New Rust Disease of Korean Willow (Salix koreensis) Caused by Melampsora yezoensis, Unrecorded Pathogen in Korea

  • During the growing season of 2015, leaf specimens with yellow rust spots were collected from Salix koreensis Andersson, known as Korean willow, in riverine areas in Cheonan, Korea. The fungus on S. koreensis was identified as the rust species, Melampsora yezoensis, based on the morphology of urediniospores observed by light and scanning electron microscopy, and the molecular properties of the internal transcribed spacer rDNA region. Pathogenicity tests confirmed that the urediniospores are the causal agent of the rust symptoms on the leaves and young stems of S. koreensis. Here, we report a new rust disease of S. koreensis caused by the rust fungus, M. yezoensis, a previously unrecorded rust pathogen in Korea.

  • Morphological and ecological comparions are made between specimens of Cobitis koreensis KIM and the geographically restricted population of the Cobitid fish which is found along the Begchon stream, Puangun, Cholla-bugdo province over a 12-month period of 1982 and 1983. The Begchon population is tentatively identified as Cobitis koreensis, but the color patterns and two morphometric proportions differ widely fromthose reported previously for this species. In addition to these character, it is easily distinguished from C. koreensis by the smaller body size, by having the larger egg size and the less egg number in female during the breeding season. As the differentiation of this population is primarily due to geographically isolation, it is supposed that Begchon cobitid population may be an undescribed distinct subspecies of K. koreensis or above the conventional subspecies level.

한국산 잉어과어류 칼납자루(Acheilognathus koreensis) 정자의 미세구조 (Ultrastructure of Spermatozoa of a Korean Bitterling, Acheilognathus koreensis (Pisces, Cyprinidae))

  • 칼납자루 Acheilognathus koreensis 정자의 미세구조를 전자현미경으로 관찰하였다. 칼납자루 정자의 미세구조는 잉어과 어류정자의 일반적인 구조와 같이 둥근 두부, 얕은 핵와, 세포의 측면에 위치한 편모구조 그리고 비 대칭적인 편모구조의 특징을 취하고 있었다. 그러나 중편에 위치한 미토콘드리아는 대부분의 잉어류에서와는 달리 융합되어 하나를 형성하고 있으며 그 주위를 막성 구조물들이 둘러싸고 있었다. 하나로 융합된 미토콘드리아를 가지는 정자의 구조는 여러개로 나타나는 것에 비하여 경골어류에서는 파생형질로 알려져 있다. 잉어류 정자에서 미토콘드리아가 융합되어 하나로 나타나는 구조는 Rodeus와 Puntungia에서 보고되어 있으며 그 주위를 막성구조물이 둘러싸고 있는 것은 납자루류에서만 보고되고 있다.

  • The genus Iksookimia (Actinopterygii: Cypriniformes: Cobitidae) is a bottom-dwelling freshwater loaches, which are well-known as their endemism and high geographic variation. However, population genetic relationships among Iksookimia spp. have remained unclear due to a shortage of genetic markers that can be applied generally in the genus. Here, we developed high-resolving microsatellite markers using I. koreensis and I. longicorpa as representatives of Iksookimia species because of their wide distribution range and phylogenetic position. Ten of polymorphic microsatellite loci were isolated from Iksookimia koreensis and were successfully cross-amplified in I. longicorpa. The mean number of observed alleles per locus was about 10.4 (range, 2-17) for I. koreensis and about 13.2 (range, 2-24) for I. longicorpa. The loci, IK03 and IK08, deviated from the Hardy-Weinberg equilibrium in I. koreensis, after applying the Bonferroni correction. The microsatellite markers obtained in the present study will be useful to evaluate population genetic structure and to establish conservation strategies for I. koreensis and related Iksookimia species.

고유종 칼납자루의 재생산 기초 연구 (Reproduction Study of Korean Endemic Species Acheilognathus koreensis)

  • 칼납자루를 중심으로 납자루아과 고유종 각시붕어, 한강납줄개, 묵답자루의 재생산을 위한 기초 연구를 하였다. 난의 형태와 부화 자어는 종별로 독특하였으며 암컷의 크기와 1회 산란량의 상관관계는 뚜렷하게 나타나지 않았다. 칼납자루 $13.0^{\circ}C$에서도 산란이 가능하였으나 수정란의 부화율은 43.5%로 낮았으며, 수온 $21.0^{\circ}C$에서 79시간 만에 부화하였고 다른 비교 종들과 같이 수온이 높을수록 부화에 이르는 시간은 단축되었다. 칼납자루는 부화 후 자어 시기를 거쳐 300일 만에 52.4 mm로 성장하여 재생산에 참여할 어미가 되기에 충분하였다. 납자루아과 어류의 재생산 연구는 고유 담수어 보존 연구의 기초자료로 활용될 것이다.

  • 한국 특산종 Cobitis kireensis의 두 아종 C. k. koreensis와 C. k. pumilus의 지리적 변이 및 아종의 위치를 규명하기 위하여 남한의 6개 집단에서 채집된 457개체를 대상으로 형태 형질 분석 및 전기영동법에 의한 유전적 변이를 조사하였다. 형태형질의 계측, 계수 및 discriminant function 분석에서 집단 간 및 아종 간의 유의한 차이는 발견되지 않았다. 각 집단 간 유전적 변이는 평균 A=1.5, P =37.3%, HD=0.053, 및 HG=0.097로 타 어류군이 변이정도보다 다소 높은 편이었으나 C. k. pumilus는 A= 1.2, P=19.0%, HD=0.029, 및 HG=0.078로 변이정도가 매우 낮았다. 두 아종 간의 평균 유전적 근연치는 S=0.894로 C. k. koreensis 5개 집단간 평균 유전적 근연치 S=0.894와 거의 동일하였으며 아종 간에 유전적 차이가 없었다. 따라서 C. k. pumilus를 아종으로 분류하는 것은 부적합하다고 사료된다.

Antibacterial activity of supernatant obtained from Weissella koreensis and Lactobacillus sakei on the growth of pathogenic bacteria

  • This study was carried out to obtain basic data for the industrial use of Weissella koreensis and Lactobacillus sakei. The antibacterial activity of supernatants obtained from W. koreensis and L. sakei were tested against pathogenic bacteria such as Escherichia coli KCCM 11234, Salmonella enteritidis KCCM 3313, Salmonella enteritidis KCCM 12021, Salmonella typhimurium KCCM 40253, and Salmonella typhimurium KCCM 15. The supernatant of L. sakei showed antibacterial activity against E. coli KCCM 11234, S. enteritidis KCCM 12021, and S. typhimurium KCCM 15, while the supernatant of W. koreensis showed antibacterial activity against E. coli KCCM 11234 and S. enteritidis KCCM 12021. The effect of pH changes and heat treatment on antibacterial activity of the supernatants was examined using the sensitive pathogenic bacteria (E. coli KCCM 11234, S. enteritidis KCCM 12021 and S. typhimurium KCCM 15). Antibacterial activity against sensitive pathogenic bacteria was maintained under heat treatment at all temperatures, but there was no antibacterial activity associated with pH modification. Furthermore, it was confirmed that the antibacterial activity of the supernatants obtained from W. koreensis and L. sakei was a result of organic acids including, lactic, acetic, phosphoric, succinic, pyroglutamic, citric, malic, and formic acids. Therefore, the present study showed that the organic acids produced by L. sakei and W. koreensis exhibited a strong antibacterial activity against pathogenic bacteria. Moreover, in the food industry, these organic acids have the potential to inhibit the growth of pathogenic bacteria and improve the quality of stored food.

  • Total 513 heterophyid flukes were collected from a carcass of wild Korean raccoon dog, Nyctereutes procyonoides koreensis, in Korea. With morphological and molecular characteristics, the flukes were identified to Cryptocotyle lata. The adult C. lata were minute, transparent, pentagonal, 522 ㎛ long by 425 ㎛ wide. Ceca extended into post-testicular region. Ventrogenital sac elliptical, 79 ㎛ by 87 ㎛ with genital pore and ventral sucker. Two testes semielliptical and slightly lobed, located in the posterior region, right testis 173 ㎛ by 155 ㎛, left testis 130 ㎛ by 134 ㎛. In a phylogenetic tree, the fluke specimen of this study was grouped with C. lata divergent from Cryptocotyle lingua. We report here N. procyonoides koreensis first as a natural definitive host of C. lata.

Draft Genome Sequence of Weissella koreensis Strain HJ, a Probiotic Bacterium Isolated from Kimchi

  • Here we report the draft genome sequence of Weissella koreensis strain HJ and genomic analysis of its key features. The genome consists of 1,427,571 bp with a GC content of 35.5%, and comprises 1,376 coding genes. In silico analysis revealed the absence of pathogenic factors within the genome. The genome harbors several genes that play an important role in the survival of the gastrointestinal tract. In addition, a type III polyketide synthase cluster was identified. Pangenome analysis identified 68 unique genes in W. koreensis strain HJ. The genome information of this strain provides the basis for understanding its probiotic properties.

Complete Genome Sequence of Weissella koreensis DMW12 Isolated from Kimchi, Traditional Korean Fermented Vegetables

  • Weissella koreensis DMW12 was isolated from kimchi added Myeongtae (Theragra chalcogramma), and its complete genome sequence was determined. The complete genome of strain DMW12 includes a single circular 1,518,288-bp chromosome without plasmids. The G+C content of the genome is 35.6 mol%. Although strain DMW12 did not showed protease and lipase activities, the genome includes 33 protease- and 3 lipase-encoding genes. The genome of strain DMW12 does not include acquired antibiotic resistance genes against ampicillin, chloramphenicol, clindamycin, erythromycin, gentamicin, kanamycin, tetracycline, and streptomycin.
