Lipolytic Effect of Supercritical Extraction from Pine Cone (Pinus koraiensis) in Mature 3T3-L1 Adipocytes (분화된 3T3-L1 세포에서 잣송이 초임계 추출물의 지방분해 효과)

  • Seeds of Korean pine cone (Pinus koraiensis) have long been consumed as an edible food in countries located in North-East Asia, On the other hand, Korean pine cone, containing various polyphenols, is discarded as a useless garbage after removing seeds. This study investigated the lipolytic effects of pine cone extract in differentiated 3T3-L1 adipocytes. Intracellular lipid accumulation was measured by Oil red O staining, free glycerol release by colorimetric reaction, and expression of genes related to lipid metabolism by real-time PCR. Compared to control, pine cone extract reduced intracellular lipid accumulation by 8.8% and increased free glycerol release by 8.2% a concentration of $5{\mu}g/mL$ in differentiated 3T3-L1 adipocytes. mRNA levels of fatty acid synthesis were not significantly different between control and pine cone extract, but mRNA levels of lipoprotein lipase (LPL) and hormone-sensitive lipase (HSL) significantly increased by 38.7% and 94.1% at a concentration of $5{\mu}g/mL$, respectively. Thus, pine cone extract is suggested to have lipolytic effects through induction of LPL and HSL gene expression.

  • Age and growth of the yellow goosefish, Lophius litulon, were studied using samples collected from the southwestern waters of Korea. Vertebrae of the fish had relatively clear annuli on their surface. The opaque zone of vertebrae was formed once a year between March and April. The oldest fish observed in this study was 8 years old for females and 5 years old for males. The relationship between the radius (R) of vertebral centrum and total length (L) was as follows: L=12.7+4.8R for females and L=9.8+5.6R for males. The relationship between total length and body weight (W) was as follows : $W=0.0089L^{3.0311}$ for females and $W=0.0329L^{2.7752}$ for males. Growth in length of the fish was expressed by the von Bertalanffy's equation as $L_t=127.60(1-e^{-0.1228(t+0.3851)})$ for females and $L_t=82.23(1-e^{-0.1832(t+0.6431)})$ for males.

  • 농업 비점오염원의 영향을 분석하고 여러 가지 대안(관리정책)이 수계의 수질에 미치는 영향을 평가하기 위해서 유역모형의 적용이 필요하다. 이러한 유역모형에는 모형의 복합성, 계산시간 등에 따라 simple method, mid-range model, detail model로 나눌 수 있다. Simple methods는 요구되는 자료가 작으나 그 결과의 정확성은 떨어진다. 반면에 detail model는 정확한 결과를 획득할 수 있으나 그만큼 방대한 자료와 모니터링을 요구하며, 보정 및 검증에 많은 시간과 노력을 필요로 한다. 특히, 복잡한 유역특성 및 축산오염원을 포함한 복잡한 특성들을 고려해서 모델링하기 위해서는 detail model이 필요하며 본 연구에서는 HSPF모델을 이용해서 그 영향을 분석하고자 하였다. 따라서 본 연구에서는 농업에 미치는 영향을 평가하기 위해서 청미천 유역 및 덕천천 유역을 대상유역으로 선정하여 그 영향을 분석하였다. 농업에 미치는 영향을 평가하기 위해서 유역내 투입된 비료사용량을 통계청 자료를 활용하여 산정후 적용하였으며, 축산 부산물중 자원화되는 축산 액비 및 퇴비량을 산정하여 그 영향에 대해 분석하였다. 또한 기존에 광범위하게 활용되고 있는 수질오염총량관리제도에서 산정되는 배출부하량에 유달율을 고려하여 점오염으로 입력하였으며, 직접방류되는 하수처리장도 함게 고려하여 구축하였다. 청미천 유역의 청미천1 지점의 BOD, T-N, T-P에 대한 보정기간의 평균 관측값은 4.205mg/L, 3.844mg/L, 0.137mg/L이며 검정기간의 관측값은 2.741mg/L, 4.638mg/L, 0.144mg/L로 나타났다. BOD, T-N, T-P에 대한 보정기간의 NSE는 -0.58, -0.20, -0.33이며, 검정기간의 NSE는 -0.26, -1.35, -3.54로 분석되었다. BOD, T-N, T-P에 대한 보정기간의 RMSE는 2.65, 1.63, 0.12이며, 검정기간의 RMSE는 1.36, 1.64, 0.07로 나타났다. 덕천천 유역의 경우 국가관측망이 없어 동진강의 제일 말단 지점인 동진강3 지점의 BOD, T-N, T-P에 대한 보정기간 평균관측값은 2.891mg/L, 3.455mg/L, 0.096mg/L이며, 검정기간의 관측값은 2.293mg/L, 3.223mg/L, 0.104mg/L로 나타났다. BOD, T-N, T-P에 대한 보정기간의 NSE는 -0.20, -0.56, -0.49이며, 검정기간의 NSE는 0.24, -0.06, -0.19으로 분석되었다. BOD, T-N, T-P에 대한 보정기간의 RMSE는 1.53, 2.12, 0.05이며, 검정기간의 RMSE는 1.05, 0.98, 0.06으로 나타났다. 본 연구의 분석결과는 점오염원, 축산계비점오염, 토지계비점오염, 시비량, 배경부하로 구분하여 분석을 실시하였으며 분석결과 청미천 유역의 비점오염 기여율이 약 65%를 차지하고 있는 것으로 평가되었으며, 덕천천 말단에서는 약 37.7%의 비점오염물질이 하천수질에 영향을 미치는 것으로 나타났다. 또한, 기존 수질오염 총량관리제도에서 점오염의 감소에 의한 비점오염비중이 커지는 것으로 평가되었으며, 그 중 축산이 차지하는 비중이 상대적으로 높은 것으로 평가되고 있지만, 본 연구의 결과에서는 그보다 적은 수준이 수계에 영향을 미치는 것으로 분석되었다.

Influence of Pre-operative Fasting Time on Blood Glucose in Older Patients (수술 전 금식이 노인환자의 혈당에 미치는 영향)

  • Purpose: This study was performed to identify changes in blood glucose at preoperative fasting time in surgical patients over 60 yr. Methods: Data collection was performed from July, 2008 through July, 2009. Participants consisted of 80 nondiabetic surgical patients. Blood glucose was checked from 3 to 5 times. The 5 times were 2-hr fasting on the pre-operative day (T1, n=80), 8 hr (T2, n=80), 10 hr (T3, n=17), 12 hr (T4, n=34) and 14 hr fasting on the day of the operation (T5, n=29). Results: Of the patients, 27.5% had a blood glucose level of less than 79 mg/dL at T2; 17.6% at T3; 32.4% at T4; and 17.2% at T5. Mean blood glucose levels were 93.8 mg/dL at T1; 88.4 mg/dL at T2; 91.7 mg/dL at T3; 87.4 mg/dL at T4: and 94.1 mg/dL at T5. Blood glucose was the lowest at T2 (p<.001). Conclusion: As 17.6-32.4% of the patients showed the blood glucose level of less than 79 mg/dL at 8-14 hr pre-operative fasting, the authors recommend that surgical patients p>60 yr-of-age be observed for hypoglycemia during pre-operative fasting of more than 10 hr and that surgical patients >60 yr-of-age with risks for hypoglycemia be scheduled for operation within 10 hr preoperative fasting.

Inhibitory Effects of Cultivated Wild Ginseng on the Differentiation of 3T3-L1 Pre-adipocytes

  • Wild ginseng has been used as a traditional medicine for thousands of years and for increase physical strength in Korea, China and Japan. This study reports that cultivated wild ginseng (CWG) inhibits adipocyte differentiation of 3T3-L1 pre-adipocytes in a concentration-dependent manner. Inhibition of adipocyte differentiation is one possible anti-obesity strategy. CWG inhibits the expression of the adipocyte differentiation regulator peroxisome proliferators-activated receptor (PPAR)${\gamma}$ and CCAAT/enhancer-binding protein ${\alpha}$mRNA. It also inhibited the expression of PPAR${\gamma}$ and adiponectin at the protein level during the differentiation of pre-adipocytes into adipocytes. Additionally, CWG blocked the cell cycle at the sub-$G_1$ phase transition, causing cells to remain in the pre-adipocyte state. These results indicate that CWG inhibits adipocyte differentiation and adipogenesis through pre-adipocyte cell cycle arrest in cultured 3T3-L1 cells.

Inhibitory Effects of Chrysanthemum boreale Makino on 3T3-L1 Preadipocyte Differentiation and Down-regulation of Adipogenesis and Lipogenesis (산국(Chrysanthemum boreale Makino) 꽃 유래 에센셜오일(Essential oil)이 지방세포 분화 및 지방생성에 미치는 영향)

  • Obesity is associated with an increased risk of many diseases including type 2 diabetes mellitus, hypertension, and hyperlipidemia. The flowers of Chrysanthemum boreale have been used as traditional medicines for the treatment of diseases such as obesity and type 2 diabetes mellitus. This study aimed to evaluate the effect of C. boreale Makino flower essential oil (CFEO) on adipocyte differentiation using preadipocyte cell line 3T3-L1. CFEO at concentrations between 0.1 and $5{\mu}g/ml$ did not affect 3T3-L1 cell viability. A CFEO concentration of between 0.1 and $1{\mu}g/ml$ significantly inhibited lipid accumulation during MDI-induced differentiation in 3T3-L1 cells in a dose-dependent manner, reaching a maximal level at $1{\mu}g/ml$ ($28.94{\pm}2.01%$; approximately 30% of control treated with MDI alone). Western blot analysis revealed that CFEO concentrations between 0.1 and $1{\mu}g/ml$ suppressed the activations of three adipogenic transcription factors in the MDI-stimulated 3T3-L1 cells: peroxisome proliferator-activated receptor ${\gamma}$; CCATT/enhancer binding protein ${\alpha}$; and sterol regulatory element binding protein-1. Moreover, the expressions of lipogenic enzymes, acetyl-CoA carboxylase, and fatty acid synthase were also inhibited by treatment with CFEO between 0.1 and $1{\mu}g/ml$. CFEO may therefore be a promising functional material for obesity prevention.

A Study on Hypoglycemic Effects of Crude Extracts of Schizandrae Fructus (오미자 추출물의 혈당 강하 효과에 관한 연구)

  • Hypoglycemic effect of Schizandrae Fructus (SF) extract containing in Okchun-san was determined on 3T3-L1 fibroblasts and adipocytes by investigating insulin-like activity, insulin sensitizing activity and ${\alpha}-glucoamylase$ suppressing activity. SF were extracted by using 70% ethanol followed by XAD-4 column chromatography with a mixture solvent of methanol and water, and the fractional extractions were utilized for assaying hypoglycemic effect. No inhibition of ${\alpha}-glucoamylase$ activity of SF was observed. Insulin-like activity 3T3-L1 adipocytes was not shown by SF. A significant insulin sensitizing activity of SF extractions was observed in 3T3-L1 adipocytes, giving SF extractions with 1 ng/ml insulin to reach glucose uptake level increased by 50 ng/ml of insulin alone. When cells were treated with SF (Fr. 4 or 5) plus 1 ng/ml insulin, glucose uptake was increased more than seven times as compared to 1 ng/ml of insulin alone, suggesting that SF extracts increased GLUT4 content by enhancing insulin signaling. These data suggest that SF extracts (especially Fr. 4 and 5) contains an effective insulin sensitizing compounds for hypoglycemic activity in 3T3-L1 adipocytes.

Inhibitory Effect of Ethyl Acetate Extract of White Peach Pericarp on Adipogenesis of 3T3-L1 Preadipocyte Cells

  • In order to determine whether peach contains compounds to regulate adipocyte differentiation, extracts of flesh/pericarp of yellow/white peach were prepared in water, ethyl acetate (EtOAc), or n-butanol solvent and determined for effects on adipocyte differentiation in C3H10T1/2 or 3T3-L1 cells. Interestingly, none of peach extracts has statistically significant stimulatory effect on the adipocyte differentiation in C3H10T1/2. Furthermore, the presence of EtOAc extract of white peach pericarp (WPP) was found to inhibit lipid accumulation in 3T3-L1 cells both by microscopic examination of Oil Red O-stained lipid droplets and by spectrophotometric quantification of extracted stain, indicating a significant inhibitory effect on adipocyte differentiation. The inhibition of lipid accumulation was accompanied by a significant decrease in the expression levels of adipocyte molecular markers-peroxisome proliferator-activated receptor $\gamma$, CAAT enhancer binding protein $\alpha$, and fatty acid-binding protein. Thus, this study determined that WPP EtOAc extract contains the inhibitory compound(s) on adipogenesis.

Antioxidant and Antiobesity Activities of Various Color Resources Extracted from Natural Plants (천연식물로부터 추출한 색소성분의 항산화 및 항비만 활성)

  • The objective of this study was to investigate the antioxidant and antiobesity activities of various color resources extracted from natural plants such as, clove, persimmon, gall nut, amur cork, gardenia, safflower, and annatto. Total phenolic content was the highest in gall nut extract (2,441.45 mg/kg) followed by clove extract (1,346.48 mg/kg). DPPH, and ABTS radical scavenging activities and ferric reducing antioxidant power (FRAP) were also higher in gall nut extract. ${\alpha}$-Glucosidase inhibitory activity ($IC_{50}$) was highest in persimmon extract ($22.83{\mu}g/mL$) followed by gall nut extract. ${\alpha}$-Amylase and lipase inhibitory activities were also higher in persimmon extract (49.45% and 61.01%, respectively). Lipid accumulation in 3T3-L1 cells was lower in persimmon, clove, and annatto extracts (81.54%, 83.36%, and 85.70% at $20{\mu}g/mL$, respectively). Triglyceride content in 3T3-L1 cells was lowest in clove extract (66.11%) followed by persimmon extract (88.88%). The results of this study suggest that gall nut extract has the highest antioxidant activity, whereas persimmon and clove extract show the antiobesity activities by inhibition of digestive enzymes and fat accumulation in 3T3-L1 cells. These extracts are useful materials for the development of antioxidant and antiobesity functional foods.

Mechanism of Formononetin-induced Stimulation of Adipocyte Fatty Acid Oxidation and Preadipocyte Differentiation

  • Decreased adipocyte fatty acid oxidation (FAO) and impaired preadipocyte differentiation characterize hypertrophic expansion of adipose tissue (AT) from obese and insulin resistant humans and are recognized as potential mechanisms for obesity-mediated dyslipidemia. Supplementation of formononetin (FMN), one of the principal isoflavones extracted from red clover or Huangqi (Astragalus roots), has been shown to have beneficial effects on obesity-related hyperlipidemia, a well-established cardiovascular risk factor. However, a target tissue and underlying mechanism(s) through which FMN acts have been under-investigated. Thus, we investigated whether FMN promotes adipocyte FAO and preadipocyte differentiation using 3T3-L1 preadipocytes to provide potential mechanisms of FMN action. We further extended this to the culture of 10T1/2 mesenchymal stem cells (MSCs) as well as mouse AT explants to reflect in vivo effects of FMN. In fully differentiated 3T3-L1 adipocytes, FMN-treatment significantly increased the expression levels of FAO-related proteins such as pAMPK, pACC, and CPT1, all of which were consistently upregulated in AT explant cultures treated with 10 μM FMN. In addition, FMN significantly enhanced the degree of differentiation of both 3T3-L1 preadipocytes and 10T1/2 MSCs into adipocytes as evidenced by Oil Red O staining of cellular lipids. This observation correlated with increased expression levels of key adipogenic transcription factors (PPARγ and C/EBPα) and their down-stream target proteins (FABP4, Glut4 and adiponectin). Moreover, FMN failed to exert its stimulatory effects on preadipocyte differentiation in both cell types in the presence of a PPARγ antagonist, suggesting a PPARγ-dependent effect of FMN. Collectively, these data provide possible mechanisms of action of FMN on lipid metabolism and further support the favorable in vivo effects of FMN in diet and obesity-induced dyslipidemia.
