Antioxidant and Antihypertensive Activities of Styela plicata according to Harvesting Time and Size (채취시기와 크기에 따른 주름 미더덕의 항산화 및 항고혈압 활성)

  • Styela plicata of large (9.82~11.66 g) and small (0.93~2.21 g) sizes harvested at different times was extracted with 4 different solvents (methanol, ethanol, acetone, and water). DPPH radical scavenging and reducing power was the highest in acetone extracts of small ones (38.98% and 1.724, respectively) harvested in November. The lowest radical scavenging activity was found in water extracts of large ones (12.03% and 0.114) in December. On the other hand, large ones harvested in September showed significantly higher inhibition rate of DNA damage (water, 56.54%; methanol, 55.83%; ethanol, 48.63%) than others. Overall, the water extraction of S. plicata tended to show a higher antigenotoxic effect. In addition, water extracts of large S. plicata from November showed the highest angiotensin converting enzyme (ACE) inhibitory activity (64.38%), while that of small ones from December exhibited the lowest (51.33%). Overall the results indicate that antioxidant and antihypertensive activities S. plicata are variable depending on harvesting time, size, and extraction solvent.

Effect of Antihypertensive Peptides Originated from Various Marine Proteins on ACE Inhibitory Activity and Systolic Blood Pressure in Spontaneously Hypertensive Rats (어육단백질로부터 분리된 항고혈압 펩타이드가 ACE 저해활성과 본태성 고혈압쥐의 혈압에 미치는 영향)

  • This study was carried out to investigate the effect of antihypertensive peptides originating from marine proteins on ACE inhibitory activity and systolic blood pressure in spontaneously hypertensive rats (SHR). Sixteen male SHR (SHR/NCrj) weighing approximately 270 g were randomly divided into few experimental groups based on diet: C (control), A (anchovy), P (pollack) and M (mackerel). The final body weights of P and M groups were higher, than those of C and A groups, but difference was not significant. Average reference blood pressure (RBP) was 224 mmHg at 12 weeks old. Compared with RBP, final systolic blood pressure of the marine peptide oops after 28 days of feeding with anchovy, pollack and mackerel fractions by gavage was decreased by 9.0% (A), 10.2% (P) and 14.3% (M), respectively, but was not different in C. Especially, final blood pressure of M was lower by 32 mmHg than RBP. These results suggested that peptide originated from mackerel hydrolysate was considered to have an antihypertensive fraction as effective lowering of blood pressure in SHR.

Quality characteristics and antioxidant activity of onion peel extracts by extraction methods (추출방법에 따른 양파껍질 추출물의 품질특성 및 항산화활성 비교)

  • This study was conducted to evaluate the antioxidant activities and the quality characteristics of 70% ethanol extracts from onion peels with different extraction methods (autoclave extraction, AE; low temperature high pressure extraction, LTPE; reflux extraction, RE; and stirrer extraction, SE). The yields of AE, LTPE, RE and SE were 9.00%, 5.39%, 13.21% and 12.41%, respectively. The total polyphenol and flavonoid contents in the AE were significantly higher than in the other extracts. The DPPH radical scavenging abilities with a concentration of 100 mg% (w/v) were : AE, 28.9%; RE, 26.07%; LTPE, 24.35%; and SE, 19.53%. The ABTS radical scavenging ability and the nitrite scavenging activity showed the same tendency as that of the DPPH radical scavenging ability. The angiotensin-converting enzyme (ACE) inhibitory activities of the LTPE and AE were higher than those of the RE and SE. The ${\alpha}$-Glucosidase inhibitory activity of the RE was higher than that of the extracts with other extraction methods. The nitrite scavenging activities with a concentration of 10 mg/mL were: AE. 33.97%; RE, 35.47%; LTPE, 21.86%; and SE, 21.71%. The ferrous ion chelating activity of the LTPE (54.73%) was significantly higher than that of the other extracts. These results suggest that AE is the superior method for the enhancement of anti-oxidant activity, and onion peel can be used as a natural antioxidant material for health foods and can be a good ingredient of functional foods.

Physiological Activity and Antioxidative Effects of Aged Black Garlic (Allium sativum L.) Extract (숙성에 의해 제조된 흑 마늘 추출물의 생리학적 활성 및 항산화 효과)

  • In this study, black garlic was produced by aging under high temperature $(70^{\circ}C)$ and high humidity (90% RH) conditions. Then, the physiological activity and antioxidative effects of its extract were compared to those of normal garlic extract. The black garlic extract had a 2.5-fold higher total polyphenol content than that of the normal garlic extract, showing levels of 10.0 mg/g and 3.7 mg/g, respectively. At the $1,000{\mu}g/mL$ concentration, the black garlic and normal garlic extracts had electron donating abilities of 101.9% and 12.9%, respectively. For their nitritescavenging effects, the normal garlic extract showed slightly higher scavenging activity than the black garlic extract at the 5 mg/mL concentration; whereas the black garlic extract had a slightly higher effect at concentrations above 20 mg/mL. In terms of their superoxide dismutase activities, the black garlic extract showed a 10-fold higher activity as compared to the normal garlic extract at the 20 mg/mL concentration. Furthermore, at 50 mg/mL, the angiotensin converting enzyme inhibitory effects of the normal garlic and black garlic extracts were approximately 52.7% and 88.8%, respectively. These results indicate that the antioxidant activity and ACE inhibitory effects of the black garlic extract were greater than those of the normal garlic extract.

Antioxidant activity and physiological properties of Moringa (Moringa oleifera Lam.) leaves extracts with different solvents (추출용매에 따른 모링가(Moringa oleifera Lam.) 잎의 항산화 및 생리활성 효과)

  • This study was conducted to investigate the antioxidant activity and physiological properties of Moringa (Moringa oleifera Lam.) leaves extracted with three different solvents (water, ethanol, and methanol). The extraction yield from water, methanol, and ethanol were 13.17, 9.54, and 7.48%, respectively. The highest total polyphenol content (58.04 mg/100 g) and total flavonoid contents (12.36 mg/100 g) were observed in water extract. The DPPH radical scavenging activity was the highest in the water extract (79.18%) at the 500 mg% level, similar to BHT (77.18%). Additionally the same tendency was observed with DPPH, ABTS radical scavenging ability, and ferreous ion chelating ability. The water extract showed relatively high antioxidant activities. The angiotensin I-converting enzyme (ACE) and the HMG-CoA reductase inhibitory activity of water extract at a concentration of 500 mg% were somewhat higher than those of the other extracts. Additionally, the HMG-CoA reductase inhibitory activity of the water extract was significantly slightly lower than that of the positive control (cholorogenic acid). These results suggest that Moringa leaves extracted with water will be useful as antioxidant-rich and functional natural foods.

Isolation and Characterization of Plant-Derived Lactic Acid Bacteria as Potential Probiotic (잠재적 생균제로서 식물 젖산균의 분리 및 특성)

  • Plant lactic acid bacteria were isolated from plant-associated fermentative foods and crops, and their probiotic properties were investigated. Isolates K27 and O2 were isolated from Kimchi and onion, and identified as Lactobacillus plantarum on the basis of 16S rRNA gene analysis. The two strains were highly resistant to acid (an MRS broth at pH 2.5), where the survival rates of L. plantarum K27 and L. plantarum O2 were 90.2% and 97.3%, respectively. L. plantarum K27 and L. plantarum O2 also showed high bile resistance to 0.5% oxgall, with a more than 70% survival rate. They showed an inhibitory effect against pathogenic strains of Escherichia coli KCCM 40880 and Pseudomonas aeruginosa ATCC 10145. The antibacterial effect of the two strains was probably due to the presence of lactic acid. ACE inhibitory activities of the two strains ranged from 72.8% to 80.6% in MRS broth. Notably, the two strains showed high ACE inhibitory activity (89.2~98.2%) in MRS broth containing 10% skim milk. Antioxidant activity was tested by DPPH radical scavenging activity, with antioxidant activities of the strains being in the range of 56.8~61.5%. The results obtained in this study suggest that L. plantarum K27 and L. plantarum O2 may be potential probiotic starter cultures with applications with fermentative products.

Bioactive Properties of Novel Probiotic Lactococcus lactis Fermented Camel Sausages: Cytotoxicity, Angiotensin Converting Enzyme Inhibition, Antioxidant Capacity, and Antidiabetic Activity

  • Fermented products, including sausages, provide several health benefits, particularly when probiotics are used in the fermentation process. This study aimed to examine the cytotoxicity (against Caco-2 and MCF-7 cell lines), antihypertensive activity via angiotensin-converting enzyme (ACE) inhibition, antioxidant capacity, antidiabetic activity via α-amylase and α-glucosidase inhibition, proteolysis rate, and oxidative degradation of fermented camel and beef sausages in vitro by the novel probiotic Lactococcus lactis KX881782 isolated from camel milk. Moreover, camel and beef sausages fermented with commercial starter culture alone were compared to those fermented with commercial starter culture combined with L. lactis. The degree of hydrolysis, antioxidant capacity, cytotoxicity against Caco-2 and MCF-7, α-amylase, α-glucosidase, and ACE inhibitory activities were higher (p<0.05) in fermented camel sausages than beef sausages. In contrast, the water and lipid peroxidation activity were lower (p<0.05) in camel sausages than beef sausages. L. lactis enhanced the health benefits of the fermented camel sausages. These results suggest that camel sausage fermented with the novel probiotic L. lactis KX881782 could be a promising functional food that relatively provides several health benefits to consumers compared with fermented beef sausage.

Inhibitory Effects of Maesaengi (Capsosiphon fulvescens) Extracts on Angiotensin Converting Enzyme and α-Glucosidase (매생이 추출물의 angiotensin converting enzyme 및 α-glucosidase 활성 저해 효과)

  • Physiological activities of hot water (MHW) and 80% ethanol (MEH) extracts from Maesaengi (Capsosiphon fulvescens) were investigated in this study. For the evaluation of antioxidant activities for MHW and MEH, 2,2-diphenyl-1-pic-rylhydrazyl (DPPH) radical scavenging activity and superoxide dismutase (SOD)-like activity were measured. DPPH radical scavenging activity and SOD-like activity of MHW, and MEH were increased weekly in a dose-dependent manner, and were about 10.8, 13.8, 62.4, and 27.1% at 10 mg/ml, respectively. The angiotensin converting enzyme (ACE) inhibitory activities of MHW and MEH were about 5.9% and 49.7% at 1 mg/ml, respectively. The ${\alpha}$-glucosidase inhibitory effect of MHW and MEH were about 1.4% and 67.3% at 1 mg/ml, respectively. To determine the influence of MHW and MEH on alcohol metabolizing activity, the generating activities of reduced-nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) were measured. Facilitating rates of ADH activity by MHW and MEH were increased weekly in a dose-dependent manner and ALDH activities were not detected. Elastase inhibitory activities of MHW and MEH were 75.9% and 51.2% at 10 mg/ml, respectively.

  • The phenolics of Sambucus sieboldiana var. pendula leaves for functional resources were examined on inhibitory activity against biological enzyme and anti-microbial activity. The amount of phenolic compounds were 11.60±0.18 and 12.43±0.07 mg/g by water and 50% ethanol extraction, respectively. The antioxidative activity of phenolic in extracts was higher than solids. The inhibition activities on angiotensin converting enzyme were 92.08 and 78.33% at 200 ㎍/mL phenolic concentration in water and ethanol extracts, respectively. The xanthine oxidase inhibitory activity were 100% at 200 ㎍/mL phenolic in water and ethanol extracts, respectively. These result was higher than 70.37% of allopurinol as positive control at 200 ㎍/mL. The inhibitory activity against hyaluronidase were each 25.35 and 43.38% in water and ethanol extracts. The water extract from S. sieboldiana var. pendula leaves showed antibacterial activity on the Propionebacterium acnes, Staphylococcus aureus and Streptococcus mutans in water extract and S. mutans in ethanol extract. This result suggests that phenolic from S. sieboldiana var. pendula leaves are suitable as functional foods with anti-hypertension, anti-gout, anti-inflammation and anti-microorganism activities.

Chemical Components and Comparison of Biological Activities on the Fruit of Natural Bogbunja (Rubus coreanus Miquel) (국내산 복분자 열매에 대한 화학적 조성 및 생리활성 비교)

  • This study was carried out to investigate biological and antioxidative activities on the fruit of bogbunja (Rubus coreanus Miquel). Total contents of phenolic compounds contents in cultivars ripened fruit and immatured wild-type fruit were 222 and 190mg/g, respectively, Polysaccharide contents of immatured wild-type fruit were the highest value of 320U. For EDA analysis, immatured wild-type fruit showed over 95% in 100$\mu\textrm{g}$/mL of sample concentration, which is the the most effective. Levels of SOD-like activities in immatured and cultivar ripened fruits were 81% and 77%, respectively, For the inhibitory effect on lipid peroxidation, all of bogbunja prepared were similar with those of $\alpha$-tocopherol as control. The inhibition of ACE activities on the water extracts of bogbunja fruit showed over 98%, especially, in immature wild-type and cultivar bogbunja.
