옥틸페놀(4-t-Octylphenol)의 Basidioradulum molare와 Schizopora paradoxa에 의한 분해 및 에스트로겐성 저감효과 (Biodegradation of 4-t-Octylphenol by Basidioradulum molare and Schizopora paradoxa and Estrogenecity Reduction of its Metabolites)

  • 계면활성제로 이용되는 난분해성 4-t-Octylphenol에 대하여 목질분해균인 큰이빨버섯과 좀구멍버섯을 선발하여 농도별 저항성 시험을 한 결과 좀구멍버섯은 200 ppm 이상의 농도에서 14일 내에 완전 균사생장(8.5 cm in petri dish)에 도달하며 큰이빨버섯은 농도의 증가에 따라 서서히 제한적인 생장을 하였다. 분해능 조사에서 좀구멍버섯은 배양 1일째 초기 투입된 4-t-Octylphenol의 95%를 분해하였으며, 큰이빨버섯은 36%의 분해능을 나타내었고 두 균주 모두 3일 이후에는 95%이상 분해하는 것으로 나타났다. 이러한 분해과정에서 좀구멍버섯은 4-t-Octylphenol 첨가에 의하여 MnP 유도효과가 있었으나 laccase는 두 균주 모두 첨가하기 전에 최고의 역가를 나타냈다. 4-t-Octylphenol 및 분해산물이 MCF-7 세포증식에 미치는 영향을 살펴본 결과 좀구멍버섯 및 큰이빨버섯 처리에 의한 분해산물의 생물학적 반응성이 하루 이후에 완전히 감소하였다. 또한, MCF-7 세포의 pS2 유전자의 발현은 처리 1일 후, 배지만 첨가한 무처리군과 동일한 양으로 감소하였다.

4-tert -octylphenol에 노출된 저서성 요각류 Tigriopus japonicus s.l.의 발생지연 현상 (Developmental Delay Effect of Harpacticoid Copepod, Tigriopus japonicus s.l. Exposure to 4-tert-octylphenol)

  • The ecotoxicological effects of 4-tert-octylphenol were observed on Harpacticoid copepoda Tigriopus japonicus s.l. gathered and cultured from tidal pool of Korean coast. There were no significant differences in survival rate (except 10 $\mu$g/L; 70.00%) and sex ratio (except 30 $\mu$g/L) on T. japonicus s.l. exposed to 4-tert-octylphenol. However, 4-tert-octylphenol induced developmental delay (copepodite emergence day and adult male mergence day) and retardation of first brooding day of adult female. Moreover the body size and biomass decreased at 4-tert-octylphenol exposure. As a result, detailed life-cycle research of T. japonicus s.l. may yield potential bioindicators for environmental monitoring and assessment.

4-t-octylphenol과 nonylphenol이 등줄쥐의 번식에 미치는 영향 (Effects of 4-t-octylphenol and Nonylphenol on the Reproduction of the Striped Field Mouse)

  • To investigate the effects of octylphenol and nonylphenol on the reproduction of the striped field mouse, the mice were subcutaneousely injected with 4-t-octylphenol 800 mg/kg (OP800), nonylphenol 900 mg/kg (NP900) or OP800 + NP900 (OPNP), respectively, in two times a week for 2 months. As the results, there were no differences between the control and the compound-treated groups in the body weight in both sexes, and the GSI (gonadosomatic index) and SVI (seminal vesicles index) in the adult males. But histological abnormalities of the reproductive organs in the mice seems to be related to the compound. This suggestion is supported by the facts that most mice treated with the compounds had only a small number of spermatozoa in the shrunken epididymal tubules. In addition, the shrunken reproductive organ in one mouse treated with the NP, suggests that the abnormalities in the wild striped field mice might be induced by the compound. Furthermore, it is indicated that the compounds must be toxicants to inhibit pregnancy, judging from the fact that all the mice treated with the compounds had failed to deliver.

Simultaneous determination of bisphenol A, chlorophenols and alkylphenols by solid-phase extraction and HPLC

  • An analytical method for determining potential endocrine disruptors (bisphenol A, 2-chlorophenol, 2,4-dichlorophenol, 2,4,6-trichlorophenol, pentachlorophenol, p-t-butylphenol, p-pentylphenol, p-hexylphenol, p-t-octylphenol, p-heptylphenol, nonylphenol) by solid-phase extraction (SPE) and High Perfomance Liquid Chromatography(HPLC) equipped with fluorescence and variable wavelength detector has been developed. The SPE process for sample concentration was performed on a commercially available Oasis HLB cartridge packed with polymeric sorbents. The effect of elution solvent and elution volume on the recoveries of the analytes were investigated with HPLC. Average recovery of >85% was achieved with 60mg sorbents using 5mL of methanol as elution solvent. Phenolic compounds in canned drinks, beverages and water samples were surveyed by this proposed method.

광양만 및 주변 하천에서의 노닐페놀 화합물 분포 (Distribution of Nonylphenol in Gwangyang Bay and the Surrounding Streams)

  • 광양만 및 주변 하천에 존재하는 페놀류 화합물과 sterol 화합물의 분포특성을 알아보기 위하여 alkylphenols (8종), chlorophenols (2종), bisphenol A (1종), coprostanols (2종) 및 cholesterol (1종) 등 화합물들을 분석대상물질로 선정하여 표층수와 표층퇴적물 시료를 분석하였다. 모든 분석대상물질 중에서 nonylphenol과 dihydrocholesterol 화합물이 가장 높은 검출빈도를 보였으며 일부 하천퇴적물 시료에서 t-octylphenol, bisphenol A, coprostanols이 검출되었다. 분석대상물질은 하천시료에서 비교적 높게 검출되었으며, 또한 광양만 내해에서 외해보다 높게 검출되었다. 표층해수와 표층해양퇴적 물에서의 nonylphenol농도는 각각 4.0∼74.0 ng $l^{-1}$ 및 3.1∼74.3 ng $g^{-1}$ dry wt.이었다. 해양표층퇴적물에 의한 nonylphenol 평균 농축배수(Log $K_{oc}$ )는 4.8이었다. 하천표층퇴적물에서의 nonylphenol농도는 4.6∼808.6 ng $g^{-1}$ dry wt.이었으며 dihydrocholesterol 농도는 78.4∼1133.6 ng $g^{-1}$ dry wt.이었다 공단지 역을 경유하는 하천시료에서는 상대적으로 높게 nonylphenol이 검출되었으며 주민 거주지역을 경유하는 하천퇴적물에서는 dihydrocholesterol 화합물이 상대적으로 높은 노출농도를 보이였다. 섬진강 표층수에서의 nonylphenol 농도는 염분도의 증가에 따라 낮아지는 경향을 보였다.

체외배양 생쥐정소세포에서 합성에스트로겐이 P450 등위효소의 발현에 미치는 영향 (Effects of Xenoestrogens on Gene Expression of Cytochrome P450 Genes in in vitro Cultured Mice Spermatogenic Cells)

  • Objective: To know the effects of xenoestrogen on spermatogenesis, we investigated the expression of cytochrome P450s enzymes (CYPscc, $CYP_{17{\alpha}}$, CYP19) and $3{\beta}$-HSD genes involved in steroidogenesis. Methods: Mouse testicular cells were prepared from 15-day-old ICR mice which had only pre-meiotic germ cells by enzyme digestion using collagenase and trypsin. Testicular cells were cultured in DMEM supplemented with FSH (0.1 IU/ml) and 10% FBS or medium with estrogen ($E_2$), bisphenol-A (BPA), octylphenol (OP; $10^{-9},\;10^{-7},\;10^{-6},\;10^{-5},\;10^{-4}M$, respectively) and aroclor 1254 (A1254) known as PCBs for 48 hours. The gene expression of cytochrome P450 enzymes were examined by semi-quantitive RT-PCR. The production of estrogen and testosterone was examined by RIA. Results: As results, expression of CYPscc mRNA was not significantly decreased, but $3{\beta}$-HSD and $CYP_{17{\alpha}}$. mRNA were significantly dose-dependent decreased. And production of testosterone and estrogen were not different except BPA and OP group ($10^{-5}M$). Conclusion: BPA, OP and A1254 might inhibit steroidogenesis by decreasing CYPscc, $3{\beta}$-HSD and $CYP_{17{\alpha}}$. mRNA expression in the mouse testis. These results suggest that BPA, OP and PCBs like as an endocrine disruptors inhibit the productions of steroidogenic enzymes and decrease the production of T and E by negative feedback mechanism. Therefore, these might disrupt steroidogenesis in Leydig cells of testis and would disturb testicular function and subsequently impair spermatogenesis.

GC/MS-SIM 방법에 의한 수돗물 중 알킬페놀, 플로로페놀과 비스페놀 A의 ppt 정량 (Part-Per-Trillion Level Determination of Alkylphenols, Chlorophenols and Bisphenol A using GC/MS-SIM in Tap Water Samples)

  • A new technique was proposed for the determination of alkylphenols, chlorophenols and bisphenol A in tap water samples. The sample preparation consists of a solid phase extraction (SPE) of alkylphenols, chlorophenols and bisphenol A from a water sample with XAD-4 and subsequent conversion to isobutyloxycarbonyl (isoBOC) derivatives or tert-butyldimethylsilyl (TBDMS) derivatives for sensitive analysis with the CC/MS SIM mode. The recoveries were 86.6 ∼ 105.2% (isoBOC derivatization) and 97.6∼484.5% (TBDMS derivatization), respectively. The limit of quantitation of alkylphenols, chlorophenols and bisphenol h for SIM were 0.001∼0.050 $\mu\textrm{g}$/1 (isoBOC derivatization) and 0.003∼0.050 $\mu\textrm{g}$/1 (TBDMS derivatization). The SIM responses were linear with the correlation coefficient varying 0.9755∼0.9981 (isoBOC derivatization), and 0.9908∼0.9996 (TBDMS derivatization). When these methods were applied to tap water samples, the range of concentrations were 22.8∼31.3 ng/1 in 2,4-dichlorophenol, 28.6∼70.3 ng/1 in pentachlorophenol, 15.2∼17.4 ng/1 in t-butylphenol, 10.8∼13.2 ng/1 in t-octylphenol and 17.6∼36.3 ng/l in bisphenol A, respectively.
