First Description of Petalonia zosterifolia and Scytosiphon gracilis (Scytosiphonaceae, Phaeophyceae) from Korea with Special Reference to nrDNA ITS Sequence Comparisons

  • Scytosiphonaceae is an acetocarpalean brown algal family, that is a recent focus of synstematics and marine biodiversity. We describe Petalonia zosterifolia and Scytosiphon gracilis from Korea for the first time. P. zosterifolia occurred on the East coast, and had flat, linear and solid thalli. S. gracilis was found in Jeju, and had cylindircal to flat and hol-low thalli. However, these two species are so similar that it is difficult to identify by morphology alone. In order to determine if the nuclear DNA reveals the distinctness of both species and to know their phylogenies, the ITS region sequences were newly detrmined in 22 samples of P. zosterifolia, Scytosiphon gracilis, and other three members of the genera from Korea. We found 0.12% variation among samples of P. zosterifolia from different locations, and no variation between S. gracilis samples from diferent years, but extensive interspecific divergences (13.62-22.83%) of each species to other members in Petalonia and Scytosiphon . The ITS sequence dta consistently showed a close relationship between P. zosterifolia and S. gracilis. This result is congruent with morphology and with the published data of plastid rbc and partial nrDNA large subunit gene sequences, and suggests that P. zosterifolia and S. gracilis might have diverged from the most recent common ancestor.

First Record of Scytosiphon gracilis Kogame (Scytosiphonaceae,Phaeophyceae) for the Pacific coast of Mexico

  • In this work we report the occurrence of Scytosiphon gracilis Kogame (Scytosiphonaceae, Phaeophyceae) on the Pacific coast of Mexico. This is the first report of S. gracilis outside its previously known distribution in the western North Pacific (Korea and Japan). The identification was based on a morphological revision and a comparison of the plastid-encoded RuBisCO spacer sequences determined for Korean and Mexican algal material. Thalli were collected from the intertidal zone of Saldamando Beach, Baja California, in January 2003. The vegetative structure, as well as habitat and geographic distribution of the species are described. Reproductive structures were not found in our specimens. The poor presence/absence of S. gracilis in previous floristic studies of the area could be due to its small size and low frequency.

Euglena gracilis 추출물의 면역조절 및 생리활성 분석 (Immunomodulatory Effects of Euglena gracilis Extracts)

  • 본 연구에서는 다양한 생리활성을 나타내는 compound를 함유하고 있는 것으로 알려진 Euglena gracilis를 hot water로 추출하고(HWE), 그 잔존물을 methanol로 추출(HWME)한 후, murine macrophage RAW 264.7 cell에 처리하여 면역활성을 측정하였다. Murine macrophage RAW 264.7 cell에서 HWE 처리로 IL-1β와 TNF-α가 농도의존적으로 증가되었고, lipopolysaccharide에 의해 유도되는 IL-6와 TNF-α 생성이 HWME 처리에 의해 유의적으로 억제되는 것을 확인하였다. 또한 α-glucosidase, protein tyrosine phosphatase (PTP1B), tyrosinase, xanthine oxidase (XO), angiotensin converting enzyme (ACE) 등에 대한 저해활성을 조사하였다. 그 결과, E. gracilis HWE 추출물에서는 α-glucosidase, tyrosinase, ACE에서 약한 억제 활성을 보였으나, HWME 추출물에서는 PTP1B와 XO에서 유의적인 억제활성을 나타냈다. 따라서 본 연구를 통하여 E. gracilis 추출물의 면역조절 활성 뿐 만 아니라 당뇨와 심혈관 질환에 대한 유의적인 억제활성을 통해 다양한 건강기능성 식품의 소재로 활용될 수 있을 것으로 사료된다.

Gracilis pull-through flap for the repair of a recalcitrant recto-vaginal fistula

  • Recto-vaginal fistulas are difficult to treat due to their high recurrence rate. Currently, no single surgical intervention is universally regarded as the best treatment option for recto-vaginal fistulas. We present a case of recurrent recto-vaginal fistula surgically treated with a gracilis pull-through flap. The surgical goals in this patient were complete excision of the recto-vaginal fistula and introduction of fresh, vascularized muscle to seal the fistula. A defunctioning colostomy was performed 1 month prior to the present procedure. The gracilis muscle and tendon were mobilized, pulled through the freshened recto-vaginal fistula, passed through the anus, and anchored externally. Excess muscle and tendon were trimmed 1 week after the procedure. Follow-up at 4 weeks demonstrated complete mucosal coverage over an intact gracilis muscle, and no leakage. At 8 weeks post-procedure, the patient resumed sexual intercourse with no dyspareunia. At 6 months post-procedure, her stoma was closed. The patient reported transient fecal staining of her vagina after stoma reversal, which resolved with conservative treatment. The fistula had not recurred at 20 months post-procedure. The gracilis pull-through flap is a reliable technique for a scarred vagina with an attenuated recto-vaginal septum. It can function as a well-vascularized tissue plug to promote healing.

  • 한국산 날개응애를 조사하던 중에 2 미기록속, Acrotocepheus 1965, Humerobates Sellnick, 1928과 2 미기록종, 산굼부리오징어응애(신칭) (A. gracilis Aoki, 1965), 콧등날개응애(신칭) (H. flechtmanni Perez-Inigo & Perez-Inigo Jr., 1993)을 발견하였기에 보고하는 바이다.

10년 이상 추시된 박 근 유리 판 이식술 (Gracilis Muscle Free Flap Transplantation Followed Up Over 10 Years)

  • Purpose: Gracilis muscle free flap transplantation is chosen in the medium sized soft tissue defect and bone exposure from trauma and chronic osteomyelitis in the lower extremity. We set a study to search for gracilis free flaps to know whether symptoms were cured or recurred in patients that have passed over 10 years from flap transplantation. Materials and Methods: From August 1995 through September 2010, we have performed 28 cases of gracilis muscle free flap in the lower extremities. Ever since no case visited to demand any discomfort, breakdown or recurrence in the flap site on outpatient basis. We made a telephone call to patients or relatives documented in the medical record and only 2 cases visited outpatient department and 9 cases postponed the visit who satisfied with the final result but 17 cases had wrong telephone numbers. Causes, area of lower extremity, recipient vessels in the lower extremity, condition of the donor thigh and overall satisfaction of the flap transplantation in activities of daily living were investigated and written down in the medical record. Results: 11 cases were reviewed after average postoperative 13.7 years. Gracilis muscle flaps were not break down at the recipient site in all cases. The wound of donor thigh wound healed good and overall activities of daily living was satisfied in all cases. Conclusion: Gracilis muscle flaps which had performed and followed up average 13.7 years revealed confident in the medium sized soft tissue defect and bone exposure from trauma and chronic osteomyelitis in the lower extremity.

Larval Development of Oregonia gracilis (Crustacea: Decapoda: Majoidea: Oregoniidae) with a Key to the Known Oregoniid Zoeae from the Northern Pacific

  • The larvae of Oregonia gracilis are described, illustrated and compared with those of other known species of the Oregoniidae. The first zoea of O. gracilis of the present study is somewhat different from that of Hart (1960) especially in having a basis and an endopod of the first maxilliped with 2, 2, 3, 3 and 3, 2, 1, 2, 5 setations, respectively and an endopod of the second maxilliped with 1, 1, 5 setation. It is found the Oregoniidae must be a homogeneous group based on the zoeal morphology. A provisional key for the identification of the known zoeae of the Oregoniidae from the northern Pacific is provided.

  • Biological fixation of carbon dioxide using microalgae is known as an effective CO$_2$reduction technology. However, many environmental factors influence microalgal productivity. Optimal cultivation factors were determined for the green alga, Euglena gracilis Z, which offers high protein and vitamin E content for animal fodder. In batch culture in a photovioreactor, it was found that theinitial pH, temperature, CO$_2$concentration in air, and light intensity during the optimal cultivating conditions were 3.5, 27$^{\circ}C$, 5-10% and 520 ${\mu}$mol/㎡/s, respectively. When tap water and freshwater were used as cultivating media unsterilized tap water was found to be effective. A kinetic model was considered to determine the relationship between the specific growth rate and the light intensity. The half-velocity coefficient (K(sub)I) in the Monod model under photoautotrophic conditions was 978.9 ${\mu}$mol/㎡/s.

  • Korean Bryocoris species, B. gracilis and B. montanus, were reviewed. Of these, B. gracilis Linnavuori, 1962 is first reported in Korea and diagnosed, based on a single female specimen collected from Mount Hwangmaesan, Gyeongsangnam-do province. A key to the Korean species and photographs on external features are given.

  • The first zoea of Pugettia gracilis is described and illustrated for the first time. Its morphological characteristics are compared with those of other known species of the genus from the northern Pacific waters. Although the Pugettia zoeas of the northwestern and the northeastern Pacifies are very similar, they can be easily distinguished by their dorsal carapace spine. In the northwestern Pacific it is spinulate with a spinous tip, while in the northeastern Pacific it is smooth with a blunt tip.
