Chemical transformation and target preparation of saponins in stems and leaves of Panax notoginseng

  • Background: Notoginsenoside Ft1 is a promising potential candidate for cardiovascular and cancer disease therapy owing to its positive pharmacological activities. However, the yield of Ft1 is ultralow utilizing reported methods. Herein, an acid hydrolyzing strategy was implemented in the acquirement of rare notoginsenoside Ft1. Methods: Chemical profiles were identified by ultraperformance liquid chromatography coupled with quadruple-time-of-flight and electrospray ionization mass spectrometry (UPLC-Q/TOF-ESI-MS). The acid hydrolyzing dynamic changes of chemical compositions and the possible transformation pathways of saponins were monitored by ultrahigh-performance LC coupled with tandem MS (UHPLC-MS/ MS). Results and conclusion: Notoginsenoside Ft1 was epimerized from notoginsenoside ST4, which was generated through cleaving the carbohydrate side chains at C-20 of notoginsenosides Fa and Fc, and vinaginsenoside R7, and further converted to other compounds via hydroxylation at C-25 or hydrolysis of the carbohydrate side chains at C-3 under the acid conditions. High temperature contributed to the hydroxylation reaction at C-25 and 25% acetic acid concentration was conducive to the preparation of notoginsenoside Ft1. C-20 epimers of notoginsenoside Ft1 and ST4 were successfully separated utilizing solvent method of acetic acid solution. The theoretical preparation yield rate of notoginsenoside Ft1 was about 1.8%, which would be beneficial to further study on its bioactivities and clinical application.

Antitumor Activity of 7-[2-(N-Isopropylamino)ethyl]-(20s)-camptothecin, CKD602, as a Potent DNA Topoisomerase I Inhibitor

  • We developed a novel water-soluble camptothecin analobue, CKD602, and evaluated the inhibition of topoisomerase I and the antitumor activities against mammalian tumor cells and human tumor xenografts. CKD602 was a nanomolar inhibitor of the topoisomerase I enzyme in the cleavable complex assay. CKD602 was found to be 3 times and slightly more potent than topotecan and camptothecin as inhibitors of topoisomerase, respecitively. In tumor cell cytotoxicity, CKD602 was more potent than topotecan in 14 out of 26 human cancer cell lines tested, while it was comparable to camptothecin. CKD602 was tested for the in vivo antitumor activity against the human tumor xenograft models. CKD602 was able to imduce regression of established HT-29, WIDR and CX-1 colon tumors, LX-1 lung tumor, MX-1 breast tumor and SKOV-3 ovarian tumor as much as 80, 94, 76, 67, 87% and 88%, respectively, with comparable body weight changes to those of topotecan. Also the therapeutic margin (R/Emax: maximum tolerance dose/$ED-{58}$) of CKD602 was significantly higher than that of topotecan by 4 times. Efficacy was determined at the maximal tolerated dose levels using schedule dependent i.p. administration in mice bearing L1210 leukemia. On a Q4dx4 (every 4 day for 4 doses) schedule, the maximum tolerated dose (MTD) was 25 mg/kg per administration, which caused great weight loss and lethality in <5% tumor bearing mouse. this schedule brought significant increase in life span (ILS), 212%, with 33% of long-term survivals. The ex vivo antitumor activity of CKD602 was compared with that of topotecan and the mean antitumor index (ATI) values recorded for CKD602 were significantly higher than that noted for topotecan. From these results, CKD602 warrants further clinical investigations as a potent inhibitor of topoisomerase I.

Factors Predicting the Interface Pressure Related to Pressure Injury in Intensive Care Unit Patients (중환자실 환자의 욕창 관련 경계압력 예측요인)

  • Purpose: Interface pressure is a factor that contributes to the occurrence of pressure injuries. This study aimed to investigate interface pressure at common sites of pressure injury (occipital, gluteal and peritrochanteric areas), to explore the relationships among risk factors, skin condition and interface pressure, and to identify risk factors influencing interface pressure. Methods: A total of 100 patients admitted to the intensive care unit were enrolled at a tertiary teaching hospital in Korea. Interface pressure was recorded by a scanning aid device (PalmQ). Patient data regarding age, pulmonary disease, Braden Scale score, body mass index, serum albumin, hemoglobin, mean blood pressure, body temperature, and oxygen saturation were included as risk factors. Data collected from July to September 2016 were analyzed using binary logistic regression. Results: The mean interface pressure of the occipital, gluteal, and right and left peritrochanteric areas were 37.96 (${\pm}14.90$), 41.15 (${\pm}16.04$), 53.44(${\pm}24.67$), and 54.33 (${\pm}22.80$) mmHg, respectively. Predictive factors for pressure injuries in the occipital area were age ${\geq}70$ years (OR 3.45, 95% confidence interval [CI]: 1.19~9.98), serum albumin deficit (OR 2.88, 95% CI: 1.00~8.26) and body temperature ${\geq}36.5^{\circ}C$ (OR 3.12, 95% CI: 1.17~8.17); age ${\geq}70$ years (OR 2.81, 95% CI: 1.10~7.15) in the right peritrochanteric area; and body temperature ${\geq}36.5^{\circ}C$ (OR 2.86, 95% CI: 1.17~6.98) in the left peritrochanteric area. Conclusion: Our findings suggest that old age, hypoalbuminemia, and high body temperature may be contributory factors to increasing interface pressure; therefore, careful assessment and nursing care of these patients are needed to prevent pressure injury. Further studies are needed to establish cutoff values of interface pressure for patients with pressure ulcers.

Litter Production and Nutrient Input via Litterfall in Quercus variabilis Forest at Mt. Worak National Park (월악산 굴참나무림의 낙엽생산과 낙엽을 통한 영양염류 이입량)

  • Litter production, nutrient concentration of each component of litterfall and amount of nutrients input to forest floor via litterfall were investigated for four years from May 2005 through April 2009 in Quercus variabilis forest at Mt. Worak National Park. Amount of litterfall in 2005, 2006, 2007, 2008 were 5.181, 5.372, 5.314, 5.361 ton $ha^{-1}yr^{-1}$, respectively. Average amount of litterfall for four years was 5.307${\pm}$0.09 ton $ha^{-1}yr^{-1}$. Average percentage of leaf litter, branch and bark, reproductive organ and the miscellaneous for four years were 67.5${\pm}$1.9, 13.3${\pm}$0.8, 10.1${\pm}$0.5 and 9.1${\pm}$1.2%, respectively. Average amount of N, P, K, Ca and Mg returned to forest floor via litterfall for four years in this Q. variabilis forest were 44.47${\pm}$6.70, 2.50${\pm}$0.25, 12.26${\pm}$0.30, 17.23${\pm}$2.32 and 9.56${\pm}$0.54 kg $ha^{-1}yr^{-1}$, respectively.

Molecular Isolation and Characterization of the 2CysPrx Gene from Salicornia herbacea (퉁퉁마디로부터 2CysPrx 유전자 분리 및 특성 분석)

  • This study is focused on the investigation of the genes which are induced by various stresses of the halophyte Salicornia herbacea. One of the factors influencing in the germination of Salicornia herbacea is salt stress. The highest germination rate was found in the condition without NaCl, and the upper limit of the NaCl concentration for the germination of Salicornia herbacea was 7%. The optimal temperature of $20^{\circ}C$showed a germination rate of 98%. Among genes induced by stress the 2CysPrx gene was cloned and analyzed for this study. The 2CysPrx gene has two cysteine conserved residues and is composed of 275 amino acids with molecular weight of 30.1kDa. The 2CysPrx gene appeared to be one copy in the genome and consists of 6 introns and 7 exons. Quantitative real-time PCR revealed that the highest transcription rate induced by NaCl and $H_2O_2$ appeared to be at the concentration of 3.5% NaCl and 40mM $H_2O_2$, respectively. The amount of transcript induced by high temperature($40^{\circ}C$) and $75{\mu}M$ of ABA was respectively highest. The gene at low temperature ($4^{\circ}C$) appeared not to be expressed. We are conducting to clone other peroxyredoxin genes induced by various environmental stresses.

The association of PBX1 polymorphisms with overweight/obesity and metabolic alterations in the Korean population

  • Pre-B-cell leukemia transcription factor 1 (PBX1), which is located on chromosome 1q23, was recently reported to be associated with type 2 diabetes mellitus. We examined whether single nucleotide polymorphisms (SNPs) of the PBX1 gene are associated with overweight/obesity in a Korean population. We genotyped 66 SNPs in the PBX1 gene and investigated their association with clinical phenotypes found in 214 overweight/obese subjects and 160 control subjects using the Affymetrix Targeted Genotyping chip array. Seven SNPs (g.+75l86C>T, g.+78350C>A, g.+80646C>T, g.+138004C>T, g.+185219G>A, g.+191272A>C, and g.+265317T>A) were associated with the risk of obesity in three models (codominant, dominant, and recessive) (P=0.007-0.05). Haplotype 1 (CAC) and 3 (TAC) of block 3 and haplotype 2 (GGAAT) of block 10 were also strongly associated with the risk of obesity. In the control group, subjects that had homozygote for the major allele for both g.+185219G>A and g.+191272A>C showed lower high density lipoprotein-cholesterol (HDL-C) level compared to those possessing the minor allele, suggesting that the association between the homozygote for the major allele for both g.+185219G>A and g.+191272A>C and HDL-C is attributable to the increased risk of obesity. This study suggests that the PBX1 gene is a possible risk factor in overweight/obese patients.

A Numerical Study for Stability of Tunnel in Jointed Rock Using Barton-Bandis Model (BB절리모델을 활용한 절리암반속 터널안정성의 수치해석적 연구)

  • For the pertinent use of NMT method, both characteristics of joints (JRC, JCS and ${\phi}_r$) and characteristics of rock mass (Q-Value) must be investigated carefully. The main objective of the study presented is to investigate how sensitive the predicted behaviour of an underground excavation is to various realistic assumptions about some input parameter for the jointed rock mass. Joint pattern in the tunnel is predicted by statistical approach (chi-square test). In this paper, sensitivity studies involving in joint characteristics were carried out. The parametric studies involving change in Barton-Bandis joint model have shown that JCS is relatively insensitive to JRC and ${\phi}_r$. An increase in JRC value may not, according to the Barton-Bandis model, necessarily lead to a decrease in displacement. The importance of dilation in predicting the behaviour of a rock mass around an excavation is emphasized from a comparison of the Barton-Bandis joint behaviour model with the Mohr-Coulomb model. The Barton-Bandis model predicted higher stress, which allow for the build-up of stress caused by dilatant behaviour.

Vegetation Type and Stand Structure of Pinus densiflora Forests in Kangwon Northern Region in Korea (강원북부지역 소나무림의 식생유형과 임분구조)

  • The objectives of this study were to analyze vegetation type and stand structure of the red pine (Pinus densiflora) in Kangwon northern region for stable and sustainable management forests. The pine forests in study sites were classified into 3 communities, 5 groups, and 2 subgroups, total 7 vegetation units. Species with constance degree of more than 61% were P. densiflora, Quercus mongolica, Lindera obtusiloba, Spodiopogon sibiricus, Atractylodes japonica, Rhododendron mucronulatum, and Carex humilis. They were showing different characteristics by pattern according to their growing district. In the importance value (I.V.) analysis of each layer, P. densiflora showed highly in tree layer while in other layers Quercus spp. was high. Especially, young P. densiflora tree hardly appeared in the herb layer, but broad-leaved trees and shrub species showed high I.V.. Furthermore, the annual ring growth of P. densiflora was reduced while that of Q. spp. increased. According to analysis of stand structures and annual ring growths, it is considered that appropriate silvicultual practice methods should be employed to remove rival broad-leaved species for maintenance of sustainable red pine forests considering the characteristics of each stand.

Effects of ${\beta}2-Adrenergic$ Receptor Polymorphism on Obesity (베타 2-아드레날린 수용체의 유전자 변이형이 비만도에 미치는 영향)

  • Objectives : The lipolytic effects of catecholamines in adipose tissue are mediated by members of adrenergic receptors. This study was conducted to examine the effects of ${\beta}2-AR$ Gln27Glu (Q27E) polymorphism on obesity indices and risk among obesity clinic patients. Methods : 532 subjects, 38 men and 494 women, who attended a weight loss program in a local obesity clinic were analyzed. Height, weight, BMI, WHR and obesity index were measured or calculated. Body composition was measured by bio-impedance analysis. Genotype of ${\beta}2-AR$ polymorphism in codon 27 was analyzed by PCR-RFLP method. Serum concentrations of fasting glucose, total and HDL cholesterol, and triglyceride were determined by autobiochemical analyzer. Results: The Genotype distributions of ${\beta}2-AR$ gene were QQ type 81.3%, QE type 17.9% and EE type 8%. Therefore, the frequency of E allele of ${\beta}2-AR$ gene was 0.170 in the total subjects. The frequency of the ${\beta}2-AR$ variant genotype(QE+EE) was significantly higher in obese group($BMI{\geqq}25$) compared with non-obese group(p=0.027). Weight was significantly higher in variant(QE+EE) type compared with normal(QQ) type in total subjects(p=0.001), male(p=0.022) and female(p=0.013). BMI, obesity index and WHR were also significantly higher in QE+EE type. Body fat man was significantly higher in QE+EE type in total subjects(p=0.005) and female(p=0.027). When forward stepwise regression analysis was used to create a model of risk predictors of obesity($BMI{\geqq}25$), QE+EE type of ${\beta}2-AR$ gene was found to be a significant risk factor for obesity (p=0.042, ORs 1.597). Conclusions: QE+EE genotype of ${\beta}2-AR$ was associated with increased obesity indices and might be a significant risk factor for obesity.

Integrated Photonic Microwave Bandpass Filter Incorporating a Polymer Microring Resonator (폴리머 마이크로링 공진기를 이용한 광학적 마이크로웨이브 대역통과 필터)

  • An integrated photonic microwave bandpass filter has been proposed and demonstrated incorporating a coherently coupled microring resonator in low-loss polymers. The proposed device may feature compact site, simple structure, tuning via the thermooptic and electrooptic effect, and flexible integration with other electrical and optical devices. The resonator was designed to have an extremely small bandwidth so that it could be used to selectively pass the optical signal carrying the microwave signal to attain efficient bandpass filtering. We made and tested two resonators with a single ring and double rings, and performed a theoretical fitting of their measured transfer curves to predict the performance of the microwave filters based on them precisely. It was found that as the number of the rings used for the resonator increases, the bandwidth gets smaller, the rolloff sharper, and the band rejection higher. Finally our filter exhibited the center frequency of 10GHz, the 3-dB bandwidth of 1.0GHz, the corresponding quality (Q) factor of 10, and the rejection outside of the passband of more than 25dB.
