Development of a Multiplex PCR Method to Detect Fungal Pathogens for Quarantine on Exported Cacti

  • Major diseases in grafted cacti have been reported and Fusarium oxysporum, Bipolaris cactivora, Phytophthora spp. and Collectotrichum spp. are known as causal pathogens. These pathogens can lead to plant death after infection. Therefore, some European countries have quarantined imported cacti that are infected with specific fungal pathogens. Consequently, we developed PCR detection methods to identify four quarantined fungal pathogens and reduce export rejection rates of Korean grafted cacti. The pathogen specific primer sets F.oF-F.oR, B.CF-B.CR, P.nF-P.nR, and P.cF-P.CR were tested for F. oxysporum, B.cactivora, P. nicotinae, and P. cactorum, respectively. The F.oF-F.oR primer set was designed from the Fusarium ITS region; the B.CF-B.CR and P.nF-P.nR primers respectively from Bipolaris and Phytophthora ITS1; and the P.cF-P.CR primer set from the Ypt1protein gene region. The quarantine fungal pathogen primer pairs were amplified to the specific number of base pairs in each of the following fungal pathogens: 210-bp (F. oxysporum), 510-bp (B. cactivora), 313-bp (P. nicotinae), and 447-bp (P. cactorum). The detection limit for the mono- and multiplex PCR primer sets was 0.1 ng of template DNA under in vitro conditions. Therefore, each primer set successfully diagnosed contamination of quarantine pathogens in export grafted cacti. Consequently, our methodology is a viable tool to screen contamination of the fungal pathogen in exported grafted cacti.

  • Electro-Optical Camera(EOC) is the main payload of Korea Multi-Purpose SATellite(KOMPSAT) with the mission of cartography to build up a digital map of Korean territory including Digital Terrain Elevation Map(DTEM). This instrument which comprises EOC Sensor Assembly and EOC Electronics Assembly produces the panchromatic images of 6.6 m GSD with a swath wider than 17 km by push-broom scanning and spacecraft body pointing in a visible range of wavelength, 510 ~ 730 nm. The high resolution panchromatic image is to be collected for 2 minutes during 98 minutes of orbit cycle covering about 800 km along ground track, over the mission lifetime of 3 years with the functions of programmable rain/offset and on-board image data storage. The image of 8 bit digitization, which is collected by a full reflective type F8.3 triplet without obscuration, is to be transmitted to Ground Station at a rate less than 25 Mbps. EOC was elaborated to have the performance which meets or surpasses its requirements of design phase. The spectral response the modulation transfer function, and the uniformity of all the 2592 pixel of CCD of EOC are illustrated as they were measured for the convenience of end-user. The spectral response was measured with respect to each gain setup of EOC and this is expected to give the capability of generating more accurate panchromatic image to the EOC data users. The modulation transfer function of EOC was measured as greater than 16% at Nyquist frequency over the entire field of view which exceeds its requirement of larger than 10%, The uniformity that shows the relative response of each pixel of CCD was measured at every pixel of the Focal Plane Array of EOC and is illustrated for the data processing.

금속가공유 취급 작업장의 생물학적 인자 노출평가 (Microbial Assessment in Metal-Working Fluids Handling Industry)

  • Objectives: The objective of this study is to evaluate microbial exposure hazards in the metal-working fluids(MWF) handling industry. Methods: Air quality parameters(airborne bacteria, fungi, endotoxin and oil mist) and bulk MWF in storage tanks were evaluated at 54 points at nine sites in South Korea. Results: The geometric means(GM) of culturable airborne bacteria, fungi, endotoxin and oil mist concentration were $133CFU/m^3$(n=376, range $7{\sim}6,510CFU/m^3$), $159CFU/m^3$(n=381, range $7{\sim}8,469CFU/m^3$), $8.06EU/m^3$(n=103, range $0.34{\sim}280.4EU/m^3$) and $0.20mg/m^3$(n=104, range $0.01{\sim}2.87mg/m^3$), respectively. The ratio of indoor to outdoor concentration was 2.7 for bacteria, 6.1 for endotoxin, and 4.8 for oil mist. Even though average airborne bacteria concentration did not exceed recommended exposure limits($1,000CFU/m^3$), MWF in the storage tanks was highly contaminated with bacteria(arithmetic mean $2.1{\times}10^6CFU/ml$) and exceeded recommended bacteria limits($10^5CFU/ml$). Conclusions: It is necessary for MWF handling workplaces to conduct periodical biohazard inspection of MWF storage tanks. Additionally, further research may be necessary to establish biological occupational exposure limits.

대학 캠퍼스 주변 호프집, PC방, 당구장의 실내 PM2.5 농도를 통한 ETS 노출 수준 평가 (Evaluation of Indoor ETS Exposure Levels in Pubs, PC Game Rooms, and Billiards Halls around a University Campus using PM2.5 Concentrations)

  • Objectives: The aims of this study were to determine the indoor level of environmental tobacco smoke (ETS) and to assess the implementation rate of smoke-free laws at hospitality venues around a university campus by measuring particulate matter smaller than $2.5{\mu}m$ ($PM_{2.5}$) as an indicator of ETS. Materials and Methods: We measured indoor $PM_{2.5}$ concentrations at 20 PC game rooms, 20 pubs, and 20 billiards halls using Sidepak AM510, a direct reading portable real time monitor, from October to December 2015. Results: Smoking was observed in 65% of the PC game rooms, 10% of pubs, and 85% of billiards halls. The average $PM_{2.5}$ concentrations were $98.2{\mu}g/m^3$, $29.0{\mu}g/m^3$, and $134.2{\mu}g/m^3$ at PC game rooms, pubs, and billiards halls, respectively. $PM_{2.5}$ concentrations in PC game rooms and billiards halls were 2 to 2.7 times higher than the 24-hour exposure standard for outdoor $PM_{2.5}$ ($50{\mu}g/m^3$) by the Ministry of Environment. Conclusions: Although a smoking ban has been implemented for PC rooms and pubs, smoking is still taking place in many of these places. More stringent legal action is required for successfully protecting patrons and workers from secondhand smoke exposure. A ban on smoking in billiards halls should be introduced as quickly as possible.

감자 유전자원의 기내 장기보존 방법 (In vitro Long Term Conservation of Potato Germplasms)

  • 감자 유전자원의 기내 유식물체 장기보존시 식물체 변이발생을 최소화하면서 최장기간 배양할 수 있는 배양조건을 확립하기 위해 생장억제물질을 사용하지 않고 탄소공급원으로서 삼투조절제를 sucrose와 혼합처리하였는데, 이러한 삼투조절제에 의한 생육지연 효과는 배양온도를 낮춤으로써 효과를 증대시킬 수 있었다. 6개월 저장 후 저온하의 4배체 계통의 초장이 2배체 계통의 초장보다 다소 높게 나타났으나 배지간 차이는 나타나지 않았다. 그러나 저온저장 12개월 후에는 mannitol과 sucrose의 혼합처리인 LSM 2 배지보다 sorbitol과 sucrose의 혼합처리인 LSM 1 배지의 처리계통에서 만족할 만한 결과를 얻을 수 있었으며, 특히 2배체 근연야생종인 stn-16은 LSM 1 배지에서 75%의 상당히 높은 생존율을 보였으며 재배종인 'Atlantic'은 두 배지 모두에서 100%의 생존율을 보였다.

Spatial and Temporal Variability of Water Quality in Korean Dam Reservoirs

  • The objectives of this study were to evaluate spatial and temporal variability of water quality in 10 reservoirs and identify the key nutrients (N, P) influencing chlorophyll-a (CHL) along with analysis of empirical models and zonal patterns of total phosphorus (TP) and CHL. We analyzed total nitrogen (TN), TP, CHL, water clarity (Secchi depth, SD), and evaluated potential limiting nutrient using ambient N:P ratios and previous criteria of ambient nutrients. Water clarity and CHL varied largely depending on the seasonal monsoon and type of reservoir, but trophic state was diagnosed as eutrophy, base on mean CHL in most reservoirs. The peak of TP did not match the contents of CHL due to rapid flushing during the high run-off period. In the reservoir of DR, regression coefficient in the $P_r$ was 0.510 but was 0.159 in the $M_o$, while the TP-CHL relation in the YR increased during the monsoon compared to the premonsoon. The regression coefficient in the $P_r$ was not statistically significant but the value of $M_o$ was 0.250. TP showed similar longitudinal zonal gradients among the reservoirs of DR, YR and JR. Empirical models of TP-CHL, based on overall data, showed that CHL was determined by phosphorus($R^2=0.244$, p=0.0019). Regression analysis of CHL-SD showed a stronger linear fit ($R^2=0.638$, p<0.001) than the TP-CHL model.

  • 해양사고는 주로 충돌과 좌초에 의한 것으로 인적과실이 원인이다. 한국형 e-Navigation 사업은 인적과실에 의한 해양사고 저감 및 초고속 해상무선통신(LTE-Maritime)의 구축 방안을 마련하고 있다. 그에 따른 사고취약선박 모니터링 지원 서비스(SV10)는 육상에서 수집한 선박의 위치정보를 기반으로 선박의 충돌 좌초 위험도를 평가하여 선박이 위험상황을 인식할 수 있도록 정보를 제공하고 해양사고 발생 시 육상에서 조기 대응할 수 있도록 실시간 선박 위치 및 위험상황 정보를 유관기관과 공유하는 서비스이다. 본 연구에서는 중앙해양안전심판원에서 제공된 해양사고 통계연보 및 재결서를 분석하여 SV10 서비스로 저감될 수 있는 해양사고 비율을 확인하였다.

  • Nitrogen is a serious contaminant in natural gas because it decreases the energy density. The natural gas specification in South Korea requires a $N_2$ content of less than 1 mol%. Thus, cost-effective $N_2$ removal technology from natural gas is necessary, but until now the only option has been energy-intensive processes, e.g., cryogenic distillation. Using porous materials for the removal process would be beneficial for an efficient separation of $CH_4/N_2$ mixtures, but this still remains one of the challenges in modern separation technology due to the very similar size of the components. Among various porous materials, metal-organic frameworks (MOFs) present a promising candidate for the potential $CH_4/N_2$ separation material due to their unique structural flexibility. A MIL-53(Al), the most well-known flexible metal-organic framework, creates dynamic changes with closed pore (cp) transitions to open pores (ops), also called the 'breathing' phenomenon. We demonstrate the separation performance of $CH_4/N_2$ mixtures of MIL-53(Al) and its derivative $MIL-53-NH_2$. The $CH_4/N_2$ selectivity of $MIL-53-NH_2$ is higher than pristine MIL-53(Al), suggesting a stronger $CH_4$ interaction with $NH_2$.

  • 본 연구는 실내골프연습장의 서비스품질이 고객만족, 고객충성도가 재등록의사에 미치는 영향을 알아보기 위하여 이루어졌다. 본 연구의 대상은 실내골프연습장을 이용하고 있는 고객들을 대상으로 하였으며, 자료수집은 편의표본추출법에 의하여 수집된 설문지 중 238부를 실제 자료처리에 사용하였다. 본 연구의 결과는 다음과 같다. 첫째, 서비스품질의 모든 하위요인이 고객만족에 정적인 영향을 미쳤다. 둘째, 서비스품질 하위요인 중 시설과 지도자만 고객충성도에 영향을 미쳤다. 셋째, 서비스품질의 모든 하위요인이 재등록의사에 정적인 영향을 미쳤다. 넷째, 고객만족은 재등록의사에 정적인 영향을 미쳤다. 다섯째, 고객충성도는 재등록의사에 정적인 영향을 미쳤다. 여섯째, 고객만족은 서비스품질과 재등록의사 관계에 있어서 매개역할을 하는 것으로 나타났다. 마지막으로, 고객충성도는 서비스품질과 재등록의사의 관계에 있어서 부분적으로 매개역할을 하는 것으로 나타났다.

$CCl_4$중에서 Thioacetamide와 N,N-Dimethylacetamide사이의 수소 결합에 관한 분광학적인 연구 (Near-IR Spectroscopic Studies of the Hydrogen Bonding between Thioacetamide and N,N-Dimethylacetamide in $CCl_4$)

  • Thioacetamide(TA)-CCl$_4$와 TA-N,N-dimethlylacetamide (DMA)-CCl$_4$ 용액에서 TA의 $v_3$ + Amide II 조합띠의 근적외선 스펙트럼을 5$^{\circ}$ ~55$^{\circ}$C 에서 얻었다. 삼성분계에서 이 조합띠는 단위체 TA, 1 : 1 TA-DMA complex and 1 : 2 TA-DMA 및 1 : 2 TA-DMA 성분으로 나타나지만, 묽은 용액에서는 단위체 TA와 1 : 1 복합체만이 나타나며 이를 컴퓨터를 사용해서 각 띠의 형태를 Lorentzian-Gaussian 곱의 함수로 보아 분리하였다. 농도 및 온도에 따른 스펙트럼을 분석하여 1 : 1복합체에 대한 평형상수와 열역학적 피라미터들을 구했으며, ${\Delta}H^{\circ}$는 -14.4 KJ mol$^{-1}$이었고 ${\Delta}S^{\circ}$는 -15.6 J mol$^{-1 }deg^{-1}$이었다.
