Inhibitory Effect of Mixed Extracts Obtained from Astragali Radix and Lithospermi Radix on Matrix Metalloproteinases in IL-1β-induced SW1353 Cells and Quantitative Analysis of Active Compounds (황기, 지치 복합물의 연골세포에서의 Matrix Metalloproteinases 저해 효과 및 유효성분의 분석)

  • Background: Astragali radix (A) and Lithospermi radix (L) have long been used as traditional medicines due to their known anti-inflammatory effects. This study aimed at evaluating, their optimal mixing ratio and their functional compounds by investigating the inhibitory effects of mixed extracts of A and L and their active compounds on matrix metalloproteinases (MMPs). Methods and Results: A and L extracts were obtained by extraction at $80^{\circ}C$ using 50% and 70% fermented alcohol, respectively, and then mixed at a ratio of 5 : 5, 6 : 4, 7 : 3 and 8 : 2 (w/w). The activities of MMP-1, MMP-3, and MMP-13 were evaluated in interleukin-1beta ($IL-1{\beta}$)-induced SW1353 cells. The extract mixtures showed synergistic inhibitory effects on MMP-3 and MMP-13, higher than the effects of the individual A and L extracts. The 7 : 3 mixture (ALM16) showed the most effective MMPs inhibitory activity, while among the active ingredients, calycosin-7-O-${\beta}$-D-glucoside and lithospermic acid exhibited excellent MMPs inhibitory activity. Additionally, an HPLC method was established for simultaneous quantification of the effective components of the extract mixtures, and validated by measuring the linearity, precision and accuracy of the limit of detection (LOD) and limit of quantification (LOQ). Conclusions: ALM16 showed the most effective MMPs inhibitory activity. Calycosin-O-${\beta}$-D-glucoside, calycosin and lithospermic acid were identified as useful candidates, as they were the major functional compounds in the MMP inhibitory activity. Summarily, ALM16 might be a highly effective in osteoarthritis management, owing to its because it exhibits a protective effect on cartilage via excellent inhibition of MMPs.

New Cosmetic Agents for Anti-aging from Zostera marina L. (잘피(Zostera marina L.)의 신규 항노화 화장품 소재 응용)

  • In order to develop new anti-photoaging agents from marine natural products, Zostera marina L. was selected for its antioxidative activity and inhibition of matrix metalloproteinase-1 (MMP-1) expression. Three compounds (compounds 1, 2, and 3) were isolated from the extract, and they were identified as apigenin-7-O-${\beta}$-D-glucoside (1), chrysoeriol (2), and luteolin (3). These compounds have SC$\_$50/ values of 0.18 mM, 0.68 mM, and 0.01 mM against l,l-dipheny1-2-picrylhydrazyl radical and 0.04mM, 0.03mM, and 0.01mM against the superoxide radical in the xanthine/xanthine oxidase system, respectively. Compound 3 suppressed the expression of MMP-1 by up to 44% at 35.0${\mu}$M and inhibited the production of interleukin 6, which is known as a cytokine that induces MMP-1 expression. In addition, the wrinkle improvement effect of the formulation with Z. marina extract was measured. As a result, remarkable reduction was found in the fine wrinkle and skin roughness after application of the cream with 3.0% this extract for 8 weeks. In conclusion, the isolated compounds from Z. marina extract were good antioxidant and suppressor of MMP-1 expression and the formulation with the extract diminished the skin wrinkle. Therefore, the extract can be used as a new anti-aging agent for application in cosmetic.

Studies on the consituents of Chrysanthemum sibiricum FISHER (구절초 chrysanthemum sibiricum FISCHER의 성분 연구)

  • A yellowish microneedles, $C_{28}$ H$_{32}$ $O_{14}$ ${\cdot}$ I$_{1}$/$_2$, H$_{2}$O, m.p.262-$4^{\circ}$ , [${\alpha}$$_{D}^{20}$= -71,$43^{\circ}$(C = 0.42, pyridine), its acetate m.p.123-5.deg., were obtained in 0.3% yield from the leaves of Chrysanthemum sibiricum F$_{ISCHER}$. This substance is insoluble in water and the usual organic solvents except pyridine and ethylene glycol and, is not decomposed by dilute mineral acids but undergoes decomposition on being boiled in 60% H$_{2}$SO$_{4}$ or 35% HCl, giving one moel each of acacetin, glucose and rhamnose. It was not hydrolysed with a rhamnodiastase preparation obtained from the seeds of Rhamnus koraiensis. After permethylation of it, the uncrystallized product was hydrolysed and apigenin-5,4'-dimethyl ehter, m.p.$262^{\circ}$ was obtained, indicating that the disaccharide residue is at the 7 position of acacetin. Partial hydrolysis of this acacetin-7-rhamnoglucoside in cyclohexanol with formic acid gave acacetin-7-glucoside, m.p.246.deg. and rutinose, identifying them with authentic specimen on a paper chromatography. It was thus identified as linarin(acacetin-7-rutinoside) by means of mixed fusion, of paper partition chromatography and of its derivatives. Zemplen and Bognar suggested that the glucosidic linkage of linarin is .betha. by means of synthesis of this substance. But there is no evidence whether it is hydrolysed by emulsin or maltase or not. Linarin itself was not hydrolysed by an emulsin existing in the seed of Apricot or a maltase, but acacetin-7-glucoside(tilianin) which obtained from linarin gave acacetin and glucose on hydrolysis with the same emulsin and accordingly the glucosidic linkages of linarin and tilianin are thus regarded as ${\beta}$.

Melanogenesis Inhibitory Activities of Diarylheptanoids from Alnus hirsuta Turcz in B16 Mouse Melanoma Cell

  • Four diarylheptanoids, (5R)-1,7-bis (3,4-dihydroxyphenyl)-heptane-5-O-$\beta$-D-glucoside (1), (5R)-1,7-bis (3,4-dihydroxyphenyl)-heptane-5-ol (2), oregonin (3), hirsutanonol (4), were isolated from the bark of Alnus hirsuta Turcz and its inhibitory effects on melanogenesis by measuring the melanin level and tyrosinase activity in B16 melanoma cell were examined. Melanin level and tyrosinase activity were reduced to 75 to 85% by addition of diarylheptanoids to incubation medium of the melanoma cell. On the other hand, melanin level and tyrosinase activity were reduced to 13 to 43% by the addition of diarylheptanoids to incubation medium of the melanoma cell treated with melanogenesis stimulator, $\alpha$-MSH and forskolin. These melanogenesis inhibitory effects were significantly different compared with control.

The Protective Effects of Phenolic Constituents from Gastrodia elata on the Cytotoxicity Induced by KCI and Glutamate

  • Seven phenolic compounds (1-7) were isolated from the tubers of Gastrodia elata. Their structures were elucidated on the basis of MS and NMR spectral data. p-Ethoxymethyl phenyl-O-${\beta}$-D-glucoside (1) was proved to be a new compound, with N-(p-hydroxybenzyl)-adenosine (7) isolated from this plant for the first time. In this study, the protective effects of the six constituents (1-6) on PC12 cells against the cytotoxicity induced by KCI and glutamate were also investigated. The viability of the PC12 cells was significantly enhanced by pretreatment with the six phenolic constituents.

Biological Activities on Phenolic Compounds of Japanese anise (Illicium anisatum L) Extracts

  • In this paper, we have isolated six phenolic compounds, such as (+)-catechin (1), taxifolin (2), taxifolin-3-O-${\beta}$-D-(+)-xylose (3), quercetin (4), quercetin-3-O-${\alpha}$-L(+)-rhamnose (quercitrin) (5), apigenin-8-C-rhamnosyl-(1'''${\rightarrow}$2'')-glucoside (2''-O-rhamnosylvitexin) (6) from the EtOAc(Ethyl Acetate) and $H_2O$ soluble fractions of Japanese anise(Illicium anisatum L) leaves and twigs. Also, we have evaluated antioxidative and antiviral activity for each isolated compound. The antioxidative test was DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity. According to the experimental results, all of the isolated compounds indicated the increased radical scavenging activities as the concentration increases and most of the isolated compounds indicated generally good antioxidative values compare to the controls, ascorbic acid and ${\alpha}$-tocopherol. In the antiviral activities, all of the isolated compounds had no potentials in rhinovirus 1B (HRV 1B). But in enterovirus 71 (EV 71) and Influenza virus A/PR/8 (Influenza PR8), only quercetin (4) indicated the good antiviral activity compare to the control. Based on the above results, we found that the phenolic compounds of Japanese anise may be applied for one of the natural biomass sources that can be used as an antioxidant and an antiviral substance.

Isolation and Identification of Bioactive Compounds from the Tuber of Brassica oleracea var. gongylodes

  • Brassica oleracea var. gongylodes (red kohlrabi) is a biennial herbaceous vegetable whose edible bulbotuber-like stem and leaves are consumed globally. Sliced red kohlrabi tubers were extracted using methanol and the concentrated extract was partitioned successively with dichloromethane (CH2Cl2), ethyl acetate (EtOAc), n-butanol (n-BuOH) and water (H2O). Repeated column chromatography of EtOAc fraction through silica, sephadex LH-20 and RP-18 gel led to isolation of eleven compounds of which compound 1 was a new glycosylated indole alkaloid derivative, 1-methoxyindole 3-carboxylic acid 6-O-β-D-glucopyranoside. Others were known compounds namely, β-sitosterol glucoside (4), 5-hydroxymethyl-2-furaldehyde (5), methyl-1-thio-β-D-glucopyranosyl disulfide (6), 5-hydroxy-2-pyridinemethanol (7), (3S,4R)-2-deoxyribonolactone (8), n-butyl-β-D-fructopyranoside (9), uridine (10) and three fructose derivatives, D-tagatose (11), β-D-fructofuranose (12) and β-D-fructopyranose (13). Similarly, isolation from CH2Cl2 fraction gave two known indole alkaloids, indole 3-acetonitrile (2) and N-methoxyindole 3-acetonitrile (3). The structure elucidation and identification of these compounds were conducted with the help of 13C and 1H NMR, HMBC, HMQC, EIMS, HR-ESIMS and IR spectroscopic data, and TLC plate spots visualization. Compounds 2, 3, 4, 5, 6, 7, 8 and 9 are noted to occur in kohlrabi for the first time. Different bioactivities of these isolated compounds have been reported in literature.

Cyanidin and Cyanidin-3-O-β-D-glucoside Suppress the Inflammatory Responses of Obese Adipose Tissue by Inhibiting the Release of Chemokines MCP-1 and MRP-2

  • Obesity-induced inflammation plays a crucial role in obesity-related pathologies such as type II diabetes and atherosclerosis. Adipose tissue macrophages and the cell-derived proinflammatory chemokines are key components in augmenting inflammatory responses in obesity. Anthocyanins such as cyanidin and $cyanidin-3-O-{\beta}-D-glucoside$ (C3G) are known to elicit anti-inflammatory activities by suppressing the production of proinflammatory mediators such as tumor necrosis factor alpha and nitric oxide in LPS-stimulated macrophages. In the present study, we investigated whether cyanidin and C3G have the potential to suppress the inflammatory responses of adipose cells. Cyanidin and C3G not only suppressed the migration of RAW 264.7 macrophages induced by mesenteric adipose tissue-conditioned medium, but also inhibited the activation of the cells to produce inflammatory chemokines such as monocyte chemoattractant protein-1 (MCP-1) and macrophage inflammatory protein-related protein-2 (MRP-2) in a dose-dependent manner. Cyanidin and C3G also inhibited the release of MCP-1 and MRP-2 from adipocytes and/or macrophages. These findings suggest that cyanidin and C3G may suppress the inflammatory responses of adipose tissue in obesity.

Changes in physicochemical components of Astragalus membranaceus fermented with Phellinus linteus (상황버섯균 접종 황기의 배양 중 이화학적 성분변화)

  • This study analyzed the changes in physicochemical components of Astragalus membranaceus (AM) fermented with Phellinus linteus. Moisture content, pH, total acidity, total reducing sugar content, extraction yield, free sugar content, free amino acid and isoflavonoid (calycosin, formononetin) were investigated. The moisture content was increased during fermentation with Phellinus linteus. The pH level increased while the total acidity significantly decreased during fermentation. The reducing sugar content were in the range of 0.32~0.61%. The extraction yield using water was higher than that using 80% ethanol. The major free sugars were identified as glucose, fructose, sucrose and the content of free sugars decreased through fermentation. However, the glucose and sucrose contents of the water extracts were increased. In addition, the free amino acid increased significantly during fermentation. Finally, calycosin and formononetins contents in water extracts of after 30 days of AM fermentaion with Phellinus linteus were (3.91 mg/100 g) and (1.38 mg/100 g), respectively. These results suggest that fermentation with Phellinus linteus could be used to increase the bioactivity of AM. The mycelium-fermented AM could be a valuable source of functional material and edible resource for industry.

The Protective Effect of Quercetin-3-O-${\beta}$-D-Glucuronopyranoside on Ethanol-induced Damage in Cultured Feline Esophageal Epithelial Cells

  • Quercetin-3-O-${\beta}$-D-glucuronopyranoside (QGC) is a flavonoid glucoside extracted from Rumex Aquaticus Herba. We aimed to explore its protective effect against ethanol-induced cell damage and the mechanism involved in the effect in feline esophageal epithelial cells (EEC). Cell viability was tested and 2',7'-dichlorofluorescin diacetate assay was used to detect intracellular $H_2O_2$ production. Western blotting analysis was performed to investigate MAPK activation and interleukin 6 (IL-6) expression. Exposure of cells to 10% ethanol time-dependently decreased cell viability. Notably, exposure to ethanol for 30 min decreased cell viability to 43.4%. When cells were incubated with $50{\mu}M$ QGC for 12 h prior to and during ethanol treatment, cell viability was increased to 65%. QGC also inhibited the $H_2O_2$ production and activation of ERK 1/2 induced by ethanol. Pretreatment of cells with the NADPH oxidase inhibitor, diphenylene iodonium, also inhibited the ethanol-induced ERK 1/2 activation. Treatment of cells with ethanol for 30 or 60 min in the absence or presence of QGC exhibited no changes in the IL-6 expression or release compared to control. Taken together, the data indicate that the cytoprotective effect of QGC against ethanol-induced cell damage may involve inhibition of ROS generation and downstream activation of the ERK 1/2 in feline EEC.
