사료 내 감귤착즙박 첨가가 저수온에서 사육된 참돔(Pagrus major)의 성장, 비특이적 면역반응 및 수온자극 스트레스에 미치는 영향 (Effects of Dietary Supplementation of a Citrus By-product on Growth Performance, Innate Immunity and Tolerance of Low Water Temperature in Red Seabream Pagrus major)

  • Our aim was to determine the effects of a citrus by-product (CBP) and CBP fermented by Lactobacillus plantarum (LP-CBP), provided as dietary supplements, on the growth performance, feed utilization, innate immunity and temperature tolerance of red seabream. A diet without inclusion of CBP or LP-CBP was used as a control and four other experimental diets were formulated to replace wheat flour by 4% and 8% of either CBP or LP-CBP (designated as Con, LP-CBP4%, LP-CBP8%, CBP4% and CBP8%, respectively). Experimental diets were fed to triplicate groups of 25 fish (initial body weight, 55.0 g) for 9 weeks. Growth performance and feed utilization were not significantly different among all the groups. Bone collagen content was significantly increased by supplementation with CBP and LP-CBP. Vitamin C concentration tended to be higher in livers of fish fed the supplements than in the control group. Myeloperoxidase, lysozyme and superoxide dismutase activities were higher in fish fed CBP or LP-CBP than in fish fed the control diet. When fish were exposed to low water temperature, cumulative mortalities of those fed CBP or LP-CBP supplemented diets were lower (29%, 33%, 34% and 33% mortalities for LP-CBP4%, LP-CBP8%, CBP4% and CBP8%, respectively) than in the control group (58%). Therefore, inclusion of either CBP or LP-CBP at up to 8% in red seabream diet brings benefits through enhanced innate immunity and better tolerance of low water temperature.

Zoogloea sp.의 다당체가 Meth A 세포에 의한 종양형성 억제 효과 (Antitumor Activities of Polysaccharudes fractuibuzed from Zoogloea sp. Against Meth A Cells)

  • The antitumor activities of the cell bound polysaccharide(CBP), water soluble polysaccharide(WSP) and sulfated polysaccharide(SP) of Zoogloea sp. were observe. The results obtained were as follows : 1) The CBP, WSP, and SP showed cytotoxic effect on the Meth A cells in vitro, however, the effect of CBP and WSP was more ten-fold greater than that pf SP. 2) When CBP, WSP, and SP was inoculated into the peritoneal cavity of the Meth A cells transplanted mice, the average survival days tended to prolonged slightly as compared with the control. 3) When Meth A cells were transplanted subcutaneously into the back side of mice, and then CBP, WSP, and Sp was inoculated into the peritoneal cavity of mice, the tumor growth inhibition ratio was 46.9% for WSP, 40.4% for CBP, and 16.2% for SP. 4) The phagocytic activity of peritoneal macrophages elicited with CBP, WSP, and SP was significantly increased than that of control. 5) The production of nitric oxide in the peritoneal macrophages stimulated with CBP, WSP, SP, and LPS aloneo was not increased than that of control. The production of nitric oxide in the peritoneal macrophages stimulated with IFN-r and CBP, IFN-r and WSP and IFN-r and SP was significantly increased than that of control, but in the case of stimulated with IFN-r and WSP was increased 50% for CBP and SP. These results suggest that the CBP, WSP and SP of Zoogloea sp. showed direct cytotoxic effect and tumor growth inhibition on Meth A cells in vitro and in vivo, and induced nitric oxide production of activated macrophages.

정공수송층이 역구조 OLED의 전기 및 광학적 특성에 미치는 영향 (Effect of Hole Transport Layer on the Electrical and Optical Characteristics of Inverted Organic Light-Emitting Diodes)

  • We have developed inverted green phosphorescent organic light emitting diodes (OLEDs) using 1,1-bis[(di-4-tolylamino)phenyl]cyclohexane (TAPC) and bis(carbazole-9-yl)biphenyl (CBP) hole transport layers. The driving voltage, current efficiency, power efficiency, and emission characteristics of devices were investigated. While the driving voltage for the same current density was about 1~2 V lower in the devices with the TAPC layer, the maximum luminance was higher in the device with the CBP layer. The maximum current efficiency and power efficiency were 3.2 and 2.7 times higher in the device with the CBP layer, respectively. The higher efficiency in the CBP device resulted from the enhanced hole-electron balance although weak parasitic recombination takes place in the CBP hole transport layer.

고추과실 세포벽 다당류의 연화에 따른 변화 (Softening Related Changes in Cell Wall Polysaccharides of Hot Pepper Fruit)

  • 고추 과실의 연화와 관련된 세포벽 다당류의 변화를 조사하기 위하여 조세포벽으로부터 화학적인 방법에 의하여 IAP(ionically associated pectin), CBP(covalently bounded pectin), HF(hemicellulosic fraction) 및 CF(cellulosic fraction) 등의 다당류를 분획하여 그 조성변화를 살펴보았다. 고추 과실 세포벽 구성 IAP와 CBP는 $4{\sim}29%$의 hemicellulose와 $0.9{\sim}2.1%$의 cellulose를 함유하였으며, HF는 $8{\sim}13%$의 pectin과 $0.1{\sim}1.1%$의 cellulose를 함유하였다. 또 IAP와 CBP를 구성하는 polyuronide는 IAP쪽이 높았으나, hemicellulose는 CBP쪽이 높았다. CF를 제외한 모든 다당류는 turning stage 이후에 감소하였다. IAP와 CBP의 구성 polyuronide는 IAP에서는 큰 변화를 보이지 않았으나, CBP에서는 증가하였는데, 이것은 CBP와 결합한 HF의 감소에 기인하며, 이 감소의 주된 원인은 구성 galactose의 손실에 의한 것으로 판단되었다.

EF-hands in CBP7 are Important in the Process of Development

  • Calcium ions play an important role in development and intracellular signaling. Dictyostelium discoideum has 14 genes encoding calcium -binding proteins (CBPs), but the function of most CBPs during development has not yet been studied. In this study, we investigated the specific functions of CBP7, one of 14 CBPs, in development using RNA interference cell lines of CBP7, cell lines overexpressing CBP7, cell lines with point mutations in the EF-hand domain, and cell lines expressing fragment proteins. was intended to reveal. CBP7 consists of 169 amino acids and contains 4EF-hand domains. The CBP7-overexpressing cells showed complete loss of developmental process. These cells remained in the single-cell growth stage under development -inducing conditions, while wild-type cells formed aggregations within 6-8h of development and eventually formed fruiting bodies. The experiments using point-mutated CBP7 protein showed that all EF-hand domains of CBP7 were important for CBP7 to function during developmental process. These results suggest that CBP7 plays an important role in developmental processes across all EF-hand domains.

사과의 성숙에 따른 Pectin질의 변화 (Changes in the Pectic Substance during Maturity of Apple)

  • Fuji 과실의 성숙에 따른 pectin 질의 변화를 조사하기 위하여 LMP (low methoxy pectin), HMP (high methoxy pectin) IAP (ionically associated pectin), CBP (covalently bounded pectin), methoxyl 및 구성당의 함량변화와 DE(degree of esterification)를 조사하였다. 그 결과 LMP+HMP와 IAP+CBP는 AIS(alcohol insoluble solid)의 각각 70%, 22%를 차지하였고 이들 성분은 성숙에 따라 다소 증가하였다. Pectose 질을 구성하는 중성당중 hexose는 LMP, HMP, IAP, CBP 다같이 $20{\sim}50%$가 감소하였는데 특히 Gal의 감소율은 63%이었다. Pentose는 $1{\sim}9%$가 증가하였고 uronic acid는 $2{\sim}4%$감소하였다. 그러나 HMP, CBP에서는 uronic acid의 상대적 함유비율은$3{\sim}5%$ 증가되었다.

  • Two intracellular enzymes, cellobiose phosphorylase (CBP) and cellodextrin phosphorylase (CDP) are involved in the phosphorolytic pathway in cellulose degradation. Those enzymes are considered to be useful in syntheses of oligosaccharides because the reactions are reversible. CBP from Cellvibrio gilvus and CDP from Clostridium thermocellum YM-4 were cloned and over-expressed in Escharichia coli. Both the enzyme reactions showed ordered bi bi mechanism. Acceptor specificity of CBP in the reverse reaction was determined. Several $\beta$-l,4-glucosyl disaccharides were synthesized by using the reaction. A new substrate inhibition pattern, competitive substrate inhibition, was also found in the reverse reaction of CBP Cellobiose was produced from sucrose at a high yield by a combined action of three enzymes including CBP

감의 연화와 관련된 세포벽다당류의 변화 (Softening Related Changes in Cell Wall Polysaccharides of Persimmon)

  • 감의 연화에 따른 세포벽다당류의 조성과 함량 및 gel여과를 통한 개략적인 분자량의 변화를 조사하였다. 조세포벽의 함량은 연화된 감에서 현저히 감소되었는데 구체적으로 ionically associated pectin (IAP )은 59%, covalently bounded pectin (CBP)은 60%, $0{\sim}3N$ KOH soluble hemicellulosic fraction($HF_2$)은 74%가 감소된 반면 4N KOH soluble hemicellulosic fraction($HF_2$)과 cellulosic fraction(CF)은 큰 변화가 없었다. 신선한 감과 연화된 감의 IAP와 CBP 구성다당류의 조성은 uronic acid외에 hemicellulose에 유래한 Pentose와 hexose가 함유되어 있었고 연화에 따라 IAP에서는 pentose가 CBP에서는 hexose가 감소되었다. 개략적인 분자량은 IAP의 400만과 25만의 Peak이 130만과 6만으로, CBP의 경우는 400만이상의 Peak이 60만과 20만으로 CBP에서의 저분자화 정도가 현저하였다. $HF_1$$HF_2$ 역시 polyuronide의 함유율이 높았는데, $HF_2$가 현저하였다. 또 연화에 따라 $HF_1$은 pentose가 $HF_2는$ uronic acid와 hexose의 손실율이 높았다. $HF_1$의 연화에 따른 재략적인 분자량은 200만의 peak이 2개로 분리 되었고 $HF_2$는 수백만 단위의 거대분자가 만단위까지 저분자화 하였다. 이상의 결과에서 감의 연화는 CBP에 인결된 hemiceldlose중 특히 $HF_2$의 저분자화와 CBP자체의 분해 내지가용화 현상과 밀접한 관련이 있는 것으로 판단되었다.

한국산 꿩으로부터 분리한 열 안정성 뉴캣슬병 바이러스의 순화와 방어효과 (Attenuation and Protective Effects of a Thermostable Newcastle Disease Virus Isolated from Korean Pheasants)

  • The objective of these experiments was to develop an attenuated thermostable Newcastle disease virus(NDV), CBP-1 strain isolated from infected pheasants. Safety, pathogenicity and protective effects against velogenic NDV were also investigated to evaluate if the attenuated NDV, CBP-1 strain could be a candidate for a new NDV vaccine strain. CBP-1 strain was passaged up to the 173 times by nine days old embryonated eggs and chicken embryo fibroblast(CEF) cell cultures. Its sensitivitly to lipid solvents and low pH, thermostability, mean death time(MDT), intracerebral pathogenicity index(ICPI) of one day old chicks and intravenous pathogenicity index(IVPI) of four weeks old chicks were examined. Safety, boosting and protective effects were tested by chicks mortality. CBP-1 NDV strain had significant thermostability at 56$\^{C}$ for 30 minutes. by hemagglutinin activity and egg infectivity test, but was not resistant to lipid solvent. It showed possibility to use as a feed or water vaccine because of the resistance to low pH. MDT, ICPI and IVPI of CBP-1 were attenuated from 51.5, 1.96, 2.60 to 112.4, 1.12, 1.45. These results implied that the 173rd passages in embryonated egg and CEF cell cultures induced a substantial attenuation of the pathogenicity of the parent virus, changing the virulence from velogenic to intermediate between mesogenic and lentogenic. After vaccination with CBP-1 at one day old by drinking water mortality was 17.5%. However, spray vaccination with B1 at one day old, CBP-1 at two weeks ild and challenge with velogenic Kyojeongwon strain at four weeks old showed 93.5% survival rate. Mortality of chicks, vaccination with 173rd passaged CBP-1 strain at one day old, two weeks old and challenge with Kyokeongwon strain at four weeks old, was 20.0%. The results of these studies indicated that partial attenuated CBP-1 strain tended to be a low safety for ND of broiler chicks and would need to be more successive attenuation.
