Basic Studies for Increment of Germanium Contents in Angelica keiskei KOIDZ., and A. acutiloba KITAGAWA (명일엽(明日葉)과 일당귀(日當歸)의 Germanium 함량(含量) 증대(增大)를 위한 기초연구(基礎硏究))

  • This study was carried out to examine the germanium contents of Angelica keiskei Koidz. and A. acutiloba Kitagawa and to intend to increase its contents while those leaf explants were culturing on MS medium supplemented with organic and inorganic germanium. Ge content of Agelica keiskei Koidz. was 2.1 times higher than that of A. acutiloba Kitagawa. Digestion was done quickly at high temperature, but Ge content was decreased. Callus formation of A. acutiloba Kitagawa was better than that of A. keiskei Koidz. Callus formation of both plants was good in order of pH 5.7, pH 5.4, and pH 6.0. But shoots from callus were formed frequently in A. keiskei Koidz., especially at pH 5.7. Callus formation of both plants was good up to 5 ppm of inorganic ($germanium(GeO_2),$ retarded at 10 ppm, and rarely formed at 100 ppm, but was good up to 10ppm of organic germanium retarded at 50 ppm and formed some-what even at 100 ppm.

Antioxidative Effect of Angelica acutiloba Kitagawa Ethanol Extract (일당귀 에탄올 추출물의 항산화 효과)

  • This study was conducted to investigate the antioxidant effect of 80% ethanol extracts from Angelica acutiloba Kitagawa (A. acutiloba Kitagawa) in vitro. The extract was further fractionated subsequently by n-hexane, chloroform, ethylacetate, n-butanol and water. Antioxidative activities of different fractions were examined by 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical generation, Rancimat test, thiobarbituric acid (TBA) value, nitrite scavenging activity, inhibition of lipid peroxidation and peroxide value (POV) in linoleic acid in comparison with the commercial antioxidant butylated hydroxy toluene (BHT). Antioxidant activities of n-hexane fraction of Angelico acutiloba Kitagawa ethanol extract were the highest among fractions and were a little less than that of BHT. Nitrite scavenging activity showed the most remarkable effect at pH 12. These results suggest that ethanol extracts of A. acutiloba Kitagawa can be used in natural antioxidant source.

Seed Germination Rate and Growth Characteristics according to Ripening Stages in Angelica acutiloba Kitagawa (일당귀의 등숙에 따른 종자 발아 및 생육 특성)

  • Background: Angelica acutiloba Kitagawa belongs to the flowering plant family Apiaceae. Its young leaves are consumed as a vegetable, and its roots have medicinal properties. This study was conducted to analyze the seed germination characteristics of A. acutiloba according to its ripening stages. Methods and Results: The seeds were harvested from a research farm managed by the Department of Herbal Crop Research, Rural Development Administration in 2018 and were divided into six groups according to their specific gravities. In particular, we studied the effect of harvesting periods and umbel orders on seed characteristics. The results showed that the rates of germination, emergence, and early growth improved as the specific gravity of the seeds increased. In addition, the germination rates of the seeds harvested in mid July and early August were significantly higher than those harvested in the other seasons, and the seeds obtained from the first floret had the greatest mass and weight. Conclusions: In this study, we demonstrated that it is possible to improve the germination rate by appropriate selection of seeds and harvesting period both of which are closely related to seed maturity.

Antioxidative and Nitrite Scavenging Abilities of Medicinal Plant Extracts (한약재 추출물의 항산화작용 및 아질산염 소거작용)

  • The purpose of this study was to investigate antioxidative and nitrite scavenging abilities of extracts of medicinal plants. Three kinds of medicinal plants, Angelica acutiloba Kitagawa (AA), Akebia quinate Decaisne (AQ), and Caragana chamlagu (CC), were extracted with water and $70\%$ ethanol. The yields of water extracts were increased in the order of Akebia quinate Decaisne ($35.63\%$, Caragana chamlagu ($31.36\%$ and Angelica acutiloba ($8.98\%$). Those of ethanol extracts were Akebia quinate Decaisne ($31.82\%$, Angelica acutiloba Kitagawa ($14.15\%$, and Caragana chamlagu ($5.20\%$). The highest electron donating ability (EDA) at 300-1,000 ppm of water extract was Angelica acutiloba Kitagawa, ranged from $62\%$ to $88\%$ and that of ethanol extract was Akebia quinate Decaisne, ranged from $72\%$ to $87\%$. Nitrite scavenging ability (NSA) of extracts measured at various pH (1.2, 3.0, 4.2, 6.0) showed highest ability in all extracts at pH 1.2 and decreased with increasing pH. The highest NSA of water extracts of 1,000 ppm at pH 1.2 showed $35-40\%$, in Caragana chamlagu and that of ethanol extract was $33-42\%$, in Akebia quinate Decaisne. From these results, Akebia quinate Decaisne was found as a potential raw material baring functionality in the respects of extract yield, EDA and NSA.

Discrimation of the three Angelica species using the RADPs and Internal Root Structure (RAPD 분석과 뿌리의 내부구조 비교를 통한 당귀류의 감별)

  • Analysis of random amplified polymorphic DNAs(RAPDs) and internal morphological features were performed using three species of medicinal plants in the genus of Angelica(A. gigas Nakai, A. sinensis(Oliv.) Diels., A. acutiloba Kitagawa) to distinguish between these three species. Fifty decarmer oligonucleotide primers were screened for the RAPDs of the herbal plant species. Five primers generated distinct RAPD markers specific to the species of Angelica, In analysis of the degree of similarity, A. sinensis(Oliv.) Diels is more closely related to A. acutiloba Kitagawa than to A. gigas Nakai. Furthermore, we proved the usefulness of RAPD analysis for the discrimination of the species using dry roots and commercial plant materials. In internal morphology of three species, A. sinensis(Oliv.) Diels seemed to be more specialized in systemic than A. acutiloba Kitagawa and A. gigas Nakai

A Basic Study on Leaf and Stem Production of Angelica acutiloba

  • This study was conducted to develop effective production system in greenhouse for leaves and stems of Angelica acutiloba by using connect pots in 2003 and 2004. Seed germination rate and plant biomass of Angelica acutiloba collected in 2004 were higher than those harvested in 2003. Germination rate of Angelica acutiloba seeds collected in 2003 was 10%, while germination rate of seeds collected in 2004 was above 90%. Especially, plant growth and yield of Angelica acutiloba grown in connect pots sized with 4 ${\times}$ 4 ${\times}$ 5cm(length ${\times}$ width ${\times}$ height) were the highest. These results indicate that leaf and stem production of Angelica acutiloba can be improved by using connect pots and optimizing seed collecting time in greenhouse.

Effects of Insects on Pollination in Angelica gigas Nakai and Angelica acutiloba Kitagawa (방화 곤충에 의한 당귀의 화분매개 효과)

  • It is known that the process of variety development and maintenance of original plant is difficult in Angelica gigas and Angelica acutiloba because they are allogamous plants. The contents like this are stand in the way of theirs breeding. This study was conducted to investigate the foraging activity and pollinational efficacy by insects to flowers of A. gigas and A. acutiloba. Foraging activity by pollinators was observed every day from 10:00 to 10:30 during 3 days after September 21. Honey bee and fly were major pollinators. The rate of hymenoptera and diptera to total numbers of insects was the 56.5% and 40.6% in A. gigas, respectively. It was also 46.3% and 51.8% in A. acutiloba, respectively. The other insects were lepidoptera, coleoptera and hemiptera. The numbers of hymenoptera and diptera were more in flowers of A. gigas was 49.2% with bee, 36.0% with fly, 15.3% with human, 0.9% without anything in the nethouse and 76.6% in the open field. That of A. acutiloba was 43.6% with bee, 27.9% with fly, 20.3% with human, 0.6% without anything in the nethouse and 46.9% in the open field. Excepting open field, seed setting rate of with bee in the nethouse was the highest of all the others. However, we concluded that the use of fly was the most effective method for pollination of A. gigas and A. acutiloba considering low cost and convenient handling.

Comparison of Volatile Compounds in Plant Parts of Angelica gigas Nakai and A. acutiloba Kitagawa (참당귀와 일당귀의 부위별 휘발성 정유성분 비교)

  • Volatile flavor compounds Angelica gigas Nakai and Angelica acutiloba Kitagawa were extracted by SDE (simultaneous steam distillation & extraction) using the mixture of n-pentane and diethylether (1:1, v/v) as an extract solvent and analyzed by GC-FID and GC-MS. The amount of essential oils of top part and root in Angelica gigas were obtained in 0.063% (v/w) and 0.389% (v/w) yields as a fresh weight base, respectively. The main compounds in top parts and the root were identified as nonane (7.51% and 24.49%, respectively), ${\alpha}-pinene$ (14.64% and 31.75%), limonene+${\beta}-phellandrene$ (14.01% and 9.66%), ${\gamma}-terpinene$ (7.85% and 1.20%), germacrene-d (5.85% and 0.22%), (E,E)-${\alpha}-farnesene$ (6.05% and 1.40%), ${\beta}-eudesmol$ (5.26% and 1.84%). Although these compounds were present in both parts. The results showed large differences in. the concentrations of them much varied. The amount of essential oils stem and leaf obtained (0.068% and 0.127% in A. gigas) and (0.153% and 0.243% in A. acutiloba) yields as a fresh weight base, respectively. More than 18 and 32 components in stem and leaf have been identified, which of main components in A. gigas were ${\alpha}-pinene$, myrcene, limonene, germacrene-d, eudesmol and butylphthalide, but germacrene-d and butylphthalide contents were also different in stem and leaf. And more than 21 and 32 components in A. acutiloba were ${\gamma}-terpinene$ and butylphthalide. Volatile compounds were very different in both species.

  • The effects of various kind and amount of nitrogen fertilizers on the plant grorth. physiological , yield and extracts. ligustilide, butylidene phthalide contents of curdsdrug "Tou-ki" (AnTelica acutiloba Kitagawa) were investigated in 1989. Five plots variouskind of nitrogen fertilizers. Namely, no nitrogen, urea. ammonium nitrate, ammoniumchloride and control plot of ammonium sulfate. The other, by providing five plotsdifferent composed ratio of nitrogenous fertilizers. containing no nitrogen (No. o) .0.5-fold nitrogen (No s), control plot of 1.0-fold nitrogen(Nl.o). 1.5-fold nitrogen(Nl s)and 2.0-fold nitrogen Na. o), but nitogen was used the ammonium sulfate. The results showedthat the crude drug "Tou-ki" can ammonium chloride be produced in good yield, displayingboth good plant growth and remarkable physiologically-active conditions, and it can beproduced such that the extracts is maximized. Additionally, using methods of gaschromatography (GC) , it was established that ligustilide and butylidene phthalide, majorcomponents in the crude drug was recovered in a good yield from the fully grown plants.The other, the plant growth, the physiologically-active, the weight of whole plant, theyield of extracts and ligustilide, butylidene phthalide were seen to be best at the plotof 2.0-fold nitrogen and according to the increase or decrease of nitrogen decreasedgradually. Therefore. about 2 fold of standard quantity seems to be the most suitablequantity of nitrogen for "Tou-ki" cultivation .uot;Tou-ki" cultivation .ion .

Germination Characteristics according to GA3 Treatment and Temperature of Angelica acutiloba (Siebold & Zucc.) Kitagawa (일당귀의 GA3 처리 및 온도에 따른 발아특성)

  • 일당귀[Angelica acutiloba (Siebold & Zucc.) Kitagawa]는 산형과(Apiaceae), 당귀속(Angelica)에 속하는 다년생 식물로 일본이 원산지다. 국내에서는 참당귀(A. gigas)와 더불어 뿌리를 약용의 목적으로 재배하며, 잎과 줄기는 쌈채소로 이용하는 작물 중 하나이다. 본 연구의 목적은 일당귀에 대한 GA3 처리 및 온도에 따른 발아특성을 분석하여 약용 및 식용으로 활용도가 높은 일당귀의 재배 및 생산에 대한 기초자료로 사용함에 있다. 연구에 사용된 일당귀의 종자는 강원도 태백시에 위치하는 일당귀 재배농가에서 2021년 10월에 채종한 종자를 구입하여 실험에 사용하였고, 종자의 수분흡수율과 다온도 종자발아기를 활용하여 GA3농도 4조건(50 ppm, 100 ppm, 500 ppm, 1,000 ppm), 온도 6조건(5 ℃, 10 ℃, 15 ℃, 20 ℃, 25 ℃, 30 ℃)에 따른 발아특성을 조사하였다. 연구결과 일당귀 종자 100립의 초기 무게 0.264 g에서 증류수 침지 2시간이 경과 후 약 60 %의 무게가 증가한 0.424 g으로 증가하였고, 이후 5 %내외의 수분흡수율을 보이며 48시간 경과 후 약 120% 증가한 0.587 g을 나타냈으며, 이후 72시간 경과 후 0.582 g으로 측정되어 일당귀 종자의 수분포화도는 약 120%정도 내외로 사료된다. GA3농도 및 온도에 따른 발아율은 500 ppm의 GA3농도에서 가장 높은 75.63 %의 발아율을 나타냈으며, 15 ℃와 20 ℃의 온도에서 가장 높은 77.6 %의 발아율을 나타냈다. 하지만 각각의 GA3농도 및 온도에 따른 발아율은 500 ppm의 GA3농도, 10 ℃의 온도 처리구에서 가장 높은 89.9 %의 발아율을 나타냈으며, 5 ℃의 온도 처리구에서는 모든 GA3처리구에서 발아가 진행되지 않았다.
