Cryopreservation of Sporothalli of the Genus Porphyra (Bangiales, Rhodophyta) from Korea

  • Cryopreservation of sporothalli of the red alga Porphyra (P. seriata, P. yezoensis, P. tenera, P. pseudolinearis and P. dentata) was performed by the two-step cooling method in liquid nitrogen. The algal samples were suspended in various cryoprotective solutions, and slowly cooled to -40$^{\circ}C$ in 4 hours using a programmed freezer. After the first slow cooling the suspensions with cryoprotectants were immediately immersed in liquid nitrogen. The suspension from the programmed freezer was thawed quickly by agitation of the vial in a water bath at 40°C. When ice in the suspension of cryogenic vial was mostly melted, the vial was transferred to an ice bath for complete melting of the residual ice. The cryoprotectants in the vial were washed off by gradual dilution with seawater. The viability of the cell was assessed with neutral red staining. The viability of Porphyra samples ranged 54.6-70.9% when the mixed suspension of 10% dimethylsulfoxide and 0.5 M sorbitol in 50% seawater used as a cryoprotectant.

Changes in the number of P. gingivalis bacteria before and after professional periodontal prevention care in periodontal disease patients (전문가 치주 예방관리 전·후 치주질환자의 P. gingivalis 세균 수 변화)

  • Background: This study aimed to investigate the changes of the amount of P. gingivalis in saliva by professional preventive treatment of periodontal disease by dental hygienists. Methods: A total of 109 subjects participated in this study with informed consent. The control group (38 subjects) performed oral hygiene management individually. The periodontal treatment group (35 subjects) underwent root planning once every 3 weeks for a total of 4 times. The professional periodontal prevention group (36 subjects) underwent interdental cleaning and professional tooth brushing once every 3 weeks for a total of 4 times. Paired T test and analysis of variance were performed to compare the difference among the groups in the amount of P. gingivalis. Results: The copies of P. gingivalis in the professional periodontal prevention group decreased from 773.62±1,198.09 to 241.40±430.40 after treatment significantly. The control group decreased from 525.22±582.54 to 244.29±385.88 after treatment. The periodontal treatment group showed insignificant change of P. gingivalis. Conclusions: This study showed the professional periodontal prevention was more effective than periodontal treatment in decrease of P. gingivalis.

Evaluation of the Non-use Values of Taishan Mountain Cultural Heritage

  • The contingent valuation method (CVM) is one of the most commonly used and effective methods to evaluate non-use value of resources. Reasonable application of CVM to value the cultural heritage is the key process of evaluation. CVM was used to evaluate the non-use value of cultural heritage of Taishan Mountain combined with questionnaire survey and field research in this study. The results indicated that the importance of the degree of the three components of non-use value was heritage value ranked highest (40.22%) > followed by existence value (38.58%) >then option value (21.20%). In addition, the rate of willingness to pay was 54.52%, the average and median values of per person were 40.17 CNY·a-1 and 20.00 CNY·a-1 and the non-use values of Taishan Mountain cultural heritage was 33 million CNY·a-1. The median value of WTP was consistent with Asian countries but was lower than European and American countries. Factors influencing WTP showed that monthly income and satisfaction with Taishan Mountain were correlated to WTP, and family location and willingness to revist were correlated remarkably with WTP. In addition, monthly income was correlated remarkably with WTP value, however other factors were not. The results showed the importance and necessity of protective development of Taishan Mountain cultural heritage, which would be used as an important reference for decision makers.

Copper phthalocyanine conjugated PANI coated screen printed carbon electrode for electrochemical sensing of 4-NP

  • In this work, we synthesized a novel electrochemical sensing materials based on tetracarboxylic copper phthalocyanine (TcCuPtc) conjugated PANI (TcCuPtc@PANI). The synthesized materials were employed to modify the screen-printed carbon electrode (SPCE) for the selective sensing of 4-nitrophenol. The TcCuPtc was conjugated with conducting polymer of PANI through the electrostatic interaction and π-π electron conjugation, the polymer film of PANI to inhibit the leakage of TcCuPtc from the surface of the electrode. The prepared TcCuPtc@PANI were characterized and confirmed by scanning electron microscopy (SEM) with EDX, ATR-IR, UV-vis absorption spectroscopy, cyclic voltammetry, and differential pulse voltammetry techniques. The prepared TcCuPtc@PANI/SPCE showed an excellent electrocatalytic sensing of 4-NP in the linear concentrations from 3 to 500 nM with a LOD of 0.03 nM and a sensitivity of 8.8294 ㎂/nM cm-2. However, the prepared TcCuPtc@PANI/SPCE showed selective sensing of 4-NP in the presence of other interfering species. The practical applicability of the TcCuPtc@PANI/SPCE was employed for the sensing of 4-NP in different water samples by standard addition method and showed satisfactory recovery results.

Multilevel Analysis on Spatial Distribution and Socio-Environmental Factors of Dental Caries in Korean Children

  • This study aims to identify the regional distribution in the prevalence of dental caries and related multidimensional factors among 12-year-old children in Korea. Data from the 2018 Child Oral Health Survey were used to calculate the average DMFT index of 12-year-old children in metropolitan cities, and a multi-level regression model was applied to explain the regional distribution of dental caries prevalence and related factors. Factors were divided into two levels by administrative structure. This study finds a significant regional difference in the prevalence of dental caries in 12-year-old Korean children across metropolitan cities. Multilevel analysis showed that district-level factors (average number of pit and fissure-sealed permanent teeth, dental treatment demand rate, preventive treatment rate, sex ratio, and number of dentists per 100,000 people) and metropolitan-level factors (intakes of cariogenic beverages and number of pediatric dental hospitals and clinics per 100,000 people) had a significant effect on dental caries prevalence (p < 0.05). Individual characteristics and local socio-environmental factors influence the prevalence of dental caries. Especially considering the strong dependence on preventive treatment and accessibility to dental care services, it is necessary to provide adequate preventive treatment and expand health care resources in high-risk areas of dental caries.

Effects of Acupuncture at HapGuk$(LI_4)$ on Pain in Rats (합곡(合谷) 침자극(鍼刺戟)이 동통억제(疼痛抑制)에 미치는 영향(影響))

  • The purpose of this study was finding the pain inhibitory effect of acupuncture at $LI_4$. The pain at dentes incisivi was evoked by noxious electric stimulation and EMG changes based on the interval were measured. To do this, the opioid antagonist was administered intraperitoneally. Following are some of results. 1. The pain inhibitory effect of acupuncture at $LI_4$ was 94.35%, 84.56%, 57.62%, 54.40%, at 10, 20, 30 and 40 minutes respectively. All the data were calculated based on 100% at 0 minute. The effect was very significant at 40 minutes. 2. The continuous effect of acupuncture at $LI_4$ after taking out the needle was 58.58%, 57.62%, 66.22%, 73.18%, 83.70%, 92.68%, at 50, 50, 70, 80, 90 and 100 minutes respectively. The overall continuous effect was shown its maximum 20 minutes after taking out the needle and reached to the initial value at 60 minutes. 3. The pain inhibitory effect following the naloxone administration and acupuncture application at $LI_4$ was 95.96%, 96.04%, 94.86%, 94.92% at 10, 20, 30 and 40 minutes respectively. All values showed similar tendency to the initial data. 4. The continuous effect of acupuncture at $LI_4$ after taking out the needle which was preceded by the naloxone administration was 94.48%, 96.02%, 96.02%, 98.00%, 98.46%, 97.18% at 50, 60, 70, 80, 90 and 100 minutes respectively. This trend was not a significant fluctuation. It was concluded that effect of acupuncture at $LI_4$ was shown in conjunction with secretion of analgesic substance. Therefore it is implied that the acupuncture application will play a major role in treating many diseases with more revelation of scientific acupuncture mechanism. Further studies of acupuncture manipulation are needed in the future based on our study.

Comparative Study on the Long-Acting Thyroid Stimulator in Graves' Disease (Graves 병(病)에 있어 Long-Acting Thyroid Stimulator(LATS)에 관(關)한 연구(硏究))

  • In order to study the role of LATS in the pathogenesis of the Graves' disease, the serum activity of the LATS was determined by the bioassay of the modified McKenzie method. The subjects examined in the study consisted of 76 individuals including 12 cases of normal control, 54 cases with typical Graves' disease and 10 cases of chronic thyroiditis. The data observed in the patients with the Graves' disease were analyzed in comparison with the clinical features, laboratory findings, and responsiveness to the treatment. The results obtained are as follows: 1. None of the subjects which did not have the Graves' disease showed a positive LATS activity, except one case with the chronic thyroiditis. 2. Twenty-two out of the 54 cases with the Graves' disease showed positive results for LATS(40.7%). The positivity was significantly higher in the patients who had been treated with anti. thyroid regimen but still showed hyperthyroidism than in the patients who had not been treated. 3. The activity of LATS was gradually decreased or even became absent as the hyperthyroidism was corrected after the treatment. 4. No significant difference was noticed in age & sex between the positive and negative groups of LATS. 5. There was no evidence of significant correlation between the LATS activity and clinical features.

Maturation and spawning of female black scraper, Thamnaconus modestus in the coastal waters off Middle East Sea, Korea (동해 중부해역에 출현하는 말쥐치(Thamnaconus modestus) 암컷의 성숙과 산란)

  • The maturation and spawning of black scraper, Thamnaconus modestus were studied using samples collected monthly from March, 2015 to February, 2016 in the coastal waters off Middle East Sea, Korea. The gonadosomatic index (GSI) of female was the highest in June. The spawning periods lasted from March to September based on histological observation of female gonad development. The percentage of sexually mature females estimated from a logistic function was over 50% for the size group 18.02 cm (total length, TL). The size of eggs spawned was between 0.40 to 0.58 mm. Fecundity varied between 185,648 and 9,747,250 eggs. The relationship between the fecundity and TL of the fish was expressed in the fecundity equation as $F=0.0297TL^{5.4835}$.

Tracing the earliest phases of star formation: A pilot survey of Planck Cold Clumps

  • We observed 38 Planck Cold Clumps (PCCs) in the $850{\mu}m$ dust continuum emission using the JCMT/SCUBA-2, and detected the emission in 15 clumps containing dense cores. In this poster we present the preliminary results. The PCCs are cold, dense, and thus, they are considered as objects in the early evolutionary stages of star formation. The sources in our sample were selected based on the Purple Mountain Observatory (PMO) 13CO (1-0) integrated intensity maps. In order to examine whether these cores detected in $850{\mu}m$ continuum have potential to be prestellar cores, we compare each core mass estimated from the $850{\mu}m$ continuum with the Virial mass and Bonnor-Ebert (BE) mass calculated from the 13CO (1-0) or C18O (1-0) spectra. By comparing the two column densities from the dust continuum and the 13CO (1-0) or C18O (1-0) line, we also derive the CO depletion factor, which could be an indicator of core evolution. The moment maps of the 13CO (1-0) line are used to study the physical properties (e.g. kinematics, turbulence) of PCCs. We investigate difference between the sources with and without detectable $850{\mu}m$ emission to study the formation conditions of dense cores.

Evaluation of the Apparent Ileal Digestibility (AID) of Protein and Amino Acids in Nursery Diets by In vitro and In vivo Methods

  • The objective was to evaluate in vitro prediction of ileal digestibility of protein and amino acids (AA) for current nursery pig diets (n = 10) by using pepsin and pancreatin incubations. To compare in vivo ileal digestibility, forty nursery pigs (4 pigs per diet) with an initial BW of $12.2{\pm}2.7$ kg were surgically equipped with T-cannula in the distal ileum. In all cases, the values of in vitro digestibility were higher than those of in vivo digestibility (p<0.05). With regard to the relationships of essential and non essential AA (CP), the $r^2$ value was 0.76. With regard to AA, high relationships were observed in Ile, Thr, and Gly (0.85, 0.83, and 0.89, respectively). Also, there was a lower relationship for Arg, Met, Ala, Asp, Glu, Pro, Ser, and Tyr with $R^2$ values of 0.56, 0.54, 0.40, 0.54, 0.45, 0.24, 0.49, and 0.35, respectively between in vitro and in vivo digestibility. The EAA relationship ($R^2$ = 0.71) was generally higher than that of NEAA ($R^2$ = 0.50) numerically. In conclusion, there were strong linear relationships between in vivo and in vitro ileal digestibility (CP, Ile, Thr, and Gly). In vitro prediction of ileal digestibility (CP, Ile, Thr, and Gly) seems to have significant potential for practical application.
