강원도 유통 발효주 중 바이오제닉 아민류 함량 조사 및 위해도 평가 (Monitoring of Biogenic Amines Content in Commercial Fermented Alcoholic Beverages in Gangwon-do and Risk Assessment)

  • Objectives: This study was conducted to estimate the related health risk by investigating the content of biogenic amines (BAs) in commercial fermented alcoholic beverages. Methods: Ninety-seven samples, including takjus (34), fruit wines (26), distilled liquors (20) and beers (17) were purchased in Gangwon-do (Province) in 2019. The eight BAs (putrescine, cadaverine, spermine, spermidine, tyramine, β-phenylethylamine, histamine, and tryptamine) were analyzed by high performance liquid chromatograph (HPLC) after extraction (0.4 M HClO4), pigment removal (carbon cartridge) and derivatization (1% dansylchloride). The health risk of exposure to BAs due to ingestion of commercial fermented alcoholic beverages was estimated as the margin of exposure (MOE). Results: Various BAs were detected in takju, fruit wine, distilled liquor and beer. Putrescine (not detected (ND)-12.60 mg/L), cadaverine (ND-5.45 mg/L), tyramine (ND-8.34 mg/L), β-phenylethylamine (ND-13.73 mg/L), histamine (ND-7.96 mg/L), and tryptamine (ND-19.00 mg/L) were found in takju. Putrescine (ND-15.34 mg/L), cadaverine (ND-7.90 mg/L), β-phenylethylamine (ND-3.06 mg/L), histamine (ND-9.68 mg/L), and tryptamine (ND-14.06 mg/L) were identified in fruit wine. 태음조위탕(太陰調胃湯)의 유탕억제효능(遺瘍抑制效能)에 관(關)한 연구(硏究) (A study on the ulcerous inhibitory effects of Taeumchowetang)

  • The experimental study used in this paper was done to examine the clinical effects of Taeumchowetang for the gastric ulcer and the function of gastrointestinal tract using rats and mice which were administered orally the water extraction from Taeumchowetang. Then, the counter-action of the water extraction on the isolated ileum and gastric fundus, the inhibitory effects of pylorus-ligated ulcer and indomethacin-induced ulcer, the associations with gastric juice secretion, total acidity, pepsin output, the transportability in the small and large intestine, were studied with administering acetylcholine chloride and barium chloride. In addition, it was investigated whether the central nervous system related to pain control and sleeping time was influenced by Taeumchowetang. The following results have been obtained; 1. As resulting from injection of acetylcholine chloride and barium chloride into the isolated ileum of rats and mice, Taeumchowetang led to have an inhibitory effect on the muscle contraction of the ileum. Then, acetylcholine chloride was measured as lower effect than barium chloride 2. For the inhibitory effect on contraction for the gastric fundus strip in rats by either acetylcholine chloride or barium chloride, the one showed low inhibitory effect, on the other hand the other showed density-dependent effect. 3. The water extraction was given on the pylorus ligated ulcer with using two different administration groups of 1,300mg/kg and 2,600mg/kg, each result was measured as 22.9% and 36.5% for an ulcerous inhibitory effect (p<0.05). 4. According to the two administration groups, the preventive effect was tabulated 18.1% and 29.3% for indomethacin-induced ulcer (p<0.05, P<0.01). 5. For associations with gastric juice secretion, total acidity, pepsin output in the administration group 2,600mg/kg, Taeumchowetang was recognized as having an inhibitory effects related to suppressive actions involving gastric juice secretion(p<0.05), and free acidity(p<0.01), but there was no significant association with total acidity and pepsin output. 6. To know the transportability in the intestine, BaSO4 solution was used. The transportability of the small intestine in the administration group 2,600mg/kg was 22.2% which was statistically significant compared with the large intestine's transportability(P<0.01). 7. In the administration group 1,300mg/kg and 2,600mg/kg, analgesic effect with against acetic acid was measured as being 16.8% and 24.4% which was shown as statistically siginificant(p<0.01). 8. No statistically significant association between Taeumchowetang and sleeping time was found in both 1,300mg/kg and 2,600mg/kg by administering phenobarbital-Na. According to the results of this study, Taeumchowetang has agreed with the effects of literature review. Further more in this study, Taeumchowetang also has preventive effects on pylorus-ligated ulcer. Hence, Teaumchowetang can be significant effect such as both anti ulcer agent and increasing gastric activity for the patients who suffer from gastric ulcer.

출하용 숙주나물의 세척 유무에 따른 형태 및 색상 변화 (Washing Effect of Marketing Mungbean Sprouts on Morphological Characters and their Color)

  • 전래적으로 숙주나물은 저장성을 높이기 위하여 세척하지 않고 탈수시켜 이용하여 왔다. 본 연구는 생산된 숙주나물의 수분 함량과 관련된 세척이 제품의 저장성에 미치는 영향을 파악하기 위하여 포장하기 직전 물에 씻거나 씻지 않는 세척 유무가 5일간 저장중 숙주나물의 형태 및 무게에 미치는 영향을 조사한 결과를 요약하면 다음과 같다. 1. 무세척 숙주나물에 비하여 세척시킨 숙주나물은 저장 4일 후까지 수분 함량이 높은 것으로 나타났다. 2. 무세척 숙주나물은 세척시킨 숙주나물에 비하여 저장중 형성된 개체당 세근수가 많았던 반면, 하배축과 뿌리 길이, 하배축 중간과 hook 부분의 직경, 각부위 및 전체 생체중과 건물중에서는 세척 유무간에 차이가 없었다. 3. 무세척 숙주나물은 세척시킨 숙주나물에 비하여 하배축 및 뿌리의 탁도, 하배축의 전단력이 높았던 반면, 색도 a와 b에서 세척 유무간 차이가 없어 저장중 무세척 숙주나물의 품질 저하가 적은 것으로 나타났다. 4. 무세척 숙주나물에서 세근수, 하배축 길이, 하배축 중간부분의 직경, 하배축과 전체 생체중 및 건물중은 저장 후 3일까지 변화가 없었으나 그 이후 감소하는 경향을 보였다. 5. 무세척 숙주나물에서 하배축의 전단력은 저장 4일 후부터, 하배축의 명도와 색도 b는 저장기간이 길어질수록 감소하는 경향을 보였다.

광중합 복합레진의 화학적 분해 평가 (AN EVALUATION OF CHEMICAL DEGRADATION OF LIGHT-CURED RESTORATIVE COMPOSITES)

  • 본 연구에서는 4종의 복합레진(Palpique Estelite, Filtek P 60, Spectrum, Charisma)을 선택하였다. 각 제품당 5개의 시편을 제작하여 무게 측정을 한 후, NaOH 용액에 2주 동안 보관하였고 1.23% HCl로 중화하고 세척, 건조시켜 무게 측정을 하였다. 주사전자현미경과 공초점 레이저 주사현미경으로 복합레진의 분해층 표면과 분해깊이를 관찰하였고, 분해 저항성은 용액내로 용출된 Si, Al, Ba의 농도를 근거로 평가하여 다음과 같은 결과를 얻었다. 1. 각 제품의 무게손실량은 Palpique Estelite가 가장 많았고, Filtek P 60, Charisma, Spectrum 순이었다. 2. 각 제품의 표면하 분해층 깊이는 Filtek P 60이 가장 깊었고, Charisma, Palpique Estelite, Spectrum 순이었다. 3. Si 용출량은 Charisma가 가장 많았고, Spectrum, Palpique Estelite, Filtek P 60 순이었다. 4. 무게손실과 분해층 깊이 사이에는 높은 상관관계를 보였다(r=0.704, p<0.05). 5. 주사전자현미경 관찰시 NaOH용액에 보관한 후 레진 기질과 충전제 사이의 결합의 파괴 양상인 분해층 표면을 관찰할 수 있었고 공초점 레이저 현미경 관찰시 레진의 분해층 깊이를 관찰할 수 있었다.

가토의 대퇴부에 Calcium Metaphosphate로 코팅된 임플란트 매식후 골유착에 관한 비교 연구 (Comparative Study on Osseointegration of Calcium Metaphosphate (CMP) Coated Implant to RBM Implant in the Femur of Rabbits)

  • Purpose: This study was conducted in order to compare the efficacy of osseointegration of three different calcium metaphosphate (CMP) coated implants in the rabbit's femur. Materials and Methods: Twenty four rabbits and three different type of CMP coated implants and RBM implants were used in this study. The animals were divided into 4 groups on the basis of implant surface characteristics. Two implants were installed into the condyle of femur of each rabbits. The animals were sacrificed at 2 and 4 weeks after installation. The undecalcified specimens were prepared for histological, radiological examination and histomorphometric analysis of implant-bone contact ratios (BIC) and bone area ratio (BA). Results: Two implants were failed to osseointegrate and implant success rate was 95.2%. There were not any significant inflammatory response in all groups. Fluorescent image at 4 weeks shows that remodeling is slower in RBM group than CMP group. CMP III showed more active remodeling than CMP I, II. In histomorphologic analysis, BIC ratio at 2 weeks was lower than 4 weeks. Conclusion: The results suggest that the ratios of CMP coated implants were higher than that of RBM control group but there is no significantly difference between RBM group and CMP group. In conclusion, CMP coated implant had more clinical availability than RBM implants.

옥방(玉房) 중석광상(重石鑛床)의 성인(成因)에 관(關)한 연구(硏究) -특(特)히 남부광체(南部鑛體)에 대(對)하여- (On the Genesis of Okbang Tungsten Deposits)

  • The Nambu orebodies of the Okbang tungsten mine are hosted in the Precambrian amphibolite and Weonnam formation. These orebodies can be classified into two types; The scheelite-bearing ore vein occurring in the amphibolite (the Nambu 1, 2 adits) and tungsten-bearing quartz vein along the contact between the amphibolite and the Weonnam formation (the Young-ho, -1, -2, -3 levels). The scheelite-bearing ore vein in the amphilbolite is discontinuous, narrow, and highly irregular in geometry, occurring only within the amphibolite with which of the vein is graduational. Based on these feature of the mode of occurrence, the origin of this ore type might be attributed to a potential segregation of tungsten ore fluid in situ from hornblenditic basic magma of the host rock. Tungsten-bearing quartz vein, however, is considered to have deposited along the N30-60E trending fractures as a later hypothermal vein after the hornblendite was emplaced. The principal ore mineral is scheelite with minor amount of wolframite, and the gangue minerals are quartz, and small amounts of fluorite, pyrrhotite, chalcopyrite and calcite. Fluid inclusion study of minerals from the Nambu orebody reveals that the fluids in fluorite of the scheelite-bearning ore vein attained a temperature range of $208{\sim}256^{\circ}C$ and those in quartz from the tungsten-bearing quartz vein a temperature range of $220{\sim}357^{\circ}C$. The real formation temperatures can be somewhat higher than filling temperatures, if pressure correction is made. Chemical analysis of 8 amphibolitc samples on major and some trace elements indicate that the amphibolite is igneous origin. On a Niggli diagram (al-alk)versus c, the analytical values are plotted on an igneous field, and on a Niggli diagram mg versus c they follow a karroo igneous trend line. According to the Ba, Cr, and Ni versus Niggli mg plots suggested by Leake (1964), Okbang amphibolite fall outside a pelitic field and compare favorably with his plots form ortho-amphibolites. Analitical values of $MoO_3$ of 8 samples of scheelite minerals from the Nambu orebody indicate that the tungsten-bearing quartz vein (type n) of Nambu orebody shows a range from 1. 69% to 4.38% which is higher than 0.94%~3.25% $MoO_3$ for the scheelite-bearing ore vein (type I). This fact indicates that the type II was deposited in a lower $fO_2/higher$ $fO_2$ environment and under lower temperature than the type I. Analysis of major components $WO_3$, MnO, and FeO of 6 samples of wolframite from the type II veins revealed that they contain 73.35~76.2% $WO_3$, 7.94~11.63% MnO, and 10.53~14.82% FeO. MnO/FeO ratios of wolframite shows the range of 0.85~1.17 which suggests a slightly higher temperature type of deposits than other major tungsten deposits in the country.

LED가 고구마 바이러스 무병묘의 기내 생장에 미치는 영향 (Effects of Light-emitting Diodes on In Vitro Growth of Virus-free Sweet Potato Plantlets)

  • 고구마 바이러스 무병주의 기내 소식물체 생장에 미치는 LED의 영향을 조사하기 위하여, '맛나미', '신천미', '연황미'의 소식물체에서 10mm 크기의 마디를 잘라 $0.2mg{\cdot}L^{-1}$ BA를 첨가한 MS배지에 배양하였다. 광질은 형광등을 대조구로 하여, 적색(660nm), 청색(460nm), 백색, 적:청(8:2), 적:청(7:3) LED를 20cm 거리에서 $150{\pm}5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ 광합성유효광량자속(PPF)을 유지하였으며, 일장은 16/8(day/night)시간, 배양온도는 $25^{\circ}C$로 조절하였다. LED 광질에 대한 품종간 반응은 적색 LED에서 같은 방법으로 10품종의 마디배양을 하여 소식물체 생장을 조사하였다. 배양 3주 후에 줄기신장은 적색 LED에서 형광등보다 유의하게 증가하였으며, 청색광에서 가장 낮았다. 줄기두께는 적:청(7:3)과 백색 LED에서 증가하였다. 생체중 및 건물중도 적색광에서 가장 높았다. 또한 적색 LED에서 신초생육은 유의한 품종간 차이를 보였다. 줄기길이, 생체중 및 건물중 등은 '신천미', '맛나미', '신황미' 등에서 컸다. 따라서 LED 광원을 이용한 고구마 마디배양은 적색 LED에서 3주간 배양하여 5cm 이상 키운 소식물체를 토양이식하는 것이 대조구인 형광등보다 배양기간을 1주 이상 단축할 수 있었다.

Quality Characteristics of Functional Fermented Sausages Added with Encapsulated Probiotic Bifidobacterium longum KACC 91563

  • The present study aimed at evaluating the utilization possibility of encapsulated probiotic Bifidobacterium longum for production of functional fermented sausages. The B. longum isolated from the feces samples of healthy Korean infants encapsulated with glycerol as a cryprotectant was used for fermented sausages production as a functional bacterial ingredient, and its effect was also compared with those inoculated with commercial starter culture (CSC). Results showed that most inoculated encapsulated B. longum (initial count, 5.88 Log CFU/g) could survive after 4 days fermentation (5.40 Log CFU/g), and approximately a half (2.83 Log CFU/g) of them survived in the products after 22 days of ripening. The products inoculated with encapsulated B. longum presented the lowest lipid oxidation level, while had higher total unsaturated fatty acid content and more desirable n-6/n-3 fatty acids than those inoculated with CSC or non-inoculated control. Moreover, the odor and taste scores in the samples made with B. longum were comparable to those in the treatment with CSC. The inoculation with the B. longum had no effects on the biogenic amine contents as well as did not cause defects in color or texture of the final products. Thus, the encapsulation could preserve the probiotic B. longum in the meat mixture, and the encapsulated B. longum could be used as a functional ingredient for production of healthier fermented meat products.

Calcium Current in the Unfertilized Egg of the Hamster

  • The presence of a calcium current $(i_{Ca^{2+}})$ passed via a specific channel was examined in the unfertilized hamster egg using the whole-cell voltage clamp technique. Pure inward current was isolated using a $Ca^{2+}-rich$ pipette solution containing 10 mM TEA. This current was independent of external $Na^+$ and was highly sensitive to the $Ca^{2+}$ concentration in the bathing solution, indicating that the inward current is carried by $Ca^{2+}$. The maximal amplitude was $-4.12{\pm}0.58nA\;(n=12)$ with 10mM $Ca^{2+}$ at -3OmV from a holding potential of -8OmV. This current reached its maximum within 20ms beyond -3OmV and decayed rapidly with an inactivation time constant $({\tau})$ of 15ms. Activation and inactivation of this $i_{Ca^{2+}}$ was steeply dependent on the membrane potential. The $i_{Ca^{2+}}$ began to activate at the lower voltage of -55 mV and reached its peak at -35 mV, being completely inactivated at potentials more positive than -40 mV. These result suggest that $i_{Ca^{2+}}$ in hamster eggs passes through channels with electrical properties similar to low voltage-activated T-type channels. Other results from the present study support this suggestion; First, the inhibitory effect of $Ni^{2+}\;(IC_{50}=13.7\;{\mu}M)$ was more potent than $Cd^{2+}\;(IC_{50}=123\;{\mu}M)$. Second, $Ba^{2+}$ conductance was equal to or below that of $Ca^{2+}$. Third, $i_{Ca^{2+}}$ in hamster eggs was relatively insensitive to nifedipine $(IC_{50}=96.6\;{\mu}M)$, known to be a specific t-type blocker. The physiological role of $i_{Ca^{2+}}$ in the unfertilized hamster eggs remains unclear. Analysis from steady-state inactivation activation curves reveals that only a small amount of this current will pass in the voltage range $(-70{\sim}-30\;mV)$ which partially overlaps with the resting membrane potential. This current has the property that it can be easily activated by a weak depolarization, thus it may trigger a certain kind of a intracellular event following fertilization which may cause oscillations in the membrane potential.

고추냉이 신초의 생육에 미치는 배지의 Salt Strength, Sucrose 농도 및 NH4/NO3 비율의 영향 (Effect of Salt Strength, Sucrose Concentration and NH4/NO3 Ratio of Medium on the Shoot Growth of Wasabia japonica in Vitro Culture)

  • 식물 세포나 조직의 배양에서 배지의 salt strength, sucrose 농도, 질소 함량과 질소원의 종류 및 그 비율 등이 세포 또는 조직의 생장에 영향을 미친다. 본 연구에서는 Agar를 함유한 MS 기본배지와 BA 1 mg/L를 첨가한 MS 기본 액체배지에서 계대배양을 통하여 생산한 다신초를 액아와 본엽이 $2{\sim}3$매 되도록 분할한 신초를 이용하여 MS 액체배지의 salt strength와 sucrose 농도 및 $NH_4/NO_3$ 비율이 고추냉이 신초의 생육에 미치는 영향을 조사하여 고추냉이 유묘의 생산에 필요한 최적 배지조건을 구명하였다. 고추냉이 신초의 증식에는 MS 액체배지의 salt strength는 기본조성이 가장 적합한 것으로 나타났으며, $1/4{\sim}1/2x$ 또는 2x MS 배지에서는 고추냉이 유묘의 생장이 크게 억제 되었다. 기본조성의 MS 액체배지에 sucrose 3%를 첨가하였을 때 고추냉이 신초의 생장이 가장 우수한 것으로 나타났으나 sucrose 농도 $1{\sim}6%$ 범위에서 신초의 수와 길이는 유의성 있는 차이를 보이지 않았다. 기본조성의 MS 액체배지에서 질소원으로 ${NH_4}^+$${NO_3}^-$의 비율을 10:50으로 조절하였을 때 고추냉이 신초의 생장이 가장 우수한 것으로 나타났으며, ${NH_4}^+$의 비율이 증가할수록 생장은 억제되었다.
