망막 내 노인성 형광색소의 광산화에 미치는 Brown 착색렌즈와 UV-A 차단 안경렌즈의 영향 (The Effect of Brown Tinted or UV-A blocking Ophthalmic Lens Against the Photooxidation of A2E, a Lipofuscin in Retina)

  • 목적: 본 연구는 노인성 황반변성(age-related macular degeneration, AMD)의 병인으로 잘 알려진 A2E의 광산화가 자외선 차단렌즈나 brown 착색렌즈에 의해 억제되는가를 알아보고자 하였다. 방법: 두 분자의 all-trans-retinal과 한 분자의 ethanolamine을 합성하여 만든 A2E에 청색광(420~470 nm, $94mW/cm^2$)을 3분 동안 조사하여 광산화를 유도하였다. UV 차단 및 brown 착색 안경렌즈의 A2E의 광산화 저해 효과는 광산화되지 않고 남아있는 A2E의 흡광도와 HPLC chromatogram 분석을 통하여 확인하였다. 결과: 자외선 차단렌즈는 청색광 조사로 유발되는 A2E의 광산화를 억제하지 못하였다. 반면 Brown 착색렌즈에서는 가시광선 차단율이 15%인 경우 A2E의 광산화율에는 별차이가 없었지만, 55%와 86% 차단된 렌즈에서는 각각 9.98%, 16.55%의 A2E 광산화 억제 효과를 보였다. HPLC로 분석한 잔존 A2E량을 확인해 보면 렌즈로 차단하지 않은 A2E의 잔존량은 $199.29{\pm}26.53{\mu}M$ 이었으나, 55% 차단율을 가진 brown 렌즈는 $264.58{\pm}31.91{\mu}M$, 86% 차단율을 가진 렌즈에서는 $402.93{\pm}26.68{\mu}M$의 A2E 잔존량을 보여 A2E 광산화 억제효과가 있음을 확인하였다. 반면, 자외선 차단렌즈의 경우에는 잔존량 분석에서도 광산화 억제효과가 없는 것으로 나타났다. 결론: 본 연구결과를 통하여 brown 착색 안경렌즈는 노인성 황반변성의 원인물질인 A2E의 광산화를 억제하는 효과가 있는 것으로 알 수 있었다.

Isolation of Prenylated Isoflavonoids from Cudrania tricuspidata Fruits that Inhibit A2E Photooxidation

  • High-performance liquid chromatography coupled to an online $ABTS^+$-based assay (online HPLC-$ABTS^+$) system was used to determine the principal antioxidants in Cudrania tricuspidata fruits. Six prenylated isoflavonoids (1 - 6) were isolated from C. tricuspidata fruits according to the online HPLC-$ABTS^+$ system. The structures of isolated compounds, alpiniumisoflavone (1), 6,8-diprenylorobol (2), 6,8-diprenylgenistein (3), pomiferin (4), 4'-methylalpiniumisoflavone (5), and osajin (6) were identified by their retention time, UV spectra, ESI-MS, and NMR data. Among these compounds, 6,8-diprenylorobol (2) and pomiferin (4) reduced A2E photooxidation in a dose dependent manner.

One-step isolation of sappanol and brazilin from Caesalpinia sappan and their effects on oxidative stress-induced retinal death

  • Caesalpinia sappan is a well-distributed plant that is cultivated in Southeast Asia, Africa, and the Americas. C. sappan has been used in Asian folk medicine and its extract has been shown to have pharmacological effects. Two homoisoflavonoids, sappanol and brazilin, were isolated from C. sappan by using centrifugal partition chromatography (CPC), and tested for protective effects against retinal cell death. The isolated homoisoflavonoids produced approximately 20-fold inhibition of N-retinylidene-N-retinyl-ethanolamine (A2E) photooxidation in a dose-dependent manner. Of the 2 compounds, brazilin showed better inhibition (197.93 ± 1.59 μM of IC50). Cell viability tests and PI/Hoechst 33342 double staining method indicated that compared to the negative control, sappanol significantly attenuated H2O2-induced retinal death. The compounds significantly blunted the up-regulation of intracellular reactive oxygen species (ROS), and sappanol inhibited lipid peroxidation in a concentration-dependent manner.   • Direct photoisomerizations of benzalpyrrolinones yield the corresponding E-isomers via a singlet state, since no effect of oxygen on the reaction rates was observed. The Z-oxodipyrromethene was photoisomerized to the E-isomer in a degassed system. In an aerobic system the oxodipyrromethene 3 was photoisomerized at the early stage of the reaction and photooxygenated slowly at latter stage of the reaction. For bilirubin, other (possibly Z ${\to}$ E) than self-sensitizing $^1O_2$ reaction should have occurred because of the lack of a solvent effect on the self-sensitized photooxidation reaction rate at the early stage.

Singlet Oxygen Quenching by Deoxygadusol and Related Mycosporine-Like Amino Acids from Phytoplankton Prorocentrum micans

  • Deoxygadusol (DO) and structurally related mycosporine-like amino acids, i.e. mycosporine glycine (MO) and mycosporine taurine (MT), were isolated from phytoplankton Prorocentrum micans and studied for the reactivity toward singlet oxygen. These water-soluble compounds with a cyclohexenone chromophore were all shown to be highly effective in quenching singlet oxygen ($^1$ $O_2$), with the efficiencies being significantly larger compared with histidine, a well-known $^1$ $O_2$ quencher. The $^1$ $O_2$ reaction rate constant ( $k_{Q}$) of DG was determined to be 5.4 ${\times}$ 10$^{7}$ $M^{-1}$ $s^{-1}$ by a steady state method based on competitive inhibition of rubrene oxidation. The feasibility of this method was confirmed by estimating the $k_{Q}$ values for MG and two other quenchers, furfuryl alcohol and 1,4-diazabicyclo [2,2,2]octane, and comparing with those values determined by the time-resolved $^1$ $O_2$ decay method in the previous work.ork.

하수 2차 처리수 재이용을 위한 저압 및 중압 고도산화시스템의 성능평가 (Assessment of Advanced Oxidation Processes using Low and Medium-Pressure Lamps with H2O2 for Reclamation of Biologically Treated Wastewater Effluents)

  • In the present study, the feasibility of $UV/H_2O_2$ systems was investigated using low and medium-pressure lamps on biologically treated wastewater effluents for secondary effluent reclamation. Two types of UV lamps were used as the light sources (a 39-W low-pressure mercury lamp and a 350-W medium-pressure mercury lamp). The results from these UV systems showed that the removal of organic compounds could be achieved in the contact time of longer than 30min (i.e., low UV doses). Efficiencies of color removal and disinfection were far better than those of organic matters measured as TOC, DOC and $TCOD_{cr}$. In the low-pressure lamp UV system, it has been found that DOC and color removals were 60.9 and 86.2% with 50mg/L of $H_2O_2$ and contact times of 30 minute, respectively. Whereas, with the medium-pressure lamp UV system, TOC, DOC and color removal were 27.1, 5.6 and 95% with 14.3mg/L of $H_2O_2$ and 14 minute of contact times, respectively. Both systems could be applied for the reclamation of secondary effluent treated with biological treatment processes.
