Identification and Characterization of Nitric Oxide Synthase in Salmonella typhimurium

  • The presence of the nitric oxide synthase (NOS) enzyme from Salmonella typhimurium (S. typhimurium) was identified by measuring radiolabeled L-$[^3H]$citrulline and NO, and Western blot analysis. NOS was partially purified by both Mono Q ion exchange and Superose 12HR size exclusion column chromatography, sequentially. The molecular weight of NOS was estimated to be 93.3 kDa by Western blot analysis. The enzyme showed a significant dependency on the typical NOS cofactors; an apparent Km for L-arginine of 34.7 mM and maximum activity between $37^{\circ}C$ and $43^{\circ}C$. The activity was inhibited by NOS inhibitors such as aminoguanidine and $N^{G}$ $N^{G}$-dimethyl-L-arginine. taken together, partially purified NOS in S. typhimurium is assumed to be a different isoform of mammalian NOSs.OSs.

  • The retardation effect of fatigue crack propagation after cyclic overloading seems to be affected by strain hardening exponent. Namely, for the material with high values of n, the delay effect is found to be severe. We proposed a modified crack retardation equation which may apply the retardation of fatigue crack growth after a cyclic overloading, as (da/dN)'$_{cyc}$=($\mu$n+λ)B $\Delta$ $K^{q}$ /[(1- $R_{eff}$) $K_{cf}$$\Delta$K]. where, $R_{eff}$ is effective stress ratio [=( $K_{min}$-K, os)/( $K_{max}$$K_{res}$)] The constants $\mu$=-0.5 and λ=0.6, and the values are found to be identical for materials such as aluminum (A 1060), steel (SS 34), brass ( $B_{s}$ SIB) and stainless steel (SUS 304) used in this investigation.   • Purpose The lung segment ratio which is obtained through quantitative analyses of lung perfusion scan images is calculated to evaluate the lung function pre and post surgery. In this Study, the planar image production methods by using Q-Metrix (GE Healthcare, USA) program capable of not only quantitative analysis but also computation of the segment ratio after having performed SPECT/CT are comparatively evaluated. Materials and Methods Lung perfusion scan and SPECT/CT were performed on 50 lung cancer patients prior to surgery who visited our hospital from May 1, 2015 to September 13, 2016 by using Discovery 670(GE Healthcare, USA) equipment. AP(Anterior Posterior)method that uses planar image divided the frontal and rear images into three rectangular portions by means of ROI tool while PO(Posterior Oblique)method computed the segment ratio by dividing the right lobe into three parts and the left lobe into two parts on the oblique image. Segment ratio was computed by setting the ROI and VOI in the CT image by using Q-Metrix program and statistically analysis was performed with SPSS Ver. 23. Results Regarding the correlation concordance rate of Q-Metrix and AP methods, RUL(Right upper lobe), RML(Right middle lobe) and RLL(Right lower lobe) were 0.224, 0.035 and 0.447. LUL(Left upper lobe) and LLL(Left lower lobe) were found to be 0.643 and 0.456, respectively. In the PO method, the right lobe were 0.663, 0.623 and 0.702, respectively, while the left lobe were 0.754 and 0.823. When comparison was made by using the Paired sample T-test, Right lobe were $11.6{\pm}4.5$, $26.9{\pm}6.2$ and $17.8{\pm}4.2$, respectively in the AP method. Left lobe were $28.4{\pm}4.8$ and $15.4{\pm}5.6$. The right lobe of PO had values of $17.4{\pm}5.0$, $10.5{\pm}3.6$ and $27.3{\pm}6.0$, while the left lobe had values of $21.6{\pm}4.8$ and $23.1{\pm}6.6$, thereby having statistically significant difference in comparison to the Q-Metrix method for each of the lobes (P<0.05). However, there was no statistically significant difference in Right middle lobe (P>0.05). Conclusion The AP method showed low concordance rate in correlation with the Q-Metrix method. However, PO method displayed high concordance rate overall. although AP method had significant differences in all lobes, there was no significant difference in Right middle lobe of PO method. Therefore, at the time of production of lung perfusion scan results, utilization of Q-Metrix method of SPECT/CT would be useful in computation of accurate resultant values. Moreover, it is deemed possible to expect obtain more practical sectional computation result values by using PO method at the time of planar image acquisition.

Awareness of Major Zoonoses among Dairy Farmers in Gyeonggi Province (경기도 낙농업자의 주요 인수공통감염증 인지도)

  • Objective: We surveyed awareness levels of brucellosis, Q fever and enterohemorrhagic Escherichia coli (EHEC) among dairy farmers in Gyeonggi Province to suggest directions for public education and public relations. Methods: We designed questionnaires to evaluate awareness of 3 major zooneses. We conducted a questionnaire survey to assess knowledge of the general characteristics of them, information sources for the awareness of zooneses, and the mode of transmission. Subjects were 716 workers from 482 dairy farms in Gyeonggi province. Results: The awareness levels for brucellosis, Q fever, and EHEC were 90.2%, 2.5% and 56.6%, respectively. Awareness of brucellosis and EHEC were tended to increase with higher number of school years. Television was the most common route of information for these zoonoses. Most common responses for questions concerning the method of transmission for each zoonoses, 'Contact with parturient fluid or placenta of animal' was 63.2% for brucellosis, 'Ingestion of raw meat or residual product' was 66.7% and 64.2% for Q fever and EHEC, respectively. The most common reason why dairy farmers think that it is difficult to prevent zoonoses was the inconvenience of wearing protection. Conclusions: Education programs for zoonoses, especially Q fever, are needed for dairy farmers. In addition, publicity information activities about prevention of zoonoses are needed for high risk groups, such as the dairy farmers surveyed.

The Effects of Training in Exercise Rehabilitation on Depressive mood, BPSD in Elderly with Alzheimer's Disease (운동재활훈련이 알츠하이머 노인의 우울정서와 정신행동증상에 미치는 효과)

  • Purpose : The purpose of this study is to examine the effects of training in exercise rehabilitation on depressive emotion, behavioral and psychological symptoms(BPSD) caused by Alzheimer's disease elders. Methods : The subjects of this study were 34 patients with Alzheimer's disease over 65 and assigned randomly into one of two groups, each with 17 people. The experimental group was treated by training in exercise rehabilitation executed 60 minutes, 3 times a week for 8 weeks. The results of the experiment were analyzed using samples t-test along with averages and standard deviations as the statistical methods for data analyses. Results : The results of this study were as follows. First, the experimental group showed statistically significant improvements in the areas of depressive emotion(SGDS) compared to control group. Second, the experimental group showed statistically significant improvements in the severity of behavioral and psychological symptoms and subjective degrees of pain of care givers(NPI-Q) compared to control group. Conclusion : Based on the results of this study, training in exercise rehabilitation have an effect on depressive mood and BPSD in elderly with Alzheimer's disease.

Low Phase Noise VCO using Microstrip Square Open Loop Multiple Split Ring Resonator (마이크로스트립 사각 개방 루프 다중 SRR(Split Ring Resonator)를 이용한 저위상 잡음 전압 제어 발진기)

  • In this paper, a novel voltage-controlled oscillator (VCO) using the microstrip square open loop multiple split ring resonator (OLMSRR) is presented for reducing the phase noise property. The square-shaped multiple split ring resonator (MSRR) having the form of the microstrip square open loop is investigated to realize this property. Compared with the microstrip square open loop resonator and the microstrip square open loop split ring resonator (OLSRR) as well as the conventional microstrip line resonator, the microstrip square OLMSRR has the larger coupling coefficient value, which makes a higher Q value, and has reduced the phase noise of VCO. The VCO with 1.7V power suppIy has the phase noise of $-124.5\;{\sim}\;-122.0\;dBc/Hz$ @ 100 kHz in the tuning range, $5.746\;{\sim}\;5.84\;GHz$. The figure of merit (FOM) of this VCO is $-203.96\;{\sim}\;-201.6\;dBc/Hz$ @ 100 kHz in the same tuning range. Compared with VCO using the conventional microstrip line resonator, VCO using the microstrip square open loop resonator and VCO using microstrip square OLSRR, the phase noise property of VCO using the proposed resonator has been improved in 25.66 dB, 8.34 dB, and 4.5 dB, respectively.

Forced Convective Boiling of Pure Refrigerants in a Bundle of Enhanced Tubes (전열촉진관군의 순수냉매 강제대류비등)

  • In this study, convective boiling tests were conducted for enhanced tube bundles. The surface geometry consists of pores and connecting gaps. Tubes with three different pore sizes (d$_{p}$ = 0.20, 0.23 and 0.27 mm) were tested using R-123 and R-l34a for the following range: 8 kg/m$^2$s G 26 kg/m$^2$s, 10 kW/m$^2$ q0 40 kW/m$^2$and 0.1 $\chi$ 0.9. The convective boiling heat transfer coefficients were strongly dependent on heat flux with negligible dependency on mass flux or quality. For the present enhanced geometry (pores and gaps), the convective effect was apparent. The gaps of the present tubes may have served routes for the passage of two-phase mixtures, and enhanced the boiling heat transfer. The convective effect was more pronounced at a higher saturation temperature. More bubbles will be generated at a higher saturation temperature, which will lead to enhanced convective contribution. The pore size where the maximum heat transfer coefficient was obtained was larger for R-l34a (d$_{p}$ = 0.27 mm) compared with that for R-123 (d$_{p}$ = 0.23 mm). This trend was consistent with the previous pool boiling results. For the enhanced tube bundles, the convective effect was more pronounced for R-134a than for R-123. This trend was reversed for the smooth tube bundle. Possible reasoning is provided based on the bubble behavior on the tube wall. Both the modified Chen and the asymptotic model predicted the present data reasonably well. The RMSEs were 14.3% for the modified Chen model and 12.7% for the asymptotic model.model.

Isolation and identification of goose skeletal muscle satellite cells and preliminary study on the function of C1q and tumor necrosis factor-related protein 3 gene

  • Objective: Skeletal muscle satellite cells (SMSCs) are significant for the growth, regeneration, and maintenance of skeletal muscle after birth. However, currently, few studies have been performed on the isolation, culture and inducing differentiation of goose muscle satellite cells. Previous studies have shown that C1q and tumor necrosis factor-related protein 3 (CTRP3) participated in the process of muscle growth and development, but its role in the goose skeletal muscle development is not yet clear. This study aimed to isolate, culture, and identify the goose SMSCs in vitro. Additionally, to explore the function of CTRP3 in goose SMSCs. Methods: Goose SMSCs were isolated using 0.25% trypsin from leg muscle (LM) of 15 to 20 day fertilized goose eggs. Cell differentiation was induced by transferring the cells to differentiation medium with 2% horse serum and 1% penicillin streptomycin. Immunofluorescence staining of Desmin and Pax7 was used to identify goose SMSCs. Quantitative realtime polymerase chain reaction and western blot were applied to explore developmental expression profile of CTRP3 in LM and the regulation of CTRP3 on myosin heavy chains (MyHC), myogenin (MyoG) expression and Notch signaling pathway related genes expression. Results: The goose SMSCs were successfully isolated and cultured. The expression of Pax7 and Desmin were observed in the isolated cells. The expression of CTRP3 decreased significantly during leg muscle development. Overexpression of CTRP3 could enhance the expression of two myogenic differentiation marker genes, MyHC and MyoG. But knockdown of CTRP3 suppressed their expression. Furthermore, CTRP3 could repress the mRNA level of Notch signaling pathway-related genes, notch receptor 1, notch receptor 2 and hairy/enhancer-of-split related with YRPW motif 1, which previously showed a negative regulation in myoblast differentiation. Conclusion: These findings provide a useful cell model for the future research on goose muscle development and suggest that CTRP3 may play an essential role in skeletal muscle growth of goose.

Detection of phenobarbital adulteration in dietary supplements: simultaneous analysis of 16 sedative-hypnotics and sleep-inducers by ultra-high-performance liquid chromatography with UV detection (UPLC-UV) and quadruple Orbitrap mass spectrometry (Q-Orbitrap-MS)

  • The safety of food is occasionally questionable, as there have been some reports of products contaminated with illegal adulterants. In this study, the presence of 16 sedative-hypnotics and sleep inducers in dietary supplements was determined by ultra-high-performance liquid chromatography with UV detection (UPLC-UV) and quadruple Orbitrap mass spectrometry (Q-Orbitrap-MS). The UPLC method was validated, providing a linearity (R2) of more than 0.999, and LODs and LOQs that ranged from 0.2 to 0.5 and 0.6 to $1.5{\mu}g\;mL^{-1}$, respectively. The repeatabilities were 0.2-8.4 % (intra-day) and 0.3-4.5 % (inter-day), and the accuracies were 89.0-117.0 % (intra-day) and 87.8-111.9 % (inter-day). The mean recoveries of the spiked samples ranged from 98.7 to 107.3 %. The relative standard deviation (%RSD) of the stability was less than 2.4 %. Using the developed method, one sedative-hypnotic compound, phenobarbital, was detected in one of the nineteen samples tested. In addition, the major characteristic fragment ions of each target compound were confirmed using Q-Orbitrap-MS for higher accuracy. Monitoring the presence of these 16 sedative-hypnotics and sleep inducers in dietary supplements should be pursued in the interest of human health, and the results of this study confirmed that the developed method has value for this application.
