Analysis on Antioxidant Activity and Agronomic Characteristics of Extract from Smilacis Chinae Radix

  • The Smilacis chinae Radix refers to the root of Smilax chinae L distributed in mountain and filed of Korea, and it is a vine shrub in the Lilaceae family, called Berchemia berchemiaefolia, and is referred to as Smilacis chinae Radix in it's a natural medicine name. Antibacterial, inflammatory, and antioxidant activity were studied in Smilacis chinae Radix. In this study, biological activities such as antioxidant (DPPH, ABTs, TPC), cytotoxicity, wrinkle improvement, and whitening improvement to increase the utilization value of Smilacis chinae Radix and identify the botanical value. Therefore, we tried to explore the applicability of Smilacis chinae Radix as a functional cosmetic material. Smilacis chinae Radix (SCR) was dried and extracted with ethanol. In order to measure the biological activity of the SCR, antioxidant activity, inhibition activities of collagenase, tyrosinase and cell viability were measured. The DPPH (1,1-diphenyl-2-picryl hydrazyl) radical scavenging activity in the extract with a concentration of 400㎍/mL is 91.22% ± 0.41%%. ABTs (2,2'-azinobis-3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging activity in the extract with a concentration of 400㎍/mL is 99.60% ± 0.03%. Total polyphenol contents (TPC) are 0.203 ± 0.05 mg GAE/mg Ext when SCR was lmg/mL. And the Cell viability for HaCaT derived human keratinocyte and Raw264.7, a mouse-derived macrophage was determined using the MTT assay. When cell was treated with 100㎍/mL of SCR, HaCaT cell showed cell viability of 78.09 ± 0.1% and Raw264.7 cell showed cell viability of 91.88 ± 0.42%. From the above results, we have shown the possibility that the CSR have antioxidant ability, inhibition activity of collagenase and tyrosinase and cell safety ability which can be useful in a functional cosmetic material.

쪽의 항산화 및 항염증 활성에 대한 연구 (Study on Antioxidant and Anti-inflammatory Activities of Persicaria tinctoria)

  • Objectives : Persicaria tinctoria belongs to the Polygonaceae family and it has been used as the natural dye traditionally. Also, it is well known that the Persicaria tinctoria is used for treating the following symptoms such as fever, inflammation and edema. The purpose of this study is to investigate the effective source of antioxidants and anti-inflammatory agent from various parts of Persicaria tinctoria.Methods : We investigated the antioxidative and anti-inflammatory properties of the Persicaria tinctoria extracts. Antioxidant activities were measured by 1,1-diphenyl-2- picrylhydrazyl (DPPH), 2, 2'-Azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity, Fe2+ chelating activity and Reducing power of Persicaria tinctoria extracts. And its inhibitory effect against oxidative DNA damage was evaluated in non-cellular system using φX-174 RF I plasmin DNA. The anti-inflammatory effect of Persicaria tinctoria was measured by using the inhibitory efficacy for the amount of nitric-oxide (NO) produced in LPS induced RAW264.7 cells.Results : The extracts from stem part showed better DPPH scavenging activity compared to those of the leaf and root extracts. Their IC50s were measured as 7.17, 144.40 and 165.07 ug/ml, respectively. These results were similar to that of ABTS radical scavenging assay and reducing power. Also, Persicaria tinctoria showed the protective effects of DNA damage against oxidative stress and anti-inflammatory effect by suppression of NO production in LPS induced RAW264.7 cells.Conclusions : These results showed that various parts of Persicaria tinctoria can be used as an effective source of antioxidants and anti-inflammatory agents via antioxidative activities and anti-inflammatory effect.

마가목 줄기 열수 및 에탄올 추출물의 항산화 활성 (Antioxidant Activities of Hot Water and Ethanol Extracts from the stem of Sorbus commixta Heal)

  • Objectives : In order to investigate the possibility of Sorbus commixta Heal. stem (SC) as a natural material, antioxidant activities of the hot water and ethanol extracts were examined. Methods : The samples of SC were pulverized, and fractions were extracted repeatedly three times from hot water and 70% ethanol at room temperature for 2 hours. The antioxidant activities were analyzed from total polyphenol, flavonoid contents, DPPH, ABTS, hydroxyl radical, $Fe2^+$ chelating, and nitrite scavenging activity. Results : Total polyphenol contents were significantly higher in ethanol extract group ($504.39{\mu}g/m{\ell}$) than in hot water extract group ($364.64{\mu}g/m{\ell}$). Total flavonoid contents were also significantly higher in ethanol extract group ($160.09{\mu}g/m{\ell}$) than in hot water extract group ($124.59{\mu}g/m{\ell}$). DPPH, ABTS, $Fe2^+$ chelating were slightly higher in ethanol extract gorups than in hot water extract groups, and increased in a dose-dependent manner. The hydroxyl radical scavenging activity (18.42~23.61%) of ethanol extract groups were shown to be approximately twice higher than that (7.63~10.37%) of hot water extract groups at $12.5{\sim}50{\mu}g/m{\ell}$ concentration. Nitrite scavenging activities of both ethanol and hot-water extract groups were shown to be higher in a dose dependent manner at the concentration of $12.5{\sim}50{\mu}g/m{\ell}$ at pH 3.0 than at pH 1.2, and ethanol extract groups (86.55~96.64%) had higher activity than the hot water extract groups (42.59~92.63%), which was higher than that of the control group antioxidant BHT (72.96~80.11%). Conclusions : The extracts of SC displayed antioxidant activities which suggested a natural material can be developed to functional material.

한국의 주요 블루베리 품종의 항산화 활성 및 페놀화합물 함량 분석 (Antioxidant activity and content of phenolic compounds in fruits of mainly cultivated blueberries in Korea)

  • 블루베리 10품종의 과실특성으로는 과립중은 0.83 ~ 1.88 g의 범위를 나타냈으며, '넬슨'이 가장 큰 과실을 결실하였으며 '블루골드'의 과실이 0.83 g으로 가장 작았다. 당도는 '시에라'에서 $16.6^{\circ}Brix$로 가장 높았고, $9.7-16.6^{\circ}Brix$를 나타내었다. 산함량은 '브리지타'에서 1.75%로 가장 높았고, '선라이즈'에서 0.94%로 가장 낮았다. 총 페놀 함량은 '노스블루'($23.75mg\;GAE{\cdot}g^{-1}\;FW$)에서 가장 높았고, '코빌'($17.15mg\;GAE{\cdot}g^{-1}\;FW$)에서 가장 낮았다. 총 플라보노이드 함량은 '넬슨'($14.1mg\;QE{\cdot}g^{-1}\;FW$)에서 가장 높았고, '듀크'에서는 $10.1mg\;QE{\cdot}g^{-1}\;FW$으로 가장 낮았다. 블루베리 추출물의 항산화활성을 3종류의 방법으로 조사한 결과, $ABTS^+$ radical 소거능은 '란코카스'(82.2%), '블루골드'(79.6%), '넬슨'(77.8%) 등에서 높았으며, DPPH radical 소거능은 '란코카스'(76.0%)에서 가장 높았으나, hydroxy radical 소거능은 '넬슨'(73.0%)에서 가장 높았다. 총 페놀 및 플라보노이드 함량, 항산화활성 등의 특성을 수치화하여 다변량 해석을 수행한 결과, 'Bluegold', 'Nelson', 'Northblue', 'Rancocas'등의 품종이 기능성 특성이 가장 양호하여 블루베리 품종 육성을 위한 육종 소재로 활용될 수 있을 것으로 여겨진다.

Napier grass (Penninsetum purpureum) 에탄올 추출물의 in vitro 항산화 효과 (In Vitro antioxidant effect of ethanol extract from Pennisetum purpureum)

  • Napier grass (Penninsetum purpureum)의 50% 에탄올 추출물의 수율, 폴리페놀 함량 및 항산화 활성을 확인하였다. Napier grass 추출물의 수율은 $6.3{\pm}0.35%$이었고, 총 페놀성 화합물 함량은 $79.6{\pm}3.65{\mu}g$ gallic acid equivalents/mg-추출물로 나타났다. Napier grass 추출물의 유리라디칼과 양이온라디칼 소거능, 환원력, 아질산염 소거능 및 지질과산화 억제능은 모두 추출물의 농도에 비례하여 증가하였다. Napier grass 추출물의 DPPH 유리라디칼에 대한 $EC_{50}$값은 $1,930.0{\mu}g/mL$로 유리라디칼에 대한 소거활성이 높지 않았다. Napier grass 추출물의 ABTS 양이온라디칼 소거, 환원력 및 아질산염 소거에 대한 $EC_{50}$값은 각각 350.0, 840.0 및 $1,470.0{\mu}g/mL$로 나타나 유리라디칼 소거활성에 비해 높은 항산화 능력을 보유하고 있었다. 특히 Napier grass 추출물은 $85.0{\mu}g/mL$의 농도에서 과산화물의 생성을 74.6% 억제할 정도로 뛰어난 지질과산화 억제활성을 보여주었다.

Effect of Dietary Inclusion of Medicinal Herb Extract Mix in a Poultry Ration on the Physico-chemical Quality and Oxidative Stability of Eggs

  • A mixture of three dietary medicinal herb extracts (MHE, mulberry leaf:Japanese honeysuckle:goldthread = 48.5: 48.5:3.0) was prepared as an additive of hen's feed. One hundred-eight, 28-wk-old Lohmann Brown hens were assigned randomly with three levels of MHE in the diet (0, 0.3, and 1%). Hens were fed for 6 wks and eggs were collected in the 6th week, and stored at $4^{\circ}C$ for 14 days to investigate the effect of MHE on the quality and oxidative stability of eggs. Internal quality of the egg including weight, shell color, albumen height, yolk color, shell weight, shell thickness, and Haugh units was not different among the dietary treatments. The oxidation stability of raw and cooked egg was determined by 2-thiobarbituric acid reactive substances (TBARS) value, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and 2,2'-azinobis(3-ethylbenzonthianoline-6-sulfonic acid) ($ABTS^{+}$) radical reducing ability. Results indicated that TBARS value at day 0 and $ABTS^{+}$ radical reducing ability of eggs from hens fed MHE were higher than from the control group. However, DPPH radical scavenging activity showed no difference in both raw and cooked samples. Results of the present study indicate that dietary MHE may slightly enhance the oxidative stability of eggs.

수삼의 증숙 횟수에 따른 페놀산 함량 변화와 라디칼 소거활성 (Changes of phenolic acid contents and radical scavenging activities of ginseng according to steaming times)

  • 본 연구는 증숙 횟수에 따른 인삼의 총페놀 화합물 함량과 페놀산 획분의 DPPH, ABTS 라디칼 소거활성을 시험하고, 개별 페놀산 조성과 함량을 GC로 분석하였다. 총페놀 함량은 0.53-2.89%로 증숙 횟수에 비례하여 증가하였다. 결합 형태에 따른 페놀산 획분의 함량은 결합형 >에스테르형>유리형 순으로 나타났으며, 이들 세 획분의 총량 (1.03-1.41%)은 증숙횟수에 영향을 받지 않았다. 각 획분에서 salicylic, cinnamic, p-hydroxybenzoic, gentisic, vanillic, syingic, caffeic, ferolic acid가 확인되었으며, 이 중 gentisic acid와 ferulic acid 가 주된 페놀산으로 50% 이상을 차지하였다. 각 페놀산 획분은 50% 이상의 DPPH와 ABTS 라디칼 소거활성을 보였으며, 결합형태에 따른 페놀산 획분 간에는 큰 차이가 없었다. 반면 각 획분별로 증숙 횟수에 따라 라디칼 소거활성이 차이를 보였으며, 증숙 횟수가 증가할수록 라디칼 소거활성도 증가하는 경향을 보였다.

재배 삼나물 뿌리 추출물의 자유 라디칼소거 및 α-glucosidase 저해활성 (Free radical scavenging and α-glucosidase inhibitory effects of a roots extract of Aruncus dioicus var. kamtschaticus)

  • 삼나물 뿌리를 80% 메탄올로 침지 추출하여 얻어진 추출물에 대해 n-hexane, EtOAc 및 n-BuOH의 유기용매를 활용하여 순차 분획을 실시하였고, 얻어진 결과물에 대하여 DPPH, $ABTS^+$ 라디칼 소거능 및 ${\alpha}$-glucosidase 저해활성을 평가하였다. DPPH 라디칼 소거능은 총 페놀성 화합물의 함량이 상대적으로 높은 EtOAc 층에서 $IC_{50}$값이 $32.3{\pm}2.3{\mu}g/mL$으로 가장 우수한 DPPH 라디칼 소거능을 확인하였고, 삼나물 뿌리 의 EtOAc 가용부에 다량 존재하는 페놀성 화합물과 라디칼 소거능과의 연관성을 시사하였다. 또한 $ABTS^+$ 라디칼 소거능은 EtOAc 층의 $IC_{50}$값이 $18.9{\pm}2.1{\mu}g/mL$의 우수한 라디칼 소거활성이 확인 하였고, 우수한 라디칼 소거 활성물질의 존재가 시사되었다. 또한, ${\alpha}$-glucosidase 저해활성을 평가한 결과, 우수한 라디칼 소거능을 나타낸 EtOAc 층의 $IC_{50}$$57.8{\pm}7.8{\mu}g/mL$의 저해활성을 나타내었으며, 이는 positive control인 acarbose의 $IC_{50}$값인 $501.1{\pm}17.5{\mu}g/mL$과 비교하여 우수한 활성이었으며, 추출물 상태의 시료를 단일물질로 정제할 경우 더욱 강한 효능의 화합물이 존재할 가능성을 시사하였다. 향후 이들 활성물질 동정을 통한 활성 기작에 대한 연구가 필요하며 본 연구결과는 보다 우수한 라디칼 소거능 및 ${\alpha}$-glucosidase 저해능을 가지는 새로운 기능성 소재 발굴을 위한 기초자료로 활용 가능할 것으로 사료된다.

수박(Citrullus vulgaris L.) 외피의 항산화 활성 탐색 (Screening of Antioxidant Activity from Exocarp of Watermelon (Citrullus vulgaris L.))

  • Watermelon (Citrullus vulgaris L.) is a summer fruit typical to help fatigue systemic absorption is getting better. The goal of this study is to screen antioxidant activity to ensure the possibility as a functional material for exocarp of watermelon. Watermelon was extracted with 70% methanol, 70% ethanol, chloroform:methanol (CM, 2:1, v/v). Total phenol contents were 12.01 mg/g, 8.89 mg/g, 3.53 mg/g in the 70% methanol, 70% ethanol, CM, in that order, respectively. Total flavonoid content, DPPH radical scavenging activity, ABTS radical scavenging activity, ferric reducing antioxidant power (FRAP), ${\beta}$-carotene bleaching assay were 70% methanol extract remarkably higher than the other extracts. And these results showed the same trend of total phenol content. From the above results shows that watermelon was effective on the antioxidative activity.

Investigation of Antioxidative Activity from Apple (Malus pumila Miller) Peel by Various Extraction Solvents

  • Consumption of fruits and vegetables has been conducted to be effective in the prevention of chronic diseases. In this study, 70% methanol, 70% ethanol and chloroform-methanol mixture (CM, 2:1, v/v) were used as solvents in the extraction of apple peels. The total phenol content, total flavonoid content and antioxidant activity of various extracts were investigated using in vitro assays. The extract obtained by 70% methanol showed the highest total phenol content ($20.87{\pm}0.17mgCAE/g$), ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity and ferric reducing antioxidant power. However, 70% ethanol extract possessed the strongest antioxidant activity assayed by DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity and ${\beta}$-carotene bleaching method. And CM extract was found to show the highest total flavonoid content with the value of $9.26{\pm}0.06mgQE/g$. These results indicated that apple peels can be used in dietary applications with a potential to reduce oxidative stress.
