전통 콩발효 식품 유래 7, 8, 4'-Trihydroxyisoflavone의 피부 생리활성 연구 (7, 8, 4'-Trihydroxyisoflavone from Fermented Soybean Food and Its Biological Activity)

  • 10년 이상 자연 발효된 콩 발효 식품에서 희귀 이소플라본 성분인 7, 8, 4'-trihydroxyisoflavone을 분리하였고, 분리된 성분의 항산화 효과를 비효소적인 방법으로는 DPPH 법을, 효소적인 방법으로는 xanthine oxidase법을 이용하여 확인하였다. 그 결과 7, 8, 4'-trihydroxyisoflavone는 농도 의존적으로 비타민 C와 유사한 DPPH 라디칼 소거 효능을 가지고 있음을 보여주었고 $6.6{\pm}0.4\;{\mu}M$ 농도에서 xanthine oxidase에서 생성된 superoxide 라디칼을 50 % 수준까지 낮춤을 보여주었다. 또한, 이 희귀 이소플라본 성분은 UVB로 유도된 MMP-1의 생성을 초기 수준으로 억제하는 것으로 나타나 피부 노화 억제에 유용한 성분임을 확인할 수 있었다.

  • 된장은 한국의 독특한 발효 식품이며 장양이라 불리는 전통적인 제조 방법에 의해 제조된다. 이에 본 연구에서는 고숙성 된장에서 새롭게 생성된 성분과 그것의 생리활성을 측정하였다. 5년 숙성된 된장으로부터 새로운 o-dihydroxyisoflavone인 7,3',4'-trihydroxyisoflavone과 기존에 알려진 두 가지의 o-dihydroxyisoflavone을 각각 분리하여 다른 isoflavone과의 항산화 효과 및 미백효과를 비교 측정하였다. Diphenyl-1-picrylhydrazyl (DPPH) 활성 저해 효과는 7,8,4'-trihydroxyisoflavone (compound 1), 7,3',4'-trihydroxyisoflavone (compound 2) 그리고 6,7,4'-trihydroxyisoflavone (compopund 3)은 각각 $21.5{\pm}0.2$, $28.7{\pm}0.4$ 그리고 $32.6{\pm}0.6$ ($IC_{50}$)을 나타낸 반면 daidzein은 이들에 비해 약한 DPPH 활성 저해 효과를 나타냈다. Superoxide 소거 효과는 L-ascorbic acid에 비해 compound 1 ($IC_{50}=18.10{\pm}0.2{\mu}M$)과 2 ($IC_{50}=10.54{\pm}0.4{\mu}M$)가 보다 효과적인 반면 compound 3과 daidzein은 이들에 비해 낮은 활성을 나타냈다. 또한 melan-a cells에서 o-dihydroxyisoflavone 유도체들의 티로시나제 활성과 멜라닌 생성을 비교 측정하였다. 특히 Compound 1 ($IC_{50}=11.21{\pm}0.2{\mu}M$), 2 ($IC_{50}=5.23{\pm}0.6{\mu}M$)의 경우 compound 3과 daidzein에 비해 티로시나제 활성을 억제하는데 효과가 있었으며 또한 멜라닌 생성을 50 % 억제할 때의 결과에서 이들 화합물은 각각 $12.23{\pm}0.7{\mu}M$ (1), $7.83{\pm}0.7{\mu}M$ (2)과 같이 멜라닌 생성 억제 효능을 나타내었다. 따라서 고숙성 된장에서 유래한 7,3',4'-trihydroxyisoflavone은 항산화 및 미백효과를 가지는 화장품 소재로써 사용 가능하리라 판단된다.

7,8,4'-Trihydroxyisoflavone, a Metabolized Product of Daidzein, Attenuates 6-Hydroxydopamine-Induced Neurotoxicity in SH-SY5Y Cells

  • Daidzein isolated from soybean (Glycine max) has been widely studied for its antioxidant and anti-inflammatory activities. However, the protective effects of 7,8,4'-trihydroxyisoflavone (THIF), a major metabolite of daidzein, on 6-hydroxydopamine (OHDA)-induced neurotoxicity are not well understood. In the current study, 7,8,4'-THIF significantly inhibited neuronal cell death and lactate dehydrogenase (LDH) release induced by 6-OHDA in SH-SY5Y cells, which were used as an in vitro model of Parkinson's disease (PD). Moreover, pretreatment with 7,8,4'-THIF significantly increased the levels of superoxide dismutase (SOD), catalase (CAT), and glutathione (GSH) and decreased malondialdehyde (MDA) activity in 6-OHDA-induced SH-SY5Y cells. In addition, 7,8,4'-THIF significantly recovered 6-OHDA-induced cleaved caspase-3, cleaved caspase-9, cleaved poly-ADP-ribose polymerase (PARP), increased Bax, and decreased Bcl-2 levels. Additionally, 7,8,4'-THIF significantly restored the expression levels of phosphorylated c-Jun N-terminal kinase (JNK), p38 mitogen-activated protein kinase (MAPK), extracellular signal-regulated kinase 1/2 (ERK 1/2), phosphatidylinositol 3-kinases (PI3K)/Akt, and glycogen synthase kinase-3 beta ($GSK-3{\beta}$) in 6-OHDA-induced SH-SY5Y cells. Further, 7,8,4'-THIF significantly increased the reduced tyrosine hydroxylase (TH) level induced by 6-OHDA in SH-SY5Y cells. Collectively, these results suggest that 7,8,4'-THIF protects against 6-OHDA-induced neuronal cell death in cellular PD models. Also, these effects are mediated partly by inhibiting activation of the MAPK and PI3K/Akt/$GSK-3{\beta}$ pathways.

Phytochemical Constituents from the Leaves of Soybean [Glycine max (L.) Merr.]

  • Phytochemicals study from the leaves of soybean [Glycine max (L.) Merr.], one of Korean edible plant materials were investigated through various chromatographic procedures. The methanolic leaves extracts of soybean yielded 16 phytochemicals, including 5 isoflavones 1-5, 3 flavones 6-8, 1 flavonol 9, 2 pterocarpans 10 and 11, 2 phenolic compounds 12 and 13, 2 phytosterols 14 and 15, and 1 sugar alcohol 16. The structures were fully characterized by analysis of physical and spectral data and were defined clearly as 4,5,7-trihydroxyisoflavone (1), 4,5,7-trihydroxyisoflavone-7-O-$\beta$-D-glucopyranoside (2), 4,7-dihydroxy-6-methoxyisoflavone (3), 4,7-dihydroxyisoflavone (4), 4,7-dihydroxyisoflavone-7-O-$\beta$-D-glucopyranoside (5), 5,7,4'-trihydroxyflavone (6), 3',4',5,7-tetrahydroxyflavone (7), 3',4',5-trihydroxyflavone-7-O-$\beta$-D-glucopyranoside (8), 3,4',5,7-tetrahydroxyflavonol (9), coumestrol (10), glyceofuran (11), 4-hydroxybenzoic acid (12), methyl-4-hydroxybenzoate (13), soyasapogenol B (14), stigmasterol (15), and D-mannitol (16), respectively. Among them, phytochemicals 7-16 were reported for the first time on the isolation and confirmation from the leaves of this species. These results suggest that the leaves extracts of soybean may possess possible health related benefits to human due to the isolated phytochemicals 1-16 which have been well known potential effects on various chronic diseases.
