Major constituents and antioxidant activities of domestic onion (Allium cepa L.) cultivars (국내 육종 양파 품종별 주요성분의 함량 및 항산화활성 비교)

  • The content of major constituents and antioxidant activities of nine onion cultivars, namely yellow domestic cultivars Meapsihwang, Sinsunhwang, Yeonsinhwang, Yeongbohwang, and Yeongpoonghwang, red domestic cultivar Eumjinara, green domestic cultivar Sweetgreen, yellow-introduced cultivar Sunpower, and red-introduced cultivar Cheonjujeok, were compared. Yeongbohwang and Cheonjujeok had the highest total phenolic and total flavonoid contents among the six yellow and two red cultivars, respectively. The red cultivars displayed the highest DPPH and ABTS+ radical scavenging activities (DRSAs and ARSAs, respectively) and reducing power (RP). Among all cultivars, Cheonjujeok and Eumjinara had the highest DRSA, while both red cultivars had similar ARSAs. Among the yellow cultivars, Sinsunhwang and Yeongbohwang showed excellent DRSAs. The RP of the domestic cultivars was slightly higher than that of the introduced cultivars. Thus, the domestic cultivar Yeongbohwang exhibited excellent characteristics among the yellow cultivars, while the red cultivars Eumjinara and Cheonjujeok were similar.

Antioxidant Production by Bacillus methylotrophicus Isolated from Chungkookjang, Korean Traditional Fermented Food (한국 전통발효식품인 청국장에서 분리한 Bacillus methylotrophicus에 의한 항산화물질의 생산)

  • Although antioxidant activities of Korean traditional fermented foods were reported by many researchers, study on antioxidant activity of microorganism originated from Korean traditional fermented foods was little. Therefore, we improved condition for antioxidant production by a bacterium isolated from home-made Chungkookjang. We selected a bacterial strain, which showed the highest antioxidative activity, from Chungkookjang and then named GJ. The selected GJ strain was identified as Bacillus methylotrophicus by alignment data of 16S rRNA gene nucleotide sequences. Improved medium compositions for DPPH radical scavenging activity were 0.25% sucrose, 1% peptone, 0.01% $MgSO_4{\cdot}7H_2O$ and initial pH 6.5, respectively. Optimal culture conditions were $30^{\circ}C$, 200 rpm and 4% inoculum volume, respectively. In improved conditions, DPPH radical scavenging activity of GJ reached to 91% in a short time. The strain GJ also possessed ACE inhibition and other antioxidative activities; ACE inhibition activity (49.4%), ABTS radical scavenging activity (99.8%), metal chelating activity (67.9%), SOD-like activity (36.5%) and reducing power ($A_{700}$ = 5.982) were observed, respectively. Therefore, our results suggest that B. methylotrophicus GJ strain may be potential candidate for functional foods, cosmetic products for anti-aging and medicine for diseases caused by oxidative stress.

Antioxidant Activities and Quality Characteristics of Gravy Sauce Added with Bokbunja (Rub coreanus Miquel), Mulberry and Blueberry (복분자, 오디 및 블루베리 첨가 그레이비소스의 품질특성 및 항산화 활성)

  • Purpose: Bokbunja (Rub coreanus Miquel), mulberry and blueberry have been reported to have powerful anti-antioxidant and anti-aging activities. The purpose of this study was to compare the quality characteristics and antioxidant activities of gravy sauce added with Bokbunja, mulberry and blueberry. Methods: Quality characteristics pH, brix, color and sensory characteristics of samples were evaluated. Antioxidant activities of samples were compared using total phenol, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) cation radical scavenging and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activities. Results: Mulberry juice showed significantly (p<0.05) higher values of pH and $^{\circ}Bx$ compared to Bokbunja and blueberry juice. Bokbunja juice also had significantly (p<0.05) higher value of Color-L value than mulberry and blueberry juice. The total phenol contents and ABTS cation radical scavenging activity of Bokbunja juice were 236.50 mgGAE/100 g and 61.66%, respectively, which were significantly (p<0.05) higher than those of blueberry and mulberry juice. pH value of gravy sauce added with mulberry (MBS) was 4.67, which was significantly higher than that of the control gravy sauce (GS) and gravy sauce added with Bokbunja (BJS) which had the lowest (p<0.05) pH value 4.33. $^{\circ}Bx$ of GS (13.50) was not significantly different from that of MBS and BBS. However, BJS showed the lowest (p<0.05) value $12.33^{\circ}Bx$. Total phenol contents and ABTS cation radical scavenging activity of BJS were 78.09 mgGAE/100 g and 14.47%, respectively, which were significantly (p<0.05) higher than those of MBS or BBS. Values of sensory characteristics color, sour aroma and sour flavor of BJS were significantly (p<0.05) higher value than those of other samples. Conclusion: Bokbunja juice and gravy sauce showed higher antioxidant activities and higher acceptance test compared to blueberry and mulberry. Thus, Bokbunja has the potential to be added to as high value products in the food industry.

Antioxidant Activity and Anti-inflammatory Effect of Extracts from Paulownia tomentosa in LPS-stimulated RAW264.7 macrophage cells

  • In this study, we investigated the antioxidant and anti-inflammatory effect of the Paulownia tomentosa extracts (PTE). The total polyphenol and flavonoid contents of PTE were 148.98±1.84 mg GAE/g extract, and 115.33±4.16 mg CE/g extract, respectively. The PTE showed that strong antioxidant activity via -diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, ABTS radical scavenging activity and FRAP assay. The anti-inflammatory activity was evaluated on lipopolysaccharide (LPS)-stimulated RAW264.7 cells. PTE remarkably reduced protein expression of inducible nitric oxide (iNOS), resulting in inhibition of production of nitric oxide (NO). Additionally, pre-treatment of PTE significantly suppressed the production of inflammatory cytokines, such as tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6). Moreover, PTE significantly attenuated LPS-induced IkappaB (IκB) degradation and suppressed nuclear factor kappa B (NF-κB) nuclear translocation in macrophages. The PTE showed high antioxidant and anti-inflammatory activity. These data suggest that PTE has pharmacological activity and may be useful for the development of anti-inflammatory agents.

Chemical Constituents of Gymnopilus spectabilis and Their Antioxidant Activity

  • Gymnopilus spectabilis, a hallucinogenic mushroom belonging to the family Cortinariaceae, is found growing in dense clusters on stumps and logs of hardwoods and conifers. It contains the hallucinogenic alkaloid psilocybin and its strongly bitter taste makes it undesirable as an edible. In an effort to identify chemical constituents of Korean native wild mushrooms, 4,6-decadiyne-1,3,8-triol (1), ergosta-4,6,8(14), 22-tetraen-3-one (2), bisnoryangonin (3), and hispidin (4) were isolated from the methanolic extract of the fruiting bodies of G. spectabilis. Their structures were assigned on the basis of various spectroscopic studies. Compounds 3 and 4 displayed significant scavenging activity against the ABTS radical cation, DPPH radical, and superoxide radical anion, while 1 and 2 exhibited no antioxidant activity.

Cell Cycle Regulation and Antioxidant Activity of Psammaplin A, A Natural Phenolic Compound from Marine Sponge

  • Psammaplin A (PSA), a naturally occurring biophenolic compound has been demonstrated to deliver significant cytotoxicity to many cancer cell lines. In this article, we investigated the effect of PSA on cell cycle progression of lung cancer cells (A549). It was found that PSA could slightly perturb the cell cycle progression of A549 cells and lead to the cell cycle arrest at G2/M phase, indicating PSA might disturb the mitosis process of A549 cells. In addition, inspired by the two phenolic groups in the structure of PSA, the antioxidant activity of it has been evaluated. Although PSA was weak in scavenging the stable free radical 1,1-diphenyl-2-picrylhyrazyl (DPPH), it showed stronger ABTS radical scavenging activity than ascorbic acid in TEAC assay. Furthermore, it was found that PSA could effectively prevent DNA strand scission induced by oxidative stress. These results suggest that PSA have both cell cycle regulation and antioxidant activities. Herein, we suggest that PSA would be a very interesting and promising candidate to be developed as a multi-function drug.

Antioxidant Activity of (8E,13Z,20Z)-Strobilinin/(7E,13Z,20Z)-Felixinin from a Marine Sponge Psammocinia sp.

  • During the course of our screening for bioactive metabolites from marine sponges, EZZ, the inseparable 1:1 mixture of (8E,13Z,20Z)-strobilinin and (7E,13Z,20Z)-felixinin has been found to deliver significant cytotoxicity against some cancer cell lines. In this study, the antioxidant activity of EZZ was first time evaluated by a series of antioxidant assays. It was found that EZZ was weak in scavenging the stable free radical 1,1-diphenyl-2-picrylhyrazyl (DPPH), but it was comparable to ascorbic acid in scavenging ABTS and superoxide radicals. In addition, EZZ could protect DNA from hydroxyl radical-induced strand cleavage. The findings of the present study suggest that EZZ possess certain antioxidant activity, which might help to prevent occurrence of cancer by alleviating the oxidative stress in cells.

Anti-oxidative Activity and the Protective Effect of Donkey's Bone and Skin Extracts on SK-N-SH Cells (당나귀 사골과 껍질의 항산화기능 및 SK-N-SH세포 보호효과)

  • The aims of this study were to determine antioxidation effect and neuroblastoma cell protection effect of donkey's bone and skin extracts (DBSE). DBSE was extracted by a pressure-cooker for 48 h and lyophilized. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was significantly increased with increased doses of DBSE and 40 mg/ml of DBSE showed 95.43% of the DPPH scavenging effect, which was equivalent to 1 mg/ml of vitamin C. The 2,2'-azino-bis (3-ethylbenzothiazoline-6- sulphonic acid) (ABTS) radical scavenging activity was also increased in a dose-dependent manner, and 20 mg/ml of DBSE showed 88.73% of the ABTS scavenging effect. The oxygen radical absorbance capacity (${\mu}M$ Trolox equivalent) of DBSE was significantly increased at a concentration of 10 mg/ml, which showed $132.53{\mu}M$ TE. The viability of oxidatively stressed brain cells induced by $500{\mu}M\;H_2O_2$ was protected by DBSE at concentrations greater than $50{\mu}M$. Cell viability after DBSE treatment at 50 and $100{\mu}g/ml$ was 53.78 and $54.34{\mu}M$ TE, respectively. There was no significant difference between both doses; however, 200 and $500{\mu}g/ml$ of DBSE showed 59.74 and 66.08% of cell viability, respectively indicating that DBSE protected SK-N-SH from oxidation stress. These results suggest that DBSE may have potential to be used as natural antioxidants in food industry, while in vivo evidence is necessary to support DBSE's in vitro-based antioxidative efficiency.

Antioxidant activity analysis of Catechin compounds in Korean green tea using HPLC On-line $ABTS^{+}$ Antioxidant screening system (HPLC On-line $ABTS^{+}$ Antioxidant screening 시스템을 이용한 한국산 녹차로부터 Catechin compounds의 황산화 활성분석)

  • In this work, we describes analysis of the antioxidant potential of Korean green tea phenolics using an high-performance liquid chromatography (HPLC) on-line $ABTS^{+}$ antioxidant screening method. In conjunction with the analysis of their 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS+) radical scavenging ability, the extraction of catechine compounds from Korean green tea were performed by various temperature and time. The optimum operating conditions were experimentally determined to analyze the catechine compounds in the pretreatment extracts. From the results, the extraction temperature $60^{\circ}C$, time 3 min was selected as an optimal antioxidant activity condition. The analysis by $C_{18}$ column was performed, the flow rate of mobile phase and UV wavelength was fixed at 1.0 ml/min and 254 nm, respectively. the mobile phase was composed from acetonitrile and water, and the gradient elution mode were applied.

Comparison of Antioxidant Activities by Different Extraction Temperatures of Some Commercially Available Cultivars of Rice Bran in Korea (추출온도에 따른 국내 벼 품종 미강의 항산화 활성 비교)

  • This study was conducted to determine antioxidant compounds and antioxidant activities of rice bran at temperatures of $10^{\circ}C$, $25^{\circ}C$, and $60^{\circ}C$ using 70% ethanol solution. DPPH, ABTS radical scavenging activities and reducing power have been used to investigate the relative antioxidant activities of extracts. The contents of total polyphenol and total flavonoids in the 70% ethanol extracts were measured by spectrophotometer. The total polyphenol content in the rice bran extracts were 790.39~930.07, 782.71~1,039.57, and 848.22~1,052.96 mg gallic acid eq./100 g in $10^{\circ}C$, $25^{\circ}C$ and $60^{\circ}C$, respectively. Antioxidant activities, such as DPPH, ABTS radical scavenging activity and reducing power, in all cultivars and temperatures was the highest in Cheonghaejinmi rice bran extracted at $25^{\circ}C$. The results obtained in the in vitro models suggest that extracts from rice bran, especially Cheonghaejinmi, has significant health-promoting effects, as it exhibits antioxidant activities.
