Synthesis and Luminescent Properties of Tetrafluorophenyl Containing Poly(p-phenylenevinylene) Derivatives

  • To investigate the effect of fluoro groups substitution on poly(p-phenylenevinylene) derivatives, poly(2,3,5,6- tetrafluoro-p-phenylenevinylene-alt-N-ethylhexyl-3,6-carbazolevinylene), PCTF-PPV, and poly[2,3,5,6-tetrafluoro-p-phenylenevinylene-alt-2-methoxy-5-(2-ethylhexyloxy)-p-phenylenevinylene], PMTF-PPV, were synthesized by the well-known Wittig condensation polymerization process. To compare the influences of fluoro groups, no fluoro groups containing model polymers, poly(p-phenylenevinylene-alt-N-ethylhexyl-3,6-carbazolevinylene), PCPPPV and poly[p-phenylenevinylene-alt-2-methoxy-5-(2-ethylhexyloxy)-p-phenylenevinylene], p-PMEH-PPV, were also synthesized. The resulting polymers were completely soluble in common organic solvents and exhibited good thermal stability up to 300℃. The polymers showed UV-visible absorbance and photoluminescence (PL) in the ranges of 259~452 nm and 500~580 nm, respectively. The tetrafluorophenyl containing PCTF-PPV and PMTF-PPV showed relatively red-shifted PL peaks at 521 nm and 580 nm, respectively, compared to that of non-fluoro groups containing polymers (PCP-PPV: 500 nm and p-PMEH-PPV: 539 nm). The single-layer light-emitting diode was fabricated in a configuration of ITO/polymer/Al. Electroluminescene (EL) emissions of PCP-PPV, PCTF-PPV, p-PMEH-PPV and PMTF-PPV were shown at 507, 524, 556, and 616 nm, respectively.

Efficient Gene Targeting using Nuclear Localization Signal (NLS) and Negative Selection Marker Gene in Porcine Somatic Cells

  • The specific genetic modification in porcine somatic cells by gene targeting has been very difficult because of low efficiency of homologous recombination. To improve gene targeting, we designed three kinds of knock-out vectors with ${\alpha}1,3$-galactosyltransferase gene (${\alpha}1,3$-GT gene), DT-A/pGT5'/neo/pGT3', DT-A/NLS/pGT5'/neo/pGT3' and pGT5'/neo/ pGT3'/NLS. The knock-out vectors consisted of a 4.8-kb fragment as the 5' recombination arm (pGT5') and a 1.9-kb fragment as the 3' recombination arm (pGT3'). We used the neomycin resistance gene (neo) as a positive selectable marker and the diphtheria toxin A (DT-A) gene as a negative selectable marker. These vectors have a neo gene insertion in exon 9 for inactivation of ${\alpha}1,3$-GT locus. DT-A/pGT5'/neo/pGT3' vector contain only positive-negative selection marker with conventional targeting vector. DT-A/NLS/pGT5'/neo/pGT3' vector contain positive-negative selection marker and NLS sequences in upstream of 5' recombination arm which enhances nuclear transport of foreign DNA into bovine somatic cells. pGT5'/neo/pGT3'/NLS vector contain only positive selection marker and NLS sequence in downstream of 3' recombination arm, not contain negative selectable marker. For transfection, linearzed vectors were introduced into porcine ear fibroblasts by electroporation. After 48 hours, the transfected cells were selected with $300{\mu}g/ml$ G418 during 12 day. The G418-resistant colonies were picked, of which 5 colonies were positive for ${\alpha}1,3$-GT gene disruption in 3' PCR and southern blot screening. Three knock-out somatic cells were obtained from DT-A/NLS/ pGT5'/neo/pGT3' knock-out vector. Thus, these data indicate that gene targeting vector using nuclear localization signal and negative selection marker improve targeting efficiency in porcine somatic cells.

Effects of Different Dietary Acidifier Sources of Calcium and Phosphorus on Ammonia, Methane and Odorant Emission from Growing-finishing Pigs

  • The objective of this study was to investigate the effects of different sources of Ca and P on urine and ileal digesta pH, and ammonia ($NH_{3}$), methane ($CH_{4}$), and odor emission. In experiment 1, eight pigs (commercial three-way cross; initial BW 67$\pm$3 kg) were arranged in a repeated 4$\times$4 Latin Square design. All pigs were equipped with a T-cannula in the distal ileum. Four corn-soybean meal based diets were formulated. Diet 1 was the control in which dicalcium phosphate (DCP) and limestone ($CaCO_{3}$) were used as the sources of inorganic P and Ca. In Diets 2 and 3, ${H_{3}}{PO_{4}}$, monocalcium phosphate (MCP), and $CaSO_{4}$replaced DCP and $CaCO_{3}$ as the inorganic sources of P and Ca. Diet 4 was similar to Diet 1 except that it was fortified with HCl to provide an acid load similar to that of diet 2. Urine and ileal digesta pH were determined in pigs fed each of these diets. In Exp. 1, urine pH decreased (p<0.05) in animals consuming diets containing ${H_{3}}{PO_{4}}$-$CaSO_{4}$ (5.85$\pm$0.38) and MCP-$CaSO_{4}$(5.73$\pm$0.30) compared with the DCP-$CaCO_{3}$ diet (6.89$\pm$0.24). In the pigs consuming ${H_{3}}{PO_{4}}$-$CaSO_{4}$, ileal digesta pH decreased compared with the control (5.52$\pm$0.28 vs. 6.66$\pm$0.17; p<0.05). Based on the results of Exp. 1, a total of four trials were performed in environmental chambers for determining how $NH_{3}$, $NH_{4}$, and odor were affected by the different dietary Ca and P sources (Exp. 2). In Exp. 2, pigs fed the ${H_{3}}{PO_{4}}$-$CaSO_{4}$ diet had decreased (30%) $NH_{3}$ emissions compared with the control (p<0.05). Also, a combination of MCP-$CaCO_{3}$-$CaCl_{12}$ decreased $NH_{3}$ emission by 15% (p<0.05). Emission of $CH_{4}$ was decreased only with the ${H_{3}}{PO_{4}}$-$CaSO_{4}$ diet with 14% (p<0.05). Odorant emission of phenolics and volatile fatty acids increased roughly three-fold with the DCP-$CaSO_{4}$ diet but was not affected by other test diets. In conclusion, acidogenic Ca and P sources in swine diets can decrease the urinary pH and reduce $NH_{3}$ and $CH_{4}$ emission from swine facilities.

  • The purpose of this study is to know the change of brain wave with stress by SUKI pressure alternative therapy. The experiment group was 12 students(male 6/female 6) with complained upper trapezius pain. Calculated the brain wave before and after stimulate the upper trapezius trigger point by SUKI and the stimulate time was 3min each persons(FP2, F3, F4, FP1, T3, T4, P3, P4). the experimet periods was 5times in a weeks with EEG(QEEG-S). The Date collecting used by Telescan(LXE5209). All the data was analyzed with SPSS 22.0 for window program. To compare the differences before and after the point pressure method, a corresponding sample of t-test was performed, and the statistical significance level was p<.05. The results was followed. The points of Fp2, F3, F4(*p<0.049, *p<0.042, *p<0.019) of EEG was showed a significant differences but Fp1, T3, T4, P3, P4 points did not showed. The SUKI alternative medicine techniques had a reduced effects for the some kind of brain stress. It is need to continuous research in the future.

Experimental Evaluation of the Effect of the Mixing Design Factors of the Cementitious Composite for 3D Printer on the Printing Quality (3D 프린터용 시멘트 복합체의 배합요인에 따른 출력 품질의 실험적 평가)

  • In this paper, to evaluate the output quality of the cementitious composite mixture for printing with the ME method for construction 3D printer, visual inspection of the output appearance and the dimensional error rate, compressive strength and flexural strength of the output were measured. As a result of the test, the mixing design with excellent output appearance was P1-2, P1-4, P2-5, P2-6, and the mixing design with good output appearance was P0-1, P1-1, P1-3, P1-6, P1-7 and P2-4. Of these mixing designs, P0-1 and P2-6 had the lowest dimensional error rates As a result of evaluating the compressive strength and flexural strength of the various mixing designs, the Mixing design with excellent output designs showed good mechnical properties. However, mixing designs with excellent mechanical properties does not necessarily have excellent output quality. Therefore, in order to accurately evaluate the output quality, it is judged that visual inspection and dimensional error rate inspection should be performed first, and then the mechanical characteristics should be reviewed.

STAT3 is Activated in a Subset of Benign and Malignant Chondroid Tumors (양성 및 악성 연골 종양에서의 STAT3 활성화)

  • Purpose: STAT3 is an oncogene that regulates critical cellular processes, and its constitutive activation has been demonstrated to correlate with biological and clinical features in many types of human malignancy. Materials and Methods: In this study, STAT3 activation was assessed in variable benign and malignant chondroid tumors in bone by immunohistochemistry using a monoclonal antibody specific for $tyrosine^{705}$-phosphorylated STAT3 ($pSTAT3^{tyr705}$). Results: Among conventional chondrosarcomas (n=17), three cases(50%) of grade III chondrosarcomas were pSTAT3-positive. All grade I and II chondrosarcomas were pSTAT3-negative. This pSTAT3 positivity according to the histologic grade was statistically significant (p=0.0432). Two cases(50%) of clear cell chondrosarcomas were pSTAT3-positive. Six cases (50%) among 12 benign chondroid tumors(6 enchondromas, 3 chondroblastomas, and 3 chondromyxoid fibromas) were also $pSTAT3^{tyr705}$-positive. Conclusion: In conclusion, STAT3 activation is associated with higher tumor grade in conventional chondrosarcomas. Our results suggest that STAT3 is activated in a subset of benign and malignant chondroid tumors, and may support the extension of the cancer stem cell hypothesis to include tumors of cartilaginous lineage.

Purification of Inositol Triphosphate Kinase from Bovine Brain (소의 뇌로부터 Inositol Triphosphate Kinase의 정제)

  • Inositol 1,4,5-triphosphate($InsP_3$) is a second messenger for mobilizing intracellular $Ca^{2+}$. It can be dephosphorylated by soluble and particulate forms on $InsP_3$ 5-phosphatase, or phosphorylated to produce inositol 1,3,4,5-tetrakisphosphate($InsP_3$) by $InsP_3$ 3-kinase. These enzymes represent possible targets for the regulation of the $InsP_3/InsP_4$ signal. $InsP_3$ 3-kinase which catalyses th ATP-dependent phosphorylation of $InsP_3$ was purified from bovine brain tissue. All operation were carried out at $4^{\circ}C$. Fresh tissure was homogenized and centrifuged. The supernatant was pooled. Proteins were precipitated from 10% polyethylene glycol, and suspended solution was applied to DEAE cellulose column for chromatography. As the result of above procedure, two isozymes of $InsP_3$ 3-kinase, I and II were obtained. Each isozyme was applied to Matriz green gel, Calmodulin-Affigel 15 column and subsequent phenyl-TSK HPLC column. Specific activites(SA) and fold of puriety were observed at each purification step of chromatography. At DEAE cellulose chromatography, SA were I, 0.6 and II, 4.8 nM/min/mg, and folds were I, 17.2 and II, 16.6. At Matrix green gel chromatography, SA were I, 18 and II, 11 nM/min/mg, folds were I, 62.1 and II, 38.0. At calmodulin-Affigel 15 column chromatography, SA were I, 19 and II, 13 nM/min/mg, folds were I, 65.5 and II, 44.8. Finally $InsP_3$ kinase I and II were purified 3,103-fold and 2,310-fold, and SA were I, 900 and II, 670 nM/min/mg, respectively. SDS-polyacrylamide gel electrophoresis elucidated 3 distinct fractions of Mr of 145,000, 85,000 and 69,500 from isozyme I, and 2 distinct fractions of Mr of 79,000 and 57,000 from isozyme II.

  • For the production of poly(3-hydroxybutyrate-co-3-hydroxyvalerate)(P(3HB-co-3HV)) from butyric acid and valeric acid, 10 strains of bacteria capable of producing P(3HB-co-3HV) were isolated from soil. Among them, the strain HJ-067 showed the best ability of producing P(3HB-co-3HV), and was indentified as a Azotobacter sp. For the production of P(3HB-co-3HV), the optimum concentrations of butyric and valeric acid were 3.0g/l, respectively. The most effective nitrogen source was $(NH_4)_{2}SO_4$ at an optimum concentration of 0.75g/l, which was equivalent to 21.36 in C/N ratio. Deficiency of the cationic metal ions ($Zn^{2+},\;Co^{2+},\;Mn^{2+}$) in the proguction medium had stimulating effect on P(3HB-co-3HV) accumulation, especially in the manganese. deficient medium. The optimum temperature for P(3HB-co-3HV) production was 27$^{\circ}C$ and the optimum initial pH was 7.0. Under the optimum conditions, 1.82g/l of P(3HB-co-3HV) and 3.00g/l of dry biomass were produced after 36 hour cultivation, and the P(3HB-co-3HV) yield and HV% were 60.60% (w/w), 15.92%, respectively.

Comparative Genetic Characterization of Plasmids of Agrobacterium Species Isolated in Korea (한국산 Agrobacterium plasmid의 유전학적 성상에 관한 연구)

  • The soil bacterium Agrobacterium tumefaciens is a plant pathogen that cause3 crown gall tumors by infecting the wounded dicotyledonous plants and subsequent integration of bacterial DNA into plant nuclear DNA. Virulent A. tumefaciens strains harbor a large Ti (tumor-inducing) plasmid that carries genes essential for tumorigenesis. In the present study, 13 strains (Malus pumila Mill; $A_{1-3}$, Populus monilifera; $W_{1-6}$, Populus tomentiglandlosa; $P_{1-3}$ and Rosa species; $R_1$) of Agrobacterium isolated in korean crown gall tumors and plasmids were observed in 6 strains ($W_2$, $W_3$, $W_6$, $P_1$, $P_3$ and $A_2$). The test for crown gall tumor formation was resulted only in ATCC15955 and $KW_2$ strains inoculated into the stem of sun flower and the development was observed for 4 and 6 weeks after inoculation. Above two Ti plasmids (pTi) were purified by cesium chloride-ethidium bromide density gradient centrifugation and digested with restriction enzyme and fragments of pTiATCC 15955 and $pTiKW_2$ observed by EcoR I ; 25&27, Hind III; 23&21, BamH I ; each 20 and Hpa I ; 12&27, and sizes of pTiATCC15955 and and $pTiKW_2$ calculated as 200 and 87 kbases. Octopine was isolated from tumor tissue ($W_{1-6}$ and $P_{1-3}$) and these strains confirmed as octopine type.

Effect of the Water Soluble Extracts from Velvet Antler on Lipid Metabolism and Blood Components in Rats (수용성 녹용 추출물이 랫드의 지방대사 및 혈액성분에 미치는 영향)

  • An experiment was conducted with 80 male rats for 4 weeks to investigate the effect of water soluble extracts(WSE) from different sections of farmed Elk(Cervus elaphus) antler on lipid metabolism and hematology. Antlers were divided into four sections of tip, upper, middle and base. Water soluble extract(35g/60ml) was prepared from each section of antler, and was administered orally to male Sprague- Dawley rats(10ml/kg body weight) once a day. Administration of WSE from upper, middle and base sections of antler decreased(P<0.0027) AST values at 3 weeks of feeding, however AST value was decreased(P< 0.0136) in the WSE of base section compared to that of control at 4 weeks of feeding. Administration of antler WSE for 4 weeks decreased total cholesterol in visceral fat(P<0.0035) and liver(P<0.0003) at 4 weeks of feeding compared to those of control. Feeding antler WSE for 4 weeks increased the compositions of C14:0(P<0.0037), C16:1(P<0.0061), C18:1(P<0.0066), C18:2(P< 0.0069) and C18:3(P<0.0035) in the visceral fat compared with that of control.
