Electrochemical Determination of 6-Benzylaminopurine (6-BAP) Using a Single-wall Carbon Nanotube-dicetyl Phosphate Film Coated Glassy Carbon Electrode

  • Herein, insoluble single-walled carbon nanotube (SWNT) was successfully dispersed into water in the presence of a special kind of surfactant-dicetyl phosphate (DCP), subsequently, a SWNT-DCP composite film coated glassy carbon electrode (GCE) was fabricated. The electrochemical behaviors of 6-benzylaminopurine (6-BAP) at the unmodified GCE and SWNT-DCP modified GCE were examined. It is found that the SWNT-DCP modified GCE remarkably enhances the oxidation peak current of 6-BAP, indicating great potential in the determination of trace level of 6-BAP. Finally, a sensitive and simple voltammetric method with a good linear relationship in the range of ${\times}5.0\;\;10^{-8}\sim 2.5\;{\times}\;10^{-6}$ mol/L, was developed for the determination of 6-BAP. The detection limit is as low as $2.0\;{\times}\;10^{-8}$ mol/L for 3-min accumulation. This newly-proposed method was successfully demonstrated with practical samples.

  • Kinetin(4.7$\times$10\ulcornerM) and 6-benzylaminopurine (2.2$\times$10\ulcornerM) were found to increase ca. 1.5-fold putrescice content in corn grown in medium containing kinetin and 6-benzylaminopurine(6-BAP) for 3days whereas kinetin was found to decrase ca. 30% spermidine and spermine, respectively. KCI (3$\times$10\ulcornerM) was found to decrease more than 50% putrescine content. After germination, ornithine decarboxylase activity was observed to increase constantly whereas arginine decarboxylase activity remained constant, suggesting involvement in putrescine biosynthesis. 6-benzylaminopurine was shown to increase more activities of arginine ornithine decarboxylase than kinetin when they were added to medium.

  • In this study, a plant tissue culture system was established for Atractylodes spp., an economically valuable medicinal crop in Korea that has low domestic production and is increasingly imported. In particular, Atractylodes ovata was treated with four types of cytokinins, 6-benzylaminopurine (BA), zeatin, kinetin, and thidiazuron (TDZ), in two different concentrations (0.5 and 1.0 mg/L). Among the four types of cytokinins, the BA treatment was effective for the shoot and root growth of A. ovata. Both the 0.5 mg/L and 1.0 mg/L concentrations of BA showed similar results; however, the 1.0 mg/L concentration of BA was more effective in promoting shoot and root growth. The treatments showed that the TDZ treatment was not effective for the shoot and root growth, except for the number of shoots and the fresh weight (FW) of the root; therefore, it was unsuitable for this species. In this study, we established a mass production system of A. ovata. Our results showed that direct in vitro regeneration may make a significant contribution to improving the cultivation of the medicinal plant A. ovata.

  • Effects of light, methyl jasmonate(MJ), 6-benzylaminopurine(BA), thidiazouron(TDZ), and 2,4-dichlorophenoxyacetic acid(2,4-D) on the contents of ecdysteroids (${\beta}$-ecdysone+polypodine B) in Polypodium vulgare L. and Achyranthes japonica Nakai were studied. When the plants of P. vulgare were cultured under light control, the ecdysteroids contents in both leaves and rhizomes decreased with increasing light intensity. The ecdysteroids contents in A. japonica were also lower when cultured under light than under dark. Among the tested plant growth regulators, MJ, BA, and TDZ increased the ecdysteroids contents in both P. vulgare and A. japonica.

  • An efficient method of shoot regeneration of peanut is described. In vitro shoot organogenesis from the callus of cotyledon explants of Arachis hypogaea L. was stimulated by addition of Adenine sulphate (Ads) along with 6 - benzylaminopurine (BAP) and - napthalene acetic acid (NAA). Ads (13 ${\mu}{\textrm}{m}$) had a stimulatory effect on shoot bud differentiation when combined with BAP (13 ${\mu}{\textrm}{m}$) and NAA (2 ${\mu}{\textrm}{m}$). Shoot organogenesis was markedly higher (92%) from callus induced on Ads, BAP and NAA combined media than from those formed by the individual supplementation of Ads or BAP with NAA. The shoots elongated on the media with GA$_3$ (1 ${\mu}{\textrm}{m}$). Elongated plantlets rooted with MS media containing IBA (9 ${\mu}{\textrm}{m}$).

  • Sedirea japonica is one of the critically endangered species in South Korea mostly due to artificial harms such as illegal collection and habitat destruction. Therefore, artificial propagation through improving germination rate, increasing growth, and controlling flowering is meaningful for the conservation and reintroduction of S. japonica. It is suggested that cytokinins are one of the multi-factors that contribute to plant growth and floral responses. Especially, exogenous cytokinins have been known to induce or promote shoot growth or earlier flowering in orchids. Therefore, it was investigated how the application of 6-benzylaminopurine (BA) influenced the growth and inflorescence of S. japonica. A foliar spray containing BA at 100, 200, 300, and 400 ppm was applied from 1st July to 30th December 2021. Leaf length, leaf length growth rate, leaf width, and width and length ratio were measured as growth-related factors. Visible inflorescence rate, inflorescence length, the number of flowers per inflorescence, and the distance between the stalks were measured as flowering-related factors. Growth-related factors except for leaf growth rate were not affected by BA treatments, while leaf growth rate was significantly increased by 200 ppm of BA treatment. The visible inflorescence rate increased by 200 ppm of BA treatment, and there seems an optimal concentration and threshold of BA treatment. An iterative experiment with more seedlings and measurement factors would be helpful to figure out the effects of exogenous BA treatment on S. japonica, and it can be applied for mass propagation.

Efficient and Reliable in vitro Regeneration System for Rubus Species as the Basis of Genetic Engineering

  • Factors affecting regeneration of different Rubus varieties (blackberry, raspberry and their hybrid) were examined and a reliable regeneration system was established. Media for stock plant maintenance were tested; different explants and media were investigated to find the best circumstances for the regeneration. The effect of the commonly used antibiotics was studied to determine the most suitable one for selection of the transformants. We found that both MS and LS media supplemented by $20\;gL^{-1}$ sucrose are suitable for the stock plant maintenance. The optimal hormone content for the stock plants is $0.125\;mgL^{-1}$ 6-benzylaminopurine (BAP) with $0.01\;mgL^{-1}$ indole-3- butyric acid (IBA). The highest regeneration rate was observed on medium containing MS salts with B5 vitamins complemented with glucose, sucrose, maltose, $10\;gL^{-1}$ each, supplemented with benzylaminopurine riboside (BAR) ($2\;mgL^{-1}$) and indole-3-acetic acid (IAA) ($0.1\;mgL^{-1}$). The regenerated shoots appeared directly from the cut edges, without callus phase. Hygromycin and geneticin proved to be good selection agents for the Rubus explants, but due to their severe effect on the tissues we propose to use marker-free constructions for the transformation.

Effects of Mineral Media, Carbon Sources and Phytohormones on Micropropagation of Alnus hirsuta (물오리나무(Alnus hirsuta)의 기내증식에 미치는 기본배지, 탄소원 및 식물호르몬의 영향)

  • Shoot tip explants from germinated seeds of Alnus hirsuta were cultured on NT (Nagata and Takebe, 1971) mineral salts medium supplemented with 6% glucose, MS (Murashige and Skoog, 1962) vitamin mixture, polyvinylpyrrolidone (PVP) and $0-50\;\mu\textrm{M}$ 6-benzylaminopurine (BAP). Five $\mu\textrm{M}$ BAP was found to give the highest shoot multiplication rate. Accordingly about 200 shoots were obtained for further experiments by multiplying shoots on this medium for 4-5 months. Regardless of carbon sources, NT mineral medium produced 3-12 times of shoots than MS mineral medium did. On NT mineral medium, 3% sucrose, 3% glucose and 6% glucose yielded no significant differences. It was observed that media consisting of 1/4-1/2 strength NT mineral salts, 3% sucrose and $1-8\;\mu\textrm{M}$ IBA produced about 100% rooting rate. Almost 100% of the resulting plantlets survived after transfer to the soil by decreasing humidity stepwise.epwise.

An Efficient In vitro Propagation of Zanthoxylum piperitum DC.

  • A protocol is described for rapid multiplication of Zanthoxylum piperitum DC. (Rutaceae), an important aromatic and medicinal plant, through shoot-tip explant cultures. Murashige and Skoog (MS) medium supplemented with various concentrations of N-6-benzyladenine (BA), N-6-benzylaminopurine (BAP) and thidiazuron (TDZ), in single or in combination with ${\alpha}-naphthaleneacetic$ acid (NAA), was used to determine the rate of shoot proliferation. N-6-benzyladenine (BA) used at 0.5mg/l, was the most effective in initiating multiple shoot proliferation at the rate of 23 microshoots per shoot-tip explants after 40 days of culture. Shoot multiplication increased 1.2-fold in each successive subculture. Induction of rooting (98%) was achieved by transferring the shoots to the same basal medium containing 2 mg/l indole-3-butyric acid (IBA). Plantlets went through a hardening phase in a controlled growth chamber, prior to in vivo transfer. These results represented that possible application for the mass production of plantlets through in vitro culture system of Zanthoxylum piperitum DC.
