Hepatic transcriptional changes in critical genes for gluconeogenesis following castration of bulls

  • Objective: This study was performed to understand transcriptional changes in the genes involved in gluconeogenesis and glycolysis pathways following castration of bulls. Methods: Twenty Korean bulls were weaned at average 3 months of age, and castrated at 6 months. Liver tissues were collected from bulls (n = 10) and steers (n = 10) of Korean cattle, and hepatic gene expression levels were measured using quantitative real-time polymerase chain reaction. We examined hepatic transcription levels of genes encoding enzymes for irreversible reactions in both gluconeogenesis and glycolysis as well as genes encoding enzymes for the utilization of several glucogenic substrates. Correlations between hepatic gene expression and carcass characteristics were performed to understand their associations. Results: Castration increased the mRNA (3.6 fold; p<0.01) and protein levels (1.4 fold; p<0.05) of pyruvate carboxylase and mitochondrial phosphoenolpyruvate carboxykinase genes (1.7 fold; p<0.05). Hepatic mRNA levels of genes encoding the glycolysis enzymes were not changed by castration. Castration increased mRNA levels of both lactate dehydrogenase A (1.5 fold; p<0.05) and lactate dehydrogenase B (2.2 fold; p<0.01) genes for lactate utilization. Castration increased mRNA levels of glycerol kinase (2.7 fold; p<0.05) and glycerol-3-phosphate dehydrogenase 1 (1.5 fold; p<0.05) genes for glycerol utilization. Castration also increased mRNA levels of propionyl-CoA carboxylase beta (mitochondrial) (3.5 fold; p<0.01) and acyl-CoA synthetase short chain family member 3 (1.3 fold; p = 0.06) genes for propionate incorporation. Conclusion: Castration increases transcription levels of critical genes coding for enzymes involved in irreversible gluconeogenesis reactions from pyruvate to glucose and enzymes responsible for incorporation of glucogenic substrates including lactate, glycerol, and propionate. Hepatic gluconeogenic gene expression levels were associated with intramuscular fat deposition.

Health and Nutrition Status of Elderly People with Multimorbidity: A Korea National Health and Nutrition Examination Survey (2013~2015) (국민건강영양조사 자료에 근거한 고령 복합만성질환자의 건강 및 영양상태 특성)

  • Objectives: This study investigated the health and nutritional status of the elderly according to the number of chronic diseases, using data obtained from the Korea National Health and Nutrition Examination Survey 2013~2015. Methods: Data from a total of 2,310 individuals, aged 65 years and over, were used for the analysis. The elders were divided into 0 (n=375), 1 (n=673), 2 (n=637) and 3 or more (n=625) groups, by considering the number of chronic diseases. Results: Compared to other groups, the elderly subjects who were living with their spouse had the highest ratio in group 0 (P < 0.05), whereas subjects without economic activities had highest ratio in 3 or more group (P < 0.05). The EQ-5D index of subjects in the 0 group (0.90 ± 0.01) was higher than that in the 3 or more group (0.86 ± 0.01) (P < 0.05). After adjusting for confounding factors, the energy intake of subjects was determined to be lowest in the 3 or more group (P < 0.05). Protein (P < 0.05) and riboflavin (P < 0.05) intakes of the 3 or more group were also lower than other groups. Conclusions: This study indicates that multimorbidity of the elderly is associated with their health and nutritional status. The nutrients intake of the elderly, especially energy, protein and riboflavin, tended to be lowest in the 3 or more group. Further research is required to elucidate the risk factors related to presence of multimorbidity in the elderly.

The Effect of Floating Wetland on Water Quality Improvement in a Eutrophic Lake (부유습지를 이용한 부영양수계 현장 수질개선 효과)

  • At weekly intervals, we monitored continuous changes in water quality by constructed floating wetland equipped with the four different filter media (sponge, volcanic stone, activated carbon and magnesium hydroxide) in a eutrophic lake from March 2011 to May 2012. We also investigated phyto- and zooplankton communities both in the influent and the effluent water through the floating wetland. Over a 10-month time period, average turbidity (66%), suspended solids (79%) and chlorophyll-a (80%) concentrations were remarkably reduced in the effluent water compared to the influent (P<0.001). The average removal rates of $NO_2-N$ and $NH_3-N$ were 24% and 20%, respectively (P<0.05). The average removal rates of $NO_3-N$ and TN were less than 10% (P>0.05). On the other hand, the average removal rates of $PO_4-P$ and TP were more than 65% (P<0.01). Interestingly, the abundance of phytoplankton in the effluent was decreased about 2.6 times compared to that of the influent, whereas the abundance of zooplankton in the effluent was increased about 3.5 times compared to that of the influent. Overall, particulate matters (SS, Chl-a and TP) and dissolved nutrients ($NO_2-N$, $NH_3-N$ and $PO_4-P$) were particularly reduced at high rates. Therefore, application of our constructed floating wetland in a eutrophic lake improved the water quality and demonstrated a potential for algal bloom mitigation.

Study on Estrogenic Activities of Pesticide Chemicals using E-screen Assay (E-Screen assay법을 이용한 농약화학물질의 에스트로겐 활성 연구)

  • In this study, sixty chemicals including 47 pesticides were screened for estrogenic activity using E-screen assay. MCF-7 cell, used in E-screen assay, is known to be proliferated by addition of estrogenic chemicals. Eight of the measured pesticides showed estrogenic activity at the concentration range of 100-10,000nM. Their relative proliferative effect (RPE) and the relative proliferative potency (RPP) were 20-65% and 0.01-1.0%, respectively, when compared with 1.0nM of $\beta-Estradiol-17-acetate(E_{2}).$ DDVP and diazinone showed most strong estrogenic potency(RPP; 1.0%) and effect(RPE; 65%) of the eight pesticides. These results are in agreement with estrogenic activity of bisphenol A is known as a positive endocrine disrupter. Also, in this study, paraquat, DDVP, 4-chloro-3-methylphenol, 2,4,6-trichlorophenol and diazinon of the measured pesticides are estimated to estrogenic chemical.

HEMATOLOGICAL STUDIES ON CATTLE EXPOSED TO Fasciola gigantica INFESTATION

  • Hematological value were determined from 140 Fasciola gigantica infested and 138 non-infested apparently healthy (control) zebu cattle from eight different sites of Bangladesh. The F. gigantica infested cattle have experienced of normocytic normochromic anemia indicated by the significant (p<0.01) decline of total erythrocyte count, hemoglobin level, packed cell volume and mean corpuscular hemoglobin concentration and significant (p<0.01) increase of erythrocyte sedimentation rate, as compared with non-infested control animals. Values of mean corpuscular volume and mean corpuscular hemoglobin content were not differed significantly between the infested and control animals. There observed significantly (p<0.01) higher eosinophil and neutrophil counts and significantly (p<0.01) lower lymphocyte counts in fasciola infested cattle in comparison with the control animals.

Chemical Characteristics of Commercial Korean Soy Sauce Produced by Alkali Treatment Following Acidic Hydrolysis

  • Alkali treatment following acidic hydrolysis (ATAH) is a commonly used practice for reducing the levels of monochloropropanediol in commercial Korean soy sauce. This study investigated the chemical compounds produced in commercial Korean soy sauce made by ATAB. The levels of amino-N, total acidity, pH, salinity and Brix of the soy sauce were 6.66%, 2.52%, 19.81%, 4.57 and 35.01, respectively. The major fatty acids were C18:2n-6, C16:0, Cl8:ln-9 and C18:3n-6. The concentration of polyunsaturated fatty acids were especially high, with of C18:2n-6 (49.8%) being the highest followed by C18:3n-6 (3.8%) and C18:3n-3 (2.4%) in that order. Among the non-volatile organic acids, the concentration of levulinic acid (1,206.28 mg/100 g) was the highest, while the taste value of citric acid was the highest. Among the ATP related compounds, IMP concentration (31.19 mg/100 g) was highest followed by AMP, hypoxanthine and GMP in that order. The concentrations of free and total amino acids in soy sauce were 6,136.94 mg/100 g and 8,702.76 mg/100 g, respectively. On the other hand, the taste value of glutamic acid, a major amino acid flavor determinant in soy sauce, was highest of all the amino acids, which is desirable since most free amino acids such as methionine, histidine and phenylalanine have a bitter taste that detracts from the flavor of soy sauce.

The Effect of a Potential Antiobesity-Supplement on Weight Loss and Visceral Fat Accumulation in Overweight Women (과체중인 여성에서 체중 감소 보조제를 이용한 체중 및 내장지방의 감소 효과)

  • Chitosan, hydroxycitrate and L-carnitine have been known to be antiobesity components. The purpose of this study was to evaluate the combined effects of chitosan, hydroxycitrate and L-carnitine mixture as a potential antiobesity supplement in overweight women. Pre-menopausal healthy females who were overweight (percent ideal body weight > 110) were included in this study. Forty-nine subjects randomly received a placebo (n = 25) or antiobesity-supplement (n = 24), which was a mixture of chitosan, hydroxycitrate, and L-carnitine. Before and after the eight-week experimental period, anthropometric parameters, blood components and computerized tomography were measured. At baseline, the two groups were well matched in terms of age, body mass index and lipid profile. After the eight weeks of potential antiobesity supplementation, the subjects' body fat percent had decreased significantly (p < 0.001) by 5.6% (39.1 $\pm$ 1 vs 36.9 $\pm$ 1%) while lean body mass increased (p < 0.01). Vsceral fat area at the L4 vertebra decreased significantly (p < 0.01) by 8.6% in the supplemented group and the total fat area at the L4 vertebra showed a tendency to decrease (p = 0.051) by 2.4%. Also, in the group given the antiobesity-supplement rather than the placebo, the fasting triglyceride level decreased significantly (p < 0.05) by 10.0%. In addition, serum total cholesterol levels in the antiobesity-supplement group showed a tendency to decrease (p=0.159) by 2.7% (194 $\pm$ 6 vs 189 $\pm$ 6 mg/dl). No side effects were found in either group during the intervention. In conclusion, the present study demonstrated that taking a mixture of chitosan, hydroxycitrate, and L-carnitine as a potential antiobesity supplement for eight weeks produced advantageous changes in the weight and visceral fat accumulation of overweight women without any side effects. A Comparison of the Adjustment Methods for Assessing Urinary Concentrations of Cadmium and Arsenic: Creatinine vs. Specific Gravity (요중 카드뮴과 비소의 보정방법 비교 : 요중 크레아티닌과 요비중)

  • Objectives: Biomarkers in urine are important in assessing exposures to environmental or occupational chemicals and for evaluateing renal function by exposure from these chemicals. Spot urine samples are needed to adjust the concentration of these biomarkers for variations in urine dilution. This study was conducted to evaluate the suitability of adjusting the urinary concentration of cadmium (uCd) and arsenic (uAs) by specific gravity (SG) and urine creatinine (uCr). Methods: We measured the concentrations of blood cadmium (bCd), uCd, uAs, uCr, SG and N-acetyl-${\beta}$-D-glucosaminidase (NAG) activity, which is a sensitive marker of tubular damage by low dose Cd exposure, in spot urine samples collected from 536 individuals. The value of uCd, uAs and NAG were adjusted by SG and uCr. Results: The uCr levels were affected by gender (p < 0.01) and muscle mass (p < 0.01), while SG levels were affected by gender (p < 0.05). Unadjusted uCd and uAs were correlated with SG (uCd: r = 0.365, p < 0.01; uAs: r = 0.488, p < 0.01), uCr (uCd: r = 0.399, p < 0.01; uAs: r = 0.484, p < 0.01). uCd and uAs adjusted by SG were still correlated with SG (uCd: r = 0.360, p < 0.01, uAs: r = 0.483, p < 0.01). uCd and uAs adjusted by uCr and modified uCr ($M_{Cr}$) led to a significant negative correlation with uCr (uCd: r = -0.367, p < 0.01; uAs: r = -0.319, p < 0.01) and $M_{Cr}$ (uCd: r = -0.292, p < 0.01; uAs: r = -0.206, p < 0.01). However, uCd and uAs adjusted by conventional SG ($C_{SG}$) were disappeared from these urinary dilution effects (uCd: r = -0.081; uAs: r = 0.077). Conclusions: $C_{SG}$ adjustment appears to be more appropriate for variations in cadmium and arsenic in spot urine.

Crystal Structure and Magnetic Properties of Iron Doped La-Sr-Mn-O (철을 미량 치환한 La-Sr-Mn-O의 결정학적 및 자기적 특성 연구)

  • The iron-doped perovskite La$_{0.67}$Sr$_{0.33}$Mn$_{0.99}$$^{57}$Fe$_{0.01}$O$_3$compound has been studied by x-ray diffraction, Mossbauer spectroscopy, and vibrating sample magnetometry. The single phase of the polycrystalline La$_{0.67}$Sr$_{0.33}$Mn$_{0.99}$$^{57}$Fe$_{0.01}$O$_3$powder has been prepared by a waterbased solgel method. Crystalline La$_{0.67}$Sr$_{0.33}$Mn$_{0.99}$$^{57}$Fe$_{0.01}$O$_3$was a rombohedral structure with lattice parameters a$_{0}$=5.480 $AA$, $alpha$=60.259$^{circ}$. Mossbauer spectra of La$_{0.67}$Sr$_{0.3}$/Mn$_{0.99}$$^{57}$Fe$_{0.01}$O$_3$have been taken at various temperatures ranging from 20 to 400 K. As the temperature increases toward the Curie temperature, T$_{c}$=375 K, the Mossbauer spectra show line broadening and the difference between the 1,6 and 3,4 linewidths is caused by the anisotropic hyperfine field fluctuation. The anisotropic field fluctuation of +H (P$_{+}$=0.80) is greater than -H (P$_{-}$=0.20). We calculated that the anisotropy energy was 124.01 erg/cm$^3$for T=150 K which is associated with the large line broadening.

Supplement Dose and Health-Related Life Style of Vitamin-Mineral Supplement User among Korean Middle-Aged (중년기의 비타민ㆍ무기질 보충제 사용량과 건강관련 생활습관 조사)

  • We studied daily micronutrient intake from vitamin-mineral supplements, health-related life style, clinical case of diseases and food frequency of the Korean middle-aged (40-59 yr, n = 404) to compare the characteristics of non-user (n = 270) and user (n = 134) of vitamin-mineral supplements. Rate of supplement use of the middle-aged was 33.2% and there was significant difference in education level (p = 0.0084) and family income (p = 0.0476) of user and nonuser. Smoking habit (p = 0.0844) and drinking frequency (p = 0.0606) tended to be lower in a supplement user than a non-user. The medical history of a case was significantly higher in users (67.9%) than in non-users (44.4%) (p = 0.001), which suggests that medical history is one of the important motivations of supplement use. Supplement users had the medical history of digestive disease (34.1%), anemia (11.0%) and hypertension (9.9%) in order. Vitamin C was the most frequently supplemented nutrient (81.3%) among vitamin-mineral supplement, and the next orders were vitamins E (73.1%), B$_2$(68.7%) and B$_{6}$ (60.4%). Mean intakes of vitamin B$_1$, iron, selenium, vitamin E, and vitamin C from supplement was 4,260%, 4,030%, 1,660% and 1,330% of RDA, respectively. The supplement users tended to consume most food items including milk & milk products (p < 0.01), rice (p < 0.01), grains (p < 0.05) and cookies (p < 0.01) less frequently than non-users. Conclusively, nutrient intake of vitamin B$_1$, iron, selenium, vitamin E, and vitamin C from supplement was excessively high compared to RDA. We suggest that the toxic effect of excessive supplementation should he informed to supplement user and nutritional education should be focused on the optimal supplement dose.e.
