Identification of Amino-Acids Residues for Key Role in Dextransucrase Activity of Leuconostoc mesenteroides B-742CB

  • Dextransucrase (DSRB742) from Leuconostoc mesenteroides NRRL B-742CB is a glucosyltransferase that catalyzes the synthesis of dextran using sucrose, or the synthesis of oligosaccharides when acceptor molecules, like maltose, are present. The DSRB742 gene (dsrB742) was cloned and the properties were characterized. In order to identify critical amino acid residues, the DSRB742 amino acid sequence was aligned with glucosyltransferase sequences, and three amino acid residues reported as sucrose binding amino acids in Streptococcus glucosyltransferases were selected for site-directed mutagenesis experiments. Asp-533, Asp-536, and His-643 were independently replaced with Ala or Asn. D533A and D536A dextransucrases showed reduced dextran synthesis activities, 2.3% and 40.8% of DSRB742 dextransucrase, respectively, and D533N, D536N, H643A, end H643N dextransucrases showed complete suppression of dextran synthesis activities altogether. Additionally, D536N dextransucrase showed complete suppression of oligosaccharide synthesis activities. However, modifications at Asp-533 or at His-643 retained acceptor reaction activities in the range of 8.4% to 21.3% of DSRB742 acceptor reaction activity. Thus at least two carboxyl groups of Asp-533 and Asp-536, and His-643 as a proton donor, are essential for the catalysis process.

  • The raw materials for plant-stalk-folkcrafts are cultivated in the whole country. Most Korean farmers, to increase their income, have produced mats and containers with various plants as a side line from very long ago. At first, they began from the instruments of life and then made folkart and at last get to manu\ulcornerfacturing of industrial folk craft. The folkcrafts, made of plant-stalk, which could nor conform to changing society, are partly declined and partly handed down as the traditions. The social change today, however, makes living conditions more speedy and multisided, accordingly the characteristics of demand also become in\ulcornerdividual and various. While the demend is various like this, suppliers cannot answer demendants' requirements, and consequently, the demand and profit cannot be increased. According to this, the purpose of this study is set up to give an answer to the situation that is at the traditional standstill, through an examination of the motives. I. The crafts of plant-stalk are made only in an organized relationship between agriculture, industry and art as it is compounded art of gathering raw material, manufacturing, producing, improv\ulcornering design and production conditions. It may be possible that a farmer gathers material and weaves it manually but in others, it is im\ulcornerpossible to refine, bleach and dye because the process requires a professional industrial treatment. It is impossible to make art works to a farmer as every farner does not always have aesthetic sense. Though a farmer or producer has these all abilities, it is not desirable to him from economical view. 2. The development or improvement is essential in many sides but the most important thing seems to he in design. According to reports, it is, howevt!r, fact that the crafts improved in design of existing works have more expanded the sales than newly developed works. Therefore, ir appears advisable to improve designs of existing things positively as they have merit of occupying a position already, but on the other hand, new crafts have to be also developed and the producer should grasp the proper time. 3. Building up an industrial complex to improve design with collecting the producing districts for this works scattered allover the country is very desirable for speedy communication, intensive educa\ulcorner tion or training, and effective guidance. 4. In producing for export abroad, before everything, must know the life environment, custom! and manners, main thought of the country, and then produce according to these. S. The crafts of plant-stalk are the fIrst industry in present but in the futher it should change intc second or third industry. 6. A synethetic organization for supporting side line should be established for effectiveness, and experts have to be secured and also the educational-industrial complex and activation of study should be preceded.

  • 본 연구는 효율적인 공급사슬관리(SCM)을 위한 전략 정보시스템 구축에 그 목적이 있다. 이러한 효율적 공급사슬관리를 위한 전략정보시스템 구축에 영향을 주는 요인들을 파워 프로세스의 관점에서 파악하고 이를 실증 연구를 통해 보다 효율적인 전략정보시스템으로서의 SCM 전략의 대안을 제시하고자 한다. 따라서 공급사슬관리의 효율적 운영에 영향을 주는 요인에 관한 영향요인들에 관한 기존 연구와 함께 파워 프로세스의 요소와의 연계를 통해 전략정보시스템으로서의 SCM 전략 유형을 제시하고 이의 특징을 연구의 범위로 설정하고 있다. 이러한 연구의 목적과 연구범위를 위해, 먼저 선행 연구된 파워 프로세스의 권력 주체가 권력요소, 가치 욕구, 환경, 관계성이라는 기본적인 다섯가지 권력 결정변수들간의 동적 상호작용을 통해 새로운 가치를 창출하는 과정을 살펴보며, 이러한 파워프로세스에 의한 전략이 권력 결정변수에 대한 가중치에 의해 전략의 유형이 결정되며, 이 유형에 따르는 전략 효과는 그 가중치와 결정 변수들간의 상호작용에 크게 의존되고 있음을 밝혀내고자 한다. 또한, 공급기업과 공급받는 기업들간의 이러한 관계는 기업이 사용하고 있는 전략적 정보시스템(SIS)으로서의 SCM 전략 유형과 일반적으로 높은 상호작용을 하고 있는 것으로 판단되어지며, 더욱이 기업의 정보가치에 대한 높은 인식으로 인한 디지털 경제하에서의 상호간 전략적 형성을 위해서는 더욱 파워프로세스적 관점하에서 SCM 전략 유형과 전략적 정보시스템간의 통합적으로 고찰함으로써 그 유용성을 살펴볼 수 있고 그러한 결과는 기업성과에 밀접한 영향을 주고 있음을 본 논문에서 밝히고 있다.더십을 행사할 때 이러한 리더십은 경영성과에 긍정적인 결과를 유발하는 것으로 나타났다. 그러므로, 리더십 성과 향상을 위한 지식경영정보시스템은 파워결정요소적 측면에서 고려되어야 할 것이다. 모델과 독립성 모델에서는 시스템 등급에 영향을 준다. 향후에는 더욱 더 다양한 상호의존 모델들이 정량화될 필요성이 있다고 본다. 진행하였다. 줄여서 보다 더 정확하고, 지능적인 규칙구성요소 추출 방법론을 제시하고 구현하여 지식관리자의 규칙습득에 대한 부담을 줄여 주고자 한다. 도움을 받을 수 있게 되었다.을 거치도록 되어있다. 교통주제도는 국가의 교통정책결정과 관련분야의 기초자료로서 다양하게 활용되고 있으며, 특히 ITS 노드/링크 기본지도로 활용되는 등 교통 분야의 중요한 지리정보로서 구축되고 있다..20{\pm}0.37L$, 72시간에 $1.33{\pm}0.33L$로 유의한 차이를 보였으므로(F=6.153, P=0.004), 술 후 폐환기능 회복에 효과가 있다. 4) 실험군과 대조군의 수술 후 노력성 폐활량은 수술 후 72시간에서 실험군이 $1.90{\pm}0.61L$, 대조군이 $1.51{\pm}0.38L$로 유의한 차이를 보였다(t=2.620, P=0.013). 5) 실험군과 대조군의 수술 후 일초 노력성 호기량은 수술 후 24시간에서 $1.33{\pm}0.56L,\;1.00{\ge}0.28L$로 유의한 차이를 보였고(t=2.530, P=0.017), 술 후 72시간에서 $1.72{\pm}0.65L,\;1.33{\pm}0.3L$로 유의한

  • 리더십에 관한 연구는 리더십 특성요인연구, 리더십 contingency 연구, 특성이론적 접근, 행동적 접근, 상황이론적 접근 등이 있으나, 이러한 접근방법이 언제나 긍정적인 리더십을 유발하는 것은 아닌 것으로 나타나고 있다. 따라서, 긍정적이고 일관된 리더십을 만들어 내기 위해서는 리더십에 대한 새로운 접근이 요청된다. 리더십에 대한 연구는 매우 다양하게 나타나고 있으나, 문헌연구에서 볼 때 리더십은 파워라는 공통 요소를 가지고 있다. 파워는 사람으로 하여금 목적지향적인 행동을 유발하는 능력이며, 문헌에 나타난 리더십의 개념은 조직구성원으로 하여금 목적지향적인 행동을 하게하는 능력이다. 따라서, 리더십은 권력의 한 유형으로 볼 수 있다. 그러므로, 본 연구에서는 리더십을 power process에 기초한 권력순환적 관점에서 연구하고자 한다.   • Polymer light emitting diodes (PLEDs) are expected to be commercialized as next generation displays by advantages of the fast response time, low driving voltage and easy manufacturing process for large sized flexible display. Generally, the electrical and optical properties of PLEDs are affected by the surface conditions of transparent electrode. The PLED devices with ITO/PEDOT:PSS/PVK/PFO-poss/LiF/Al structures were prepared by using the spin coating method. For this, PEDOT:PSS(poly(3,4-ethylenedioxythiophene):poly(styrene sulfolnate)) Al 4083 and PVK(N-vinylcabozole) were used as hole injection and transport layers. The PFO-poss(poly(9,9-dioctylfluorene)) was used as the emitting layer. The dependence of $O_2$ plasma treatment of ITO electrode on the electrical and optical properties of PLEDs were investigated. The sheet resistances increased slightly with an improved surface roughness of ITO electrode as the RF power increased during $O_2$ plasma treatment. The PLED devices prepared on the ITO/Glass substrates, which were plasma-treated at 40 watt in RF power for 30 seconds under 40 mtorr $O_2$ pressure, showed the maximum external emission efficiency of 0.86 lm/W and the maximum luminance of $250\;cd/m^2$, respectively. The CIE color coordinates are ranged $X\;=\;0.13{\sim}0.18$ and $Y\;=\;0.10{\sim}0.16$, showing blue color. emission.

Biographical Experience of Persons with Disabilities after an Accident : A Qualitative Study (중도장애인의 사고 이후 생애경험에 관한 질적 연구)

  • The purpose of the research is to understand the life of persons with disabilities caused by an accident by reconstructing their post-accident experiences. As a research tool, Fritz $Sch{\ddot{u}}tze's$ narrative interview was used, and three cases were analyzed in terms of a 'process structure of life'. After the researcher reconstructed each case, she sorted the cases comprehensively both according to the common features as well as to the differences among them. In doing so, the researcher came up with three characteristics of persons with disabilities caused by an accident. First, rather than identifying with their present life, the interviewees identified themselves with their life before the accident happened. Such identification problem is connected with their problem of accepting their handicap. And the interviewees identified themselves with disability groups. It showed that they feel more comfortable and secure among other people with disabilities than with families or friends. This makes them stay in the group of the disabled, in which they can understand and respect each other, rather than staying in a group in which people discriminate the disabled from able bodied and do not accept the disabled as equal members. Second, in their narration the interviewees told stories emphasizing their life after the accident happened. This feature can be compared with the experiences of those persons who underwent a trauma. Finally, it was found that our country's welfare policy for persons with disabilities caused by an accident is designed without differentiating people according to gender or educational background. Through these findings, the researcher intends to reflect on our country's current social welfare services and to look for new methods in order to implement proper social welfare practices.

Refractive index change of nonlinear polymer thin films induced by corona poling and quantitative evaluation of poling effect (코로나 극성배향이 비선형 고분자박막의 복소굴절율에 미치는 영향 및 배향효과의 정량화)

  • We prepared the side-chain type nonlinear optical NPP(N-(6-nitrophenyl)-(L)-prolinol) polymer films by spin coating method. Ellipsometric spectra were in situ collected by using spectroscopic phase modulated ellipsometer while the NPP polymer films were being corona poled at the temperature above glass transition. We calculated film thickness and the refractive index dispersion by modeling the spectro-ellipsometry data in transparent region. We also calculated the refractive index and the extinction coefficient of the polymer films by numerically inverting the spectro-ellipsometry data in absorbing region, while the previously determined film thickness was used. The independently determined extinction coefficient spectra from the analysis of transmission spectra were compared with those by spectro-ellipsometry and they showed an excellent agreement with each other. From the analysis of the complex refractive index change of the NPP polymer thin films induced by the corona poling, we could determine the vertical complex refractive index and the horizontal complex refractive index separately. Using the volume fraction of the vertical component f⊥, the degree of poling of poled NPP polymer films was quantitatively addressed. It is suggested that the present method can be used to quantitatively address the degree of poling in an absolute manner and to depth profile the poled fraction of thick polymer films. It will be useful to understand the structural change of polymer films and hence the poling mechanism during the poling process.

  • Roasting may lead to the formation of undesired compounds, such as polycyclic aromatic hydrocarbons (PAHs). In this study, green coffee beans were roasted under controlled conditions and the formation of PAHs during the roasting process was monitored. Roasting was performed in a hot air roaster, with an inlet air temperature varying from 150 to $250^{\circ}C$ for 5, 10, and 20 min. The PAH content of the roasted coffee was then evaluated by HPLC-FLD. The levels of total PAHs in Arabica (Colombia, Brazil) and Robusta (India) coffee samples were 1.26-215.07, 1.85-178.14, and 0.18-2.61 ${\mu}g$/kg, respectively.

  • The purpose of this study is to develop Family Life Education program for Marital education in Korea. In order to realize the about research purpose, this study is divided into three sections. First, nature conception and contents f Family Life Education, second, patterns and models for Family Life Education Program in USA and Korea, and Third, the future of Family Life Education. Family Life Education (FLE) is an education originated in reaction to changing social conditions, industrialization and urbanization, it deals with the perceived inadequacies of families to cope with these changes, thereby reduction social problems involved with, and improving family life. With and assumption that the studies about FLE have not been so active in Korea, the present thesis examines the FLE in Korea. The necessity of marital education in Korea Is based upon the theories of FLE in the United States of America. Also, it atempts to formulate a tentative plan for the promotion of a FLE program in Korea. The concept of FLE in Korea was manifested in the social educational law and life-span education. specified as 4th among ten presidential regulations. It is defined as " the Life-span developmental education to enhance the quality of human life, to solve family problems, to develop an individual's potentiality and to strengthen the family relations. of the FLE programs, the marital education is considered one of the furcial wubjects becaused it is the core of the family life. With this premise, FLE tries to wupport the healthy mrital relations. subsequently helping to explore the family potentiality and to strengthen family ties. The objects of the FLE program are unmarried, pre-marred , and married persons. For the married persons, the FLE program should be arranged in accordance with their rrital status divided by the family life cycle so that they may prevent possible family problems at each stage of the family life. Also, to prevent the problems occurred in the course of carrying out family functions, the FLE program should be provided with on the basis of a family unit, thereby improving the quality of the family functions. The FLE is to hep persons begin smoothly the adaption process for a successful marital relations. For the FLE to be practised more widely systematically, research on family education--that is basic study, research and production and evaluation or the FLE to be carried out by research institute wholly responsible for it-- should be done first.

Design and Performance Analysis of EU Directory Service (ENUM 디렉터리 서비스 설계 및 성능 평가)

  • ENUM(tElephon NUmbering Mapping) is protocol that brings convergence between PSTN Networks and IP Networks using a unique worldwide E.164 telephone number as an identifier between different communication infrastructure. The mechanism provides a bridge between two completely different environments with E.164 number; IP based application services used in PSTN networks, and PSTN based application services used in IP networks. We propose a new way to organize and handle ENUM Tier 2 name servers to improve performance at the name resolution process in ENUM based application service. We build an ENUM based network model when NAPTR(Naming Authority PoinTeR) resource record is registered and managed by area code at the initial registration step. ENUM promises convenience and flexibility to both PSTN and IP users, yet there is no evidence how much patience is required when users decide to use ENUM instead of non-ENUM based applications. We have estimated ENUM response time, and proved how to improve performance up to 3 times when resources are managed by the proposed mechanism. The proposition of this thesis favorably influences users and helps to establish the policy for Tier 2 name server management.
