과 HC의 spermine (Sm) 함량을 각각 유의하게 감소시켰다. CC는 PT함량에 영향을 미치지 않았으나 spermidine (Sd)와 Sm 함량은 현저히 증가시켰다. 그러나 이같은 CC의 작용을 ECS-3 시간후와 24시간후에는 전혀 볼 수 없었을 뿐 아니라, ECS-3 시간후의 HC의 Sd 함량은 정상치보다 유의한 감소로 역전되었다. 또한 CC는 $5{\times}ECS$에 의한 Pt와 Sm 함량-감소를 유의하게 억제하였다. SM은 전 부위의 Sd 함량과 EC의 Sm 함량을 유의하게 증가시켰고, 이같은 증가가 ECS-3 시간후에 더욱 상승되었으며, 이때 HC와 EC의 Pt함량도 유의하게 증가되었다. 또한 SM은 $5{\times}ECS$에 의해 Pt와 Sm 함량-감소를 유의하게 억제하였다. 그러나 DO는 HC의 Sd와 모든 부위의 Sm 함량을 유의하게 감소시켰고 또한 Pt와 Sm에 대한 $5{\times}ECS$의 감소작용을 더 상승시켰다. AG는 모든 부위의 Pt 함량을 현저히 증가시켰을 뿐 아니라 HT의 Sd와 모든 부위의 Sm도 유의하게 증가시켰고, $5{\times}ECS$에 의한 Pt와 Sm 함량-감소를 유의하게 억제하였다. 이상의 성적은 $5{\times}ECS$로 나타나는 ACR의 유지-저해에 대한 aminoguanidine의 보호작용의 일부가 polyamine 대사에 대한 그의 diamine oxidase 억제작용에 기인됨을 시사하는 것으로 사료된다.

Backbone NMR assignments of the anti-CRISPR AcrIIA5 from phages infecting Streptococcus thermophilus

  • The CRISPR-Cas system provides an adaptive immunity for bacteria and archaea against invading phages or foreign plasmids. In the type II CRISPR-Cas system, a single effector protein Cas9 and a guide RNA form an RNA-guided endonuclease complex that can degrade DNA targets of foreign origin. To avoid the Cas9-mediated destruction, phages evolved anti-CRISPR (Acr) proteins that neutralize the host bacterial immunity by inactivating the CRISPR-Cas system. Here we report the backbone 1H, 15N, and 13C resonance assignments of AcrIIA5 that inhibits the endonuclease activity of type II-A Streptococcus thermophilus Cas9 and also Streptococcus pyogenesis Cas9 using triple resonance nuclear magnetic resonance spectroscopy. The backbone chemical shifts of AcrIIA5 predict a disordered region at the N-terminus, followed by an αββββαβββ fold.
