Induction of an Experimental PKU-Like Condition in Infant Rats During the First Two Weeks After Birth (신생(新生)쥐의 생후(生後) 2주간(週間)에 있어서 Phenylketonuria 적(的) 조건(條件)의 실험적(實驗的) 유도(誘導))

  • The objective of this study is to induce the primary characteristics of phenylketonuria in infant rats during the first 2 weeks after birth. The critical biochemical parameter in the development of phenylketonuria is the elevation of plasma phenylalanine while tyrosine is maintained at a relatively low level. A PKU-like condition was induced in infant rats during the first 2 weeks after birth using a modification of our previously published procedure for the development of a temporary (1 to 3 days) PKU-like condition. Phenylalanine was administered by stomach intubation every 6 hours (starting at 6:00 a.m.) at a dose level of 400mg per kg body weight (after birth-day 2 thru 5) and 500mg per kg body weight (day 6 thru 14). Amethopterin was given at 0.00625 or 0.0125mg per kg body weight (day 3 thru 14) and p-chlorophenylalanine at 50 mg per kg body weight (day 5 thru 14) at 9:00 a.m. and 9:00 p.m. At the times measured (6,10 and 14 days) plasma phenylalanine/tyrosine (P/T) ratios were elevated from a normal value of the or less to values ranging from 6 to 15. During the second week after birth a staggering gait, abnormal stance and decreased social behavior were also observed. None of these PKU-like characteristics were apparent in the three control groups receiving (a) no phenylalanine or inhibitors, (b) phenylalanine alone, or (c) inhibitors alone. The establishment of these primary biochemical characteristics of phenylketonuria by stomach intubation of phenylalanine and a combination of low dose levels of enzyme inhibitors to infant rats provides an experimental system which should he valuable for extensive biochemical, histological and behavioral studies in phenylketonuria.

  • The methanol extract of perilla leaves contained compounds to reduce the mutagenicity of aflatoxin B$_1$(AFB$_1$) in Salmonella typhimurium TA98 and 100. They were separated by solvent extractions and identified by GC and GC-MS as 2-ethoxy acetate, 1, 2, 3, 4-tetramethyl-cis-cyclobutene, two isomers of methyl 11, 14, 17-eicosatrienoate, 12-acetyl-9-octadecanoic acid, and phytol. The antimutagenicities of phytol and methyl 11, 14, 17-eicosatrienoate were dependent on the mutagens tested. Phytol reduced the mutagenicity of Trp-p-2 but not of AFB$_1$and methyl 11, 14, 17-eicosatrienoate reduced the activities both of the mutagens.

Effect of Pile Temperature Control on Changes of Physicochemical Parameters of Composted Poultry Waste (계분의 콤포스터 처리시 내부온도 조절이 생산물의 물리·화학적 성상에 미치는 영향)

  • When broiler litter was composted under the control of peak temperature of piles(uncontrolled, controled below $70^{\circ}C$ and below $60^{\circ}C$), changes of physical and chemical parameters were determined throughout the processing period. Broiler litter was composted in each of three $1.0{\times}1.0{\times}1.2m$ dimensional facilities for 8 weeks. After 5 to 6 weeks of composting, broiler litter was converted into the final compost with no ammonia odour, rice hull size of particle, and faint brown color. Central temperature of piles reached to the peak(about $69{\sim}70^{\circ}C$) within 3 to 4 days after composting and gradually decreased thereafter. The final product contained 26.5% of moisture, 9.0~9.1 of pH, and 14.0~14.3 of C/N ratio. The increase of C/N ratio with processing resulted from the considerable loss of N. The total wet weight of the final composts was an average of 38.3% of the initial weight, the dry weight of those 64.1%, and the organic matter weight of those 34.8%. Treatments of central temperature of composts did not affect changes of moisture, pH, C/N ratio, total wet weight, total dry weight, and total organic matter weight. In general, composted broiler litter was converted into the final product with little change in physical and chemical parameters after 5 to 6 weeks of processing. Nitrogen losses during the composting should be prevented for the improvement of the composting efficiency of broiler litter.

Distillers Dried Grain as a Partial Replacement for Wheat Flour in the Diet of Juvenile Rockfish Sebastes schlegeli

  • A 9-week feeding experiment was designed to test the effects of the dietary inclusion of distillers dried grain (DDG) on the growth performance, body composition, and antioxidant activity of juvenile rockfish Sebastes schlegeli. Five isonitrogenous and isocaloric diets were formulated to contain 0% DDG (DDG0), 7% and 14% DDG from rice (diets DDG-R7 and DDG-R14, respectively), as well as 7% and 14% DDG from rice and wheat flour (diets DDG-RW7 and DDG-RW14, respectively). Three replicate groups of juvenile rockfish averaging $68.0{\pm}0.4g$ were fed one of the diets to apparent satiation twice a day. Survival, weight gain, feed efficiency, and the protein efficiency ratio of fish were not affected by dietary DDG (P > 0.05). Proximate and amino acid compositions of the whole body were not affected by dietary DDG (P > 0.05). Plasma total protein, glucose, total cholesterol, glutamate oxaloacetate transaminase, glutamate pyruvate transaminase, phospholipid, and triglyceride were not affected by dietary DDG (P > 0.05). Radical scavenging activity on 1,1-diphenyl-2-picryl-hydrazyl (DPPH) in the plasma of rockfish fed the DDG-RW14 diet was higher than that of fish fed the DDG0 diet (P < 0.05). Hydroxyl radical scavenging activity in the liver of rockfish fed diets containing DDG was higher than that of fish fed the DDG0 diet (P < 0.05). Alkyl radical scavenging activity in the liver of rockfish fed the DDG-R7 diet was higher than that of fish fed the DDG0 diet (P < 0.05). These results suggest that DDG is a suitable ingredient for the partial replacement of wheat flour and can be used at a rate of up to 14% in the diet without incurring negative effects on the growth performance or body chemical composition of juvenile rockfish.

  • Phytochemical investigation of the 80% MeOH extract from the leaves of Hosta longipes resulted in the isolation of sixteen compounds (1 - 16). The structures of the compounds were elucidated by spectroscopic methods to be methyl 10,10-dimethoxydecanoate (1), methyl 10-hydroxy-8E,12Z-octadecadienoate (2), methyl coriolate (3), trans-phytol (4), phytene-1,2-diol (5), phyton (6), (3S,5R,6S,7E,9R)-7-megastigmene-3,6,9-triol (7), (3S,5R,6S,9R)-3,6,9-trihydroxymegastigman-7-ene (8), shikimic acid (9), p-coumaramide (10), trans-N-p-coumaroyltyramine (11), cis-N-coumaroyltyramine (12), tryptophan (13), thymidine (14), adenosine (15), and deoxyadenosine (16). Compound 1 was synthesized, but not yet isolated from natural source, and compounds 2-16 were isolated for the first time from this plant source.

The Effects of Different Crude Protein Levels in Same Methionine and Lysine Diet on the Performance of Laying Hens (동일한 Methionine과 Lysine수준의 사료에서 단백질수준이 산란계의 생산성에 미치는 영향)

  • The purpose of this study was to investigate the effects of dietary protein levels on laying hen performance. The level of methionine and lysine were 0.32% and 0.64%, respectively and the levels of protein were 12%, 13%, 14% or 15%. Total 384 laying pullets of 22weeks age were reared from January 28, 1989 to March 23, 1990 for 60 weeks. The results obtained were summarized as follows : 1 Egg productions was highest at 15% of protein in phase I, 14% in phase II, and 13% in phase III, and there was significantly different egg Production among treatments during phase I and phase II (P<0.05). 2. Egg weight was heaviest in 14% of protein treatment in three phases and they showed significantly different egg weight among different levels of protein in phase I (P<0.01), phase II and III (P<0.05) , but there was not significantly different between 14% and 15% of protein. 3. Daily egg mass tends to increase followed by increasing of protein level and showed signifiant differences among treatments in phase I and phase II (P<0.01). 4. The 14% of protein treatment showed the highest daily feed intake and it showed significant difference in phase I and phase II (P<0.01) , but there was no significant difference between 14% and 15% of protein. 5. Feed efficiency was improved significantly followed by increasing of protein level in phase I (P<0.01) and phase II (P<0.05), but there was no significant difference among treatments in phase III. 6. Viability tends to increase as increasing of protein level, but there was no significant difference among treatments. 7. Utilizabilities of dry matter, crude protein and ether extract of experimental diets were not different among treatments, but the utilizability of carbohydrate tends to increase as increasing of protein level (P<0.05). 8. Eviscerated yield and abdominal fat accumulation was not difference among treatments. 9. Egg shell quality and chemical composition of egg content were not different among treatments. 10. The feed cost per kg egg mass showed the cheapest in 13% of protein treatment in all phase, but there were no significant differences among treatments.

Synthesis and Light-emitting Properties of Random Copolymers Composed of Phenylsilyl- and Alkoxy-Sustituted Phenylenevinylene

  • New random copolymers of phenylsilyl- and alkoxy-substituted phenylenevinylene, DMPS(9)-MEH(1)-PPV, DMPS(5)- MEH(5)-PPV and DMPS(1)-MEH(9)-PPV, have been synthesized by the Gilch dehydrohalogenation route, and the light-emitting properties of these polymers have been studied. The synthesized polymers were completely soluble in common organic solvents, and exhibited good thermal stability, almost up to $380^{\circ}C$. They showed UV-visible absorbance and photoluminescence (PL) in the ranges of 422-510 and 513-590 nm, respectively, according to their feed ratios. Electroluminescent devices were fabricated with these polymers as emitting layers, and ITO and Al as anode and cathode, respectively. DMPS(1)-MEH(9)-PPV, DMPS(5)-MEH(5)-PPV and DMPS(9)-MEH(1)-PPV exhibited EL emission maxima at 575 nm, 565 nm, and 541 nm, respectively.

The Effects of Bed Baths with 2% Chlorhexidine on the Incidence of Methicillin-resistant Staphylococcus aureus and Blood Stream Infection in Intensive Care Units (2% 클로르헥시딘 침상목욕 간호가 중환자실 입원환자의 메티실린 내성 황색포도상구균과 혈류감염 발생에 미치는 효과)

  • The purpose of this study was to identify the effects of bed baths with 2% chlorhexidine on the incidence of Methicillin-resistant Staphylococcus aureus (MRSA) and blood stream infection (BSI) and to suggest guidelines on the therapeutic bed baths using skin cleaner. This study was designed to compare the incidence of MRSA and BSI of the experimental group(n=188) who received the bed baths with 2% chlorhexidine with the incidence of MRSA and BSI of the control group(n=199) who received the existing bed baths with soap and skin cleaner. A research design used in the study was a randomized control group posttest-only design. The experimental group had 6.7% decrease in MRSA acquisition than the control group (7.4% vs 14.1%, p=.036). The experimental group was decreased in the incidence density of MRSA than the control group (9.32 cases per 1,000 patient-days at risk of experimental group vs 15.44 cases per 1,000 patient-days at risk of control group; p=.099). The experimental group had 4.5% decrease in the rate of BSI than the control group (0.5% vs 5.0%, p=.011). The experimental group was decreased in the incidence density of BSI than the control group (0.67 cases per 1,000 patient-days at risk of experimental group vs 5.52 cases per 1,000 patient-days at risk of control group; p=.052). These finding indicated that bed baths with 2% chlorhexidine is an effective nursing intervention to decrease the incidence of MRSA and BSI.

Physico-chemical Characteristics of Yogurt by Lactobacillus spp. from Pickles (젓갈에서 분리한 Lactobacillus spp.로 제조한 요구르트의 이화학적 특성)

  • Three strains of lactic acid bacteria were isolated from fish and shrimp pickles. Two strains were identified as Lactobacillus casei, and one strain as L. Pentosus, respectively. All three strains were used as starters in producing yogurts. The physico-chemical characteristics of yogurts were examined. The original pH, titratable acidity, visicosity and viable cell counts of the yogurts were $4.03{\sim}4.26,\;1.049{\sim}l.217%,\;1,772{\sim}2,232\;cps\;and\;1.4{\times}10^9{\sim}1.6{\times}10^9\;cfu/ml$, respectively. In evaluating buffer capacity, $12.50{\sim}14.06\;ml$ 1.0N HCl was consumed to titrate 100 ml of yogurt to pH value 2 units below the original pH value and $9.46{\sim}13.06\;ml$ of 1.0N NaOH was consumed to pH value 4 units above the original pH value. The ${\beta}-galactosidase$ activity reached maximum at 48 hrs, and reduced gradually during fermantation. After 2 hr incubation of yogurts at $37^{\circ}C$ under different pH conditions, ${\beta}-galactosidase$ activities of three strains were reduced to 50% at pH 3.5, but there were no remaining activities neither at pH 2.5 nor at pH 1.5. Under the same pH conditions the number of viable cells decreased to $1.9{\times}10^6{\sim}1.8{\times}10^8\;cfu/ml$ at pH 2.5 and $1.0{\times}10^3{\sim}2.4{\times}10^5$ at pH 1.5, respectively. However, no significant difference was found at pH 3.5.

Degradation of Trichloroethylene by a Growth-Arrested Pseudomonas putida

  • A toluene-oxidizing strain of Pseudomanas mendocina KR1 containing toluene-4-mono-oxygenase (TMO) completely degrades TCE with the addition of toluene as a co-substrate in aerobic condition. In order to construct in situ bioremediation system for TCE degradation without any growth-stimulating nutrients or toxic inducer such as toluene, we used the carbon-starvation promoter of Pseudomonas putida MK1 (Kim, Y. et al., J. bacteriol., 1995). Upon entry into the stationary phase due to the deprivation of nutrients, this promoter is strongly induced without further cell growth. The TMO gene cluster (4.5 kb) was spliced downstream of the carbon starvation promoter of Pseudomonas putida MK1, already cloned in pUC19. TMO under the carbon starvation promoter was not expressed in E. coli cells either in stationary phase or exponential phase. For TMO expression in Pseudomonas strains, tmo and carbon starvation promoter region were recloned into a modified broad-host range vector pMMB67HES which was made from pMMB67HE(8.9 kb) by deletion of tac promoter and lacIq (about 1.5 kb). Indigo was produced by TMO under the carbon starvation promoter in a Pseudomonas strain of post-exponential phase on M9 (0.2% glucose and 1mM indole) or LB. 18% of TCE was degraded in 14 hours after entering the stationary phase at the initial concentration of 6.6 ${\mu}$M in liquid phase.
