pH Affects the In vitro Formation of cis-9, trans-11 CLA and trans-11 Octadecenoic Acid by Ruminal Bacteria When Incubated with Oilseeds

  • The effect of pH on the fermentation characteristics and the formation of cis-9, trans-11 conjugated linoleic acid (CLA) and trans-11 octadecenoic acid by mixed ruminal bacteria was examined in vitro when incubated with linseed or rapeseed. Concentrate (1%, w/v) with ground linseed (0.6%, w/v) or rapeseed (0.5%, w/v) was added to 600 ml mixed solution of strained rumen fluid with artificial saliva (1:1, v/v), and was incubated anaerobically for 12 h at $39^{\circ}C$. The pH of culture solution was maintained at level close to 4.5, 5.3, 6.1 and 6.9 with 30% $H_2SO_4$ or 30% NaOH solution. pH increment resulted in increases of ammonia and total volatile fatty acid (VFA) concentration in culture solutions containing both oilseeds. Fermentation did not proceeded at pH 4.5. Molar proportion of acetate decreased but that of propionate increased as pH increased when incubated with oilseeds. While the hydrogenating process was very slow at the pH range of 4.5 to 5.3, rapid hydrogenation was found from the culture solutions of pH 6.1 and 6.9 when incubated with linseed or rapeseed. As pH in culture solution of linseed or rapeseed increases proportions of oleic acid (cis-9 $C_{18:1}$) and trans-11 octadecenoic acid increased but those of linoleic acid and linolenic acid decreased. The CLA proportion increased with pH in culture solution containing rapeseed but CLA was mostly not detected from the incubation of linseed.

The Processing Conditions of Extracts from Rapid Fermented Anchovy Sauce (속성멸치간장엑기스분의 가공조건)

  • AS a part of investigation to use anchovy more effectively as food source, this work was undertaken the processing conditions of extracts from rapid fermented anchovy sauce. To prepare rapid fermented products, the chopped anchovy was mixed with 50% water (w/w), and then hydrolyzed by autolysis or addition of koji under different conditions of hydrolysis. The optimal conditions for hydrolysis of chopped anchovy were revealed $50^{\circ}C$, 6 hours, pH 8.0 by autolysis, and ,$50^{\circ}C$, 6 hours, pH 7.0 by addition of 10% koji, respectively. But, pH control was not much beneficial in increasing yield. The effect of soybean protein isolote for improvement of bitter taste was also tested. The reasonable amount of added soybean protein isolate was revealed 5% on the weight basis of the chopped anchovy. The reaction mixture hydrolyzed under optimal conditions were added with 1% onion powder (w/w), 1% garlic powder(w/w) and 1% red pepper powder(w/w) for masking fishy odor, inactivated for 20 min at $100^{\circ}C$, and then centrifuged for 20 min at 4,000 rpm. The supernatant liquor was filtrated and evaporated to 50%(v/v). finally, table salt was added for bateriostatic effect and characteristic taste of rapid fermented products. the reasonable amount of added table salt was reversed 15% on the volume basis of the evaporated liquor. The hydrolysis ratio of product made by addition of water, product made by addition of koji and water, and product made by addition of soybean protein isolate, koji and water hydrolyzed under optimal conditions were 58.4%, 82.1% and 86.2%, respectively.

  • In this study investigations have been carried out for the evaluation of small DMFCS under passive operation conditions for use in portable powers. Under passive conditions, a maximum performance was obtained at a methanol concentration of 4 M and at a catalyst loading of $8mg/cm^2$ on both electrodes. By optimizing various parameters, we could achieve the highest performance of $55mW/cm^2$ at 1 attn and at R.T.A monopolar stack consisting of 6 unit cells with active area of $4.5cm^2/cell$ was prepared and it showed a uniform voltage distribution all over the cells and it had a power output of 1 watt and a power density of $37mW/cm^2$ A monopolar stack which consisted of 16 cells and produced a 2.4W power was also fabricated and was tested for operation of a miniature car.

Effect of Administration Method on Blood Garcinia cambogia Concentration and Antioxidative Activity (투여 방법에 따른 가르시니아 캄보지아 혈중 농도 변화와 항산화 효과 변화)

  • The objective of this study was to investigate the effects of administration methods for on Garcinia cambogia on blood Garcinia cambogia concentration and antioxidative levels. Rats were divided into three groups: G1 (normal group), G2 (one administration of Garcinia cambogia extract 2,800 mg/kg b.w.), G3 (three separate administrations every 6 h of Garcinia cambogia extract 750, 850, and 1,200 mg/kg b.w. for 18 h). Blood samples were collected every hour, and animals sacrificed 18 h after the oral administration of Garcinia cambogia extract. We examined changes in the serum concentration of Garcinia cambogia by HPLC analysis. Two hours following an oral administration of Garcinia cambogia extract (2,800 mg/kg b.w.), serum Garcinia cambogia levels reached their maximum, but gradually decreased until 10 hours when it was no longer detectable. In contrast, serum Garcinia cambogia levels under G3 administration were maintained above a certain level after 18 h. To determine whether this level of Garcinia cambogia could affect blood oxidative levels, we measured serum lipid peroxidation by TBARS levels. TBARS levels from G3 treatment were significantly lower than G1 and G2. To analyze other antioxidative activities, radical scavenging activities were measured by the DPPH and ABTS radical scavenging assays. There were no significant differences between the groups in DPPH radical scavenging activity. However, ABTS radical scavenging activity significantly increased with G3 treatment compared with G1 and G2. In conclusion, our data show that three times administration of Garcinia cambogia every 6 h may helpful for maintaining serum Garcinia cambogia levels and antioxidative effects.

Solvent Effects on the Isotropic NMR Shifts in Quinuclidine and Pyridine-Type Ligands Coordinated to the Paramagnetic Polyomometalate, $[SiW_{11}Co^{II}o_{39}]^{6-}$

  • The solvent effects on the isotropic NMR shifts in conformationally rigid ligands such as quinuclidine, pyridine, and 4-aminopyridine coordinated to the paramagnetic polyoxometalate, [SiW11CoⅡO39]6- (SiW11Co), are reported. For these complexes the ligand exchange is slow on the NMR time scale and pure 1H NMR signals have been observed at room temperature. The signals for the SiW11Co complexes are shifted upfield whe dimethyl sulfoxide-d6 (DMSO) is added to a D2O solution. The isotropic shifts are separated into contact and pseudocontact contributions by assuming that the contact shifts are proportional to the isotropic shifts of the same ligands coordinated to [SiW11NiⅡO39]6-. It is shown that both the contact and pseudocontact shifts decrease (the absolute values of the pseudocontact shifts increase), when D2O is replaced by DMSO. It is suggested that D2O, a strong hydrogen bond donor, withdraws electron density from [SiW11CoⅡO39]6-, increasing the acidity of the cobalt ion toward the axial ligand. When D2O is replaced by DMSO, the acidity of the cobalt ion in SiW11Co decreases, weakening the Co-N bond. Then both the contact and pseudocontact shifts are expected to decrease in agreement with the observed solvent effects.

Concept Design of a H.A.U.'s Subsonic Wind Tunnel (H대학교 아음속 풍동 개념설계)

  • A closed-circuit type wind tunnel is designed, which has a test section with the dimensions $1.2(W){\times}1.2(H){\times}3.4(L)$. A subsonic wind tunnel is designed to improves educational circumstances and promote ground tests. It is constituted of an exchangeable test section, first and second diffusers, a fan, a settling chamber, a contraction, and 4 corners. The maximum velocity in the test section is 70m/s and the contraction ratio is 6.25:1. Input power in the wind tunnel is about 96.1 kw (128.8 hp) and its energy ratio is 3.89. It has the dimension of about $7.4(W){\times}3.6(H){\times}21.7m(L)$. The wind tunnel designed in this investigation will be an effective educational and investigational equipment.

Oligosaccharide Production by Leuconostoc lactis CCK940 Which Has Glucansucrase Activity (Leuconostoc lactis CCK940의 Glucansucrase 활성에 의한 올리고당 생산 최적화)

  • Glucansucrase is an enzyme classified as a glycoside hydrolase (GH) 70 family, which catalyzes the synthesis of glucooligosaccharides with a low molecular weight using sucrose as a donor of D-glucopyranose and maltose as a carbohydrate acceptor. In this study, glucansucrase-producing lactic acid bacteria strain was isolated from the fermented foods collected in traditional markets, and the optimum conditions for the oligosaccharide production were investigated. The strain CCK940 isolated from Chinese cabbage kimchi was selected as an oligosaccharide-producing strain due to its high glucansucrase activity, with 918.2 mU/mL, and identified as Leuconostoc lactis. The optimum conditions for the production of oligosaccharides using Leu. lactis CCK940 were to adjust the initial pH to 6.0, add 5% (w/v) sucrose and 10% (w/v) maltose as a donor and acceptor molecules, respectively, and feed 5% (w/v) sucrose at 4 and 8 h of cultivation. When Leu. lactis CCK940 was cultured for 12 h at optimum conditions, at least four oligosaccharides with a polymerization degree of 2-4 were produced.

Improvement of the adhesion and resistivity of low-pressure chemical vapor deposited tungsten films by controlling deposition parameters (LPCVD로 증착한 텅스텐 박막의 증착 조건 제어에 의한 접착성 및 저항 특성 향상)

  • Tungsten(W) thin films with good adhesion property and low resistivity (~10 $\mu$\Omega$$.cm) were deposited directly on $SiO_2$ by LPCVD. The adhesion property of W thin films on $SiO_2$ improves as the temperature and/or $SiH_4/WF_6$ gas ratio increase. Specifically tungsten thin films could be deposited on $SiO_2$ even at $350^{\circ}C$ if the gas ratio of 2 was employed. The resistivity of tungsten thin films deposited at $350^{\circ}C$ was high due to the presence of $\beta$-W. However, the resistivity can be minimized by increasing the amount of $H_2$ gas flow. Therefore, it is shown in this work that the adhesion of tungsten thin films on $SiO_2$ can be improved simply by controlling the process parameters (e.g., temperature, gas ratio and $H_2$ flow rate) without employing complex deposition methods or additional glue layers.

Encapsulation of Bromelain in Liposome

  • Bromelain has been used as a meat-tenderizing agent in food processing. To increase the availability of bromelain, microencapsulation into liposome was carried out by the dehydration and rehydration method. Small unilamellar vesicles prepared by sonication treatment showed higher encapsulation efficiency (EE) than by the French press method. In the preparation of liposome, the effect of pH and centrifugal force on EE was also investigated and it showed a higher EE at acidic pH than at alkaline pH with centrifugation at 80, 000$\times$g. The actual EEs except unencapsulated bromelain which bound on the outside surface of liposome by electrostatic interaction also were investigated, and the optimal EE was at pH 4.6, at 0.6 of a ratio of bromelain to phosholipid(18.2%, w/w). Release of bromelain from liposomes was stimulated as the temperature increased at neutral pH.

Comparison of Characteristics of Acid-catalyzed Hydrothermal Fractionation for Production of Hemicellulose Hydrolyzate from Agricultural Residues (농경잔류물로부터 헤미셀룰로오스 가수분해물 생산을 위한 산촉매 열수 분별공정의 특성 비교)

  • The objective of this work was to investigate the feasibility of acid-catalyzed hydrothermal fractionation for maximum solubilization of the hemicellulosic portion of two typical agricultural residues. The fractionation conditions converted into combined reaction severity (CS) in the range of 1.2-2.9 was used to establish a simple reaction criteria at glance. The hemicellulosic sugar yield of 56.6% was shown when rice straw was fractionated at the conditions at the conditions; 160 ℃ of temperature 0.75% (w/v) of H2SO4, 20 min of reaction time, 1:15 solid/liquid ratio. The hemicellulosic sugar yield of 83.0%, however, was achieved when barley straw was fractionated at the conditions at the conditions; 150 ℃ of temperature 0.75% (w/v) of H2SO4, and 15 min of reaction time, 1:10 solid/liquid ratio. For barley straw, acid-catalyzed hydrothermal fractionation could be effectively performed. After the fractionation process, the remaining fractionated solids were 48.5% and 57.5% from raw rice and barley straws, respectively. The XMG contents in the solid residues decreased from 17.3% and 17.6% to 6.0% and 2.6%, which corresponded to 16.7% and 8.5% on the basis of the raw straws, respectively. In another way, only 5.6% of cellulose and 8.5% of XMG were lost due to excessive decomposition during the acid-catalyzed hydrothermal fractionation of barley straw, compared to cellulose and XMG losses of 6.4% and 26.6% in rice straw. Hemicellulosic sugars from the rice straw were considered more over-decomposed due to the somewhat higher reaction severity at the acid-catalyzed hydrothermal fractionation.
