Formulation of Surimi and Surimi-based Products with Acceptable Gelling Ability from Squid Muscle (가열 젤 형성능을 가진 오징어 Surimi와 Surimi-based 제품을 위한 첨가물의 최적화)

  • We investigated the optimum formulation to improve the gelling ability of squid, Dosidicus gigas, surimi. The solubility of minced squid muscle was highest at pH 10.7, and lowest at pH 5.0. The yields of conventional surimi and protein recovery after alkaline pH-shift processing were $68.1{\pm}2.4%$ and $65.3{\pm}2.6%$, respectively, whereas the protein recovery with acidic pH-shift processing was only $21.2{\pm}1.6%$. The addition of 5% starch decreased the breaking force regardless of the kind of starch, while the mixture of corn, potato, and wheat starch (total 15%) increased the breaking force by up to 1.9 fold. The addition of 5% egg white, 5% porcine plasma protein, 0.3% $CaCl_2$, and 0.3% Polymix GA significantly increased the breakingforce (P<0.05). None of the ingredients examined in this study significantly affected the deformation value (P<0.05). The optimum concentrations of egg white and $CaCl_2$ to obtain a breaking force of 55 g and a whiteness of 70 were 2.69% and 0.22%, respectively.

A study on the frequencies of medicinal herb combinations in the prescriptions of "Bangyakhappyeon(方藥合編)" ("방약합편(方藥合編)" 수록(收錄) 처방(處方) 내(內)의 약물(藥物) 조합 빈도(頻度) 연구)

  • Through analyzing the frequencies of medicinal herb combinations in the prescriptions of "Bangyakhappyeon(方藥合編)", we could understand the characteristics of prescription composition and author's medical thoughts. So we conducted this study that analyze combination of 324 medicinal herbs in 467 prescriptions, and found meaningful combinations of 5,045. Among these, the most meaningful combination is the one of 10 herbs, Glycyrrhiza uralensis Fischer, Angelica gigas Nakai, Ziziphus jujube Mille, Atractylodes japonica Koidzumi, Poria cocas Wolf, Zingiber officinale, Rehmanniae radix preparata, Cinnamomum loureirii, Panax ginseng C.A.Meyer, Astragalus membranaceus. This combination is Sibjeondaebo-tang(Shiquandabu-tang) without Cnidium officinale and Paeonia japonica which compose Samul-tang(Siwu-tang). It means this combination is kind of modified Sibjeondaebo-tang(Shiquandabu-tang), which is more effective in tonifying qi than tonifying blood. Prescriptions of "Bangyakhappyeon(方藥合編)" involve herbs mostly used to improve weak and frail health and the herbs which are more effective in tonifying qi than tonifying blood. Through this study, we know the author' s one of medical view points on tonifying qi and blood, attaching importance to tonifying qi relatively to tonifying blood.

AMOEBA INFECTION IN OYSTER (CRASSOSTREA GIGAS) (굴에 기생하는 Amoeba에 대하여)

  • A species of enigmatic amoeba was found front the oysters culcured in Jinhae Bay and the Goje Bay in September-October 1978 and October 1979. Occurrence and the morphological characteristics of the amoeba were studied. Based on their peculiar morphological characters the enigmatic body was a species of parasitic amoeba. The amoebas were morphologically very similar to the amoeboid parasite described by Sawyer(1977). Occurrence of the oysters infected with the amoeboid ranged from zero to $13.3\%$. The infection rate did not show any relations to the depth of water where the oysters were suspended. The longest diameter of the amoeba is $24\;\mu$. The thin outer membrane is lightly stained with eosin and PAS positively. Initially one amoeboid was found in an ovum, and later it propagated to 3 to 4 individuals.

Studies on the Composition of Sterol and the Presence of Cholesterol ${\Delta}^7$-dehydrogenase in Marine Animal Products (동물성 수산식품 중의 Sterol 조성과 Cholesterol ${\Delta}^7$-dehydrogenase의 존재에 관한 연구)

  • Levels of sterols including ${\Delta}^7$-dehydrogenase isolated from the tissues of marine animal products (20 species) were determined on 1.5% OV-17 columm of gas-liquid chromatography. The composition showed that the mussels and clams contained various sterols in their tissues : cholesterol, brassicasterol. 24-methylenecholesterol with some minor components such as 22-trans-norcholesta-5,22-dien-3${\beta}$-ol, 22-cis-dehydrocholesterol, 22-trans-dehydrocholesterol, desmosterol, 7-dehydrocholesterol, campesterol, stigmasterol, ${\beta}$-sitosterol, isofucosterol, and 7-dehydrocholesterol which could be converted into vitamin $D_3$ in the skin tissue of animal was present in the muscle of oyster, Crassostrea gigas. On the other hand, the others including gastropoda were predominantly composed of cholesterol. The minor sterols such as 24-methylenecholesterol, stigmasterol and ${\beta}$-sitosterol in the fish intestines are supposed to be derived from dietary plankton. Cholesterol ${\Delta}^7$-dehydrogenase which could convert cholesterol into ${\Delta}^7$-dehydrogenase was present in the pickles of Tricurus haumela intestine.

Protective Effect of HP08-0106 on Ligature-induced Periodontitis in Rats

  • Periodontitis is an inflammatory disorder of the periodontium, characterized by destruction of the tooth supporting tissues including alveolar bone and mediated by various pro-inflammatory mediators. Here, we demonstrated that HP08-0106, composed of four crude drugs-Gardenia jasminoides Grandiflora, Angelica gigas Nakai, Rehmannia glutinosa, and Schizonepeta tenuifolia in a weight ratio of 2:2:1:2, perturbs inflammatory responses, osteoclast formation in LPS-induced RAW 264.7 cells and alveolar bone resorption in ligature-induced periodontitis. HP08-0106 decreased the protein level of iNOS and COX2 as well as the secreted level of IL-$1{\beta}$, indicating that HP08-0106 has antiinflammatory effects. HP08-0106 also inhibited the expression of genes associated with osteoclastogenesis including c-Fos, MMP-9 and TRAP. Moreover, HP08-0106 exhibited a protective effect from alveolar bone loss in ligature-induced periodontitis animal models. Our results strongly suggest that HP08-0106 represent an important therapeutic tool to treat inflammatory disorders associated with bone loss such as periodontitis.

Comparative Study on Physicochemical Characteristics of Oyster Shells, Crassostrea gigas, Cultured in Various Waters in Korea (해역별 굴 패각의 특성비교)

  • Shell of oyster has a function of shelter from predator as well as growth potential. We investigated the physicochemical characteristics of oyster shells from three difference oyster farms. The shells from Wando waters characterized high stability of nanostructure, ticker nacreous layer, high thermal decomposition temperature and low content of sulphur as well. These results represent that Wando coast is pristine water for oyster farming.

A Literature Review of Herbal Medicines on Osteoporosis Studies - Reviewing Articles Published after Year 2000 (골다공증 연구에 사용되는 처방(處方) 및 본초(本草)에 관한 문헌 연구 - 2000년 이후 발표된 논문을 중심으로)

  • Objectives : The purpose of this study was to investigate which herbs and herbal formulae are frequently used for the treatment of osteoporosis since 2000. Methods : We searched the CENTRAL, MEDLINE, AMED, CINAHL, Scopus, KISS, Korea MED, KERIS, Kmbase, Dbpia, J-STAGE, CNKI, J Oriental Rehab Med., J Korean Oriental Med., J Oriental Gynecol. using search key words osteoporosis, osteopenia, bone density and bone loss combined with herbal, botanical plant and phytomedicine. Results : 73 studies were reconfirmed and 83 herbs have been found. For single herb, most of them were categorized into Boyik-yak(補益藥, Buyi-yao). For herbal formulae, various herbal formulae are used. Calculating each herb in consisted in each herbal formula, Angelica Gigas Radix(當歸) and Rehmanniae Radix Preparat(熟地黃) turned out to be the most frequently used herb. Conclusions : Further studies regarding herbal medicines on osteoporosis are needed.

Mercury Contents of Medicinal Plants and the Cultivated Soils in Korea

  • This study was conducted to identify transition characteristics of mercury in several selected medicinal plants and to find the appropriate management for production of safety food. Cultivated soils and medicinal plants were collected at 29 sites for Angelica gigas (Korean angelica root), 68 sites for Platycodon grandiflorum (Balloon flower), 35 sites for codonopsis lanceolata (Deoduck), 36 sites for Dioscorea batatas (Chinese yam), 32 sites for Rehmannia glutinosa (Foxglove), 16 sites for Cnidium officinale makino (cnidium), and 26 sites for Astragalus membranaceus (milk vetch root) during the harvest season of 2013. Mercury in the soils and medicinal roots were analyzed with a Direct Mercury Analyzer. Average content of mercury in soils cultivated medicinal plants was $0.023mg\;kg^{-1}$ (range: from 0.003 to $0.074mg\;kg^{-1}$) and average content of mercury in medicinal plants was $0.003mg\;kg^{-1}$ (range: from 0.001 to $0.011mg\;kg^{-1}$), indicating that mercury in the surveyed soils and medicinal plants were not exceeded the Korean regulation.

Transferability of Cupped Oyster EST (Expressed Sequence Tag)-Derived SNP (Single Nucleotide Polymorphism) Markers to Related Crassostrea and Ostrea Species

  • Single nucleotide polymorphisms (SNPs) are widely acknowledged as the marker of choice for many genetic and genomic applications because they show co-dominant inheritance, are highly abundant across genomes and are suitable for high-throughput genotyping. Here we evaluated the applicability of SNP markers developed from Crassostrea gigas and C. virginica expressed sequence tags (ESTs) in closely related Crassostrea and Ostrea species. A total of 213 putative interspecific level SNPs were identified from re-sequencing data in six amplicons, yielding on average of one interspecific level SNP per seven bp. High polymorphism levels were observed and the high success rate of transferability show that genic EST-derived SNP markers provide an efficient method for rapid marker development and SNP discovery in closely related oyster species. The six EST-SNP markers identified here will provide useful molecular tools for addressing questions in molecular ecology and evolution studies including for stock analysis (pedigree monitoring) in related oyster taxa.

참당귀 뿌리 배양에 있어서 root segmentation과 식물생장조절제가 뿌리 생장과 decursinol angelate 생산에 미치는 영향

  • Decursinol angelate, a new anticancer agent, was produced by root cultures of Angelica gigas Nakai. In addition, difference of specific yields between primary and secondary root was investigated. It was found that specific yield of secondary root was much higher than that of primary root at various conditions, so that it was thought that the formation and growth of secondary root were feasible. From this point of view, effects of root segmentation and plant growth regulators (NAA, IBA) on root morphology and decursinol angelate production were examined. Root segmentation increased secondary root mass and product formation. On the other hand, addition of NAA or IBA at various concentrations promoted secondary root formation and production of decursinol angelate significantly. Five-fold increase of production was obtained at 4 mg/L of IBA compared to control without NAA and IBA.
