Synthesis of New Pyrimidinylaminobenzene Derivatives and Their Antiproliferative Activities against Melanoma Cell Line

  • A series of new diarylamide and diarylurea derivatives possessing pyrimidinylaminobenzene scaffold was synthesized. Their in vitro antiproliferative activities were tested against A375P human melanoma cell line. Among them, compounds 1a-c, k and 2b-d, f, g, j, l showed superior potencies against A375P human melanoma cell line to Sorafenib. In particular, compound 2f possessing 3-fluoro-5-trifluoromethyl moiety exhibited the highest potency with $IC_{50}$ value in nanomolar scale.

Inorganic sulfur reduces the motility and invasion of MDA-MB-231 human breast cancer cells

  • This study investigated the effects of inorganic sulfur on metastasis in MDA-MB-231 human breast cancer cells. MDA-MB-231 cells were cultured in the absence or presence of various concentrations (12.5, 25, or 50 ${\mu}mol$/L) of inorganic sulfur. Cell motility, invasion, and the activity and mRNA expression of matrix metalloproteases (MMPs) were examined. Numbers of viable MDA-MB-231 cells did not differ by inorganic sulfur treatment from 0 to 50 ${\mu}mol$/L within 48 h. Inorganic sulfur significantly decreased cell motility and invasion in the MDA-MB-231 cells in a dose-dependent manner (P<0.05), as determined using a Boyden chamber assay and a Matrigel chamber. The activities of MMP-2 and MMP-9 were significantly reduced by inorganic sulfur in a dose-dependent manner (P<0.05). The inorganic sulfur also significantly inhibited MMP-2 and MMP-9 expression in the cells (P<0.05). These data suggest that inorganic sulfur can suppress cancer cell motility and invasion by inhibiting MMP-2 and MMP-9 activity and gene expression in MDA-MB-231 cells.

The Influence of Empathy and Leadership on Happiness in Nursing College Students (간호대학생의 공감과 리더십이 행복감에 미치는 영향)

  • This study was attempted to find out the effect of empathy and leadership of nursing college students on happiness. For this study, from November to December 2020, 142 students were participated from C University in U City, using self reported questionnaire. Descriptive statistics, t-test, ANOVA, Scheffe test, Pearson's correlation analysis, and multiple regression analysis were used for data analysis. As results of this study, it was revealed that there was a significant positive correlation between the happiness of nursing students on empathy and leadership(r=.424, p<.001, r=.635, p<.001). Also, the factors affecting the happiness of nursing students were personality(β=-0.284, p<.001), health status(β=-0.375, p<.001, β=-0.178, p<.001), empathy(β=0.351, p<.001) and leadership(β=0.133, p=.039) with a total explanatory power of 58.1%. As the results of this study, in order to increase the happiness of nursing students, it is necessary to develop their empathy skills and leadership. The specific strategies are to develope and apply special programs to develop the empathy skills and leadership.

Floral Design Courses to Build Self-Esteem in Children Enrolled in the Community Child Care Center for Low-Income Families: Focused on the Accreditation Process of the Junior Florist Certification

  • The purpose of this study was to look into the influence of the junior florist training course on self-esteem of children in low-income families at the community child care center. Accordingly, the program was carried out by dividing 46 children into two groups from June 2 to August 30, 2017. The program was designed into 12 sessions of floral arrangement classes, followed by the certification test. For the program, this study set 11 flower forms focusing on line, surface, and mass arrangements as the basic forms of the western flower arrangements. For the certification test, this study had the participants take the junior florist course implemented by the Korea Ggotggozi Daegyeong Association. Seventy four percent of the total participants in this program acquired a Junior Florist Certificate. As a result, self-esteem of the experimental group significantly improved (p=.000). By items of self-esteem, the experimental group showed significant changes in social self-esteem (p=.002), familial self-esteem (p=.016), and academic self-esteem (p=.031). General self-esteem did not show a statistically significant difference, but there was an improvement by 1.9%, and this study found that there was a 5.4% change on average in social, familial, and academic self-esteem. By gender, it was found that both male (p=.003), and female participants (p=.013) showed a significant change. By grade, there were significant changes in both the lower grades (p=.014), and higher grades (p=.003).

A Structural Model for Chemotherapy Related Cognitive Impairment and Quality of Life in Breast Cancer Patients (유방암 환자의 항암화학요법 관련 인지기능 변화와 삶의 질의 구조모형)

  • Purpose: This study aimed to develop and test a structural model for chemotherapy-related cognitive impairment of breast cancer patients based on a literature review and Hess and Insel's chemotherapy-related cognitive change model. Methods: The Participants consisted of 250 patients who were ${\geq}19$ years of age. The assessment tools included the Menopause Rating Scale, Symptom Experience Scale, Hospital Anxiety and Depression Scale, Everyday Cognition, and Functional Assessment of Cancer Therapy-Breast Cancer. Data were analyzed using the SPSS 21.0 and AMOS 21.0 programs. Results: The modified model was a good fit for the data. The model fit indices were ${\chi}^2=423.18$ (p<.001), ${\chi}^2/df=3.38$, CFI=.91, NFI=.91, TLI=.89, SRMR=.05, RMSEA=.09, and AIC=515.18. Chemotherapy-related cognitive impairment was directly influenced by menopausal symptoms (${\beta}=.38$, p=.002), depression and anxiety (${\beta}=.25$, p=.002), and symptom experiences (${\beta}=.19$, p=.012). These predictors explained 47.7% of the variance in chemotherapy-related cognitive impairment. Depression and anxiety mediated the relations among menopausal symptoms, symptom experiences, and with chemotherapy related cognitive impairment. Depression and anxiety (${\beta}=-.51$, p=.001), symptom experiences (${\beta}=-.27$, p=.001), menopausal symptoms (${\beta}=-.22$, p=.008), and chemotherapy-related cognitive impairment (${\beta}=-.15$, p=.024) had direct effects on the quality of life and these variables explained 91.3%. Conclusion: These results suggest that chemotherapy-related toxicity is highly associated with cognitive decline and quality of life in women with breast cancer. Depression and anxiety increased vulnerability to cognitive impairment after chemotherapy. Nursing intervention is needed to relieve chemotherapy-related toxicity and psychological factor as well as cognitive decline for quality of life in patients undergoing chemotherapy.

A Study on the Influence of Human Resource Management of Franchise Restaurant Employees on Business Performance (프랜차이즈 요식업 종사자의 인적자원관리가 경영성과에 미치는 영향)

  • This study analyzed the effect of human resource management on the performance of franchise restaurant workers. First, employment management of franchisees did not have statistically significant effect on business performance. Second, evaluation management (p<.05, ${\beta}=1.887$) showed a significant effect on job improvement, but did not affect financial performance significantly. Third, career management (p<.05, ${\beta}=1.715$) was found to have an impact on financial performance, but it did not significantly affect job performance. Fourth, there was a somewhat higher correlation between job improvement and job management (.441), job improvement and evaluation management (.476), p<.01, and financial performance and employment management (.375) (.356), and financial performance and career management (.371) were found to be correlated with p<.05 significance level. This study analyzes the effect of human resources management on the performance of franchise restaurant workers, and finds out the importance of characteristics of factors such as recruitment management, career management, and evaluation management. Research has significance.

A plasma circulating miRNAs profile predicts type 2 diabetes mellitus and prediabetes: from the CORDIOPREV study

  • We aimed to explore whether changes in circulating levels of miRNAs according to type 2 diabetes mellitus (T2DM) or prediabetes status could be used as biomarkers to evaluate the risk of developing the disease. The study included 462 patients without T2DM at baseline from the CORDIOPREV trial. After a median follow-up of 60 months, 107 of the subjects developed T2DM, 30 developed prediabetes, 223 maintained prediabetes and 78 remained disease-free. Plasma levels of four miRNAs related to insulin signaling and beta-cell function were measured by RT-PCR. We analyzed the relationship between miRNAs levels and insulin signaling and release indexes at baseline and after the follow-up period. The risk of developing disease based on tertiles (T1-T2-T3) of baseline miRNAs levels was evaluated by COX analysis. Thus, we observed higher miR-150 and miR-30a-5p and lower miR-15a and miR-375 baseline levels in subjects with T2DM than in disease-free subjects. Patients with high miR-150 and miR-30a-5p baseline levels had lower disposition index (p = 0.047 and p = 0.007, respectively). The higher risk of disease was associated with high levels (T3) of miR-150 and miR-30a-5p ($HR_{T3-T1}=4.218$ and $HR_{T3-T1}=2.527$, respectively) and low levels (T1) of miR-15a and miR-375 ($HR_{T1-T3}=3.269$ and $HR_{T1-T3}=1.604$, respectively). In conclusion, our study showed that deregulated plasma levels of miR-150, miR-30a-5p, miR-15a, and miR-375 were observed years before the onset of T2DM and pre-DM and could be used to evaluate the risk of developing the disease, which may improve prediction and prevention among individuals at high risk for T2DM.

Developmental Toxicity by Exposure to Bisphenol A Diglycidyl Ether during Gestation and Lactation Period in Sprague-dawley Male Rats

  • Objectives : Bisphenol A diglycidyl ether (BADGE) is the major component in commercial liquid epoxy resins, which are manufactured by co-reacting bisphenol A with epichlorohydrin. This study was performed to show the developmental effects of prenatal and postnatal exposures to BADGE in male rat offspring. Methods : Mated female rats were divided into four groups, each containing 12 rats. The dosing solutions were prepared by thoroughly mixing BADGE in corn oil at the 0, 375, 1500 and 3000 mg/kg/day concentrations. Mated females were dosed once daily by oral gavage on gestation day (GD) 6 - 20 and postnatal day (PND) 0 - 21. Pregnant female dams were observed general symptoms and body weight. Also, male pups were observed the general symptoms, body weight, developmental parameters (e.g. anogenital distance, pina detachment, incisor eruption, nipple retention, eye opening, testis descent), organ pathologic changes and hormone levels of plasma. Results : Pregnant rats treated with BADGE died at a rate of about 70% in the 1500 mg/kg/day group and all rats treated with 3000 mg/kg/day died. Body weight, for male pups treated with doses of 375 mg/kg/day, was significantly lower than in the control group at PND 42, 56, and 63 (p<0.05). Evaluation of body characteristics including; separation of auricle, eruption of incisor, separation of eyelid, nipple retention, descent of testis, and separation of the prepuce in the BADGE treated group showed no difference in comparisons with the control group. AGD and adjusted AGD (mm/kg) for general developmental items in BADGE 375 mg/kg/day treated pups tended to be longer than in controls, however, these differences were not statistically significant. Relative weights of adrenal gland, lung (p<0.05), brain, epididymis, prostate, and testis (p<0.01) were heavier than in control in measures at PND 9 weeks. There were no significant changes in comparisons of histological findings of these organs. Loss of spermatids was observed in the seminiferous tubule at PND 9 weeks, but no weight changes were observed. The plasma estrogen levels were similar in the control and treatment groups at PND 3, 6 and 9 weeks. The plasma testosterone levels in the control group tended to increase with age. However, in the BADGE 375 mg/kg/day treated male pups it did not tend to increase. Conclusions : These findings suggest that BADGE is a chemical that has developmental effects consistent with it being an endocrine disruptor.

Chemical Constituents of Bark of Beilschmiedia kunstleri Gamble with Anticancer, Anti-Streptococcus pyogenes, Anti-Bacillus cereus and Anti Plesiomonas shigelloides Activities

  • Lauraceae is a family medicinal plant whose tubers possesses antimicrobial, and cytotoxic, such as antiparasitic and anti-inflammatory special effects and has been used for the medicine in the cure of hepatitis and rheumatism. The antimicrobial activities of bioactive compounds including one neolignan; kunstlerone (1) and two alkaloids include isocaryachine (2) and noratherosperminine (3) as well as crude hexane, methanol and dichloromethane extracts were evaluated. Additionally, the effect of compounds 1, 2 and 3 were evaluated on A549, PC-3, A375, HT-29 and WRL-68 cell lines. In conclusion, kunstlerone 1 showed moderate cytotoxicity against various cancer cell lines such as A549, PC-3, A375, HT-29 and WRL-68, respectively with $EC_{50}$ values of 28.02, 26.78, 33.78, 33.65 and $16.46{\mu}g/mL$. The crude methanol extract showed antigrowth activity against S. pyogenes II and B. cereus, with MICs of $256{\mu}g/mL$. The compounds kunstlerone (1), isocaryachine (2) and noratherosperminine (3) showed complete inhibition against P. shigelloides, with MIC ${\leq}60{\mu}g/mL$ compare to ampicillin, as a positive control, which showed antigrowth activity against P. shigelloides at MIC $10{\mu}g/mL$.

The Factors Affecting Self-Confidence in Performing Patient Safety Management Nursing students (간호대학생의 환자안전관리 수행자신감에 미치는 영향요인)

  • This study was conducted to identify the factors affecting the self-confidence in performing patient safety management in nursing students. As a result of the analysis, 7.0 points in knowledge of patient safety management, 3.86 points in attitude of patient safety management, 3.47 points in clinical decision making ability, and 4.15 points in self-confidence in performing patient safety management. Self-confidence in performing patient safety management was positively correlated with knowledge of patient safety management(r=.216, p<.01) and attitude of patient safety management(r=.375, p<.01), clinical decision making ability(r=.467, p<.01). The self-confidence in performing patient safety management was influenced by clinical decision making ability(β=.409, p<.001) and attitude of patient safety management(β=.198, p=.011), explanatory power was 26.3%.
