식이단백질조성이 흰쥐의 성장과 지방대사에 미치는 영향 (Effects of Dietary Protein on Growth and Lipid Metabolism in Growing Rats)

  • The effect of dietary protein on growth and lipid levels of plasma and liver was studied in weanling male rats fed diets differing protein sources and amino acid balance. Rats were devided into 9 experimental diets which were grouped into 3 categories ; 1) Simple protein category includes gluten-, soy protein isolate-, and casein-containing diet groups, 2) Supplemented category includes casein supplemented with methionine, soy protein isolate supplemented with methionine, and gluten supplemented with lysine and methionine, 3) Mixed protein category includes diet groups containing gluten (2/3), casein (1/3), soy protein isolate (2/3) and casein (1/3), and casein (1/3), soy protein isolate (1/3) and gluten (1/3). The experimental diets composed of 15% protein, 65.8% carbohydrate, 10% fat and 1% cholesterol. The body wt. gain and P.E.R. were greater in rats of supplemented and mixed protein groups than simple protein groups. No statistical differences were found in plasma cholesterol among gluten, soy protein isolate and casein groups. Consumption of diets supplemented with limiting amino acid to gluten or soy protein isolate reduced the plasma cholesterol level by 23.2% and 34.2% respectively. However there was no difference between casein and the supplemented casein groups. The mixed protein groups shows relatively high plasma cholesterol concentration and low liver cholesterol levels. On the other hand gluten group showed low plasma cholesterol and high liver cholesterol levels, which means body cholesterol pool may not have been changed by the dietary protein. Feeding soy protein meal and the supplemented soy protein isolate resulted in lower plasma cholesterol, plasma triglycerides, liver cholesterol and liver triglycerides levels. This hypolipidemic effect is considered to see unique to soy protein isolate. Rats in gluten and the supplemented gluten groups showed lower plasma protein levels and a tendency of fatty liver.

Effects of Synchronization of Carbohydrate and Protein Supply on Ruminal Fermentation, Nitrogen Metabolism and Microbial Protein Synthesis in Holstein Steers

  • Three rumen-cannulated Holstein steers were fed three diets, each with a different synchrony index (SI) (LS: 0.77, MS: 0.81, and HS: 0.83), in order to examine the effect of diet on rumen fermentation, nitrogen balance, and microbial protein synthesis. Synchrony index was calculated based on the carbohydrate and crude protein fractions of each ingredient and their degradation rates. Feeding the steers diets with different SIs did not influence dry matter, crude protein, NDF, or ADF digestibility. The concentrations of total and individual VFA in the rumens of steers that were fed the two higher-SI diets were higher than in those fed the low-SI diet (p<0.05), but there was no significant difference between the two higher-SI diets. One hour after feeding, steers on the LS diet had lower ruminal pHs than did those fed the MS or HS diets (p<0.05), and animals on the LS diet generally showed higher ruminal $NH_3$-N levels than did animals on the other diets, with the 4-h post-feeding difference being significant (p<0.05). Steers receiving the LS diet excreted more nitrogen (N) in their urine than did those on the two higher-SI diets (p<0.05), and the total N excretion of those on the LS diet was also higher (p<0.05). Microbial N levels calculated from the concentration of urinary purine derivatives were generally higher when the SI was higher, with the highest microbial protein synthesis being produced by steers on the HS diet (p<0.05). In conclusion, in the current study, ingestion of a synchronous diet by Holstein steers improved microbial protein synthesis and VFA production and decreased total N output.

Growth performance and nitrogen excretion of broiler chickens fed low protein diets supplemented with crystalline amino acids

  • This study was conducted to determine the effects of amino acid (AA) supplementation in low-protein (LP) diets on growth performance and nitrogen (N) excretion. A total of 175 7-day-old Ross 308 male broilers, with a mean body weight (BW) of 165 g (standard deviation = 11.2 g), were grouped into five blocks by BW and allocated to seven treatments according to a randomized complete block design with five replicate cages at five birds per cage. Dietary treatments comprised a control diet containing 20.0% crude protein (CP) and six LP diets containing either 18.5% or 17.0% CP. These LP diets were supplemented with either no AA supplementation, indispensable AA, or both indispensable and dispensable AA (glutamic acid and glycine). Birds were fed experimental grower diets from day 7 to 21 and then commercial finisher diets until day 28. During the grower period (day 7 to 21), birds fed LP diets supplemented with indispensable AA exhibited greater (p < 0.05) BW, body weight gain (BWG), feed intake (FI), and gain-to-feed ratio (G:F) than birds fed LP diets without crystalline AA and were comparable to birds fed the control diet. During the finisher period (day 21 to 28), birds fed LP diets supplemented with indispensable AA showed greater (p < 0.05) BW than birds fed LP diets without crystalline AA, and their growth performance was comparable to birds fed the control diet. Throughout the overall period, supplementing indispensable AA in LP diets resulted in elevated (p < 0.05) BWG, FI, and G:F more than those of LP diets without crystalline AA and were comparable to those of the control diet. Supplementing indispensable AA in LP diets decreased amount and coefficient of N excretion as much as the control diet. Dispensable AA supplementation in LP diets did not influence growth performance and N excretion. In conclusion, supplementing indispensable AA in LP diets maintains growth performance and N excretion until the dietary CP lowers from 20.0% to 17.0% during the grower period. As long as dietary CP is above 17.0%, dispensable AA may not be deficient in LP diets during the grower period.

어린병아리에서 저단백질사료내 제한아미노산의 규명과 함유황아미노산의 요구량 결정 (Identification of Limiting Amino Acids and Determination of Requirement of Total Sulfur-containing Amino Acids in a Low Protein Diet in Young Chicks.)

  • 저단백질(13% C. P.) 사료는 어린 병아리에서 단백질급원의 Net Protein Utilization가 조사하거나 또는 아미노산의 불균형을 연구할 때 적절한 방법으로 인정되고 있다. 이때의 기준 단백질로서 정제된 대두단백을 많이 이용하고 있으나 이런 사료의 제한 아미노산에 대한 연구가 되어 있지 않다. 본 연구에서는 사료내 유일한 단백질급원으로서 isolated soy protein을 15% 수준으로 함유한 purified-type의 기초사료에서 제한 아미노산을 규명하고 이의 요구량을 조사하였다. 어린병아리의 기초사료에 methionine, lysine, threonine 및 tryptophan 등을 단독으로 또는 두개 이상을 같이 첨가해서 급여한 결과. methionine만이 결핍된 것으로 나타났다. 총함유황아미노산(TSAA)의 요구량을 dose-response curve 방법에 의해 반응선과 plateau선과의 상호교차점에서 구하였는데, 최대의 성장과 사료섭취량을 위한 요구량은 각각 사료단백질의 4.73%와 3.73% 수준이였다. 이들 수치를 TSAA의 섭취량을 기준으로 표시해 보면 최대성장율, 사료섭취량 및 사료효을을 위해 각각 167.7, 136.8 및 159.7mg/bird/day의 양이 필요하였다.

Hypocholesterolemic metabolism of dietary red pericarp glutinous rice rich in phenolic compounds in mice fed a high cholesterol diet

  • BACKGROUND/OBJECTIVES: The purpose of the current study was to investigate the effect of red pericarp glutinous rice rich in polyphenols (Jakwangchalbyeo, red rice) on serum and hepatic levels of cholesterol and hepatic protein expression linked to synthesis and degradation of cholesterol in a hypercholesterolemic mice diet as compared with brown rice. MATERIALS/METHODS: C57BL/6 male mice were randomly divided into four groups (n = 5 each), which were fed different diets for a period of 12 weeks: American Institute of Nutrition (AIN)-93G diet, AIN-93G diet with 2% cholesterol, brown rice with 2% cholesterol, or red rice with 2% cholesterol. RESULT: Consumption of red rice resulted in a significant decrease in serum level of low-density lipoprotein cholesterol and hepatic levels of triglyceride and total-cholesterol. Expression of acyl-coenzyme A cholesterol acyltransferase-2 (ACAT-2), sterol regulatory element binding protein-2 (SREBP-2), and 3-hydroxyl-3-methylglutaryl coenzyme A (HMG-CoA) reductase was decreased, while expression of phosphorylated adenosine monophosphate activated protein kinase (p-AMPK)/AMPK ratio, cholesterol 7-${\alpha}$-hydroxylase (CYP7a1), and sterol 12-${\alpha}$-hydroxylase (CYP8b1) was increased in mice fed red rice. Brown rice had similar effects on cholesterol metabolism, but the effect of red rice was significantly greater than that of brown rice. CONCLUSIONS: The current study suggested that red rice had a hypocholesterolemic effect by lowering hepatic cholesterol synthesis through ACAT-2, HMG-CoA reductase, and SREBP-2, and by enhancing hepatic cholesterol degradation through CYP7a1 and CYP8b1 in mice fed a hypercholesterolemic diet.

식이 단백질 수준의 차이가 흰쥐에서 무균 배양된 원충 감염시에 Caecal Content의 pH와 혈청 단백질에 미치는 영향 (Effect of Dietary Protein Level on Caecal Content pH and Serum Protein in Rats to Axenically Cultured Protozoa)

  • Cell structure is based on proteins. Since the antibody is proteous substance, the continous low protein feeding decreases the resistance of host against pathogenic agents. The present study was designed to investigate the infectivity of protozoa to rats which were fed with variously prescribed diets. Experimental group was divided into 4 groups according to the level of casein in the diet, group I: casein 0%, group II: casein 5%, group III: casein 15%, group IV: casein 30%. Each animal was fed for 5 weeks followed by inoculation of protozoa in cecum and sacrified each 1 week later of the infection. Each diet group, non infected with protozoa was recognized as the control. Result are summerized as follows : 1. All the rats of group I died in 2∼4 weeks and 2 of 12 rats in group II were also died in the period. 2. The growth rate and FER were high in group III and IV compared with group II. Therefore low protein feeding decrease growth and feed efficaly ratio(FER). 3. The pH of caecal contents between the infected group and control showed no difference, but the values of group III and IV were higher than the group II. Low pH of the caecal contents provides a suitable condition for determining their susceptibility to Entameoeba histolytical trophozoite. 4. Amounts of serum total protein in group II, III and IV showed no significant difference with the control and infected group, but amounts in group III and IV were higher than the group II. Therefore, continuous low protein feeding decrease serum total protein. 5. Albumin, ${\alpha}$1, gloulin, ${\alpha}$2 globulin, ${\beta}$ globulin, ${\gamma}$ gloulin of group III and IV were all high to compare than the group II. Albumins of group III and IV of control was higher than infected group, but there was no difference in ${\gamma}$ globulin between the infected and control group.

전화 상담을 통한 비만 여성의 치료 효과 (Effectiveness of a consulting program through telephone in treating obese women)

  • The trial was conducted to assess the impact of consulting through telephone on body weight reduction among 18 obese adult women. The degree of obesity, food intake pattern, and life style were examined through phone calls. Subjects visited our clinic and their body fat percentages were measured using impedance fat meter (model SIF-891), and were recommended and recorded to have a low calory diet with changes in the food habit and behavior. The study results are as follows: 1. There is a positive relationship between food habit, lifestyle, obese degree, and fat percentage. 2. 8-10kg and 5kg reductions in the body weight were observed among 14 subjects and 2 subjects, respectively. However, 2 subjects maintained their body weights. 3. The body reduction curve looked like a stairway (type) with low slope and was related with food intake amount. 4. It was possible to have the adequate protein intake only through the vegetable diet. 5. 15 out of 18 subjects were on the prescribed low calory diets for 3 days. Therefore, there is a time limitation to follow the diet treatment. 6. A body weight reduction altered dietary intake of foods which were rich in sweet taste, and the favorite foods were chocolate and candy.

Effects of different levels of crude protein and protease on nitrogen utilization, nutrient digestibility, and growth performance in growing pigs

  • This study was conducted to evaluate the effects of different levels of crude protein (CP) and protease on nitrogen (N) utilization, nutrient digestibility, and growth performance in growing pigs. A total of six crossbred ([Landrace × Yorkshire] × Duroc) barrows were individually accepted in 1.2 m × 0.7 m × 0.96 m stainless steel metabolism cages. The pigs (average initial body weight of 27.91 ± 1.84 kg) randomly assigned to six diets with six weeks (6 × 6 Latin square design). The experiment was carried out in an environment with a temperature of 23 ± 1.5℃, a relative humidity of 83 ± 2.3% and a wind speed of 0.25 ± 0.03 m/s. The dietary treatments were arranged in a 2 × 3 factorial design with two levels of CP (15.3% or 17.1%) and three levels of protease (0 ppm, 150 ppm, or 300 ppm). The average daily gain and gain to feed ratio (G:F) tended to increase (p = 0.074) with increasing amounts of protease. The low CP level diet reduced (p < 0.050) urinary and fecal N concentrations, the total N excretion in feces, and increased (p < 0.050) N retention. Different protease levels in the diet did not affect (p > 0.05) at N intake, but supplementation of the diets with 300 ppm protease decreased (p < 0.050) the N concentration in urine and feces and tended to increase (p = 0.061) the percentage of N retention retained of the total N intake. The dietary CP level did not affect (p > 0.050) the apparent total tract digestibility (ATTD) of dry matter, digestible energy (DE), and metabolic energy (ME), but diet supplementation with 300 ppm protease showed higher (p < 0.050) ATTD of DE and ME than in the protease-free diet. Therefore, a low protein diet with protease could improve the utilization of nitrogen, thereby reducing the negative effect of N excretion into the environment while maintaining or increasing growth performance compared to a high protein diet.

Effects of Levels of L-Leucine Supplementation with Sub-optimal Protein in the Diet of Grower-finisher Broiler Chickens on Carcass Composition and Sensory Characteristics

  • An experiment involving 180 straight run one-day-Cobb broilers was conducted to evaluate the effect of supplementation of L-leucine with different levels of crude protein (CP) on carcass composition and sensory characteristics of broiler grower-finisher chickens. Six experimental diets comprising two levels of crude protein (CP) i.e., 20 and 20% with three levels of L-leucine i.e. 0, 0.5 and 0.67%, were offered to birds from 21-42 d of age. The birds were randomly divided into 36 experimental pens, 5 chickens in each pen, and there were 6 replicates under each diet. L-leucine supplementation did not affect the bone and lean, whereas fat was decreased (p<0.05) when L-leucine was added at 0.5%. Similarly, there were no significant differences (p>0.05) in the lean, fat and bone among chickens fed two levels of CP. No significant differences between dietary treatments were observed on any sensory characteristics affected by dietary L-leucine and CP. From this study, it is obvious that supplementation of up to 0.5% L-leucine reduced fat. However, other characteristics were not affected by supplementation of L-leucine. Similarly, reduction of body composition and sensory characteristics were not apparent on a diet low in CP.

사료내 동물성 단백질원들이 조피볼락의 성장과 체조성에 미치는 영향 (Effects of Dietary Animal Protein Sources on Growth and Body Composition in Korean Rockfish, Sebastes schlogeli)

  • 본 연구는 조피볼락 사료에 있어서 동물성 단백질원들의 영양적 평가(체조성과 성장)를 결정하기 위하여 수행되었다. 6가지 실험사료 중 북양어분구(WFM), 넙치근육구(FMM), 오징어간분구(SLP)는 한가지, 혈분구(BM)는 혈분과 북양어분을, 카제인구(CG)는 카제인, 젤라틴 및 북양어분을, 그리고 난단백구(EWA)는 난단백, 젤라틴 및 북양어분을 단백질원으로 이용하여 사료내 조단백질 함량을 50%로, 가용에너지는 15.9 KJ/g (physiological fuel value ; 단백질 : 지방 : 탄수화물 = 16.7 KJ/g : 37.7 KJ/g : 16.7 KJ/g)으로 조정하였다. 성장률, 사료효율 및 단백질효율은 북양어분구보다 넙치육분, 오징어간분구보다 혈분구 그리고 난백구보다 카제인구가 유의적으로 우수한 결과를 보였으며(P<0.05), 혈분구는 북양어분을 혼합하여 혈분의 기호성을 높인 결과로 사료된다. 6가지 실험구의 전어체 분석, HSI, Hb, Hematocrit은 다양하게 유의적인 차이를 나타냈다. 따라서, 상기 결과는 조피볼락에 있어서 동결 건조된 넙치근육분이 가장 좋았고, 난단백은 가장 저조한 성장을 보여 주었다.
