Fluorescent siderophore 생산균주, TS3-7에 의한 풋마름병 발병 억제 (Suppression of Bacterial Wilt with Fuorescent Pseudomonads, TS3-7 strain)

  • Among the root colonizing and plant growth promoting bacteria isolated from the bacterial wilt suppressive soil, five strains were detected to produce siderophores by CAS agar assay. The most effective isolate, TS3-7 strain induced significant suppression of bacterial wilt disease in tomato and pepper plants. Seed treatment followed by soil drench application with this strain resulted in over 80% reduction of bacterial wilt disease compared with the control. Significant disease suppression by TS3-7 strain was related to the production of siderophore. Besides iron competition, induction of resistance of the host plant with siderophore was suggested to be another mode of action that suppress bacterial wilt, based on the lack of direct antibiosis against pathogen in vitro. According to Bergey's Manual of Systemic Bacteriology and 16S rDNA sequence data, TS3-7 stain was identified as Pseudomonas sp. TS3-7.

약모밀(Houttuynia cordata) 뿌리에서 추출한 순차분획물의 항균활성

  • 약모밀(Houttuynia cordata)의 뿌리에서 추출한 ethanol, chloroform, ethylacetate, butanol 및 수용성 추출물의 항균활성을 9 종의 Gram 양성균과 13 종의 Gram 음성균에 대하여 disc diffusion 방법으로 조사하였다. Ethanol, ethylacetae, butanol 추출물은 B. megaterium C. xerosis, S. aureus, S. epidermidis 및 E.coli 에 대하여 항균활성을 나타내었고, chloroform과 수용성 추출물은 활성이 나타나지 않거나 미약하였다. 그러나 Ethylacetate와 butanol 추출물은 상대적으로 낮은 농도에서 강한 항균활성을 나타내었다.

Antibacterial Activity of Triterpenoids from Clerodendron trichotomum

  • The aim of this research was to investigate the antibacterial activity of Clerodendron trichotomum. Antibacterial activities of the n-hexane, methylene chloride (MC), ethyl acetate, and n-butanol fractions from C. trichotomum were tested against Staphylococcus aureus, Escherichia coli, and Helicobacter pylori. The n-hexane and MC fractions showed antibacterial activity against H. pylori at a concentration of 1.7 mg/mL and showed inhibition zones of 10 and 11 mm in disc assay, respectively. Further testing of 22-dehydroclerosterol and ${\beta}$-amyrin (each 3.4 mg/mL) from the MC fraction of C. trichotomum revealed moderate antibacterial effects against E. coli, S. aureus, and H. pylori. In particular, ${\beta}$-amyrin showed clear zones of 12 and 13 mm against E. coli and H. pylori, respectively, suggesting its potential as an antibacterial agent. The active compounds from C. trichotomum might provide a promising therapeutic agent against infections by E. coli, S. aureus, and H. pylori.

  • The present study was performed to elucidate desmutagenic principles from Ecklonia stolonifera extracts against 2-amino-1-methyl-6-phenylimidazo[4,5-b] pyridine(PhIP) and 2-amino-3,8-dime-thylimidazo[4,5-f]duinoxaline(MeIQx) with Salmonella/mammalian-microsome mutagenicity test. Alginate, phenols, chlorophyll and carotenoids from Ecklonia stolonifera were extracted and their desmutagenicities were assayed. Alginate hydroysates showed desmutagenic activities against PhIP and MeIQx at high level dose. Phenol fractions and bromophenol showed desmutagenic activity of about MeIQx at high level dose. Phenol fractions and bromophenol showed desmutagenic activity of about 90% per 0.5mg against PhIP and MeIQx. Chlorophyllin among chlorophyll derivatives exhibited remarkable desmutagenic activities of 92.9% and 82.7% at 20uM against PhIP and MeIQx, respectively. Carotenoids, such as lutein and $\alpha$-cryptoxanthin isolated from Ecklonia stolonifera exerted also high desmutagenic activity. Major desmutagenic substances from Ecklonia stolonifera are considered to be chlorophyllin, phenols, lutein, $\alpha$-cryptoxanthin and low molecular alginates.

  • 냉각탑수 중의 Legionella 속 균을 저감시킬 수있는 방법개발의 일환으로 L. pneumophila에 강한 항균성물질을 생산하는P. seruginosa KLP-2 균주를 냉각탑수로부터 분리 동정하였다 P.aeruginosa KLP-2 균주를 냉각탑수로부터 분리 동정하였다. P. aeruginosa KLP-2 균주의 배양여액은 L. pneumophila의 Vibro cholerae non-Ol Bacillus cereus, Bacillus subtilis 및 Staphylococcus aureus에 대해서도 항균성을 나타내었다. P. seruginosa KLP-2 균주 배양여액의 항균활성을 증가시키기 위한 최적조건은 0.1%의 $K_2$$HPO_4$와 0.05%의 $MgSO_4$$7H_2$O로 조성되 무기염배지에 탄소원으로써 1%의 glycerol, 질소원으로써, 0.6%의 proteose peptone이 첨가된 pH 7.0의 배지 조건에서 가장 높은 항균 활성을 나타내었으며 $35^{\circ}C$에서 20시간 진탕배양(50 rpm)한 후 4일간 정치 배양하였을 때 최대의 항균활성을 나타내었다.

PC12 손상 세포 및 전뇌허혈 유발 Gerbil에 대한 지실의 세포보호효과 연구 (Protective Effect of Aurantii Immaturus Fructus on Hypoxia Reperfusion Induced by PC12 Cell Damage and Global Ischemia in Gerbil)

  • Object : This research was performed to investigate the protective effect of Aurantii Immaturus Fructus against ischemic damage using PC12 cells and global ischemia in gerbils. Methods : To observe the protective effect of Aurantii Immaturus Fructus on ischemia damage, viability and changes in activities of superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase and production of malondialdehyde (MDA) were observed after treating PC12 cells with Aurantii Immaturus Fructus during ischemic insult. Gerbils were divided into three groups : a normal group, a 5-min two-vessel occlusion (2VO) group, and an Aurantii Immaturus Fructus administered after 2VO group. The CCAs were occluded by microclip for 5 minutes. Aurantii Immaturus Fructus was administered orally for 7 days after 2VO. The histological analysis was performed at 7 days after the surgery. For histological analysis, the brain tissue was stained with 1% cresyl violet solution. Results : The results showed that 1. Aurantii Immaturus Fructus had a protective effect against ischemia in the CAI area of the gerbil hippocampus 7 days after 5-minute occlusion, 2. In the hypoxia/reperfusion model using PC12 cells, the Aurantii Immaturus Fructus had a protective effect against ischemia in the dose of $0.2{\;}\mu\textrm{g}/ml,{\;}2{\;}\mu\textrm{g}/ml{\;}and{\;}20{\;}\mu\textrm{g}/ml$ 3. Aurantii Immaturus Fructus increased the activities of glutathione peroxidase and catalase, 4. The activity of superoxide dismutase (SOD) was increased by ischemic damage, which might represent self protection. This study suggests that Aurantii Immaturus Fructus has some neuroprotective effect against neuronal damage following cerebral ischemia in vivo with a widely used experimental model of cerebral ischemia in Mongolian gerbils, and it also has protective effects on a hypoxia/reperfusion cell culture model using PCq2 cells. Conclusions : Aurantii Immaturus Fructus has protective effects against ischemic brain damage at the early stage of ischemia.

패혈증 치료제 개발을 위한 황백이 포함된 생약혼합제제 ABHC의 항균 효능 (Antibacterial Activity of Herbal Complex ABHC for Development of Novel Therapeutic Agent Against Sepsis)

  • Sepsis, an infectious disease, is a life-threatening condition that arises when the response to infection causes injury to tissues and organs. The purpose of this study was to demonstrate whether ABHC-1 and ABHC-2, two functional extracts from herbal complex, have an anti-bacterial effect against Escherchia coli in vivo, in vitro experimental model. ABHC-1 and ABHC-2 showed the antibacterial activity against the bacteria by paper disc method. The minimum inhibitory concentration (MIC) was measured using alamar blue reagent. The MIC was shown at $60{\mu}g/ml$ from ABHC-1 and $500{\mu}g/ml$ from ABHC-2 against E. coli. We next examined the effect of ABHCs on the production of inflammatory cytokine, such as tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), which is related to the induction of inflammation, in RAW 264.7 cell. ABHC-1 and ABHC-2 increased $TNF-{\alpha}$ production of RAW 264.7 cell in a dose-dependent manner while two extract decreased $TNF-{\alpha}$ production in lipopolysaccharide (LPS)-stimulated RAW 264.7 cell in a dose-dependent manner. At a dose of $1{\times}10^8$ E. coli. i.p., non-treated mice were succumbed, while most of mice treated with ABHC-1 were survived. Therefore, our results suggest that ABHC-1 has anti-bacterial activity and can be a novel therapeutic agent against infectious diseases.

황련과 Pelargonium sidoides 복합제제의 호흡기 감염 세균에 대한 항균 효과 (Anti-bacterial effect of fusion formulation of Coptis rhizoma and Pelargonium sidoides on the growth of bronchial diseases bacteria)

  • Objective : Coptis rhizoma is traditional herb in Korean medicine, and Pelargonium sidoides extract has been used for relief of acute bronchitis in Western medicine. The present study examined the antibacterial effect of fusion formulation of Coptis rhizoma extract and Pelargonium sidoides extract against bronchial diseases bacteria. Methods : Test sample, fusion formulation of Korean and Western medicine, was prepared by mixing Coptis rhizoma extract and Pelargonium sidoides extract at a ratio of 1:2 (w/w). Antimicrobial properties of test sample were determined by agar diffusion assay and minimum inhibitory concentration (MIC) against bronchus diseases bacteria. Results : In agar diffusion assay, the highest amount of test sample (4 mg/disk) exhibited antibacterial activity against all microorganisms tested. Test sample showed the high activity for S. aureus (19.5 mm), C. diptheriae (16.5 mm), A. fumigatus (19.3 mm), F. nucleatum (22.7 mm) and Mycobactrium sp. (17.3 mm), whereas it showed a low activity for K. pneumonia (9.7 mm). The MIC value was determined as $250{\mu}g/m{\ell}$ against C. diptheriae. Test sample showed better growth inhibitory effects against S. aureus and A. fumigatus with the MIC valus of $125{\mu}g/m{\ell}$. Conclusion : These results suggest the possibility of application to chronic respiratory diseases of fusion formulation of Korean and Western medicine, which was prepared with Coptis rhizoma extract and Pelargonium sidoides extract.
