수종 한약 복합물의 역형성갑상선암세포 SNU-80에 대한 항암효과 (Antineoplastic Effect of Several Herbal Medicine Mixtures on SNU-80 Anaplastic Thyroid Carcinoma Cell Line)

  • Objectives: The purpose of this study was to investigate the antineoplastic effect of several herbal medicine mixtures (compositions of Astragalus membranaceu, Angelica gigas, Trichosanthes kirilowii, Panax ginseng, Rhus verniciflua Stokes) on the SNU-80 anaplastic thyroid carcinoma cell line. Methods: MTT assay was used to examine whether our herbal medicine mixtures decreased cell growth rate of SNU-80. Wound healing assay and Transwell invasion assay was performed to investigate whether our herbal medicine mixtures affect the migration and invasion of anaplastic cancer cells, SNU-80. ELISA assay was performed to know if our herbal medicine mixtures suppressed the expression of pro-invasive molecules, such as vascular endothelial growth factor (VEGF) and matrix metalloproteinase-2 (MMP-2) secreted from SNU-80. Results: MTT assay demonstrated that A. membranaceus:A. gigas:T. kirilowii=1:1:1 or 3:1:1, A. membranaceus:A. gigas :T. kirilowii:P. ginseng=1:1:1:1 or 3:1:1:1, A. membranaceus:A. gigas:T. kirilowii:P. ginseng:R. verniciflua Stokes=1:1:1:1:1 or 3:1:1:1:1 strongly suppressed the growth of SNU-80. Wound healing assay demonstrated that A. membranaceus:A. gigas=3:1, A. membranaceus:A. gigas:T. kirilowii=1:1:1 or 3:1:1, A. membranaceus:A. gigas:T. kirilowii:P. ginseng=1:1:1:1 or 3:1:1:1, A. membranaceus:A. gigas:T. kirilowii:P. ginseng:R. verniciflua Stokes=1:1:1:1:1 or 3:1:1:1:1 inhibited the migration of SNU-80. Transwell invasion assay demonstrated that A. membranaceus:A. gigas=1:1, A. membranaceus:A. gigas:T. kirilowii =1:1:1 or 3:1:1, A. membranaceus:A. gigas:T. kirilowii:P. ginseng=1:1:1:1, A. membranaceus:A. gigas:T. kirilowii:P. ginseng :R. verniciflua Stokes=1:1:1:1:1 or 3:1:1:1:1 inhibited the invasion of SNU-80. ELISA assay demonstrated that A. membranaceus :A. gigas:T. kirilowii=1:1:1 or 3:1:1, A. membranaceus:A. gigas:T. kirilowii:P. ginseng:R. verniciflua Stokes=1:1:1:1:1 suppressed the expression of VEGF. Also, A. membranaceus:A. gigas=1:1, A. membranaceus:A. gigas:T. kirilowii=1:1:1 or 3:1:1, A. membranaceus :A. gigas:T. kirilowii:P. ginseng=1:1:1:1 or 3:1:1:1, A. membranaceus:A. gigas:T. kirilowii:P. ginseng:R. verniciflua Stokes =1:1:1:1:1 or 3:1:1:1:1 suppressed the expression of MMP-2. Conclusions: The results obtained in this study suggest that several herbal medicine mixtures suppresse the growth and inhibit the migration and invasion of SNU-80, which is anaplastic thyroid cancer cells. Especially, A. membranaceus:A. gigas: T. kirilowii=1:1:1 mixture had a stronger anti-cancer effect.

Angelica gigas 에탄올 추출물의 Hyaluronic acid 합성 효과에 대한 실험적 연구 (An Experimental Study on the Effect of Angelica gigas Ethanol Extract on Hyaluronic Acid Synthesis)

  • Objectives : Hyaluronic acid(HA) is a mucopolysaccharide, occuring naturally in living organisms. It is one of the most hydrophilic molecules, so it has been known as being related to skin hydration and skin aging. The purpose of this study is to examine the effects of Angelica gigas(A. gigas) ethanol extract on hyaluronic acid synthesis. Methods : To determine cytotoxicity and hyaluronic acid synthase 2 gene expression, hyaluronic acid production in HaCaT cells, MTT assay and RT-PCR ELISA was used. Results : There were no cytotoxicity in $50{\mu}g/ml$ concentration A. gigas extract in MTT assay. Hyaluronic acid synthase 2(HAS2) gene expression was increased by all treated concentration A. gigas extract. Hyaluronic acid production was higher than control group in $50{\mu}g/ml$ & $100{\mu}g/ml$ concentration A. gigas extract. Conclusions : Hyaluronic acid production was increased by A. gigas extracts. Therefore, We suggest that A. gigas can make a contribution to the moisturizing effect on human skin.

참당귀 추출물을 첨가한 쿠키의 품질 특성 (Quality Characteristics of Cookies Containing Powder of Extracts from Angelica gigas Nakai)

  • The principal objective of this study was to develop processed food with the effect of extract from Angelica gigas Nakai by assessing the quality characteristics of cookies. Cookie samples were prepared with shredded extract from Angelica gigas Nakai at varied levels of 0, 1, 3, and 5%. The pH of dough decreased significantly with the added extract from Angelica gigas Nakai and the density increased significantly in the 5% added group(p<0.05). The spread factor decreased significantly as the added extract from Angelica gigas Nakai increased(p<0.05). The Hunter color's L value in cookies significantly decreased and the a, b values in cookies increased(p<0.05). The hardness of cookies significantly increased added extract from Angelica gigas Nakai increased(p<0.05). The acid value and peroxide value were lower in added Angelica gigas Nakai extract groups than in the control group as the storage period of cookies passed. The results of sensory characteristics demonstrated that 1% added Angelica gigas Nakai extract showed the highest degree of preference among all the items of added Angelica gigas Nakai extract(color, odor, taste, hardness, crispness and overall quality) and the added Angelica gigas Nakai extract indicated possibilities for developments of Angelica gigas Nakai extract cookies.

UHPLC-DAD 및 다변량분석법을 이용한 참당귀의 산지감별법 연구 (Geographical Classification of Angelica gigas using UHPLC-DAD Combined Multivariate Analyses)

  • Geographical classification of A. gigas was performed in the present study using UHPLC-DAD combined with multivariate data analysis techniques. Six active constituents were isolated from A. gigas; nodakenin, marmesin, decursinol, demethylsuberosin, decursin and decursinol angelate. One hundred sixty eight A. gigas samples were simultaneously determined using UHPLC-DAD. A principal component analysis (PCA) and partial least square discriminant analysis (PLS-DA) was used to classify the samples according to geographical origins (Korea and China). The origins of A. gigas from Korea and China were correctly classified by 81.6% and 93.8% using PLS-DA Y prediction. This result demonstrates the potential use of UHPLC-DAD combined with multivariate analysis techniques as an accurate and rapid method to classify A. gigas according to their geographical origin.

The effect of extract from leaves and stalks of Angelica gigas on the innate immunity

  • The dried root of Angelica gigas (A. gigas) has been traditionally used as an oriental medicine, which is known to improve blood circulation and blood stasis. In the present study, leaves and stalks of A. gigas were used to investigate their effects on the innate immunity. The extracts were prepared from leaves and stalks of A. gigas and were fed to mice. The numbers of blood cells, total WBCs, neutrophils, lymphocytes, eosinophils and basophils were increased by 50% in mice fed with leaves extract of A. gigas compared to control mice. However, the numbers of blood cells were decreased when treated with stalks extract of A. gigas. The level of cholesterol and triglyceride in serum was markedly reduced in both mice group fed with leaves extract and stalks extract of A. gigas compared to control group (P<0.01). There was no significant change in the level of albumin, total protein, phosphate and calcium in serum. Activity of cationic peptide was found to be diffused in the testicles of mice fed with leaves extract of A. gigas compared to control group, which might be due to increased lysozyme in testicle. The lysoplate assay and immunohistochemistry assay suggest that the extract of leaves and stalks of A. gigas are immunogenic, but the effects might be related with acquired immune response rather than innate immunity.

백련초분말과 승검초분말 첨가에 따른 식빵의 품질 특성 (Quality Characteristics of Bread with Added Angelica plant (Bakluncho) and Angelica Gigas (Senggumcho) Powders)

  • The effects of adding Angelica plant (Bakluncho) and Angelica gigas (Senggumcho) powders on the quality characteristics of bread were investigated. The moisture, crude protein, ash, reducing sugar and vitamin C contents of Angelica plant (Bakluncho) powder were 7.78%, 6.07%, 7.37%, 6.13% and 50.10mg%, respectively, and of Angelica gigas (Senggumcho) powder were 8.69%, 12.28%, 5.15%, 2.21% and 108.00mg%, respectively. The lightness value decreased but the redness and yellowness values increased with the addition of Angelica plant (Bakluncho) and Angelica gigas (Senggumcho) powders. The mechanical hardness of bread decreased with the addition of 3% of Angelica plant (Bakluncho) powder, but increased with the addition of 1% and 5% of Angelica plant (Bakluncho) powder. The mechanical hardness of bread decreased with the addition of 1%, 3% and 5% of Angelica gigas (Senggumcho) powder. In sensory characteristics, the crust color, crumb color, moistness and springiness decreased, but aroma and hardness increased with the addition of Angelica plant (Bakluncho) and Angelica gigas (Senggumcho) powders. The savory taste and overall quality of breads with 1% Angelica plant (Bakluncho) and 1% Angelica gigas (Senggumcho) powders were higher than without either powder. A negative correlation was observed between mechanical hardness and overall bread quality with the addition of Angelica plant (Bakluncho) powder, while a positive correlation (p<0.01) was observed with the addition of Angelica gigas (Senggumcho) powder.

강원도 재배 참당귀 및 일당귀의 활성에 대한 비교연구 (Comparison of Activity of Angelica Gigas and Angelica Acutiloba from Kangwon)

  • In genus Angelica, three species have been used and cultivated for medical material in orient, A. gigas in korea, acutiloba in Japan and sinensis in China. The plant material of Angelica spp. is used for the treatment of women's disease as a hematic. The extracts from A. gigas and acutiloba were fractionated aqueous partitions. And study was performed to examine DPPH scavenging activities, BSA degradadion, anti-apoptosis and NO scavenging. DPPH radical scavenger activity was measured by DPPH method, it was shown dose-dependently effect. and BSA degradadion was shown same result. Treatment of cells with hydrogen peroxide, a reactive oxygen species was to indiced cell death and pretreatment with Angelica gigas and angelica acutiloba extract attenuated the occurrence of $H_2O_2$-induced cell death. In vitro nitric oxide (NO) scavenging effect on Angelica gigas and angelica acutiloba extracts. All extracts effectively reduced the generation of NO radicals in a dose-dependant manner.

  • Spermatogenesis, ultrastructure, and sperm morphology of the Pacific oyster (Crassostrea gigas) were investigated with TEM and SEM. C. gigas sperm were primitive consisting of a head midpiece and tail. Sperm size (head and midpiece) was about 1.78 ${\mu}m$. Sperm morphology was similar to a sharp of a small water jar with a rough surface. Sperm had both anterior nuclear fossa (anf) and posterior nuclear fossa (pnf). Acrosome forms had a hat-like shape. The axial rod was projected in front of the acrosome. C. gigas sperm had four large mitochondria in the midpiece.

Cytotoxic Coumarins from the Roots of Angelica gigas NAKAI

  • Some known coumarins, decursin, nodakenetin, umbelliferone, 7-demethylsuberosin, columbianetin, decursinol angelate and decursinol, showing significant cytotoxic activities against P388 cell lines,were isolated from the roots of Angelica gigas (Umbelliferae) . 7-Demethylsuberosin and columbianetinwere obtained from Angelica gigas for the first time. Chernotaxonornic difference about coumarins com-ponents between the roots of Angelica gigas and those of A. acutiloba is also discussed.

참당귀에서 분리된 내생균의 다양성과 병원균 억제 효과 (Species Diversity and Antifungal Activity of Endophytic Fungi Isolated from Angelica gigas Nakai)

  • 다양한 지역에서 채취한 참당귀(Angelica gigas Nakai)의 잎, 줄기, 뿌리에서 내생균을 분리하였다. 내생균 균주는 형태적특징과 ITS (internal transcribed spacer) 지역의 염기서열을 이용하여 동정하였다. 총 35종의 내생균이 동정되었다. 내생균의 다양성은 참당귀의 조직에 따라 다르게 나타났다. 분리된 내생균은 참당귀 점무늬병균 Phoma sp. Y11 균주와 대치배양하여 항균활성을 측정하였다. 총 14종의내생균이균주 Y11에대해억제효과를나타내었다. 본 연구는 참당귀에서 분리한 내생균이 참당귀의 점무늬병에 대한 생물학적 방제제로 사용될 수 있다는 것을 제시하고 있다.
