Effects of Dopamine and Haloperidol on Morphine-induced CREB and AP-1 DNA Binding Activities in Differentiated SH-SY5Y Human Neuroblastoma Cells

  • In the present study, we first examined whether the changes in the DNA binding activities of the transcription factors, cAMP response element binding protein (CREB) and activator protein-1 (AP-1) mediate the long-term effects of morphine in differentiated SH-SY5Y human neuroblastoma cells. The increases in CREB and AP-1 DNA binding activities were time-dependent up to 6 days of morphine treatment (1, 4, and 6 days). However, the significant reduction in the DNA binding activities of CREB and AP-1 was observed after 10 days of chronic morphine $(10\;{\mu}M)$ administration. Secondly, we examined whether the changes of CREB and AP-1 DNA binding activities could be modulated by dopamine and haloperidol. Dopamine cotreatment moderately increased the levels of the CREB and AP-1 DNA binding activities induced by 10 days of chronic morphine treatment, and haloperidol cotreatment also resulted in a moderate increase of the CREB and AP-1 DNA binding activities. However, dopamine or haloperidol only treatment showed a significant increase or decrease of the CREB and AP-1 DNA binding activities, respectively. In the case of acute morphine treatment, the CREB and AP-1 DNA binding activities were shown to decrease in a time-dependent manner (30, 60, 90, and 120 min). Taken these together, in differentiated SH-SY5Y cells, morphine tolerance seems to involve simultaneous changes of the CREB and AP-1 DNA binding activities. Our data also suggest the possible involvement of haloperidol in prevention or reversal of morphine tolerance at the transcriptional level.

Vasodilation Effect of the Water Extract of Gingko biloba, Acanthopanax senticosus and Augmented Four-Substance Decoctions in Rat Thoracic Aorta (백과엽(白果葉) 및 자오가(刺五加) 가미사물탕(加味四物湯)이 백서(白鼠)의 흉부대동맥(胸部大動脈) 혈관이완(血管弛緩)에 미치는 영향)

  • We have examined the relaxational response to the water extract of Angelica gigas $N_{AKAI}$ (AG), Gingko biloba $L_{INNE}$ (GB), Acanthopanax senticosus $H_{ARMS}.$ (AP) and Augumented-Four-Substance Decoction (AG-FSD, GB-FSD, AP-FSD) in isolated thoracic aorta from sprague dawley(SD) rat. Rat thoracic aorta was investigated in vessel segments suspended for isometric tension recording by polygraph. Responses to AG, GB, AP and AG-FSD, GB-FSD, AP-FSD were investigated in vessels precontracted with 5-hydroxytryptamine(5-HT) were compared in vasodilation effect. We found that the thoracic aorta segments responded to AG, GB, AP and AG-FSD, GB-FSD, AP-FSD with a dose-dependent vasodilation. The 5-hydroxytryptamine induced contraction at $10^{-4}M$ were inhibited by 26.3%, 75.8%, 87.5% and 6.9%, 22.6%, 30.8% after addition of the 0.1 g/mL water extract of AG, GB, AP and AG-FSD, GB-FSD, AP-FSD. In conclusion, AG, GB, AP and AG-FSD, GB-FSD, AP-FSD induced relaxation in the isolated rat thoracic aorta were composed of dose-dependent relaxation. AP-FSD has very potent vasodilation.

Effects of Bacillus SW1-1 coated diets on innate immunity and disease resistance of olive flounder Paralichthys olivaceus against Edwardsiella tarda infection

  • Bacillus SW1-1 is a probiotic isolated from shrimp intestines. We investigated the effects of Bacillus SW1-1 coated diets on the growth, feed utilization, innate immunity, hematological parameters and resistance to Edwardsiella tarda in olive flounder (Paralichthys olivaceus). A commercial diet was used as the control (AP0) and two other diets were prepared by coating 0.25% (AP25) or 0.50% (AP50) probiotic powder which contains 1.0 × 107 CFU/g Bacillus SW1-1. Four replicate groups of olive flounder (153 ± 2 g) were fed one of the diets for 12 weeks. Growth performance and feed utilization of the fish were not significantly affected by the dietary Bacillus SW1-1. After the challenge with E. tarda, AP50 group showed significantly higher survival than AP0 and AP25 groups. Innate immunity and anti-oxidant capacity of the fish were not significantly affected after the feeding trial. However, after the E. tarda challenge, the innate immune parameters (immunoglobulin, lysozyme and anti-protease) were significantly improved in fish fed AP25 and AP50 diets compared to those in fish fed AP0 diet. After the challenge test, significantly lower glucose level was observed in AP50 group compared to AP0 group. These results indicate that dietary supplementation of Bacillus SW1-1 could increase the disease resistance of olive flounder against E. tarda infection. The optimum coating levels of Bacillus SW1-1 needs to be further elucidated.

  • This study was performed in order to examine the statistical quality control of environmentally-friendly agricultural products(EF AP) and they using school meal service whether to be influences to EF AP recognition of it's parents. As a results of above research, we can make a conclusion the elementary and middle school meal service using the EF AP leads to the recognition change to parents affirmatively in Jeollanam-do.

Effects of Aralia canescens and Phellodendron amurense Extracts on Streptozotocin Induced Diabetic ICR Mice (Streptozotocin으로 당뇨를 유도한 생쥐의 간과 체장에서 황백피와 두릅나무 추출물이 지질과 산화물 생성과 글루타티온 의존성 효소의 활성에 미치는 효과)

  • The effects of Aralia canescens and Phellodendron amurense(AP) extracts on the experimental diabetes in ICR mice were investigated. 96male ICR mice were induced diabetes mellitus by intrape-ritoneal streptozotocin injection(75mg/kg B.W.) and divided into two injection groups which are 5 day injection and 10 day injection group. Then, each injection group was subdivided into 8 groups of 6 animals repspectively. CIC served as control and CI1, CI2 and CI3 were treated with 50, 150, 250mg/kg B.W. of AP extracts powder in 0.9% NaCl solution. Animals of groups DIC, DI1, DI2 and DI3 were strepto-zotocin-induced diabetes. DIC served as diabetic control and the rest groups received 50, 150, 250mg/kg B.W of AP extracts powder in saline solution respectively. The body weight, liver and kiney weight changes and blood levels of glucose, cholesterol and triglyceride were measured. Thiobarbituric acid reactive substance(TBARS), and glutathione reductase(GR) and glutathione peroxidase(GPx) activities were also measured for determining antioxidant effects. AP extracts increased the body weight in diabetic groups. The liver and kidney weight/100g B.W. in DIC group were greater than those of normal ICR group but after AP extracts injection, liver and kidney weight were decreased significantly. These effects were more efficient at 10 days injection group. The total, LDL, VLDL cholesterol and triglyceride levels were significantly higher in DIC group and the extent of decrement responded to AP injection dose. The contents of TBARS and antioxidant enzyme activities were relatively decreased after AP extracts injection. These results suggest that the intraperitoneally administered AP extracts may have not only hypoglycemic effect but act as antioxidants by reducing lipid peroxidation.

A Study on Feedstuff Utilization of Agricultural By-Products as a Major Feed Source in Korean Native Goat (농산부산물을 이용한 한국재래산양의 사료개발에 관한 연구)

  • This study was carried out to investigate the amounts of voluntary intake, digestibility and nitrogen retention in Korean native goats (KNG) fed agricultural by-products containing rice straw (RS) and apple pomace (AP) and to obtain a basic information for establishing the feeding system of KNG. The result are as follows. 1. Among the chemical composition of experimental diets, the highest values in crude protein (CP ; 18.6%) and crude ash contents (10.7%) were observed in alfalfa hay. Those of RS+AP treatment were significantly low 6.0 and 4.8%, respecitively. Acid detergent fiber (ADF), ether extract and non-structural carbohydrate (NSC) contents were shown an adverse tendency. 2. Dry matter (DM) intakes per day in KNG fed RS+commercial diet (CD), RS+AP and RS+AP+CD were significantly lower (P<0.05) 210.3, 228.3, respectively than 358.1g in alfalfa hay. 3. DM intakes per basal weight expressed as DM g/kg of BW0.75 and DM g/kg of BW(%) were highest (P<0.05) 60.5g and 3.3%, respectively in KNG fed alfalfa hay, any other treatments showed 35.6 to 42.5g and 2.0 to 2.3%, respectively and this result was similar to those of DM intake per day. 4. Digestibilities of DM, organic matter, CP, ADF, neutral detergent fiber (NDF). crude ash and ether extract in alfalfa hay and RS+AP+CD treatment were significantly higher (P<0.05) than RS+CD treatment. Those of CP, ADF and NDF of RS+AP treatment in not significantly different with these treatments. 5. NSC digestibility was higher in alfalfa hay (66.7%) than those of other treatments (44.2~52.0% ; P<0.05). 6. Nitrogen retentions expressed as N retention(g) and N retention (%) in KNG were highest (P<0.05) 0.6g and 15.6%, respectively in RS+AP+CD treatment and RS+CD treatment was lowest (P<0.05) -06g and -21.4%, respectively.

Drug Resistance and R. Plasmids of Enterobacteria Isolated from Bovine Udder Infections (젖소 유방염유래(乳房炎由來) 장내세균(腸內細菌)의 약제내성(藥劑耐性) 및 R. Plasmids)

  • A total of 250 enteric bacteria (148 Escherichia coli, 41 Klebsiella pneumoniae, 46 Enterobacter spp. and 15 Proteus spp.) isolated from bovine udder infections in 1979 through 1980 were examined for drug resistance and prevalence of R. plasmids. The drug tested were streptomycin (SM), kanamycin (KM), ampicillin (AP), chloramphenicol (CP), tetracycline (TC), gentamicin (GM), oxolinic acid (OA) and nalidixic acid (NA). The detection of R. plasmids was performed with Escherichia coli ML 1410 NAr as the recipient. Of the 148 Escherichia coli isolated, 68(45.9%) were found to be resistant to one or more drugs tested, and about 50% of the resistant strains were multiply resistant. of the 68 drugresistant strains, 13(19.1%) were found to carry R. plasmids which were capable of performing a conjugal transfer. CP resistance was transfered together with the other resistance. Of 41 strains of Klebsiella pneumoniae isolated, 90.2% were resistant to the drugs, alone or in combination thereof. Strains resistant to AP and TC were 63.4%, and 48.8%, respectively. R. plasmids were detected in 78.4% of the drug-resistant strains, and these strains transfered all or a part of their drug resistance pattern. AP and CP resistance were transfered in 100% of AP and CP-resistant strains. Eleven (37.9%) of 29 R. plasmids showed a thermosensitive transfer. Of the 46 strains of Enterobacter spp. isolated, 37(80.4%) were resistant to the drugs tested. A high percentage of resistance was noted for AP(65.2%). All strains resistant to four or more drugs transferred their resistances to Escherichia coli ML 1410, but strains resistant to three or fewer drugs did not transfer the resistances. All of the 15 Proteus strains isolated were resistant to more than two drugs. of them, 6 were quadruple resistance to SM, KM, CP and TC, and 9 were double one to AP and TC. Three (20.0%) of the drug-resistant isolates had R.plasmids conferring AP and TC resistance. GM, OA and NA of the drugs tested were very active to all of 250 Gram-negative enteric bacteria isolated from bovine udder infections.

Combined Effects of Acidification, Zeolite, and Biochar on Ammonia Emission and Nitrate Leaching from Pig Slurry

  • This study aimed to evaluate the efficiency of combining acidification with adsorbents (zeolite and biochar) to mitigate the environmental impacts of pig slurry, focusing on ammonia (NH3) emission and nitrate (NO3-) leaching. The four treatments were applied: 1) pig slurry (PS) alone as a control, 2) acidified PS (AP), 3) acidified pig slurry with zeolite (APZ), and 4) acidified pig slurry with biochar (APB). The AP mitigates NH3 emission and NO3- leaching compared to PS alone. Acidification reduced the cumulative NH3 emission and its emission factor by 35.9% and 12.5%, respectively. The APZ and APB increased NH4+-N concentration, with the highest level in APB, compared to AP. The NH4+ adsorption capacity of APB (0.90 mg g-1) was higher than that of APZ (0.63 mg g-1). The APB and APZ treatments induced less NH3 emission compared to AP. The cumulative NH3 emission was reduced by 12.2% and 27.6% in APZ and APB, respectively, compared to AP treatment. NO3- leaching began to appear on days 12 and 13, and its peak reached on days 16 and 17, which were later than AP. The cumulative NO3- leaching decreased by 17.7% and 25.0% in APZ and APB, respectively, compared to AP treatment. These results suggest that combining biochar or zeolite with acidified pig slurry is an effective method to mitigate NH3 emission and NO3- leaching, with biochar being particularly effective.

Calcium release and physical properties of modified carbonate apatite cement as pulp capping agent in dental application

  • Background: Carbonate apatite ($CO_3Ap$) and silica-calcium phosphate composite (SCPC) are bone substitutes with good prospect for dental application. SCPC creates a hydroxyapatite surface layer and stimulate bone cell function while, $CO_3Ap$ induce apatite crystal formation with good adaptation providing good seal between cement and the bone. Together, these materials will add favorable properties as a pulp capping material to stimulate mineral barrier and maintain pulp vitality. The aim of this study is to investigate modification of $CO_3Ap$ cement combined with SCPC, later term as $CO_3Ap-SCPC$ cement (CAS) in means of its chemical (Calcium release) and physical properties (setting time, DTS and pH value). Methods: The study consist of three groups; group 1 (100% calcium hydroxide, group 2 $CO_3Ap$ (60% DCPA: 40% vaterite, and group 3 CAS (60% DCPA: 20% vaterite: 20% SCPC. Distilled water was employed as a solution for group 1, and $0.2mol/L\;Na_3PO_4$ used for group 2 and group 3. Samples were evaluated with respect to important properties for pulp capping application such as pH, setting time, mechanical strength and calcium release evaluation. Results: The fastest setting time was in $CO_3Ap$ cement group without SCPC, while the addition of 20% SCPC slightly increase the pH value but did not improved the cement mechanical strength, however, the mechanical strength of both $CO_3Ap$ groups were significantly higher than calcium hydroxide. All three groups released calcium ions and had alkaline pH. Highest pH level, as well as calcium released level, was in the control group. Conclusion: The CAS cement had good mechanical and acceptable chemical properties for pulp capping application compared to calcium hydroxide as a gold standard. However, improvements and in vivo studies are to be carried out with the further development of this material.

Improved cryptanalysis of lightweight RFID mutual authentication Protocols LMAP, $M^2AP$, EMAP (경량 RFID 상호인증 프로토콜 LMAP, $M^2AP$, EMAP에 대한 향상된 취약성 분석)

  • In this paper, we present a security analysis of Lightweight RFID Mutual Authentication Protocols-LMAP[10], $M^2AP$[11], EMAP[12]. Based on simple logic operations, the protocols were designed to be suitable for lightweight environments such as RFID systems. In [8,9], it is shown that these protocols are vulnerable to do-synchronization attacks with a high probability. The authors also presented an active attack that partially reveals a tag's secret values including its ID. In this paper, we point out an error from [9] and show that their do-synchronization attack would always succeed. We also improve the active attack in [9] to show an adversary can compute a tag's ID as well as certain secret keys in a deterministic way. As for $M^2AP$ and EMAP, we show that eavesdropping $2{\sim}3$ consecutive sessions is sufficient to reveal a tag's essential secret values including its ID that allows for tracing, do-synchronization and/or subsequent impersonations.
