Cytotoxicity of Ergosterol Derivatives from the Fruiting Bodies of Hygrophorus russula

  • Bioassay-guided fractionation of the $CHCl_3$-soluble fraction of a MeOH extract of the fruiting bodies of Hygrophorus russula led to the isolation of five ergosterol derivatives (1 - 5). The structures of these compounds were identified as ergosterol peroxide (1), ergosta-4,6,8(14),22-tetraen-3-one (2), ergosta-7,22-diene-3${\beta}$,5${\alpha}$,6${\alpha}$-triol (3), ergosta-7,22-diene-3${\beta}$,5${\alpha}$,6${\beta}$,9${\alpha}$-tetraol (4), and 5${\alpha}$,6${\alpha}$-epoxy-ergosta-8(14),22-diene-3${\beta}$,7${\alpha}$-diol (5) by comparing their physicochemical and spectral data with those in the literature. These compounds were evaluated for in vitro cytotoxicity against A549 and XF498 cancer cell lines. Most of the tested compounds, except for compound 3, exhibited moderate cytotoxicity against both A549 and XF498 cell lines with $IC_{50}$ values ranging from 10.2 to 18.3 ${\mu}g/ml$ and from 11.4 to 24.6 ${\mu}g/ml$, respectively.

  • A series of (-)-9-fluoro-2,3-dihydro-3(S)-methyl-10-(4-substituted-1-piperazinyl)-7-oxo-7H-pyrido [1,2,3-de] [1,4]-benzoxazine-6-carboxylic acid (5-8) was synthesized and evaluated for antibacterial activity against gram(+) and gram(-) bacteria. These synthesized compounds showed a lower activity than levofloxacin.

  • 5-Arylamino-6-chloro-4.7-dioxoindazoles (DZs) were newly synthesized for the evaluation of antifungal activities. The compounds DZs were prepared by regioselective nucleophilic substitution of 5.6-dichloro-4.7-dioxoindazoles with appropriate arylamines in high yield. DZs were tested for their growth inhibitory activities against Candida species and Aspergillus niger. The MIC values were determined by the two-fold dilution method. In general. DZs showed in vitro antifungal activities. Among the tested compounds, DZ1, 3, 6, 7 and 12 showed potent antifungal activities against Candida species and Aspergillus niger. DZ7 was the most effective in preventing the growth of Candida species.

Control Efficacy of Entomopathogenic Microagent against Spodoptera exigua on Organic Chinese Cabbage (유기재배 배추포장 내 파밤나방에 대한 곤충병원성 미생물의 방제 효과)

  • This study was carried out to identify the control effect of entomopathogenic microagent against Spodoptera exigua on organic chinese cabbage. In laboratory condition, insecticidal activity of 4 commercial BT pesticides against S. exigua were lower than 10% against second instar S. exigua. The insecticidal activity of entomopathogenic nematode were 33.3%, 83.3% and 100% at the concentration of $1{\times}10^2$, $3{\times}10^2$, $1{\times}10^3nematodes/ml$, respectively. Mixture of BT and nematode showed growth inhibition against S. exigua larvae. S. exigua nucleopolyhedrovirus (SeNPV) of $10^5PIBs/ml$ showed more than 70% insecticial activity. The yield of SeNPV was increased as in higher initial inoculation concentration of NPV, food supply, and growth temperature increased. In greenhouse experiment, the control value of BT and nematode mixture treatment was higher than BT and nematode treatment alone against S. exigua. In treatment of $10^7PIBs/ml$ of SeNPV, S. exigua was controlled completely. In farm condition, mixture of microbial agent and organic agricultural material showed higher control value against lepidopteran pest including S. exigua than BT single treatment.

Protective Effects of Red Ginseng Saponins against to 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) Induced Toxicity in Guinea Pigs (기니피그에서 홍삼 사포닌의 2,3,7,8-TCDD 독성 방어 효과)

  • This study was carried out to investigate the protective effect of Red Ginseng Saponins on 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) induced toxicities in guinea pigs ($200{\pm}10$ g). Normal control (NC) group guinea pigs ($200{\pm}10$ g) received vehicle and saline, while the TCDD-treated (TT) group was given water-extract (WE), saponin fraction (SF) and non-saponin fraction (NSF). Korean red ginseng fractions were administered from 1 week before TCDD-exposure for 4 weeks. Body weight loss and deteriorated clinical parameters related to sugar metabolism and liver function such as lipase and AST, respectively, these were significantly reduced by both saponin and non-saponin fractions. However, increase of lipase was attenuated by the saponin fraction in a dose-dependent manner. Only AST was affected by the saponin fraction. The results suggest that saponins are active substances in the Korean red ginseng water extract against TCDD induced toxicities in Guinea pigs.

Screening for in vitro Cytotoxic Activity of Seaweed, Sargassum sp. Against Hep-2 and MCF-7 Cancer Cell Lines

  • Discovery of anticancer drugs that kill or disable tumor cells in the presence of normal cells without undue toxicity is a potential challenge for therapeutic care. Several papers in the literature have emphasized the potential implications of marine products such as seaweeds which exhibit antitumor activity. Study attempts to screen the antitumor effect of Sargassum sp, against chosen cell lines such as MCF-7 (Breast cancer) and Hep-2 (Liver Cancer). Ethanol extract of Sargassum sp. was concentrated using a Soxhlet apparatus and dissolved in DMSO. In vitro cytotoxic activity of Sargassum sp at various concentrations ($100{\mu}g/ml-300{\mu}g/ml$) screened for antitumor effect against the chosen cell lines using MTT assay (3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide, a yellow tetrazole). The study documented that the percentage of cell viability has been reduced with increased concentration, as evidenced by cell death. Sargassum sp extract shows potential cytotoxic activity ($P{\leq}0.05$) with $IC_{50}$ of $200{\mu}g/ml$ and $250{\mu}g/ml$ against Hep-2 and MCF-7 cell lines respectively. The ethanol fraction of Sargassum sp induced cell shrinkage, cell membrane blebbing and formation of apoptotic bodies with evidence of bioactive components as profound influencing factors for anti-tumor effects. Further research need to be explored for the successful application of Sargassum sp as a potent therapeutic tool against cancer.

Study on the Anti-oxydative, Anti-tumor effect of Whakijogyungtang (화기조경탕(化氣調經湯)이 항산화 및 항암효과)

  • Objectives : This study was carried out to evaluate anti-oxydative, anti-tumor effect for clinical application of Whakijogyungtang (WJT) Results : 1. DPPH radical scavenging activities of WJT water extracts(Exts) were in proportion as concentration of WJT.(3 ${\mu}g/ml:12.6{\pm}2.3$ %) 2. ABTS+ scavenging activities of WJT water Exts were more effective in high density.(3 ${\mu}g/ml:4.3{\pm}1.6$ %, 10 ${\mu}g/ml$: $11.8{\pm}2.5$ %, 30 ${\mu}g/ml:45.3{\pm}3.2%$ 100 ${\mu}g/ml$: $62.7{\pm}4.8%$) 3. Hydrogen peroxide$(H_2O_2)$ scavenging activities of WJT water Exts were effective.(3 ${\mu}g/ml:4.7{\pm}0.8$ %, 10 ${\mu}g/ml: $8.2{\pm}1.6$ %, 30 ${\mu}g/ml:19.5{\pm}3.2$ % 100 ${\mu}g/ml$: $24.6{\pm}3.8$ %) 4. Anti oxidative effects against linoleic acid were not effective. 5. The generation of $O_2\;^-$ in S-180 cells were according to the concentration of WJT water Exts, specially effective over 100 ${\mu}g/ml$ concentration. 6. The SOD activities in S-180 cells were in proportion as cytotoxicity against S-180 cells of WJT water Exts. 7. The GPx activities in S-180 cells were in proportion as cytotoxicity against S-180 cells of WJT water Exts(more effective on 300 ${\mu}g/ml$ and 1000 ${\mu}g/ml$ concentration), but the catalase activities in S-180 cells were not effective. 8. The results of activites against multi-drug-resistance(MDR) of WJT were as follows. 1) In water Exts from WJT, cytotoxicity against AML-2/D100 with vincristine($IC_{50:}39.78$ ${\mu}g/ml$) was more effective than without vincristine($IC_{50:}$ 183.58 ${\mu}g/ml$), Cross resistance(CR:3.85) was not effective, and anti-MDR activites(RF) was effective.(RF:3.85) 2) In hexane fraction, cytotoxicity against AML-2/D100 with vincristine ($IC_{50:}130.88$ ${\mu}g/ml$) was more effective than without vincristine ($IC_{50:}293.10$ ${\mu}g/ml$) and anti-MDR activites(RF) was effective.(RF:4.61) 3) In chloroform fraction, the cytotoxicity against AML-2/WT and AML-2/D100 was not effective. 4) In ethyl acetate fraction, cytotoxicity against AML-2/D100 with vincristine($IC_{50:}36.43$ ${\mu}g/ml$) was more effective than without vincristine ($IC_{50:}73.07$ ${\mu}g/ml$), Cross resistance(CR:0.53) was not effective, and anti-MDR activites(RF) was effective.(RF:2) 5) In butanol fraction, the cytotoxicity against AML-2/WT and AML-2/D100 was not effective. 6) In $H_2O$ fraction, the cytotoxicity against AML-2/WT and AML-2/D100 was not effective. Conclusion : These result suggest that WJT has antioxidative effects, anti-tumor effects by apoptosis of free radical$(O_2\;^-)$ activity, and anti-MDR activites(especially hexane and ethyl acetate fraction).

In Vitro Antibacterial Effects of Tunong-san and Kwaruwoobang-tang Aqueous Extracts against Staphylococcus aureus (투농산(透膿散) 및 과루우방탕(瓜蔞牛蒡湯)의 Staphylococcus aureus에 대한 In Vitro 항균력 평가)

  • Objectives: Recently Ciprofloxacin, used in the treatment of mastitis, showed many serious side effects. The object of this study was to recognize whether TNS and KWT can be used in the treatment of mastitis by observing the in vitro antibacterial effects of TNS and KWT aqueous herbal extracts against S. aureus. Methods: Antibacterial activities of TNS and KWT aqueous extracts against S. aureus ATCC 25923 were detected using standard agar microdilution methods. In addition, the effects on the bacterial growth curve were monitored at MIC and $MIC{\times}2$ levels. The effects on the intracellular killing and bacterial invasion of individual test materials were also observed using Raw 264.6 and MCF-7. The results were compared with Ciprofloxacin, a second generation of quinolone antibiotics in the present study. Results: MIC of aqueous extracts of TNS and KWT against S. aureus were detected as ($0.313{\pm}0.107$) and ($0.137{\pm}0.053$) mg/ml, respectively. MIC of Ciprofloxacin was detected as ($0.469{\pm}0.297$) ${\mu}g/ml$ at same conditions. In addition, TNS, KWT aqueous herbal extracts and Ciprofloxacin were also showed marked dosage-dependent inhibition of bacterial growth, and dramatical inhibitions on the both intracellular killing assays and bacterial invasion using Raw 264.6 and MCF-7 cells were detected. Conclusions: The results obtained in this study suggest that TNS and KWT aqueous herbal extracts showed antibacterial effects against S. aureus, and they also showed dosage-dependent inhibitory effects on the bacterial growth. And they showed the significant intracellular killing and bacterial invasion effects. It means, KWT and TNS may show more potent anti-infectious effects against S. aureus in vivo.

Antimicrobial Activity of Garlic Juice against Escherichia coli O157:H7 (마늘즙의 Escherichia coli O157:H7에 대한 항균작용)

  • The antimicrobial activity of fresh garlic juice against Escherichia coli O157:H7 was investigated. When E. coli O157:H7 was cultured for 18 hr in the trypticase soy broth containing 1%, 3%, and 5% garlic juice, viable cell number of E. coli O157:H7 was reduced to $2.3{\times}10^2\;CFU/mL$ at 5% from $7{\times}10^8\;CFU/mL$ at the non-treated culture, respectively. The inhibitory effects of the ground beef treated with 3%, 6%, and 10% garlic juice against E. coli O157:H7 was significantly enhanced with approximate 2 log-reduction compared to that of ground beef without garlic. There was no significant difference in the inhibition of E. coli O157:H7 among the groups with different amounts of garlic juice (p<0.05). These results suggest that garlic juice may function well as a natural preservative in food system.

Production of monoclonal antibodies against marine birnavirus (Marine birnavirus (MABV)에 대한 단클론 항체 생산)

  • We developed and subsequently characterized mouse monoclonal antibodies (mAbs) against marine birnavirus (MABV). Eight hybridoma clones secreting mAbs against MABV were established. All eight mAbs (8G6, 11C3, 15E3, 17H6, 32A6, 35A7, 38B5, and 47E3) were reacted with viral protein 3 of MABV in MABV-infected CHSE-214, whereas, no reactivity was observed in normal CHSE-214 by western blot analysis. Moreover, these eight mAbs were strongly reacted with MABV, and no cross-reactivity has been observed against other five fish viruses (hirame rhabdovirus, infectious hematopoietic necrosis virus, nervous necrosis virus, spring viraemia of carp virus, and viral hemorrhagic septicemia virus), although five mAb (11C3, 15E3, 17H6, 32A6, and 38B5) reacted with both MABV and infectious pancreatic necrosis virus by enzyme linked immunosorbent assay (ELISA). These results indicate that the mAbs can be of value in MABV detection.
