Potentiation of Apoptin-Induced Apoptosis by Cecropin B-Like Antibacterial Peptide ABPs1 in Human HeLa Cervical Cancer Cell Lines is Associated with Membrane Pore Formation and Caspase-3 Activation

  • Apoptin, a chicken anemia virus-encoded protein, induces apoptosis in chicken or human tumor cells, localizing in their nuclei as opposed to the cytoplasm of non-transformed cells. The present study was undertaken to investigate whether ABPs1 could potentiate apoptin-induced apoptosis in HeLa cells. ABPs1 and the apoptin genes were successfully cloned into pIRES2-EGFP expression vector and expressed in HeLa cells. We report that ABPs1 augments apoptin cell growth inhibition in a concentration- and time-dependent manner. The DAPI staining and scanning electron microscopy observations revealed apoptotic bodies and plasma membrane pores, which were attributed to apoptin and ABPs1, respectively. Further, ABPs1 in combination with apoptin was found to increase the expression of Bax and to decrease the expression of survivin compared with either agent alone or the control. The apoptotic rate of HeLa cells treated with ABPs1 and apoptin in combination for 48 h was 53.95%. The two-gene combination increased the caspase-3 activity of HeLa cells. Taken together, our study suggests that ABPs1 combined with apoptin significantly inhibits HeLa cell proliferation, and induces cell apoptosis through membrane defects, up-regulation of Bax expression, down-regulation of survivin expression, and activation of the caspase-3 pathway. Thus, the combination of ABPs1 and apoptin could serve as a means to develop novel gene therapeutic agents against human cervical cancer.

Biological Control Effect of Treating Avirulent Bacteriocin-Producing Strain of Pseudomonas sozanacearum Adapted to Low Temperature on Tobacco Bacterial Wilt (비병원성 Bacteriocin 생성 Pseudomonu solonueomm의 저온성 균주를 이용한 담배 세균성마름병 방제효과)

  • Effect of an avirulent bacteriocin-producing strain(ABPS) of Pseudomonas solanacearum adapted to low temperature on the control of tobacco bacterial wilt was examined under the natural field conditions. The ABPS of p. solanacearum were succeeding-cultured at gradually low temperature, 3$0^{\circ}C$ to 13$^{\circ}C$. The isolates adapted to low temperature grew faster than the wild type either in artificial media or on the tobacco rhizoplane. The control effect of one of the isolates on bacterial wilt was higher than that of the wild type when the bacterial suspension had been poured onto the tobacco rhizosphere soil on 1 day before and 15 days after transplanting to the field. It was suggested that ABPS of p. solanaceamm adapted to the low temperature, might be more effective biological control agent than the wild type.

  • Control effect of an avirulent bacterlocin-producing strain(ABPS) Y6l-1 of Pseudomonas solanacearum on bacterial wilt was 58.8 % when the bacterial suspension had poured onto the rhizosphere soil of tobacco cultivar NC82 on one day before transplanting to the field and of hilling time. Until eight weeks after inoculation, survival of the strain on rhizoplane and in stem of the plants inoculated was better than that of other four strains tested. It suggested that survival of the ABPS in and on the plants should be supported for the sufficient protection.

Effects of Astragalus Polysaccharides, Achyranthes bidentata Polysaccharides, and Acantbepanax senticosus Saponin on the Performance and Immunity in Weaned Pigs

  • Two trials were conducted to study the effects of two Chinese herbal polysaccharides, Astragalus polysaccharides (APS) and Achyranthes bidentata polysaccharides (ABPS), and one Chinese herbal saponin, Acantbepanax senticosus saponin (ASS), on the immunity and growth performance of weaned pigs. Experiment 1 was a 14-day growth assay, in which 32 weaned pigs were randomly allocated to one of four dietary treatments: i) 0.05% talcum powder control; ii) 0.05% APS; iii) 0.05% mixture of APS and ASS in a 1:1 ratio by weight; and iv) 0.05% mixture of APS, ASS, and ABPS in a ratio of 1:1:1 by weight. Blood samples were collected on day 14 to determine plasma parameters. Feed intake, body weight gain, and feed efficiency were also determined. Experiment 2 was a 21-day immunity assay, in which 16 weaned pigs were randomly allotted to one of two dietary treatments: i) 0.05% talcum powder control; and ii) 0.05% mixture of APS and ASS in a 1:1 ratio by weight. On day 21, pigs were challenged with lipopolysaccharide (LPS) and 3 h later blood samples were collected and analyzed for lymphocyte proliferation as well as interleukin 6 (IL-6), insulin-like growth factor 1 (IGF-1), growth hormone (GH), and cortisol levels. In Experiment 1, feeding Chinese herbal polysaccharides and saponin increased growth performance of the pigs. The effects of the mixture of APS and ASS were especially notable, as there was a significant improvement in growth performance compared with the control (p<0.05). The plasma concentration of immunoglobulin G (IgG), nitric oxide (NO), and nitric oxide synthase (NOS) were increased in all treatments groups, with the mixture of APS and ASS increasing the level of IgG and NOS significantly (p<0.05), compared with the control. There was no difference in the NO level between the control and treatment groups (p>0.05). In Experiment 2, Chinese herbal polysaccharides and saponin showed immunostimulating effects. The level of cortisol, GH, and IGF-I were significantly increased (p>0.05), and the level of IL-6 showed a significant decrease (p<0.05) in the APS and ASS treatment after the LPS challenge. The mixture of APS and ASS could stimulate the blood lymphocyte proliferation significantly whether the LPS was injected or not (p<0.05). These results show that Chinese herbal extracts can improve growth performance and stimulate immunity of weaned pigs. A mixture of APS and ASS, compared with APS alone, could be a new kind of immunostimulant for weaned pigs, which could result in greater positive effects on their growth performance and immunity.

  • Microstructural characteristics of the Complex Perovskite (1-x) $(Li_{1/2}Sm_{1/2})TiO_3-x (Na_{1/2}Sm_{1/2})TiO_3$ (LNST) system have been investigated using the transmission electron microscopy (TEM). When $0.0{\leq}x{\leq}0.6$, the vacancy ordering forming the 1/2 (001) superlattice reflections due to the A-site cation deficiencies has apperaed. It could be confirmed by presence of ABPs. But it was difficult to form the vacancy ordering since vacancy concentration gradually lowered as the amount of the substituted Li ions decrease. Antiphase boundaries (APBs) were presented in microstructures of LNST when $0.8{\leq}x{\leq}1.0$. It was considered that these boundaries were caused by the 1:1 chemical ordering of A-site cations, Na and Sm ions. LNST had not only the antiphase tilting of oxygen octahedron but also the inphase tilting of oxygen octahedron and the antiparallel shift of cations all of them.
