항공수심라이다 데이터 해저면 포인트 클라우드 분리를 위한 CSF 알고리즘 적용에 관한 연구 (Segmentation of Seabed Points from Airborne Bathymetric LiDAR Point Clouds Using Cloth Simulation Filtering Algorithm)

  • 항공수심라이다(ABL: Airborne Bathymetric LiDAR)는 녹색 레이저(green laser)를 사용하여 연안 및 하천에 대해 해저지형과 수심에 대한 관측을 동시에 수행하는 첨단측량 기술이다. 항공수심라이다를 활용하여 해저지형 정보를 구축하기 위해서는 취득된 포인트 클라우드로부터 해수면과 해저면 점들을 분리하고 추출하는 과정이 필요하다. 기존의 해저면 점을 추출하기 위한 연구는 주로 waveform 분석(analysis)을 기반으로 수행되었다. 하지만 일반 사용자의 경우 waveform 데이터에 대한 접근성이 낮으며, waveform 분석 기반 해저면 추출 방법론에 대한 보완도 필요하다. 본 연구는 항공수심라이다 데이터의 지형학적 정보를 사용하여 해저면 점들을 추출하기 위한 연구를 수행하였다. 이를 위해 지면분리(ground filtering) 기법인 CSF (Cloth Simulation Filtering) 알고리즘을 RIEGL VQ880 항공수심라이다 시스템으로부터 취득된 데이터에 적용하고 효용성을 분석하였다. 실험결과 CSF 알고리즘을 항공수심라이다 데이터의 해저면 포인트 추출에 효과적으로 적용할 수 있음을 확인하였다.

Camptothecin 에 의한 ST1571 의 항암 활성 증강 (Synergistic antitumor activity of ST1571 and camptothecin in human cancer cells)

  • 본 연구에서는 전이성 암세포와 항암제 다제내성 세포에 있어서 항암제 내성에 영향을 미치는 것으로 알려진 DNA-dependent protein kinase (DNA-PK) 가 Abl protein-tyrosine kinases저해제인 STl571 내성에도 연관되어 있는지에 대하여 조사하였다. 또한 STl571 과 topoisomerase I 저해제인 camptothecin (CPT) 의 단독 및 병용처리에 의한 항암 활성을 전이성 암세포와 항암제 다제내성 세포를 대상으로 조사하였다. 세포의 전이도와 내성정도에 따라 STl571 의 감수성이 다르게 나타났다. 이와 함께 ST1571의 처리후 농도에 따라 전이도가 낮은 KMl2, PC3 세포와 항암제 감수성인 CEM, MCF-7 세포에서는 DNA-PK 의 발현이 감소하는 반면, 전이도가 높은 KML4a, PC-MM2 세포와 다제내성 CEM/MDR 및 MCR/MDR 세포에서는 그 발현이 증가되어 있음을 알 수 있었다. 이는 DNA-PK 의 발현이 STl571 의 내성에 관여한다는 것을 시사한다. 이와 같은 결과에 근거하여 DNA-PK 의 발현을 감소시키는 CPT를 STl571 내성을 나타내는 암세포에 대하여 STl571 과 병용처리 하였다. 그 결과 DNA-PK의 발현이 감소되고 세포증식이 억제됨으로써 ST1571 의 감수성이 CPT에 의해 증가하는 것을 알 수 있었다. 따라서 본 연구에서는 DNA-PK가 STl571 의 내성을 극복하는데 있어서 새로운 표적이 될 수 있으며, STl571 의 치료내성 극복에 CPT 와의 병용처리가 유효함을 알 수 있었다.

Growth Inhibitory and Pro-Apoptotic Effects of Hirsuteine in Chronic Myeloid Leukemia Cells through Targeting Sphingosine Kinase 1

  • Chronic myeloid leukemia (CML) is a slowly progressing hematopoietic cell disorder. Sphingosine kinase 1 (SPHK1) plays established roles in tumor initiation, progression, and chemotherapy resistance in a wide range of cancers, including leukemia. However, small-molecule inhibitors targeting SPHK1 in CML still need to be developed. This study revealed the role of SPHK1 in CML and investigated the potential anti-leukemic activity of hirsuteine (HST), an indole alkaloid obtained from the oriental plant Uncaria rhynchophylla, in CML cells. These results suggest that SPHK1 is highly expressed in CML cells and that overexpression of SPHK1 represents poor clinical outcomes in CML patients. HST exposure led to G2/M phase arrest, cellular apoptosis, and downregulation of Cyclin B1 and CDC2 and cleavage of Caspase 3 and PARP in CML cells. HST shifted sphingolipid rheostat from sphingosine 1-phosphate (S1P) towards the ceramide coupled with a marked inhibition of SPHK1. Mechanistically, HST significantly blocked SPHK1/S1P/S1PR1 and BCR-ABL/PI3K/Akt pathways. In addition, HST can be docked with residues of SPHK1 and shifts the SPHK1 melting curve, indicating the potential protein-ligand interactions between SPHK1 and HST in both CML cells. SPHK1 overexpression impaired apoptosis and proliferation of CML cells induced by HST alone. These results suggest that HST, which may serve as a novel and specific SPHK1 inhibitor, exerts anti-leukemic activity by inhibiting the SPHK1/S1P/S1PR1 and BCR-ABL/PI3K/Akt pathways in CML cells, thus conferring HST as a promising anti-leukemic drug for CML therapy in the future.

The Lichen Genus Polychidium New to South Korea

  • The lichen species Polychidium muscicola was found in South Korea for the first time and this is the first record of the genus Polychidium from the Korean peninsula. This species is characterized by a dichotomously branched minute thallus with clustered cells of Nostoc as the photobiont. A detailed description and illustrations of this species are provided.

Discrimination between RNAP IIA and IIO in Preinitiation Complex Assembly and Tyrosine Phosphorylation of the Carboxy Terminal Domain

  • Multiple phosphorylation of the carboxy-terminal domain (CTD) of the largest subunit in RNA polymerase II (RNAP II) is thought to play an important role in the transcription cycle. The preinitiation complex in a partially purified complete transcription system suggested that RNA polymerase IIA containing unphosphorylated CTD is involved in complex assembly, whereas RNA polymerase IIO containing Ser and Thr phosphorylated CTD is not involved in preinitiation complex assembly. Recently a minimal transcription system was developed which requires chemically defined minimal components for its transcription: TBP, TFIIB, TFIIF, RNAP II and a supercoiled adenovirus-2 major late promoter (Ad-2 MLP). It would be using interesting to determine the consequence of CTD phosphorylation on preinitiation complex formation using the minimal transcription system. Contrary to the results from the partially purified complete transcription system, both RNA polymerase IIA and IIO are equally recruited in the preinitiation complex formation. The discrepancy may result from the two different assays used to determine complex formation, the use of chemically undefined complete and defined minimal transcription systems. This implicates that some factors in the complete transcription system are involved in the distinction between RNAP IIA and IIO in complex assembly. In addition multiple tyrosine phosphorylation of the CTD of RNAP II was prepared with c-Abl kinase and its recruiting ability in the preinitiation complex was examined. Compare with Ser and Thr phosphorylated RNAP IIO, Tyr phosphorylated RNAP IlOy forms a stable preinitiation complex in both the minimal and complete transcription systems. Based on these results, it seems that tyrosine phosphorylation of the CTD is important in the transcription cycle on the special subset of class-II promoter or has a different role in the transcription process.

Selective miRNA Expression Profile in Chronic Myeloid Leukemia K562 Cell-derived Exosomes

  • Background: Chronic myeloid leukemia (CML) is a myeloproliferative disorder of hematopoietic stem cell scarrying the Philadelphia (Ph) chromosome and an oncogenic BCR-ABL1 fusion gene. The tyrosine kinase inhibitor (TKI) of BCR-ABL1 kinase is a treatment of choice for control of CML. Objective: Recent studies have demonstrated that miRNAs within exosomes from cancer cells play crucial roles in initiation and progression. This study was performed to assess miRNAs within exosomes of K562 cells. Methods: miRNA microarray analysis of K562 cells and K562 cell-derived exosomes was conducted with the 6th generation miRCURYTM LNA Array (v.16.0). Gene ontology and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses were also carried out. GO terms and signaling pathways were categorized into 66 classes (including homophilic cell adhesion, negative regulation of apoptotic process, cell adhesion) and 26 signaling pathways (such as Wnt). Results: In exosomes, 49 miRNAs were up regulated as compared to K562 cells, and two of them were further confirmed by quantitative real-time PCR. There are differentially expressed miRNAs between K562 cell derived-exosomes and K562 cells. Conclusion: Selectively expressed miRNAs in exosomes may promote the development of CML via effects on interactions (e.g. adhesion) of CML cells with their microenvironment.

Comparison of Univariate and Multivariate Gene Set Analysis in Acute Lymphoblastic Leukemia

  • Background: Gene set analysis (GSA) incorporates biological with statistical knowledge to identify gene sets which are differentially expressed that between two or more phenotypes. Materials and Methods: In this paper gene sets differentially expressed between acute lymphoblastic leukaemia (ALL) with BCR-ABL and those with no observed cytogenetic abnormalities were determined by GSA methods. The BCR-ABL is an abnormal gene found in some people with ALL. Results: The results of two GSAs showed that the Category test identified 30 gene sets differentially expressed between two phenotypes, while the Hotelling's $T^2$ could discover just 19 gene sets. On the other hand, assessment of common genes among significant gene sets showed that there were high agreement between the results of GSA and the findings of biologists. In addition, the performance of these methods was compared by simulated and ALL data. Conclusions: The results on simulated data indicated decrease in the type I error rate and increase the power in multivariate (Hotelling's $T^2$) test as increasing the correlation between gene pairs in contrast to the univariate (Category) test.

Successful Rechallenge with Imatinib in a Patient with Chronic Myeloid Leukemia Who Previously Experienced Imatinib Mesylate Induced Pneumonitis

  • Imatinib mesylate is a targeted therapy that acts by inhibiting tyrosine kinase of the bcr-abl fusion oncoprotein, which is specific to chronic myeloid leukemia (CML), and the c-transmembrane receptor, which is specific to gastrointestinal stromal tumors. Interstitial pneumonitis is a rare adverse event of imatinib therapy. It is clinically difficult to distinguish from infectious pneumonia, which can frequently occur due to the underlying disease. The standard treatment for imatinib-induced pneumonitis is to discontinue the medication and optionally administer corticosteroids. However, there are a few cases of successful retrial with imatinib. We describe a case of successful rechallenge of imatinib in a patient with imatinib-induced interstitial pneumonitis and CML without a recurrence of the underlying disease after 3 months of follow-up.

Partial turbulence simulation and aerodynamic pressures validation for an open-jet testing facility

  • This paper describes partial turbulence simulation and validation of the aerodynamic pressures on building models for an open-jet small-scale 12-Fan Wall of Wind (WOW) facility against their counterparts in a boundary-layer wind tunnel. The wind characteristics pertained to the Atmospheric Boundary Layer (ABL) mean wind speed profile and turbulent fluctuations simulated in the facility. Both in the wind tunnel and the small-scale 12-Fan WOW these wind characteristics were produced by using spires and roughness elements. It is emphasized in the paper that proper spectral density parameterization is required to simulate turbulent fluctuations correctly. Partial turbulence considering only high frequency part of the turbulent fluctuations spectrum was simulated in the small-scale 12-Fan WOW. For the validation of aerodynamic pressures a series of tests were conducted in both wind tunnel and the small-scale 12-fan WOW facilities on low-rise buildings including two gable roof and two hip roof buildings with two different slopes. Testing was performed to investigate the mean and peak pressure coefficients at various locations on the roofs including near the corners, edges, ridge and hip lines. The pressure coefficients comparisons showed that open-jet testing facility flows with partial simulations of ABL spectrum are capable of inducing pressures on low-rise buildings that reasonably agree with their boundary-layer wind tunnel counterparts.

Detection of BCR/ABL Fusion Gene by Hematological and Cytogenetical Analysis in Chronic Myeloid Leukemia Patients in Quetta, Pakistan

  • Background: Chronic myeloid leukemia (CML) is a myeloproliferative disorder of pluripotent stem cells, caused by reciprocal translocation between the long arms of chromosomes 9 and 22, t(9;22)(q34;q11), known as the Philadelphia chromosome. Materials and Methods: A total of 51 CML patients were recruited in this study. Complete blood counts of all CML patients were performed to find out their total leukocytes, hemoglobin and platelets. FISH was performed for the detection of BCR-ABL fusion and cryptogenic tests using bone marrow samples were performed for the conformation of Ph (9;22)(q34;q11) and variant translocation mechanisms. Results: In cytogenetic analysis we observed that out of 51 CML patients 40 (88.9%) were Ph positive and 4 (8.88%) had Ph negative chromosomes. Mean values of WBC 134.5 $10^3/{\mu}l$, hemoglobin 10.44 mg/dl, and platelets 288.6 $10^3/{\mu}l$ were observed in this study. Conclusions: In this study, Ph positive translocation between chromosome (9:22)(q34;q11) were observed in 40 (88.9%) CML patients.
