Blockade of Kv1.5 by paroxetine, an antidepressant drug

  • Paroxetine, a selective serotonin reuptake inhibitor (SSRI), has been reported to have an effect on several ion channels including human ether-a-go-go-related gene in a SSRI-independent manner. These results suggest that paroxetine may cause side effects on cardiac system. In this study, we investigated the effect of paroxetine on Kv1.5, which is one of cardiac ion channels. The action of paroxetine on the cloned neuronal rat Kv1.5 channels stably expressed in Chinese hamster ovary cells was investigated using the whole-cell patch-clamp technique. Paroxetine reduced Kv1.5 whole-cell currents in a reversible concentration-dependent manner, with an $IC_{50}$ value and a Hill coefficient of $4.11{\mu}M$ and 0.98, respectively. Paroxetine accelerated the decay rate of inactivation of Kv1.5 currents without modifying the kinetics of current activation. The inhibition increased steeply between -30 and 0 mV, which corresponded with the voltage range for channel opening. In the voltage range positive to 0 mV, inhibition displayed a weak voltage dependence, consistent with an electrical distance ${\delta}$ of 0.32. The binding ($k_{+1}$) and unbinding ($k_{-1}$) rate constants for paroxetine-induced block of Kv1.5 were $4.9{\mu}M^{-1}s^{-1}$ and $16.1s^{-1}$, respectively. The theoretical $K_D$ value derived by $k_{-1}/k_{+1}$ yielded $3.3{\mu}M$. Paroxetine slowed the deactivation time course, resulting in a tail crossover phenomenon when the tail currents, recorded in the presence and absence of paroxetine, were superimposed. Inhibition of Kv1.5 by paroxetine was use-dependent. The present results suggest that paroxetine acts on Kv1.5 currents as an open-channel blocker.

Study on the Counterflow Regenerative Evaporative Cooler with Finned Channels (대향류 핀삽입형 재생증발식 냉방기 연구)

  • The regenerative evaporative cooler(REC) is to cool a stream of air using evaporative cooling effect without an increase in the humidity ratio. In the regenerative evaporative cooler, the air can be cooled down to a temperature lower than its inlet wet-bulb temperature. Besides the cooling performance, for practical application of the regenerative evaporative cooler, the compactness of the system is also a very important factor to be considered. In this respect, three different configurations, i.e., the flat plate type, the corrugated plate type, and the finned channel type are investigated and compared for the most compact configuration. The optimal structure of each configuration is obtained individually to minimize the volume for a given effectiveness within a limit of the pressure drop. Comparing the three optimal structures, the finned channel type is found to give the most compact structure among the considered configurations. The volume of the regenerative cooler can be reduced to 1/8 by adopting the finned channel type as compared to that of the flat plate type.

Analysis of temperature-dependent abnormal bursting patterns of neurons in Aplysia

  • Temperature affects the firing pattern and electrical activity of neurons in animals, eliciting diverse responses depending on neuronal cell type. However, the mechanisms underlying such diverse responses are not well understood. In the present study, we performed in vitro recording of abdominal ganglia cells of Aplysia juliana, and analyzed their burst firing patterns. We identified atypical bursting patterns dependent on temperature that were totally different from classical bursting patterns observed in R15 neurons of A. juliana. We classified these abnormal bursting patterns into type 1 and type 2; type 1 abnormal single bursts are composed of two kinds of spikes with a long interspike interval (ISI) followed by short ISI regular firing, while type 2 abnormal single bursts are composed of complex multiplets. To investigate the mechanism underlying the temperature dependence of abnormal bursting, we employed simulations using a modified Plant model and determined that the temperature dependence of type 2 abnormal bursting is related to temperature-dependent scaling factors and activation or inactivation of potassium or sodium channels.

DESIGN OPTIMIZATION OF UPPER PLENUM OF PBMR USING RESPONSE SURFACE APPROXIMATION (반응면기법을 이용한 PBMR 기체냉각형 고온가스로 상층부의 최적설계)

  • Shape optimization of an upper plenum of a PBMR type gas cooled nuclear reactor has been performed by using three-dimensional Reynolds-Averaged Navier-Stokes (RANS) analysis and surrogate modeling technique. The objective function is defined as a linear combination of uniformity of flow distribution in the core and pressure drop in the upper plenum and the core. The ratio of thickness of slot to diameter of rising channels, ratio of height of upper plenum to diameter of rising channels, and ratio of height of the slot at inlet to outlet, are used as design variables for optimization. Design points are selected through Latin-hypercube sampling. The optimal point is determined through surrogate-based optimization method which uses 3-D RANS analyses at design points. The results show that the optimum shape represent remarkably improved performance in flow uniformity and friction loss than the reference shape.

Involvement of Ca2+ and K+ channels in the action of NO on gastric circular muscle (기니피그 유문부 윤상근의 자발적 수축 및 서파에 대한 nitric oxide의 억제적 작용과 Ca2+ 및 K+ 통로의 관련성)

  • It was investigated whether $Ca^{2+}$ and $K^+$ channels were involved in the inhibitory action of nitric oxide (NO) on the contractile and slow wave activity of guinea pig gastric antral circular muscle. The gastric antral circular muscle showed spontaneous phasic contraction and slow wave. NO donors, 3-morpholinosydnonimine hydrochloride (SIN-1, $0.01{\sim}100{\mu}M$) and S-nitroso-L-cysteine (CysNO, $0.001{\sim}10{\mu}M$), reduced not only the amplitude of phasic contraction but also that of slow wave in a concentration-dependent manner. Both the perfusion of $Ca^{2+}$-free solution and the administration of $Ni^{2+}$, a nonselective $Ca^{2+}$ channel blocker, reduced the phasic contraction as well as the amplitude and frequency of the slow wave. The effects of these treatments were similar to those of NO donors. Nifedipine ($10{\mu}M$), a specific L-type $Ca^{2+}$ channel blocker, abolished the phasic contraction and remarkably reduced the plateau of slow wave but had no profound effect on the upstroke of slow wave. In the whole-cell patch clamp mode, CysNO shifted the steady-state activation curve for L-type $Ca^{2+}$ current to the right and the steady-state inactivation curve to the left. Pretreatment of various $K^+$ channel blockers such as tetraethylammonium (1 mM), 4-aminopyridine (0.5 mM), glibenclamide (10 mM), apamin ($0.1{\mu}M$), and iberiotoxin ($0.1{\mu}M$) did not affect the inhibitory action of SIN-1. These results suggest that NO donors suppress mechanical and electrical activity of guinea pig gastric antral circular muscle by inhibition of L-type $Ca^{2+}$ channel rather than by activation of $K^+$ channels.

Role of $Ca^{2+}$ and Calmodulin on the Initiation of Sperm Motility in Salmonid Fishes

  • $K^+$ efflux through a certain type of $K^+$ channels causes the change of membrane potential and leads to cAMP synthesis in the transmembrane cell signaling for the initiation of sperm motility in the salmonid fishes. The addition of $Ca^{2+}$ conferred motility to the trout sperm that were immobilized by external $K^+$ and other alkaline metals, $Rb^+$ and $Cs^{2+}$, suggesting the participation of external $Ca^{2+}$ in the initiation of sperm motility. L-type $Ca^{2+}$ channel blockers such as nifedipine, nimodipine, and FS-2 inhibited the motility, but N-type $Ca^{2+}$ channel blocker, w-conotoxin MvIIA, did not. On the other hand, the membrane hyperpolarization and cAMP synthesis were suppressed by $Ca^{2+}$ channel blockers, nifedipine, and trifluoroperazine. Furthermore, these suppressions were relieved by the addition of $K^+$ ionophore, valinomycin. Inhibitors of calmodulin, such as W-7, trifluoperazine, and calrnidazol-C1, inhibited the sperm motility, membrane hyperpolarization, and cAMP synthesis. The results suggest that $Ca^{2+}$ influx through $Ca^{2+}$ channels that are sensitive to specific $Ca^{2+}$ channel blockers and calmodulin participate in the changes of membrane potential, leading to synthesis of cAMP in the cell signaling for the initiation of trout sperm motility.

A semispherical SQUID magnetometer system using high sensitivity double relaxation oscillation SQUIDs for magnetoencephalographic measurements

  • We designed and constructed a multichannel superconducting quantum interference device (SQUID) magnetometer system to measure magnetic fields from the human brain. We used a new type of SQUID, the double relaxation oscillation SQUID (DROS). With high flux-to-voltage transfers of the DROS, about 10 times larger than the dc SQUIDs, simple flux-locked loop circuits could be used for SQUID operation. Also the large modulation voltage of the DROS, typically being 100 $mutextrm{V}$, enabled stable flux-locked loop operation against the thermal offset voltage drift of the preamplifier. The magnetometers were fabricated using the Nb/AlOx/Nb junction technology. The SQUID system consists of 37 signal magnetometers, distributed on a semispherical surface, and 11 reference channels were installed to pickup background noises. External feedback was used to eliminate the magnetic coupling with the adjacent channels. The liquid helium dewar has a capacity of 29 L and boil-off rate of about 4 L/d with the total 48 channel insert. The magnetometer system has an average noise level of 3 fT/√Hz at 100 Hz, inside a shielded loon, and was applied to measure auditory-evoked fields.

Dendritic localization and a cis-acting dendritic targeting element of Kv4.2 mRNA

  • Kv4.2, a pore-forming $\alpha$-subunit of voltage-gated A-type potassium channels, is expressed abundantly in the soma and dendrites of hippocampal neurons, and is responsible for somatodendritic $I_A$ current. Recent studies have suggested that changes in the surface levels of Kv4.2 potassium channels might be relevant to synaptic plasticity. Although the function and expression of Kv4.2 protein have been extensively studied, the dendritic localization of Kv4.2 mRNA is not well described. In this study, Kv4.2 mRNAs were shown to be localized in the dendrites near postsynaptic regions. The dendritic transport of Kv4.2 mRNAs were mediated by microtubule-based movement. The 500 nucleotides of specific regions within the 3'-untranslated region of Kv4.2 mRNA were found to be necessary and sufficient for its dendritic localization. Collectively, these results suggest that the dendritic localization of Kv4.2 mRNAs might regulate the dendritic surface level of Kv4.2 channels and synaptic plasticity.

Design of a Cost-Effective Hybrid-Type PBEx Providing a High Power Budget in an Asymmetric 10G-EPON

  • This paper proposes a cost-effective hybrid-type power budget extender (PBEx) that can provide a high power budget of over 45 dB in an asymmetric 10-Gb/s Ethernet passive optical network (10/1G-EPON). The hybrid-type 10/1G-EPON PBEx comprises a central office terminal (COT) and remote terminal (RT) module supporting four channels and uses a coarse wavelength division multiplexing (CWDM) technology between the COT and RT for a reduction of fiber cost and efficient access network design. The proposed 10/1G-EPON PBEx can provide over a 40-km reach and 128-way split per CWDM wavelength with no modification of a legacy 10/1G-EPON system and can satisfy the error-free service in $10^{10}$ packet transmission.

Development of a 1500N-thrust Swirling-Oxidizer-Flow-Type Hybrid Rocket Engine

  • We have been developing a 1500N-thrust Swirling-Oxidizer-Flow-Type hybrid rocket engine. In order to put the engine into practical use, we conducted long duration burning experiments up to 25s to examine the influence of configuration change of fuel grain on the engine performance and designed an LOX vaporization nozzle to supply GOX for the 1500N-thrust engine. The experiment with a small hybrid rocket engine showed that combustion was stable and the engine performance was approximately constant during combustion. There was no essential problem to with increasing combustion time. The LOX vaporization nozzle designed had 30 rectangular channels with a depth of 0.5mm. During passing through the nozzle, the LOX increased in temperature and vaporized sufficiently.
