Effects of Oxygen-Derived Free Radicals on Brain Microsomal $Na^+-K^+-ATPase$ Activity (산소유리라디칼이 뇌조직 미크로좀분획의 $Na^+-K^+-ATPase$ 활성도에 미치는 영향)

  • The effects of xanthine-xanthine oxidase reaction on brain microsomal $Na^+-K^+-ATPase$ activity were studied to see possible involvement of oxygen free radicals in pathologic change occurring in ischemic state of CNS accompanied by cerebral vascular occlusion or impact injury. When microsomal fraction was incubated with xanthine ana xanthine oxidase, $Na^+-K^+-ATPase$ activity of the fraction was markedly inactivated (80% inactivation) whereas btssl $Mg^{++}-ATPase$ was much less sensitive (less than 10% inactivation) compared to that of $Na^+-K^+-ATPase$. The inactivation was observed only in the presence of both xanthine and xanthine oxidase, not either of them alone, and the extent of inactivation was dependent on the concentration of xanthine. In an attempt to determine which of the oxygen species was responsible for the inactivation, the ability of various scavengers to overcome the inactivation was tested. Superoxide dismutase, catalase and 1,4-diazabicyclo(2,2,2)octane were shown to reverse the inactivation of the ATPase in dose-dependent manner. In contrast, mannitol as well as other $OH{\cdot}$quenchers were ineffective in limiting oxygen radical-induced inactivation. Thus $O_{\bar{2}}{\cdot},\;H_2O_2$ and $^1O_2$ were implicated to be mediators involved in the inactivation. Since oxygen radicals are suspected as being a cause of the peroxidative damaging process in train ischemia, the ATPase inactivation by oxygen radicals may be a possible contributing factor which gives rise to functional derangement of nerve cells observed in the pathologic process.

Dynamics of Phytoplankton and Zooplankton of a Shallow Eutrophic Lake (lake llgam) (수심이 얕은 부영양 인공호(일감호)의 동 ${\cdot}$ 식물플랑크톤 동태학)

  • This study was attempted to understand seasonal dynamics of phyto- and zooplankton communities in shallow, eutrophic Lake llgam and to compare them with the PEG (Plankton Ecology Group) model. Seasonal succession pattern of phytoplankton community was similar to PEG model as Chlorophyceae and Baciliphyceae increase during spring and autumn fellowed by increase of Cyanophyceae. However, based on the cell density and biomass, a dominant phytoplankton community differed with PEG model: Cyanophyceae had been a dominant community throughout a year, except for ice-cover period during which Chlorophyceae was a dominant group. In spring, when ice melted and dissolved nutrients in water column increased, the increase of Chlorophyceae occurred: when nutrients (DIN and DIP) rapidly decreased, Cyanophyceae increase occurred. Microcystis, Oscillatoria, Lyngbya, Merismopedia were maior dominant species of Cyanophyceae and their cell density and/or biomass was the highest in October 2000 (12.9${\pm}$5.8${\times}10^5$ cells/ml, 3.5${\pm}$0.9${\times}10^3{\mu}gC/l$). Cyanophyceae biomass showed positive relationship with chlorophyll a ($r^2$ = 0.71,P< 0.001) and TP concentration ($r^2$ = 0.62, P< 0.001). Small-sized rotifers such as Keratella cochlearis, increased between March and May when Chlorophyceae increased. Both high standing crop of copepods and cladocerans, such as Diaphanosoma brachyrum and Bosmina longirostris occurred between June and September accompanied with the increase of Dinophyceae and Bacillariophyceae. There was no evidence that clear-water phase was caused by zooplankton grazing. The diversity and evenness index of phyto- and/or zooplankton increased with chlorophyll a concentration. These results suggest zooplankton grazing and limiting nutrient deficiency could lead to change of phytoplankton biomass, but not the phytoplankton community in Lake llgam.

  • In Summer 2008, a multidisciplinary survey was conducted onboard R/V Haeyang 2000 to understand plankton response to the three distinct physico-chemical settings that developed in the Ulleung Basin of the East Sea. Baseline settings of hydrographic conditions included the presence of the thin (<20 m) Tsushima Surface Water (TSW) on top of the Tsushima Middle Water (TMW). It extends from the Korea Strait to $37^{\circ}N$ along the $130^{\circ}E$ and then turns offshore and encompasses the relatively saline (T>$26^{\circ}C$, S>33.7) Ulleung Warm Eddy surface water centered at $36.5^{\circ}N$ and $131^{\circ}E$. A relatively colder and saline water mass appeared off the southeastern coast of Korea. It was accompanied by higher nutrient and chlorophyll-a concentrations, suggesting a coastal upwelling. Most of the offshore surface waters support low phytoplankton biomass (0.3 mg chl-a $m^{-3}$). A much denser phytoplankton biomass (1-2.3 mg $m^{-3}$) accumulated at the subsurface layer between 20-50 m depth. The subsurface chlorophyll-a maximum (SCM) layer was closely related to the nutricline, suggesting an active growth of phytoplankton at depth. The SCM developed at shallow depth (20-30 m) near the coast and deepened offshore (50-60 m). A fucoxanthin/zeaxanthin ratio was high in coastal waters while it was low in offshore waters, which indicated that diatoms dominate coastal waters while cyanobacteria dominate offshore waters. The community structure and biomass of phytoplanktonare closely related to nitrogen availability. Zooplankton biomass was higher in the coastal region than in the offshore region while species richness showed an opposite trend. Zooplankton community structure retained a coastal/offshore contrast. These suggest that summer hydrography is a stable structure, lasting long enough to allow a hydrography-specific plankton community to evolve.

The Induction of ROS-dependent Autophagy by Particulate Matter 2.5 and Hydrogen Peroxide in Human Lung Epithelial A549 Cells (미세먼지와 산화적 스트레스에 의한 인간 폐 상피 A549 세포에의 ROS 의존적 자가포식 유도)

  • Recently, interest in the harmful factors of particulate matter (PM), a major component of air pollution, has been increasing. In particular, PM2.5 with a diameter of less than 2.5 ㎛ is well known to induce oxidative stress accompanied by autophagy in human lung epithelial cells. However, studies on whether PM2.5 increases autophagy under oxidative stress and whether this process is reactive oxygen species (ROS)-dependent are insufficient. Therefore, in this study, we investigated whether PM2.5 promotes autophagy through the generation of ROS in human alveolar epithelial A594 cells. According to our results, cells co-treated with PM2.5 and hydrogen peroxide (H2O2) showed a lower cell viability than cells treated with each alone, which was associated with increased total and mitochondrial ROS production. The co-treatment of PM2.5 and H2O2 also increased autophagy induction, which was confirmed through Cyto-ID staining, and the expression of autophagy biomarker proteins increased. However, when ROS generation was artificially blocked by N-acetyl-L-cysteine pretreatment, the reduction in cell viability and induction of autophagy by PM2.5 and H2O2 co-treatment were markedly attenuated. Therefore, the present results suggest that PM2.5-induced ROS generation may play a critical role in autophagy induction in A549 cells.

Isoalantolactone Inhibits the Formation of Multicellular Tumor Spheroids Derived From Human Hepatocellular Carcinoma Hep3B Cells Through the Induction of ROS-dependent Apoptosis (ROS 의존적 세포사멸 유도를 통한 isoalantolactone의 인간 간세포암종 Hep3B 세포 유래 다세포 종양 spheroid 형성의 억제)

  • Although two-dimensional (2D) monolayer cell culture models are still widely used as the optimal models for anticancer activity research, three-dimensional (3D) multicellular tumor spheroid (3D MTS) models that can better approximate the tumor environment can offer an alternative to bridge the gap between in vitro and animal model studies. Isoalantolactone is among the sesquiterpene lactones found in medicinal plants, including the roots of Elecampane (Inula helenium L.), and is known to have various pharmacological activities, including anticancer activity. In this study, we investigated whether the anticancer activity of isoalantolactone observed in 2D models could be reproduced in a 3D MTS model derived from human hepatocellular carcinoma (HCC) Hep3B cells. According to our results, isoalantolactone inhibited the formation of MTSs in a manner dependent on the treatment concentration, which was accompanied by an increase in reactive oxygen species (ROS) generation. In particular, as isoalantolactone treatment and the culture time increased, the area of proliferating cells was replaced by cells in which apoptosis was induced. Additionally, in MTSs, isoalantolactone increased the expression of death-receptor-related proteins and the activity of caspase-3, and it decreased the expression of the Bax/Bcl-2 expression ratio and total poly(ADP-ribose) polymerase. However, when the production of ROS was artificially blocked, all these changes caused by isoalantolactone were attenuated and the cell survival rate of MTS cells was restored. Therefore, the results of this study suggest that the induction of apoptosis in Hep3B cell-derived MTSs by isoalantolactone is achieved through the activation of extrinsic and intrinsic pathways and is ROS-dependent.

Studies on the Fatty Acid Composition of Lipids from Some Seeds of the Cucurbitaceae Family (박과식물(科植物) 종자유(種子油)의 지방산(脂肪酸) 조성(組成))

  • Levels of total, neutral and polar lipids from the seeds of eight species of the Cucurbitaceae f Cucurbita moschata, Lufa cylindrica, Citrullus vulgari, Cucumis melo var. makuwa, Cucumis satvus, Lag leucantha. Trichosanthes kirilowii and Momordica charantia, were determinded, and their fatty compositions were also analyzed by gas-liquid chromatography. The results were summarized as foll. Lipid contents of the seeds range from 21.9 to 50.7%, which contained 98% up of neutral lipi the fatty acid compositon of ottal lipids from the seeds of Cucurbita moschata, Lufa cylindrica, Ci vulgari, Cucumis melo var. makuwa, Cucumis sativus and Lagenaria leucantha, linoleic acid is the mos dominant component(56.8${\sim}$84.0%) followed by oleic acid(5.7${\sim}$22.2%) and palmitic acid(6.1${\sim}$1) with a trace amount of ${\alpha}-linolenic$ acid(below 0.6%). On the contrary, the seed oils of Tricho kirilowii and Momordica charantia are characterized by presence of considerable amounts of con trienoic acid such as punicic acid($_{9c.11t.13c-}C_{18:3}$) and ${\alpha}-eleostearic$ acid($_{9c.11t.13c-}C_{18:3}$). For example total lipids of T. kirilowii seeds were mainly composed of linoleic acid(40.5%) and punicic acid(3) in the fatty acid composition, while those of M. charantia seeds predominantly comprised ${\alpha}-eleos$ acid as a main component(66.9%), accompanied by oleic acid(11.7%) and linoleic acid(10.4%). oil ${\beta}-eleostearic$ acid($_{9t.11t.13c-}C_{18:3}$) was checked as a trace. Fatty acid profiles of neutral lipids close resemblance to those of total lipids in all the seed oils, but are different from those of polar In particular, conjugate trienoic acids including punicic acid and ${\alpha}-eleostearic$ acid which are oc as the most abundant component in both neutral lipids of T. kirilowii and M. charantia seed oils, ar ent in a extremely small amount in both polar lipids. The fatty acid distribution in the polar lipid the samples except for T. kirilowii and M. charantia seed oils, showed a tendency of consid increased level of saturated fatty acids(25.0${\sim}$29.4%) compared with that in the neutral lipids(9.9%).
