Roles of Neutral Sphingomyelinase 1 on CD95-Mediated Apoptosis in Human Jurkat T Lymphocytes

  • CD95 receptor is a member of tumor necrosis factor receptor family that mediates apoptosis in many cell types when bound by CD95 ligand or cross-linked by agonistic anti-CD95 antibodies. To determine the role of neutral sphingomyelinase (nSMase) on CD95-mediatd apoptosis, human Jurkat T lymphocytes were exposed to recombinant human CD95 ligand. Treatment with CD95 ligand induced cell death in a concentration and time-dependent manner. CD95-induced cell death was suppressed by inhibitors of SMase such as AY9944 or desipramine. Transfection with human nSMase1 siRNA plasmid into CD95 ligand-treated cells significantly prevented CD95-mediated cell death. CD95-mediated elevation of intracellular ceramide level detected by FACS analysis with anti-ceramide antibody was also decreased by nSMase1 siRNA. Knock-down of nSMase1 expression also blocked cytochrome c release into cytosol and caspase-3 cleavage in CD95-treated cells. Taken together, these results suggest that nSMase1 may play an important role in CD95-mediated apoptotic cell death in Jurkat T cells.

Risk Factors for Suicidal Ideation across the Life Cycle among Korean Adults: Korean Psycho-social Anxiety Survey (한국 성인의 생애주기별 자살생각의 위험요인: 한국 사회·심리적 불안 조사)

  • Purpose: To identify the risk factors for suicidal ideation across the life cycle among Korean adults using data from 2015 Korean Psycho-social Anxiety Survey. Methods: The data were statistically analyzed using hierarchical logistic regression analysis. Results: The factors associated with suicidal ideation among the young adult group were education level (odds ratio [OR] 1.62, 95% confidence interval [CI] 1.05~2.49), self-esteem (OR 1.19, 95% CI 1.12~1.27), stress (OR 3.26, 95% CI 2.07~5.15), anger control problems (OR 3.58, 95% CI 2.34~5.50), and depression (OR 2.59, 95% CI 1.66~4.04) whereas among the middle-aged adults the factors were education level (OR 1.58, 95% CI 1.12~2.23), existence of a spouse (OR 2.55, 95% CI 1.72~3.78), self-esteem (OR 1.13, 95% CI 1.08~1.18), stress (OR 2.46, 95% CI 1.81~3.33), anger control problems (OR 1.70, 95% CI 1.22~2.36), and depression (OR 2.64, 95% CI 1.91~3.64). Among the older adult group the findings were the existence of a spouse (OR 2.16, 95% CI 1.37~3.39), self-esteem (OR 1.22, 95% CI 1.13~1.32), stress (OR 2.21, 95% CI 1.38~3.54), anxiety (OR 2.42, 95% CI 1.46~4.02), and depression (OR 2.48, 95% CI 1.30~4.72). Conclusion: The findings suggest that there may be a need for different suicide intervention programs to decrease suicidal ideation across the life cycle.

Procedural outcomes of laparoscopic caudate lobe resection: A systematic review and meta-analysis

  • A systematic review was conducted in compliance with PRISMA statement standards to identify all studies reporting outcomes of laparoscopic resection of benign or malignant lesions located in caudate lobe of liver. Pooled outcome data were calculated using random-effects models. A total of 196 patients from 12 studies were included. Mean operative time, volume of intraoperative blood loss, and length of hospital stay were 225 minutes (95% confidence interval [CI], 181-269 minutes), 134 mL (95% CI, 85-184 mL), and 7 days (95% CI, 5-9 days), respectively. The pooled risk of need for intraoperative transfusion was 2% (95% CI, 0%-5%). It was 3% (95% CI, 1%-6%) for conversion to open surgery, 6% (95% CI, 0%-19%) for need for intra-abdominal drain, 1% (95% CI, 0%-3%) for postoperative mortality, 2% (95% CI, 0%-4%) for biliary leakage, 2% (95% CI, 0%-4%) for intra-abdominal abscess, 1% (95% CI, 0%-4%) for biliary stenosis, 1% (95% CI, 0%-3%) for postoperative bleeding, 1% (95% CI, 0%-4%) for pancreatic fistula, 2% (95% CI, 1%-5%) for pulmonary complications, 1% (95% CI, 0%-4%) for paralytic ileus, and 1% (95% CI, 0%-4%) for need for reoperation. Although the available evidence is limited, the findings of the current study might be utilized for hypothesis synthesis in future studies. They can be used to inform surgeons and patients about estimated risks of perioperative complications until a higher level of evidence is available.

Methylenetetrahydrofolate Reductase Polymorphisms and Susceptibility to Esophageal Cancer in Chinese Populations: a Meta-analysis

  • Although many epidemiologic studies investigated the methylenetetrahydrofolate reductase (MTHFR) polymorphisms and their associations with esophageal cancer, definite conclusions could not be drawn. To clarify the effects of MTHFR polymorphisms on the risk of esophageal cancer, a meta-analysis was here performed in Chinese populations. A total of 16 studies including 3,040 cases and 4,127 controls were involved in this metaanalysis. Overall, significant associations were found between the MTHFR C677T polymorphism and esophageal cancer risk when all studies in Chinese populations were pooled into the meta-analysis (T vs. C, OR = 1.19, 95% CI = 1.06-1.34; TT vs. CC, OR = 1.35, 95% CI = 1.07-1.70; TT+ CT vs. CC, OR = 1.29, 95% CI = 1.08-1.54; TT vs. CC + CT, OR = 1.19, 95% CI = 1.03-1.37). In subgroup analyses stratified by ethnicity and source of controls, the same results were found in Kazakh (TT vs. CC, OR = 1.38, 95% CI = 1.02-1.87; TT + CT vs. CC, OR = 1.50, 95% CI = 1.03-2.18), in not stated populations (T vs. C, OR = 1.24, 95% CI = 1.08-1.42; TT vs. CC, OR = 1.47, 95% CI = 1.10-1.96; TT + CT vs. CC, OR = 1.30, 95% CI = 1.05-1.60; TT vs. CC + CT, OR = 1.32, 95% CI = 1.12-1.56), and in hospital-based studies (T vs. C, OR = 1.34, 95% CI = 1.19-1.51; TT vs. CC, OR = 1.81, 95% CI = 1.37-2.39; TT + CT vs. CC, OR = 1.51, 95% CI = 1.26-1.83; and TT vs. CC + CT, OR = 1.39, 95% CI = 1.13-1.70). In conclusion, this meta-analysis provides evidence that the MTHFR C677T polymorphism contributes to esophageal cancer development in Chinese populations.

Blood Pressure and the Risk of Death From Non-cardiovascular Diseases: A Population-based Cohort Study of Korean Adults

  • Objectives: The objective of this study was to assess the relationship between systolic and diastolic blood pressure (SBP, DBP) and the risk of death from specific causes other than cardiovascular diseases. Methods: We calculated the risk of specific death by SBP and DBP categories for 506 508 health examinees in 2002-2003 using hazard ratios (HRs) and 95% confidence intervals (CIs) in a Cox proportional hazards model. Results: Compared to normal levels (SBP <120 or DBP <90 mmHg), stage I systolic and diastolic hypertension (SBP 140-159, DBP 85-89 mmHg, respectively) were associated with an increased risk of death from diabetes mellitus, alcoholic liver disease, and renal failure (HR, 1.83; 95% CI, 1.51 to 2.22; HR, 1.24; 95% CI, 1.06 to 1.46; HR, 2.30; 95% CI, 1.64 to 3.21; HR, 1.67; 95% CI, 1.27 to 2.20; HR, 1.99; 95% CI, 1.41 to 2.81; HR, 1.31; 95% CI, 0.99 to 1.73, respectively), but a decreased risk of death from intestinal pneumonia (HR, 0.64; 95% CI, 0.42 to 0.98; HR, 0.59; 95% CI, 0.39 to 0.91). Only stage II systolic hypertension (SBP ${\geq}160mmHg$) was associated with an increased risk of death from pneumonia, liver cirrhosis, and intestinal ischemia (HR, 1.54; 95% CI, 1.19 to 1.98; HR, 1.46; 95% CI, 1.00 to 2.15; HR, 3.77; 95% CI, 1.24 to 11.40, respectively), and stage I and II diastolic hypertension (SBP 140-159 and ${\geq}160mmHg$) were associated with an increased risk of death from intestinal ischemia (HR, 3.07; 95% CI, 1.27 to 7.38; HR, 4.39; 95% CI, 1.62 to 11.88, respectively). Conclusions: An increase in blood pressure levels may alter the risk of death from certain causes other than cardiovascular diseases, a well-known outcome of hypertension, although the mechanism of these associations is not well documented.

Subjective health status and relative factors of old-old elderly of more than 75-year-old -Based on 2011 Korea National Health and Nutrition Examination Survey- (75세 이상 후기노인의 주관적 건강상태와 관련 요인 - 2011년 국민건강영양조사 자료를 이용하여 -)

  • This study examined the health status and relative factors of 468 people of the latter stage of more than 75-year-old in 2011 the Korean National Health and Nutritional Examination Survey. The subjective health status were influenced significantly by sex(OR=0.456, 95%CI=0.257-0.805), occupation(OR=1.437, 95%CI=0.963-2.149), spouse(OR=0.673, 95%CI=0.443 -1.022), degree of stress(OR=0.476, 95% CI=0.309-0.730), depression(OR=0.410, 95% CI=0.238-0.704), subjective oral status(OR=1.874, 95% CI=1.332-2.643), smoking(OR=0.738, 95% CI= 0.523-1.039), drinking(OR=1.251, 95% CI=1.017-1.540), and waking practice(OR=1.698, 95% CI=1.188 -2.431). The results suggest that health education of active participations and publicity policies should be established and help improve the subjective health status in the latter stage of a 75-year-old.

Factors Associated with Falls in the Elderly : Based on 2014 the Korean Elderly Survey (노인의 낙상에 영향을 미치는 요인 : 2014년 노인실태조사를 중심으로)

  • The purpose of this study was to identify factors associated with falls among older adults. This cross-sectional study used the data of 10,272 older adults who participating in the Korean Elderly survey, including demographic and health related characteristics. The data were analyzed with complex samples in SPSS ver. 23.0. The results of multiple logistic regression analysis showed that age (OR=1.262, 95%CI 1.111-1.433), education (OR=0.846, 95%CI 0.764-0.937), subjective health status (OR=1.172, 95% CI 1.041-1.320; OR=1.422, 95%CI 1.289-1.570), number of chronic illness (OR=1.438, 95%CI 1.050-1.968), number of medications (OR=0.798, 95%CI 0.659-0.967), muscle strength of low extremities (OR=1.260, 95%CI 1.101-1.441), and fear of falling (OR=3.576, 95%CI 3.166-4.039; OR=14.236, 95%CI 12.637-16.038)were significantly associated with fall accidents in the elderly. Thus, it is necessary to develop fall prevention program to reduce fear of falling and improve health status in the elderly.

Association of Coronary Artery Calcium Scores with Cadiovascular Disease Risk Factors in an Asymptomatic Adults (무증상 성인에서 심혈관질환 위험요소와 관상동맥 석회 수치와의 관계)

  • Coronary artery calcium scores(CACS) has been used as surrogate marker for coronary atherosclerosis. We evaluated 1042 patients who visited the Department of Health Promotion Center in Chonnam National University Hospital and had a test of the CACS from January 2006, to December 2008. This study was performed to evaluate the relation of the CACS with Cadiovascular disease(CVD) risk factors and FRS. CACS and FRS was a significant difference between the group whose calcium score was 0 and the group whose calcium scores were 1 in case of men 2.38(95% CI, 1.83-3.11), women 2.12(95% CI, 1.03-4.35). The age-and sex-adjusted odds ratios for predictor of CVD risk factors to women with age was 1.10(95% CI, 1.06-1.15), HDL-cholesterol was 2.38(95% CI, 1.04-5.44), Fasting plasma glucose was 2.89(95% CI, 1.16-7.21), to men with age was 1.11(95% CI, 1.08-1.14), LDL-cholesterol was 2.12(95% CI, 1.28-3.50), gamma-GTP was 1.73(95% CI, 1.17-2.55), Diabetes mellitus medication was 3.92(95% CI, 1.73-8.89). The CACS seems to be a siginificant factor to evaluate the CVD risk factors.

Risk Factors for Lung Cancer in the Pakistani Population

  • Background: Lung cancer is one of the most prevalent malignancies in the world and both incidence and mortality rates are continuing to rise in Pakistan. However, epidemiological studies to identify common lung cancer determinants in the Pakistani population have been limited. Materials and Methods: In this retrospective case-control study, 400 cases and 800 controls were enrolled from different hospitals of all provinces of Pakistan. Information about socio-demographic, occupational, lifestyle and dietary variables was extracted by questionnaire from all subjects. Odd ratios (ORs) and 95% confidence intervals (CIs) were calculated. and dose-response associations were also assessed for suitable factors. Results: Strong associations were observed for smoking (OR=9.4, 95%CI=6.9-12.8), pesticide exposure (OR=5.1, 95%CI=3.1-8.3), exposure to diesel exhaust (OR=3.1, 95%CI=2.1-4.5), red meat consumption (OR=2.9, 95%CI=1.8-4.7) and chicken consumption (OR=2.8, 95%CI=1.7-49). Other associated factors observed were welding fumes (OR=2.5, 95%CI=1.0-6.5), sedentary living (OR=2.0, 95%CI=1.6-2.6), family history (OR=2.0, 95%CI=0.8-4.9), wood dust (OR=1.9, 95%CI=1.2-3.1), tea consumption (OR=1.8, 95%CI=1.2-2.6), coffee consumption (OR=1.8, 95%CI=1.1-2.8), alcoholism (OR=1.7, 95%CI=1.1-2.5) and asbestos exposure(OR=1.5, 95%CI=0.5-4.4). Consumption of vegetables (OR=0.3, 95%CI=0.2-0.4), juices (OR=0.3, 95%CI=0.3-0.4), fruits (OR=0.7, 95%CI=0.5-0.9) and milk (OR=0.6, 95%CI=0.5-0.8) showed reduction in risk of lung cancer. Strongest dose-response relationships were observed for smoking ($X^2=333.8$, $p{\leq}0.0000001$), pesticide exposure ($X^2=50.9$, $p{\leq}0.0000001$) and exposure to diesel exhaust ($X^2=51.8$, $p{\leq}0.0000001$). Conclusions: Smoking, pesticide exposure, diesel exhaust and meat consumption are main lung cancer determinants in Pakistan. Consuming vegetables, fruits, milk and juices can reduce the risk of lung cancer risk, as in other countries.
