수성이상계에서 Pluronic F-68과 산소전달물질이 참당귀 현탁세포 증식에 미치는 영향 (Effects of Pluronic F-68 and Oxygen Vectors on the Cell Growth of Angelica gigas Nakai in Aqueous Two-Phase System)

  • 본 연구에서는 수성이상계에서 Pluronic F-68과 산소전달물질을 적용하여, 물질전달과 산소전달을 증가시켜 당귀세포의 증식을 향상시켰다. 특히 산소전달물질인 n-hexadecane이 Pluronic F-68보다 수성이상계에서의 당귀 세포증식에 더 긍정적임을 확인하였다. 따라서 Pluronic F-68과 적절한 산소 전달물질의 첨가는 수성이상계 뿐만 아니라 대량배양을 위한 고농도배양 등에 효과적으로 적용가능하리라 사료된다.

  • By-products produced from medicinal plants (MP) were evaluated as feed additives on growth performance, blood chemistry, and immune responses of the juvenile red lip mullet Liza haematocheila. A commercial diet was used as the control. Four experimental diets were prepared by combining 0.5% of MPs such as Panax ginseng (PG), Schisandra chinensis (SC), Angelica gigas (AG), and Rehmannia glutinosa (RG), via water absorption, to the control diet. Triplicate groups of fish averaging 61.8 g were fed one of experimental diets to apparent satiation for 8 weeks. Dietary supplementation of MPs did not affect the growth performance, biological indices and whole body composition of fish compared to the control. The fish fed with AG diet had an increased weight gain, and specific growth rate compared to fish in SC group. Decreased level of plasma glucose was observed in fish fed diet containing MPs. Lysozyme activity was significantly increased in the fish fed AG diet compared to that of fish in PG group. These results indicate that dietary supplementation of Angelica gigas may enhance growth performance and immune responses in juvenile red lip mullet.

용남광도 해역의 굴(Crassostrea gigas) 및 육상오염원에서 분리한 대장균(Escherichia coli)의 항생제 내성 (Antimicrobial Resistance in Escherichia coli Isolated from Oyster Crassostrea gigas and Inland Pollution Sources in the Yongnam-Gwangdo Area, Korea)

  • We isolated and characterized Escherichia coli (E. coli) from oyster Crassostrea gigas and inland pollution sources on Yongnam-Gwangdo island and neighboring areas on the southern coast of Korea in 2014-2015. A total of 222 strains of E. coli were isolated from 132 oysters and 88 samples from inland pollution sources. These 222 isolates were tested for their susceptibility to 24 antimicrobial agents, and 221 isolates were found resistant to the tested antibiotics. Of these 99.5% and 70.7% of the isolates showed very high resistance to rifampin and cephalothin respectively, followed by tobramycin (23.4%), streptomycin (20.2%), tetracycline (19.4%), cefepime (18.9%), ceftazidime (18.9%) and nalidixic acid (16.7%). The resistance rate of E. coli isolated from oysters was higher than that from inland pollution sources. In addition, multiple resistance to at least four antibiotics were present in 73.2% and 26.5% of E. coli isolates from oysters and inland pollution source samples, respectively.

Effects of Ovarian Parasite Marteilioides chungmuensis on the Reproduction of the Pacific Oyster Crassostrea gigas Assessed by Histology and Enzyme-Linked Immunosorbent Assay (ELISA)

  • The paramyxean parasite Marteilioides chungmuensis infects the cytoplasm of the eggs of Pacific oysters Crassostrea gigas , resulting in spawning failure of the infected females. Such infected eggs appear as bump-like nodules on the body in late fall when most of the uninfected females complete spawning. In this study, we estimated the quantity of the infected eggs using an enzyme-linked immunosorbent assay (ELISA), which is destroyed by M. chungmuensis parasitism. In December, the infected oysters collected from Tongyoung on the south coast exhibited numerous yellowish bump-like nodules as signs of infection. In histology, the infected oysters exhibited mature eggs in the follicle, which were heavily infiltrated by hemocytes. ELISA indicated that the infected egg mass accounted for 7.52±5.50 percent of the body weight, suggesting the ovarian parasite causes substantial reproductive loss. Histology also indicated that the infected oysters are in a poor nutritional condition, as the digestive gland atrophy (DGA) level is comparatively higher than the uninfected oyster. The total carbohydrate contents in the infected oysters (108.68±44.41 mg/g dry wt) were significantly lower than in uninfected oysters (269.76±50.97 mg/g dry wt), suggesting that M. chungmuensis parasitism also affected the energy storage capacity of the host during the resting stage.

생굴(Crassostrea gigas)의 선도 변화에 포장용기가 미치는 영향 (Effect of the Packaging Container on the Freshness of Raw Oysters Crassostrea gigas)

  • The shelf life of oysters Crassostrea gigas, in two different types of packaging containers, polyethylene (PE) and polyethylene terephthalate (PET), was determined by evaluating the pH, glycogen and soluble protein content, turbidity, and viable cell count. After 7 days of storage, the pH of the packing water in the PE container decreased to 5.88, while the pH in the PET container decreased to 6.03. In the PE container, the glycogen content of the oysters decreased by 0.85 g/100 g and the soluble protein content and turbidity of the packing seawater increased by 1,927.21 mg/100 g and 3.24 McF, respectively. In the PET container, the glycogen content of the oysters decreased by 0.96 g/100 g and the soluble protein content and turbidity of the packing seawater increased by 1,674.75 mg/100 g and 0.98 McF, respectively. The reaction rate constants (K) were as follows: glycogen content, -0.18 (PE) and -0.10 (PET); soluble protein content, 0.29 (PE) and 0.26 (PET); and turbidity, 0.41 (PE) and 0.06 (PET). These results suggested that PET can be used as a new packaging container material for raw oysters because the quality is maintained and it offers more convenient handling during distribution.

  • Pacific oysters Crassostrea gigas are a popular shellfish in the Republic of Korea. However, due to their abundant moisture and nutrient content, oysters are susceptible to microbiological growth and biochemical changes, which lead to quality degradation. Therefore, the present study aimed to investigate the effectiveness of modified atmosphere packaging (MAP) in maintaining the quality of raw oysters during storage. Microbiological and physicochemical parameters such as pH, glycogen content, soluble protein, turbidity, and volatile basic nitrogen (VBN) were analyzed for oysters stored under various gas compositions and storage periods. The results showed that there was no significant increase in viable cell count in MAP oysters after six days in MAP oysters. Moreover, the physicochemical quality of non-MAP oysters deteriorated rapidly, whereas the quality of MAP oysters were maintained during storage. This study suggests that MAP can be an effective technique for maintaining the freshness of raw oysters during distribution and storage, and may also be useful for extending the shelf-life and maintaining the quality of other seafood products.

RBL-2H3 세포에서 당귀(當歸) 및 산수유(山茱萸) 복합추출물의 알레르기 개선에 대한 효과 (Anti-Allergic Effects of Angelica gigas Nakai and Corni fructus extract (AC) on degranulation and production of cytokine in RBL-2H3 mast Cells)

  • Objectives : Recently, research has been actively conducted on the efficacy of complexes based on oriental medicine prescriptions for improving immune activity and allergies. In this study, In this study, we aimed to examine the effect of Angelica gigas Nakai and Corni fructus extract (AC), medicinal herbs, among candidate drugs derived through preliminary experiments with various components of oriental medicine prescriptions for allergies, on allergies in RBL-2H3 cells. Methods : We evaluated the effect of the ethanol extract of Ulmus on the allergic inflammatory response in anti-DNP-IgE activated DNP-HSA in RBL-2H3 cells. Cell toxicity was determined by WST-1 assay and the markers of degranulation such as beta-hexosaminidase, histamine, TNF-α and IL-6 production of inflammatory mediators and FcεRI-mediated expression. Results : The results showed that treatment with AC extract (20, 40 and 80㎍/㎖) noncytotoxic levels and significantly inhibited the release of β-hexosaminidase, histamine and the production of TNF-α and IL-6 in RBL-2H3 by the antigen stimulation. Conclusions : These results indicate that AC extract exhibits anti-allergic activity through inhibition of degranulation and inhibition of inflammatory mediators and cytokine release. These findings suggest that AC extract may have potential as a prophylactic and therapeutic agent for the treatment of various allergic diseases.

Agglutination Activity of Fasciola gigantica DM9-1, a Mannose-Binding Lectin

  • The DM9 domain is a protein unit of 60-75 amino acids that has been first detected in the fruit fly Drosophila as a repeated motif of unknown function. Recent research on proteins carrying DM9 domains in the mosquito Anopheles gambiae and the oyster Crassostrea gigas indicated an association with the uptake of microbial organisms. Likewise, in the trematode Fasciola gigantica DM9-1 showed intracellular relocalization following microbial, heat and drug stress. In the present research, we show that FgDM9-1 is a lectin with a novel mannose-binding site that has been recently described for the protein CGL1 of Crassostrea gigas. This property allowed FgDM9-1 to agglutinate gram-positive and -negative bacteria with appropriate cell surface glycosylation patterns. Furthermore, FgDM9-1 caused hemagglutination across all ABO blood group phenotypes. It is speculated that the parenchymal located FgDM9-1 has a role in cellular processes that involve the transport of mannose-carrying molecules in the parenchymal cells of the parasite.

Negative Correlation between the Prevalence of Norovirus and High Bacterial Loads of Escherichia coli in Oysters Crassostrea gigas

  • Seasonal variation in the prevalence of norovirus in oysters Crassostrea gigas was investigated and compared to levels of Escherichia coli, a fecal indicator in oysters. Oysters were collected from Iwon-myeon, Taean-gun, Korea, a primary production area for European Union export of oysters between 2013 and 2014. We observed seasonality in the prevalence of norovirus in oysters, with a higher prevalence and viral load detected during winter months. Oysters taken from production areas that complied with the European Union standard for raw consumption (< 230 MPN/100 g of E. coli) had 22.1% of the samples test positive for norovirus (15/68 samples). However, norovirus was not detected in any of the samples (0%, 0/4 samples) that were collected from production sites that exceeded the standard fecal contamination level for raw consumption (> 230 MPN/100 g of E. coli). These results indicated that there is a negative correlation between the prevalence of norovirus and high levels of E. coli in oysters. Therefore, our results suggest that current food safety guidelines using only a bacterial fecal contamination indicator, E. coli, may not adequately assess shellfish production areas for viral and bacterial contamination.
