열처리가 천마의 Gastrodin과 Gastrodigenin 및 라디칼 소거능에 미치는 영향 (Influence of Heat Treatment on Gastrodin, Gastrodigenin, and Free Radical Scavenging Activity of Gastrodia elata Blume)

  • This study evaluated the effects of heat treatment on gastrodin and gastrodigenin content, and antioxidant activities, in Gastrodia elata Blume. Gastrodin and gastrodigenin content was analyzed post-method validation, and antioxidant activity evaluation, including assessing total polyphenol content, DPPH, and ABTS radical scavenging activities, was done. The validation of the analysis method demonstrated excellent linearity. The limits of quantification of gastrodin and gastrodigenin were 2.89 and 3.47 ㎍/mL, respectively. Moreover, the results of intra- and inter-day precision analysis demonstrated relative standard deviation values, within 5%. The recovery rates for gastrodin and gastrodigenin were 97.22~98.85 and 97.99~99.91%, respectively, indicating good accuracy. Under different heat treatment conditions, gastrodin and gastrodigenin content significantly increased (p<0.05), ranging from 91.15 to 310.27 and 559.66 to 830.02 mg/100 g DW, respectively. Additionally, the total polyphenol content exhibited a significant (p<0.05) increasing trend, ranging from 1,444 to 1,798 mg/100 g DW, as the temperature and time of heat treatment increased. The DPPH and ABTS radical scavenging abilities demonstrated an increasing trend at 120℃ during heat treatment. These research findings are expected to enhance our understanding of the changes in gastrodin and gastrodigenin content, and antioxidant effects in Gastrodia elata Blume during heat treatment.

충북산 토종고추의 계통별 품질 분석 (Quality Characteristics of by Lines of Native Pepper Grown in Chungbuk Province)

  • This study examined the quality characteristics of 10 different lines of native peppers collected from organic farms in Chungbuk province. The study found a strong correlation between the redness (a*) and ASTA values, which both contribute to the perceived quality of peppers. The highest values were observed in the Iyugsa line. The content of capsaicinoid and beta-carotene also showed a positive correlation, with Chilsungcho having the highest statistically significant value. While the total polyphenol content did not correlate with the other indicators, Chilsungcho again had the highest levels. The Yuwolcho line exhibited the highest ABTS radical scavenging ability, while the Eumseongcho line showed the highest DPPH radical scavenging ability. Taking into account the overall bioactivity quality, Chilsungcho had the highest values in terms of total polyphenol, beta-carotene, capsaicinoid, and redness. It also had the second highest total flavonoid content, ABTS, and DPPH radical scavenging activity, all statistically significant. Therefore, Chilsungcho can be considered an excellent choice when considering physiological activity. Furthermore, this study provides valuable information about the unique characteristics of these 10 native pepper lines, which can assist in selecting the appropriate pepper for food manufacturing and serve as a helpful resource for future research.

Chemical composition, antioxidant and antifungal activities of rhizome essential oil of Kaempferia parviflora Wall. ex Baker grown in Vietnam

  • The aim of the present study was to evaluate the chemical composition and antioxidative activity of rhizome essential oil of Kaempferia parviflora Wall. ex Baker. The essential oil extracted by hydrodistillation was chemically profiled by GC/MS analysis. The antioxidative activity was determined and evaluated spectroscopically by the 2,2-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging assays. According to the results, the major essential oil components were camphene (18.03%), β-pinene (14.25%), a-pinene (12.38%), endo-borneol (10.23%), β-copaene (8.38%), and linalool (8.20%). K. parviflora rhizome oil possessed antioxidant potential, exhibiting DPPH and ABTS radical scavenging activities as high as 80.90 and 94.04%, respectively, at a concentration of 10 mg/mL. The corresponding IC50 values were 0.451±0.051 and 0.527±0.022 mg/mL, respectively (IC50 values for ascorbic acid, as the standard, were 0.209±0.016 and 0.245±0.022 mg/mL, respectively). The mycelium of F. oxysporum was distorted and collapsed when treated with 0.5 mg/mL of the EO of K. parviflora rhizome for 3 days treatment, which may provide an important information for exploring the metabolism of the fungicide K. parviflora rhizome and its derived compounds against F. oxysporum. This study provides the chemical properties of the essential oil of K. parviflora rhizome grown in Vietnam and their potential antioxidant and antifungal activities.

홍삼 추출물을 첨가한 요구르트의 항산화능 (Antioxidant Activity of Yogurt Supplemented with Red Ginseng Extract)

  • The antioxidant properties of red ginseng extracts prepared under different extraction conditions were evaluated by a variety of antioxidant assays, including $DPPH^{\cdot}$ radical scavenging, $ABTS^{+\cdot}$ radical scavenging, superoxide anion scavenging, nitrite scavenging and reducing power activities. The contents of total phenolic compounds and flavonoids were also determined. The various antioxidant activities were compared to positive controls such as Trolox, tannic acid and ascorbic acid. The antioxidant activities of all of the extracts were shown to be the highest in the ethanol extract. The antioxidant activities of the red ginseng powder were the lowest among the samples. The amounts of total phenolic compounds and flavonoids were at a maximum in the ethanol extract. Correlation analysis demonstrated the existence of a linear relationship between free radical scavenging activities and the phenolic compounds contents of extracts. The antioxidant activity of yogurt was increased as the result of the addition of red ginseng extract. The quality characteristics of the yogurt to which red ginseng extract was added were similar to those of yogurt without red ginseng extract. The overall sensory score and color of yogurt made from 0.5% red ginseng was the best of the tested yogurts. In accordance with the antioxidant activity and quality characteristics, the optimal concentration of red ginseng extract was approximately 0.5%.

살균 및 비가열압착한 들깨오일의 영양성분 및 항산화 분석 (Analysis of nutrients and antioxidants of sterilized and non-heat-pressed perilla oil)

  • 본 연구에서는 살균 및 비가열압착한 들깨오일(superilla)을 활용하여 영양성분을 확인하고 기능성 식품으로서의 가능성을 알아보기 위해 시판되는 들기름과 비교실험을 수행하였다. 수분 함량과 pH는 CPPO에서 높았고, 구리, 코발트, 칼슘 함량은 SRPO에서 높았다. 총 비타민 E, 토코페롤, 리놀렌산 함량은 LPPO에서 더 높았다. 총 폴리페놀 함량과 ABTS 라디칼 소거활성은 SRPO에서 가장 높은 수치를 보였으며, 아질산염 소거능 측정값은 CPPO에서 가장 높았다. L 및 a 값은 CPPO에서 더 높았고 b값은 LPPO에서 더 높았다. 이상의 결과를 통해 살균 및 비가열압착한 들깨오일(SRPO)의 무기질 함량, 총 폴리페놀 함량, ABTS 라디칼 소거활성 및 아질산 소거능이 있음을 확인하였다. 또한 시간이 경과할수록 SRPO는 과산화물가가 낮았고, LPPO는 산가가 유의하게 증가하였다. 따라서 본 연구를 종합해 볼 때, 살균 및 비가열압착한 들기름의 경우 기존의 방식으로 생산된 들기름과 영양소 성분이 유사하였다. 이는 새로운 들기름 제조방법을 제공하는 계기가 되었으며, 이를 통해 들기름의 상품화의 기초가 마련되었다고 판단된다.

마이크로파처리에 의한 심황색소의 화학안정성 및 세포독성 변화 (Changes in chemical properties and cytotoxicity of turmeric pigments by microwave treatment)

  • 다양한 생리활성을 가진 심황과 쿠쿠미노이드가 가공식품류 등에 광범위하게 적용되는 바, 실제 식품 중에 이용되는 oleoresin turmeric 색소에 대해 마이크로파처리에 의한 화학안정성 및 생리활성의 변화를 분석하였다. 심황색소는 마이크로파 가열처리에 의해 황색도와 화학안정성이 감소하였으며 특히 3종의 쿠쿠미노이드 중 쿠쿠민이 마이크로파처리에 가장 민감하였고 BMC가 가장 안정하였다. 마이크로파처리 후 심황색소의 ABTS, AAPH peroxyl radical 및 아질산염 제거활성은 증가하였으며, DPPH 라디칼 제거활성에는 변화가 없었다. 심황색소에서 생성되는 활성 산소종은 마이크로파처리 후 증가하였으나, 정상장관계 세포 INT 407과 대장암 세포 HCT 116을 대상으로 한 세포독성 효과는 두 종류의 세포에서 모두 약화되었다. 본 연구에서는 다양한 식품 중에 첨가되는 심황색소가 마이크로파처리에 의해 화학적변화 및 생리활성에 영향을 가져올 수 있음을 보여주며, 이를 함유한 식품의 적절한 가공 및 조리방법이 고려되야 함을 제시하고 있다.

머루 과피와 종자 에탄올 추출물의 항산화 활성 및 항돌연변이 활성 분석 (Analysis of Antioxidative and Antimutagenic Activities of Ethanol Extracts from Pericarp and Seeds of Wild Grape (Vitis coignetiea))

  • The antioxidative activity and antimutagenic activity of the ethanol extracts from pericarp and seeds of wild grape (Vitis coignetiea) were analyzed in this study. The antioxidative activity of the extracts from wild grape was determined using 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assay. The antimutagenic activity of the extracts was evaluated on Salmonella typhimurium TA98 and TA100 by Ames test using 4-nitroquinoline 1-oxide (4-NQO) and sodium azide as mutagens. In the antioxidative activity determination, $IC_{50}$ values of the DPPH radical scavenging activity of the extracts from pericarp and seeds were 27.16 ppm and 7.61 ppm, respectively. Additionally, ABTS radical scavenging activities of pericarp and seed extract were 99.75% and 98.87% at 200 ppm, respectively. In the antimutagenic activity determination, pericarp extract at 5 mg/plate inhibited 72.6% and 74.3% of mutagenicity of S. typhimurium TA98 induced by 4-NQO and sodium azaid, respectively. Also, the mutagenicity inhibition rates of seed extract at 5 mg/plate were 77.8% and 74.5% in S. typhimurium TA100 induced by 4-NQO and sodium azaid, respectively. These results demonstrate that the ethanol extract from wild grape has remarkable antioxidant activity and antimutagenicity.

국내 자생 민들레 5종의 항산화 활성 및 Nitric Oxide 생성억제 활성 (Effect of Five Korean Native Taraxacum on Antioxidant Activity and Nitric Oxide Production Inhibitory Activity)

  • The effect on the antioxidant activity and Nitric Oxide activity production inhibitory activity of Taraxacum has not been known. Therefore, phenolics and flavonoid contents were investigated from the ethanol extracts of five different Taraxacum species. The results showed that, among the five Taraxacum, T. hallaisanensis contains the highest total phenolic and flavonoid contents. When the antioxidant activity was measured by DPPH, $ABTS^+$ and reducing power activity, the free radical scavenging activity of T. hallaisanensis was also the highest among five Taraxacum species. However, measurement by CCK-8 assay in Raw264.7 cells indicated that the extracts of Taraxacum species have no effect on cell viability. Moreover, we also investigated the effect of Taraxacum species on NO scavenging activity in lipopolysaccharide (LPS)-stimulated Raw264.7 cells. The results clearly showed that Taraxacum species inhibited NO production, and the inhibitory effect of T. hallaisanensis was the strongest. The above results suggested that Taraxacum species affected the antioxidant and NO scavenging activity, and among the five species, antioxidant and NO scavenging activity assay of T. hallaisanensis was significantly higher than those of other four Taraxacum species. Therefore, T. hallaisanensis could be used as a potential drug with anti-oxidant and anti-inflammatory effect.

고초균을 이용한 조직대두단백의 고체 발효 기간에 따른 라디칼 소거 활성 및 물성 평가 (Evaluation of Radical Scavenging Activity and Physical Properties of Textured Vegetable Protein Fermented by Solid Culture with Bacillus subtilis HA According to Fermentation Time)

  • 조직대두단백을 이용하여 B. subtilis HA에 의한 장기간 동안 고체 발효를 통해 얻어진 발효물의 품질, 항산화 활성 및 물성 변화를 평가하였다. 발효시간에 따라 발효물의 색도는 L값은 감소하고 a, b 값은 증가하였으며 수용성 추출물의 갈색도는 발효 48시간까지는 서서히 증가하나 그 후에는 변화가 없는 것으로 나타났다. TVP 발효물의 물과 70% 에탄올 추출물의 DPPH radical 소거 활성 측정 결과 발효 24시간째 70% 에탄올 추출물의 $ IC_{50}$값이 0.99 mg/mL로 가장 활성이 우수하였고 발효시간이 길어져도 활성이 증가하지 않았다. 또한 ABTS radical 소거 활성도 70% 에탄올 추출물이 발효 72시간째 $ IC_{50}$값이 1.68 mg/g으로 가장 높았다. TVP 발효물의 점조도는 발효 48시간째 7.89 $Pa{\cdot}s^n$으로 가장 높았고 그 후에는 감소하는 것으로 나타났으며 점질물 함량은 발효 24시간째 26.49%로 가장 높은 함량을 보인 후 감소하는 것으로 나타났다. 발효물의 점탄성은 발효 48시간째 가장 높았으며 이후 감소하였고 탄성(G')보다 점성(G")이 높은 것으로 나타났다. $\gamma$-PGA 함량은 발효시간이 증가하면서 증가하는 경향을 보이면서 168시간에 37.72% 생산되었고, 분자량은 발효시간이 경과될수록 감소하였다. Levan 함량은 발효 초기인 12시간째 7.83%에서 발효시간이 경과할수록 감소하는 것으로 나타났다.

추출방법 및 용매에 따른 청나래고사리의 항산화 활성 (Antioxidant Activities of Ostrich Fern by Different Extraction Methods and Solvents)

  • 본 연구는 청나래고사리 성엽 추출물의 항산화물질의 함량과 항산화능을 향상시킬 수 있는 효과적인 추출방법을 개발하기 위하여 시행하였다. 동결건조한 청나래고사리 성엽의 분쇄시료 1 g을 메탄올, 80% 에탄올 및 물 등 3가지 용매와 섞은 후 6시간 동안 상온 침지, $60^{\circ}C$ 가열 및 200 rpm에서 교반하여 추출하거나 42 kHz의 초음파 수조에서 15, 30, 45분 동안 추출하였다. 추출물은 여과한 다음 가용성고형분, 총 폴리페놀 및 총 플라보노이드의 함량을 측정하였으며, 항산화활성은 DPPH와 ABTS radical 소거활성을 $RC_{50}$으로 환산하여 측정하였다. 80% 에탄올을 용매로 하여 30분 동안 초음파추출 하였을 때 가용성고형분(0.317 $g{\cdot}g^{-1}$ db), 총 폴리페놀(70.90 $mg{\cdot}g^{-1}$ db) 및 총 플라보노이드(41.53 $mg{\cdot}g^{-1}$ db)의 추출수율이 가장 우수하였으며, DPPH와 ABTS radical 소거활성 또한 가장 우수하였다(각 $RC_{50}$=0.14 $mg{\cdot}ml^{-1}$와 0.09 $mg{\cdot}ml^{-1}$). 상기의 연구결과에 따라 청나래고사리의 성엽은 천연항산화소재로 활용가능하며, 80% 에탄올을 용매로 15-30분 동안 초음파추출하는 것이 추출물의 항산화효과를 증가시키며 추출에 소요되는 시간을 줄일 수 있는 효과적인 추출방법으로 생각되었다.
