  • A new phenylbutenoid dimmer, ( )-trans-3-(3'-methoxy-4'-hydroxyphenyl)- 4- [(E)- 3',4'-dimethoxystyryl] cyclohexene (1), were isolated from the rhizomes of Zingiber cassumunar along with three known phenylbutenoids, ( )-trans-3-(3', 4'- dimethoxyphenyl)-4-[(E)-3'",4"'-dimethoxystyryl]cyclohexene (2), 4-(3' ,4'- dimethoxyphenyl)but-1,3-diene (3) , and 4-(2',4',5'-trimethoxy-phenyl)but-1,3-diene (4), and a known heptanoid, curcumin (7), as cytotoxic constituents against several human cancer cell lines. YOLOv4 Grid Cell Shift Algorithm for Detecting the Vehicle at Parking Lot (노상 주차 차량 탐지를 위한 YOLOv4 그리드 셀 조정 알고리즘)

  • YOLOv4 can be used for detecting parking vehicles in order to check a vehicle in out-door parking space. YOLOv4 has 9 anchor boxes in each of 13x13 grid cells for detecting a bounding box of object. Because anchor boxes are allocated based on each cell, there can be existed small observational error for detecting real objects due to the distance between neighboring cells. In this paper, we proposed YOLOv4 grid cell shift algorithm for improving the out-door parking vehicle detection accuracy. In order to get more chance for trying to object detection by reducing the errors between anchor boxes and real objects, grid cells over image can be shifted to vertical, horizontal or diagonal directions after YOLOv4 basic detection process. The experimental results show that a combined algorithm of a custom trained YOLOv4 and a cell shift algorithm has 96.6% detection accuracy compare to 94.6% of a custom trained YOLOv4 only for out door parking vehicle images.

  • The cycloaddition reaction of N-phenyl-C-cinnamonitrilimine 4 to coumarin leads to the for-mation of 3-styrylbenzopyrano[4,3-c]pyrazole derivative 6, whereas 3-phenylsulfonylcoumarin 9 or 3-bromocoumain 10 or 3-cyanocoumarin 11 gives 1-styrylbenzopyrano[3,4]pyrazole derivative 7. Also, the cycloaddition of 4 to 3-acetylocoumarin 15 and 3-benzoylocumain 16 gives the corresponding dihydropyrano[3,4-c]pyrazole adducts 17 and 18 respectively. Oxidation of 17 and 18 gives 7.

  • The application of the pervaporation process in biotechnology is rapidly growing. A two stage pervaporation process can be applied to the downstream processing of ethanol fermentation. In this paper, the second stage process in which the water-ethanol composition was 50:50 wt.% was investigated in more detail by using Nylon 4 and its blended membranes containing poly(vinyl alcohol) (PVA) and poly(acrylic acid) (PAA). Nylon 4 membranes were tested for compositions at 25, 30, and $35^{\circ}C$. Nylon 4 membranes had a separation factor of 4.18 with a permeability of $0.69kg/m^2hr$ at water-ethanol composition of 50:50 wt.%, while nylon 4-PVA blended membranes crosslinked by 5 Mrad gamma-ray irradiation showed a higher separation factor of 10.56 with permeability $0.55kg/m^2hr$ at the same composition. Nylon 4 also showed a high separation factor 27.8 at the ethanol-water azeotropic composition among the homopolymer membranes.

The Influence of Pair Programming on 4Cs Improvement in Middle School Informatics (중학교 정보교과에서 짝 프로그래밍이 4Cs 향상에 미치는 영향)

  • A model of cooperative learning, Pair Programming was applied to regular class for middle school students. Algorithm and programming skill were taught through the pair programming for two week and the achievement was analyzed based on 4Cs(Critical thinking, Communication, Collaboration, Creativity) proposed by the NEA(National Education Association). The control group was statistically significant on Critical Thinking between pre-test and post-test1, while the experimental group was statistically significant on all measurements of 4Cs. Between pre-test and post-test2, the control group showed overall little difference in range of 0.2 points on the 4Cs average scores. while the experimental group was statistically significant on all measurements of 4Cs. Between post-test1 and post-test2, there was no significant difference on 4Cs average scores in both groups. The experimental group showed higher score results on all 4Cs than the control group. This study verifies that the benefits of Pair Programming on 4Cs.

Optimization and Elucidation of Interactions between Ammonium, Nitrate and Phosphate in Centella asiatica Cell Culture Using Response Surface Methodology

  • The effects of macronutrients $(NO_3^-,\; NH_4^+\;and\;PO_4^{3-})$ on cell growth and triterpenoids production in Centella asiatica cell suspension cultures were analyzed using the Box­Behnken response surface model experimental design. In screening and optimization experiments, $PO_4^{3-}$ as a single factor significantly influenced cell growth where increasing the phosphate level from 0.1 to 2.4 or 2.6 mM, elevated cell growth from 3.9 to $14\~16g/L$. The optimum values predicted from the response surface model are 5.05mM $NH_4^+$, 15.0mM $NO_3^-$ and 2.6mM $PO_4^{3-}$, yielding 16.0g/L cell dry weight with $99\%$ fitness to the experimental data. While the $NH_4^+-NO_3^-$ interaction influenced cell growth positively in the optimization experiment, $NH_4^+$ and $NO_3^-$ as single factors; and interactions of $NO_3^--PO_4^{3-},\;NH_4^+-PO_4^{3-}$ and $NH_4^+-NO_3^-$ were all negative in the screening experiment. Cell growth and the final pH level were positively affected by $PO_4^{3-}$, but negatively affected by $NH_4^+\;and\;NH_4^+-PO_4^{3-}$ interactions. The different effects of factors and their interactions on cell growth and final pH are influenced by a broad or narrow range of macronutrient concentrations. The productions of triterpenoids however were lower than 4mg/g cell dry weight.

The Role of Nuclear Receptor Subfamily 1 Group H Member 4 (NR1H4) in Colon Cancer Cell Survival through the Regulation of c-Myc Stability

  • Nuclear receptor subfamily group H member 4 (NR1H4), also known as farnesoid X receptor, has been implicated in several cellular processes in the liver and intestine. Preclinical and clinical studies have suggested a role of NR1H4 in colon cancer development; however, how NR1H4 regulates colon cancer cell growth and survival remains unclear. We generated NR1H4 knockout (KO) colon cancer cells using clustered regularly interspaced short palindromic repeats (CRISPR)-CRISPR-associated protein-9 nuclease (CAS9) technology and explored the effects of NR1H4 KO in colon cancer cell proliferation, survival, and apoptosis. Interestingly, NR1H4 KO cells showed impaired cell proliferation, reduced colony formation, and increased apoptotic cell death compared to control colon cancer cells. We identified MYC as an important mediator of the signaling pathway alterations induced by NR1H4 KO. NR1H4 silencing in colon cancer cells resulted in reduced MYC protein levels, while NR1H4 activation using an NR1H4 ligand, chenodeoxycholic acid, resulted in time- and dose-dependent MYC induction. Moreover, NR1H4 KO enhanced the anti-cancer effects of doxorubicin and cisplatin, supporting the role of MYC in the enhanced apoptosis observed in NR1H4 KO cells. Taken together, our findings suggest that modulating NR1H4 activity in colon cancer cells might be a promising alternative approach to treat cancer using MYC-targeting agents.

  • Ninty four strains of lysine producing micro-organisms in culture broth during fermentation have been isolated from soil and other sources. From the comparison of the amounts of lysine produced, 6 strains have been selected as the potentially useful strains, and identified tentatively as Micrococcus sp. (S-16-4), Corynebactcrium sp. (S-27-12, S-281-3, CBY-4) and Brevibacterium sp. (M-6-71, F-629-2), respectively. From the further studies with Corynebacterium sp., S-27-12, its maximum yield was found to be 4mg lysine/ml of synthetic medium, consist of glucose(7.5%), urea(0.6%), $KH_2PO_4(0.2%)$, $Na_2HPO_4(0.05%)$, $MgSO_4{\cdot}7H_2O(0.03%)$, $MnSO_4{\cdot}4H_2O(0.001%)$ and $FeSO_4{\cdot}7H_2O(0.0005%)$ at pH 7.2 and $30^{\circ}C$ after 4 days.
