Secretory Expression of Human Growth Hormone in Saccharomyces cerevisiae Using Three Different Leader Sequences

  • A recombinant human growth hormone(hGH) was expressed as a secretory product in the yeast Saccharomyuces cerevisiae. There different leader sequences derived from the mating fac-tor $\alpha$1(MF$\alpha$1) inulinase and invertase were used to direct the secretion of hGH into the extracel-lular medium. Among three leader sequences tested, the inulinase leader sequence was found to be the most efficient in the secretory expression of hGH. In contrast, no hGH was detected in the ex-tracellular medium with the invertase leader sequence. After 48 h shake-flask culture, the yields of hGH secreted into th emedium by the invertase. MF$\alpha$1 inulinase and invertase leader sequences were approximately 0, 0.3 and 0.9 mg/L, respectively. The secretion efficiencies were also found to be 0, 3.8 and 13% for the invertase , MG$\alpha$1 and inulinase leader sequences, respectively.

학생4-H 지도교사의 자기효능감 및 역할갈등에 관한 연구 (A Study on Self-Efficacy and Role Conflict of 4-H Leaders)

  • The objectives of this study were to measure role conflict of 4-H leaders and to verify the differences of role conflict and self-efficacy according to individual characteristics. The data were collected from 99 leaders who took charge of guiding 4-H youth organization in school as teacher advisor. The statistical techniques used for this study were frequency, t-test, one-way analysis of variance and correlation. The major findings of this study were as follows; 1) The 4-H leaders had some role conflict and role ambiguity in guiding youth activities in the school. 2) The leader advisor group with teaching experience of $5{\sim}9$ years showed higher role conflict and lower role ambiguity. 3) The 4-H leaders with age over 45 felt higher role conflict and lower role ambiguity. 4)The leaders' self-efficacy was positively correlated with self esteem, and negatively associated with role ambiguity.

미국의 4-H 캠핑지도자의 역할에 대한 고찰 (A Study on the 4-H Camping Leader's Role in USA)

  • The 4-H camping programs in USA have been providing various resources so that members can achieve their physical, intellectual, social and spiritual potential. The camping programs have been carried out members to participate in programs as responsible citizens in their local, national and international communities. One of the key requirements of successful youth camping was the leadership role of leaders with more conscious and systematic for preparing, equipping, and carrying out programs. This paper was intended to explore the goals of 4-H camping and leaders' role in USA, and give implications to Korean outdoor activities. To perform the 4-H camping effectively, the camping leaders need to focus on the following steps: First of all, they need to recognize the specific purposes and missions of participation in the 4-H camping. Secondly, they should make sense of the site-based teaching skills and its application in the field. Thirdly. they should set up the principles of effective camping strategies and abide by the steps in the process of activities. In addition, they should be qualified to handle the risk management skills, research and survey technology, and environment conservation efforts throughout the camping.

The Effect of Leadership Types of Managers Perceived by ICT Organization Members on Leadership Trust and Job Satisfaction

  • We study to confirm the relationship between leadership trust and job satisfaction on the leadership style of managers recognized by ICT organization members. To this end, the research hypothesis established by using SPSS23.0 for the survey results of 230 ICT organization members was analyzed as follows. First, the hypothesis 1 person leadership type (individual consideration: H1-1, intellectual stimulation: H1-2, charisma: H1-3, conditional compensation: H1-4, exceptional reward: H1-5) had a significant effect on leader trust. The results were obtained. Second, the hypothesis 2 manager's leadership type (individual consideration: H21-1, intellectual stimulation: H2-2, charisma: H2-3, conditional compensation: 2-4, expected compensation: H2-5) is based on job satisfaction. As a result of verifying the impact relationship, all showed significant effect on job satisfaction. Third, it acted as a positive factor in the job satisfaction of the leader of the hypothesis 3-member organizational member. It was confirmed that the job satisfaction of organizational members started based on the trust of actual leaders. Fourth, the role of leadership trust was partially confirmed between leadership type and job satisfaction. Through this study, in order to improve job satisfaction of ICT organizational members, the importance of exerting competency on the leadership type of managers and trusting the leaders on the characteristics consistent with ICT organizational characteristics was emphasized.

친환경농업육성을 위한 영농 4-H조직의 활성화 방안 (A Study on the Activation of 4-H Farming Organization for development of Environmental-Friendly Agriculture)

  • In this article the author provided a brief introduction to the function and activation of 4-H fanning organization. The strategies developed to improve the environmental-friendly Agriculture are as following : the efficiency cooperatives of 4-H fanning organization, invitation of the upbringing plan of organization leader for promotion of the pilot region project of environmental-friendly Agriculture connected with green tourism, the establishment of reasonable management plan and strategies. In conclusion, there must be strategic measures to increase the income of environmental-friendly farmers. All of this information could be useful for the further development of the strategy for environmental-friendly Agriculture in organization and management.

Secretion of Bovine $\beta$-Casein by Saccharomyces cerevisiae

  • Yeast expression plasmids containing an appropriate leader sequence and bovine $\beta$-casein cDNA were constructed to produce $\beta$-casein for the study of its functional characteristics. Two kinds of expression systems for $\beta$-casein were constructed using pCGYl444 as a precursor plasmid. This plasmid is a yeast-E. coli shuttle vector which contains the chelatin promoter. The plasmid pISB202 contains the invertase leader sequence and $\beta$-casein gene. The plasmid pDEB303 contains the original bovine $\beta$-casein leader sequence gene. These two plasmids were introduced into S. cerevisiae AB116 which is a strain deficient in the major yeast proteinases. Each clone was grown in minimal media for 24 h before induction by $CuSO_4$. The cells were thus grown under expression conditions. Both strains harbouring pISB202 and pDEB303 expressed bovine $\beta$-casein. The $\beta$-casein was detected using immunochemical staining after western blot. Secretion of $\beta$-casein was detected in the culture broth. The estimated amount of secreted $\beta$-casein was approximately 50 ${\MU}g$/l.

대장균에서 과잉생산된 Bacillus licheniformis B1의 ${\beta}$-1,4-Glucanase 특성 (Characterization of Bacillus licheniformis B1 ${\beta}$-1,4-Glucanase Overproduced in Escherichia coli)

  • Bacillus licheniformis B1의 ${\beta}$-1,4-glucanase 유전자는 Escherichia coli BL21에서 발현되어, 이로부터 수용성의 50 kDa 단백질이 과량생산되었다. 반면에 B. licheniformis에서는 37 kDa의 형태가 분비되었다. E. coli에서 발현된 ${\beta}$-1,4-glucanase는 leader peptide가 제거되지 않고 포함되어 있고 Bacillus에서는 효소의 carboxy 말단에서 processing이 일어난 것으로 보인다. E. coli에서 생산된 ${\beta}$-1,4-glucanase의 최적온도는 $40^{\circ}C$이었지만, $60^{\circ}C$에서도 최대치의 76% 활성을 유지하였다. 효소의 최적 pH는 7이었고, 효소의 활성은 전체적으로 보면 약산성, 중성 및 약알칼리 영역에 널리 걸쳐 있었다. Cellulase는 식품, 세제, 펄프, 제지, 섬유산업 등 다양한 분야에서 주로 산성의 곰팡이계 효소가 이용되고 있으나, 중성 및 알칼리성 cellulase 연구 및 개발은 미흡한 편이다. 본 연구에서 개발된 중성 cellulase가 바이오 연료 개발 등의 분야에서 활용되기를 기대해 본다.

독도 심해토 메타게놈 유래 신규 내열성 에스테라아제의 생화학적 특성규명 (Biochemical Characterization of a Novel Thermostable Esterase from the Metagenome of Dokdo Islets Marine Sediment)

  • 독도 해저 2,000 미터 퇴적토를 이용한 메타게놈 유전자 은행의 60,672 클론을 기름 성분 tributyrin이 첨가된 배지에서 스크리닝 하였다. 활성을 가진 클론에서 EstES1 유전자를 선발하였다. EstES1은 553개 아미노산으로 구성된 분자량 59.4 kDa 단백질로, 가장 높은 유사성은 Haliangium ochraceum의 carboxylesterase와 44%이었다. EstES1 서열 내부에는 carboxylesterase의 전형적인 penta-peptide motif, catalytic triad 및 N-terminal 부위 37개의 leader sequence가 존재했다. 서열기반 계통분석 결과, EstES1은 신규한 esterase 임을 보여주었다. EstES1 효소의 leader 서열을 제거한 재조합 수용성 RLES1 효소는 탄소 2-12까지 포함된 long acyl ethyl ester 기질을 모두 이용할 수 있지만, p-Nitrophenyl butyrate (C4)에 가장 높은 활성과 turn-over 값을 보였다. 최적 활성은 $45^{\circ}C$, pH 9.0이다(specific activity 255.4 U/mg). 또한 강알칼리 상태인 pH 10.5까지 80% 이상의 활성이 유지되었다. EstES1의 활성은 $60^{\circ}C$에서 내열성을 보여, 1시간 동안 활성을 100% 유지할 수 있다. 효소 활성은 여러 종류의 유기용매 하에서도 안정하게 유지되었다. 따라서, EstES1은 배양이 불가능한 난배양 미생물로부터 유래된 신종 효소 유전자로서, 고온의 지방산 가수분해, 알칼리 상태나유기용매가 존재하는 여러 공정분야에 활용될 수 있다.

  • tRNA and 7SL RNA based antisense vehicles were prepared by inserting conserved anti-viral and anti-viroid domains. Anti-PVS coat protein leader sequence (ACPL) and antistructural antihairpin domain of PSTVd (AHII) were inserted in tRNA cassette; anti- zing finger domain of PVS, AHII and anti hop latent viroid ribozyme were inserted in 7SL RNA gene isolated from A. thaliana. These constructs were shown to be transcribed both, in in vitro and in in vivo conditions. However, it followed from our work that closely linked position of PoIII reference genes and PoIIII antisense genes within T-DNA lead to the impairment of RNA expression in transgenic plants. To assay in vivo transcription of antisense genes, hairy root potato cultures were established using h. tumefaciens A4-24 bearing both, Ri plasmid and PoIII-promoterless plant expression vectors with antisense RNA genes. Expression of antisense RNA in transgenic potato tissues was proven by specific RT-PCR reactions.

Exergetic design and analysis of a nuclear SMR reactor tetrageneration (combined water, heat, power, and chemicals) with designed PCM energy storage and a CO2 gas turbine inner cycle

  • The tendency to renewables is one of the consequences of changing attitudes towards energy issues. As a result, solar energy, which is the leader among renewable energies based on availability and potential, plays a crucial role in full filing global needs. Significant problems with the solar thermal power plants (STPP) are the operation time, which is limited by daylight and is approximately half of the power plants with fossil fuels, and the capital cost. Exergy analysis survey of STPP hybrid with PCM storage carried out using Engineering Equation Solver (EES) program with genetic algorithm (GA) for three different scenarios, based on eight decision variables, which led us to decrease final product cost (electricity) in optimized scenario up to 30% compare to base case scenario from 28.99 $/kWh to 20.27 $/kWh for the case study. Also, in the optimal third scenario of this plant, the inner carbon dioxide gas cycle produces 1200 kW power with a thermal efficiency of 59% and also 1000 m3/h water with an exergy efficiency of 23.4% and 79.70 kg/h with an overall exergy efficiency of 34% is produced in the tetrageneration plant.
