Protective Effect of Red ginseng Saponin on Decrease of Femur Weight in Female Guinea Pigs Acutely Exposed to 2,3,7,8-tetrachlorodibenzo-ρ-dioxin (TCDD) (TCDD (2,3,7,8-tetrachlorodibenzodioxin)로 급성독성을 유도한 자성 기니픽의 대퇴골 무게감소에 대한 홍삼사포닌의 방어효과)

  • To investigate the protective effect of saponin from red ginseng extract, 2,3,7,8-tetrachlorodibenzo--dioxin (TCDD) was exposed to female guinea pigs and then femur weights was measured. Forty eight female guinea pigs ($820{\pm}25\;g$) were divided into 6 groups. Normal control group (NC) received vehicle and saline; only TCDD-treated group (TT) received TCDD ($5.0\;{\mu}g/kg$, single dose) intraperitoneally; pretreated group of saponin 10 (PE 10) received 10 mg/kg of saponin i.p. for 4 weeks from 1 week before TCDD-exposure; pretreated group of saponin 20 (PE 20) also received 20 mg/kg of saponin i.p. for 4 weeks from 1 week before TCDD-exposure. While, post-treated group of saponin 10 (CE 10) received 10 mg/kg of saponin i.p for 3 weeks after TCDD-exposure. Post-treated group of saponin 20 (CE 20) received 20 mg/kg saponin i.p for 3 weeks after TCDD-exposure. Body weight of TT group was significantly decreased after TCDD-exposure. However, body weight in all saponin-treated groups increased throughout the experimental period, although the increasing rate was slower than that of NC group. Body weights of PE 10 and 20 groups showed more higher increase than those of CE groups during the experimental period. Decrease of femur weights in female guinea pigs by TCDD intoxification was significantly recovered by the saponin treatment. Decrease of $Ca^{2+}$ level of femurs in female guinea pigs exposed TCDD also recovered by the treatment of saponin from red ginseng extract. Especially, PE20 group showed the highest increase of the $Ca^{2+}$ level in femur among the saponin treated groups. These results suggest that ginseng saponin might be a useful protective agent against femur damage caused to decrease of $Ca^{2+}$ by TCDD.

Effects of Environmental Factors on Growth and Nitrogen Fixation Activity of Autumn Olive (Elaeognus umbellata) Seedlings (보리수나무 유식물의 생장과 질소고정 활성에 대한 환경요인의 영향)

  • Effects of environmental factors of light, temperature, nitrogen sources and water stress were analyzed quantitatively on the nodule formation and nitrogen fixation activity of autumn olive plant (Elaeagnu$ umbellala Thunb.) during the seedling growth. Seedlings showed the maximum nitrogenase activity of $72.5\;\mu\textrm{M}\;C_2H_4{\cdot}g\;fr\;wt\;nodule^{-1}{\cdot}h^{-1}$ in the early nodulation stage. The relative growth rate and T/R ratio changed from $1.60%{\cdot}d^{-1}$ and 1.12 in the earlier stage to $3.75%{\cdot}d^{-1}$ and 2.31 in the later stage, respectively. light conditions of 20-25, 1015 and 4-6% resulted in decreases of 41, 54 and 71% of the nitrogenase activity, respectively. Nodules incubated in 15, 20, 25 and $30^{\circ}C$ showed the activities of 5.4, 24.7, 51.6 and $58.5\;\mu\textrm{M}\;C_2H_4{\cdot}g\;fr\;wt\;nodule^{-1}{\cdot}h^{-1}$ respectively. Pretreatment with low temperature ($15^{\circ}C$) followed incubation at $30^{\circ}C$ attained higher nitrogenase activity ($66.5\;\mu\textrm{M}\;C_2H_4{\cdot}g\;fr\;wt\;nodule^{-1}{\cdot}h^{-1}$) than that with higher temperature ($35^{\circ}C$). The oxygen pressure above 16 kPa is necessary for saturation of the nodule activity, but the activity was inhibited severely by physical impact such as the exision or isolation of nodules from the root. The relative activities of early nodules grown in pH 5.5, 6.5 and 8.0 were 89, 100 and 40% and those grown in 1 and 3 mM of $NO_3\;and\;NH_4$ were 6, 1 and 68, 50%, respectively. Watering levels of 20, 50 and 100 mL during the seedling growth resulted in 35, 120 and 8 mg of nodule formation and 33.6, 58.4 and $8.4\;\mu\textrm{M}\;C_2H_4{\cdot}g\;fr\;wt\;nodule^{-1}{\cdot}h^{-1}$ of the nitrogenase activity, respectively. Water stress with 86% decrease of soil water content caused temporary wilting point of leaf and a complete disappearance of nitrogenase activity of nodules, though the water content and transpiration rate in plant were reduced to 90 and 53%, respectively.tively.

Comparison of Chemical Characteristics of Korean Mountain Ginseng Different Parts According to Extract Conditions (장뇌삼의 부위별 추출조건에 따른 이화학적특성 비교)

  • This study was conducted to investigate extracting solution effect on the chemical compositions in different parts of Korean mountain Ginseng. Water, 80% EtOH and 80% MeOH are used as extraction solutions, and extracting conditions were 2 hr at $85^{\circ}C$ in water bath. The Brix(%) of the extract were ranged from $0.42{\sim}22.58%$, 80% EtOH extract for leaf is the highest level as 22.58%. The pH ranges of the extracts were $4.43{\sim}7.41$ and brown color of the extract was the highest with 1.803 in 80% EtOH extract for leaf, respectively. In case of hunter's color value of the extract, L value is the highest with 24.35 in 80% EtOH extract of seed, a and b value were the highest with 0.41 in 100% water extract of leaf and 3.69 in 80% MeOH extract of stem. Sucrose is the major free sugar of the extinct it highest content with 3673 mg% in 80% MeOH extract of mot and fructose is the highest with 1897 mg% in 80% MeOH extract of leaf, Major organic acids are identified as malic, tartaric and citric acid, and total organic acid content is the highest with 5,254 mg% in 80% MeOH extract of leaf and 1,527 mg% in 80% EtOH extract of leaf, The extracted major minerals ate P and K, P content highest with 15,563 ppm in 100% water extract of stem, K is 4,952 ppm in 80% MeOH extract of leaf, and Ca is the highest with 3,052 ppm in 1011% water extract of leaf. These results suggest that extracting solvent (80% MeOH) is concerned with the extract preparation of Korea Mountain Ginseng.

Effect of calcium chloride on the texture of pickled radish wrap (염화칼슘 첨가에 따른 쌈무의 조직감 개선 효과)

  • This study was conducted to evaluate the quality characteristics of pickled radish wrap supplemented with calcium chloride ($CaCl_2$) at different concentrations. With $CaCl_2$ supplementation, initial pH and acidity were 3.8~4.0 and 2.7~3.0% (w/v), respectively. During the 28 day-preservation at $4^{\circ}C$ or $50^{\circ}C$, the initial pH and acidity were maintained. However, when the pickled radish wrap was preserved at $25^{\circ}C$, it had lower pH (by 0.5) and higher acidity (by 1.9~2.2%) when compared with the initial values. The difference in color (${\Delta}E$-value) of the pickled radish wrap preserved at $50^{\circ}C$ was significant (${\Delta}E$-value 23~27), though the hardness of the sample prepared at $4^{\circ}C$ or $25^{\circ}C$ showed no distinguishable differences at various concentrations of $CaCl_2$ supplementation (0.1, 0.2, 03, 0.4, and 0.5%). However, once preserved at $50^{\circ}C$, it resulted in the greater hardness with the higher concentrations of $CaCl_2$ supplementation. In terms of softness, the overall trend in changes was similar to the hardness. A sensory evaluation using the samples preserved at $4^{\circ}C$ for 28 days demonstrated that the preparation with 0.4% $CaCl_2$ ranked highest in taste, hardness and overall acceptability. It wass suggested that supplementation with 0.4% $CaCl_2$ would be most appropriate to improve the textural properties of pickled radish wrap while maintaining its organoleptic quality.

Antioxidant activities of blueberry hot water extracts with different extraction condition (추출조건에 따른 블루베리 열수추출물의 항산화 활성 비교)

  • Five extraction conditions (AE, autoclave extraction; OE, oven extraction; HWSE, hot water and sonication extraction; HWASE, hot water acidified with 0.5% (v/v) acetic acid and sonication extraction; and BE, boiling extraction) were examined to compare the effects of different hot water extraction methods on the antioxidant properties of blueberries. The extraction yields of the AE, OE, HWSE, HWASE, and BE were 7.94%, 8.35%, 8.55% 9.15%, and 8.50%, respectively. The polyphenol and flavonoid contents of AE were 3.47 mg GAE/g and 1.59 mg RE/g, respectively, which were highest centents among others. Those of OE were ranked second to the highest. The total anthocyanin content of HWSE (5.29 mg/g) was significantly higher than that of others whereas that of AE showed the lowest content (0.96 mg/g). The order of ABTS radical and alkyl radical scavenging activities was as follows: AE > BE > OE > HWSE > HWASE. The antioxidant properties were considerably correspondent with the total polyphenol and flavonoid content. DPPH radical scavenging activity was quite high in HWSE, AE, and BE extraction, however, there were no significant differences among the five extraction methods in the aspect of $Fe^{2+}$ ion chelating activities. Moreover, AE showed the highest SOD activity, and protected the dermal fibroblast the best against $H_2O_2$-induced cytotoxicity. In conclusion, it was suggested that the autoclave extraction (AE) would be the most effective method for preparing blueberry hot water extracts with relatively high antioxidant activities.

Effect of seasonal cabbage cultivar (Brassica rapa L. ssp. Pekinesis) on the quality characteristics of salted-Kimchi cabbages during storage period (계절별 배추 품종에 따른 절임배추의 저장중 품질 특성)

  • This study investigates the physicochemical, microbiological and sensory characteristics of seasonal salted-Kimchi cabbages to provide basic data on its uniform quality. Generally, seasonal salted-Kimchi samples had different pH values at initial storage periods, but there was no difference in pH between the seasonal samples when stored for longer periods. The samples from the fall and winter seasons were relatively low in acid and high in solid soluble content compared to samples from other seasons. Salted-Kimchi cabbages in the summer showed the highest microbiological number compared to samples from other seasons. In the sensory evaluation, there were differences in the appearance, aroma, and taste, depending on seasonal samples at different storage periods. The correlation coefficient between the quality characteristics in the seasonal samples showed a positive or negative correlation between the quality characteristics at 1% significant level. In the principal component analysis, F1 and F2 were shown the 51.81% and 14.23% of the total variance (66.21%), respectively. In the PCA pattern of seasonal salted-Kimchi cabbages during storage periods, winter samples were distributed on the top of F2, spring samples were in the middle of F2, while the rest of the samples were distributed on the bottom of F2. According to increasing storage periods, initial storage samples were distributed at the left of F1, while other samples were located at the right of F2.

Physicochemical properties of spray-dried rice flour with Lactobacillus plantarum CGKW3 (분무건조공정을 이용한 유산균포집 미분의 제조 및 물리화학적 특성)

  • The physicochemical properties of spray-dried rice flour with Lactobacillus plantarum CGKW3 were investigated. Amylose and damaged starch contents of spray-dried rice flour (S10, S20, S30, and S50) with L. plantarum CGKW3 were 14.18~17.75% and 24.65~34.08%, respectively. The particle size of spray-dried rice flour was $82.28{\sim}131.17{\mu}m$. The rice flour with L. plantarum CGKW3 showed a good powder flowability. The water absorption and water solubility of spray-dried rice flour were 1.96~2.13 and 9.91~21.95%, respectively. Thermal properties measured by differential scanning calorimeter revealed that the enthalpy (${\Delta}H$) for starch gelatinization were highest in the rice flour (S50) with L. plantarum CGKW3. When compared, the viable cell number of spray-dried rice flour were found to be in the following order: S10 (5.78 log CFU/g) < S20 (6.38 log CFU/g) < S30 (6.69 log CFU/g) < S50 (7.11 log CFU/g). The survaival rate of L. plantarum CGKW3 was 60.02-73.85%, which reflected the improvement in the quality of rice flour with an increase in treatment concentration. Based on our results, spray-dried rice flour with L. plantarum CGKW3 could be used in various types of rice foods.

Physicochemical Characteristics of Yanggaeng with Pear Juice and Dried Pear Powder Added (배즙과 배 건조분말을 첨가한 양갱의 물리화학적 특성)

  • This study was conducted to investigate the physicochemical characteristics of yanggaeng prepared with pear juice and dried pear powder. The proximate composition and total polyphenol content, antioxidant activities, Hunter's color values, texture, sensory properties of pear yanggaeng variants [pear juice (PJ), pear juice and hot-air-dried pear powder (PJH), pear juice and freeze-dried pear powder (PJF) yanggaeng] were examined. The moisture content was highest (27.9%) in the control (C) yanggaeng, but the latter's energy and carbohydrate content were lower than in the pear yanggaeng variants. The differences between the pear yanggaeng variants were not significant.The total polyphenol content was highest in the 20.7 mg/100g PJF yanggaeng. The antioxidant activities of PJF yanggaeng were higher than those of the other pear yanggaeng varients. The Hunter color value results showed that the lightness (L) values were highest in C yanggaeng whereas the redness(a) and yellowness(b) values were highest in PJH yanggaeng. The springiness in texture was lowest in C yanggaeng and not significant in the others. The hardness and chewiness were highest in PJH and PJF yanggaeng, but adhesiveness was lowest therein. The results of the sensory test showed that PJF yanggaeng was the highest in flavor, color, taste, hardness, chewiness, and overal quality. Based on these results, it can be concluded that pear juice and freeze-dried pear powder yanggaeng has excellent physicochemical and antioxidant activities.

Quality Changes of Minimally Processed Sliced Deodeok (Codonopsis lanceolata) during Storage by Packaging Method (포장방법 차이에 따른 신선편이 슬라이스 더덕의 저장 중 품질 변화)

  • In this study, the changes that occurred in the quality of minimally processed sliced Deodeok (Codonopsis lanceolata) in relation to the packing method during storage at $7^{\circ}C$ were investigated. The storage tests were conducted for seven days using PE sealing, but PP sealing and vacuum packaging preserved the Deodeok for 14 days. On the seventh day, the vinyl-packaged Deodeok showed a remarkable fall in quality with 4.5 °Brix, but the PP-sealed and vacuum-packaged Deodeok showed slight falls with 6.4 and $6.8^{\circ}Brix$, respectively. The PE- and PP-sealed Deodeok did not show significant differences in texture and moisture content for two days, and the moisture content was highest in the vacuum-packaged Deodeok during storage. In relation to the total viable cell and the coliform count, the vacuum-packaged Deodeok showed the lowest rate of increment during storage, followed by the others. Thus, the bubble-washed and vacuum-packaged minimally processed sliced Deodeok was found to have the best quality.

Evaluation of Muscle Quality of Olive Flounder (Paralichthys olivaceus) Fed Extruded Pellets Containing Different Protein and Lipid Levels, and Raw Fish-based Moist Pellet (단백질 및 지질함량이 다른 배합사료와 생사료로 사육한 넙치의 육질평가)

  • This study was conducted to investigate the effect of extruded pellets (EP) containing different levels of protein (51%-55%) and lipid (9%-15%) on flesh quality of flounder (Paralichthys olivaceus) comparing with raw fish-based moist pellet (MP). Two replicate groups of 40 fish per each tank (initial mean weight 106 g) were fed one of three experimental EP (EP1, EP2 and EP3) containing different protein and lipid levels, a commercial EP (EP4) and MP for 16 weeks. Moisture content of fish fed MP was significantly higher than that of fish fed all EP. Significantly higher contents of 16:1n-7, 20:4n-6, 20:5n-3 and lower 18:2n-6 were observed in fish fed EP4 and MP compared with fish fed EP1, EP2, EP3. The 22:6n-3 content was not significantly different among all groups. Taurine content of fish fed MP was significantly higher than that of fish fed all EP. Asparagine content of fish fed EP1 was significantly higher than that of fish fed EP2, EP, EP4 and MP. Significant difference were observed in lysine and serine contents of fish fed experimental diets. In nucleotides and their related compounds, ATP and AMP content was not affected by diets. Significant difference were observed in IMP contents of fish fed experimental diets. Textural properties, no significant difference was observed among the fish groups fed different diets.
