The Effect of Glutamine on Production of Tissue-type Plasminogen Activator from Recombinant Human Melanoma Cells in Glutamine-limited Fed-batch Cultivation

  • Under glutamine-limited condition, $2\times10^6$ (viable cells/ml) of maximum cell density and 13.5 ($\mu g$/ml) of tissue-type Plasminogen Activators (tPA) production were maintained by spike feeding fresh medium in fed-batch cultivation of human recombinant melanoma cells. It showed that tPA production was much seriously affected than cell growth according to initial glutamine concentrations. Above 3.4 (mmol/I) of glutamine concentration both cell growth and tPA production were not much affected by increasing initial glutamine concentration. Glutamine depleted situation was occurred at latter periods of batch and fed-batch cultivations below 5.4 (mmole/I) of initial glutamine concentration. It also showed that maximum glutamine consumption and ammonia evolution rates were closely related to initial glutamine concentrations. Maximum specific tPA production rate was estimated as $8.1\times19^{-6}$ ($\mu g$/cells/h) at 3.4(mmol/I) of glutamine concentration, which is higher than that from other batch and fed-batch processes.

The Phase-Shift Method for the Langmuir Adsorption Isotherms at the Noble Metal (Au, Rh) Electrode Interfaces (귀금속(Au, Rh) 전극계면에서 Langmuir 흡착등온식에 관한 위상이동방법)

  • The Langmuir adsorption isotherms of the over-potentially deposited hydrogen (OPD H) fur the cathodic $H_2$ evolution reaction (HER) at the poly-Au and $Rh|0.5M\;H_2SO_4$ aqueous electrolyte interfaces have been studied using cyclic voltammetric and ac impedance techniques. The behavior of the phase shift $(0^{\circ}{\leq}{-\phi}{\leq}90^{\circ})$ for the optimum intermediate frequency corresponds well to that of the fractional surface coverage $(1{\geq}{\theta}{\geq}0)$ at the interfaces. The phase-shift profile $({-\phi}\;vs.\;E)$ for the optimum intermediate frequency, i.e., the phase-shift method, can be used as a new electrochemical method to determine the Langmuir adsorption isotherm $({\theta}\;vs.\;E)$ of the OPD H for the cathodic HER at the interfaces. At the poly-Au|0.5M $H_2SO_4$ aqueous electrolyte interface, the equilibrium constant (K) and the standard free energy $({\Delta}G_{ads})$ of the OPD H are $2.3\times10^{-6}$ and 32.2kJ/mol, respectively. At the poly-Rh|0.5M $H_2SO_4$ aqueous electrolyte interface, K and ${\Delta}G_{ads}$ of the OPD H are $4.1\times10^4\;or\;1.2\times10^{-2}$ and 19.3 or 11.0kJ/mol depending on E, respectively. In contrast to the poly-Au electrode interface, the two different Langmuir adsorption isotherms of the OPD H are observed at the poly-Rh electrode interface. The two different Langmuir adsorption isotherms of the OPD H correspond to the two different adsorption sites of the OPD H on the poly-Rh electrode surface.

Bioethanol Production from Seaweed Undaria pinnatifida Using Various Yeasts by Separate Hydrolysis and Fermentation (SHF) (갈조류 미역(Undaria pinnatifida)의 분리당화발효와 다양한 효모를 이용한 바이오에탄올의 생산)

  • Bioethanol was produced using the separate hydrolysis and fermentation (SHF) method with macroalgal polysaccharides from the seaweed, Undaria pinnatifida as biomass. This study focused on the pretreatment, enzymatic saccharification, and fermentation of yeasts in co-culture. Ethanol fermentation with 14.5% (w/v) seaweed hydrolysate was performed using the yeasts, Saccharomyces cerevisiae KCTC 1126 alone, Pichia angophorae KCTC 17574 alone, and their co-cultures with the yeasts either adapted to mannitol or not. Among the combinations, the co-culture of non-adapted S. cerevisiae and P. angophorae adapted to mannitol showed high bioethanol production of 12.2 g/l and an ethanol yield ($Y_{EtOH}$) of 0.41. Co-culture in the SSF process was employed in this study, to increase the ethanol yields of 35.2% and reduction of 33.3% in fermentation time. These results provide suitable information on ethanol fermentation with marine seaweeds for bioenergy production.

  • Recent observations of Sloan Digital Sky Survey (SDSS) for the ugriz filters have provided the largest and the most homogeneous photometric database. Especially, the ugriz systems have been used in many telescopes, such as SDSS, CFHT, and Gemini, and will be applied next generation large telescopes. In addition, many stellar evolution models, like as PADOVA, BaSTI and DSEP, have introduced theoretical isochrones in ugriz system, to apply the isochrones into the photometry of stars in globular clusters. In this study, we transformed Yonsei-Yale($Y^2$) isochrones to ugriz photometric system and fit the isochrones to the (g-r, r), (g-i, r), and (r-i, r) CMDs of 13 globular clusters (Chun et al. 2009, Clem et al. 2008, An et al. 2008). We found that the derived ages from $Y^2$-isochrones are 1~3Gyr younger and the estimated distance moduli are 0.3mag larger than the values inferred from the other isochrones in r-band. Also, the E(B-V) are 0.02~0.04mag less than those estimated from the other models. The ages of each globular cluster estimated from $Y^2$-isochrones are 12~14Gyr and distance moduli show good agreement with previous studies. From this result, we confirmed the availability of the $Y^2$-isochrones in gri filters. However, it is a problem which will improve that E(B-V) values obtained from $Y^2$-isochrones are too small. Finally, the result of this study is expected to be used research of globular cluster in ugirz photometric system.

The Effect of Solidification Rates and Thermal Gradients on Directionally Solidified Microstructure in the Ni-base Superalloy GTD111M (GTD111M 초내열합금에서 응고속도 및 온도구배가 일방향응고 조직 에 미치는 영향)

  • Morphological evolution and growth mechanism at the solid/liquid interface during solidification were investigated in the Ni-base superalloy GTD111M by directional soldification and quenching(DSQ) technique. The experiments were conducted by changing solidification rate(V) and thermal gradient(G) which are major solidification process variables. High thermal gradient condition could be obtained by increasing the furnace temperature and closely attaching the heating and cooling zones in the Bridgeman type furnace. The dendritic/equiaxed transition was found in the G/V value lower than $0.05$\times$10{^3}^{\circ}C$s/$\textrm{mm}^2$, and the planar interface of the MC-${\gamma}$ eutectic was found under $17 $\times$ 10{^3}^{\circ}C$ s/$\textrm{mm}^2$. It was confirmed that the dendrite spacing depended on the cooling rate(GV), and the primary spacing was affected by the thermal gradient more than solidification rate. The dendrite lengths were decreased as increasing the thermal graditne, and the dendrite tip temperature was close to the liquidus temperature at $50 \mu\textrm{m}$/s.

Genetic Variations of Trichophyton rubrum Clinical Isolates from Korea

  • Trichophyton rubrum is one of the well-known pathogenic fungi and causes dermatophytosis and cutaneous mycosis in human world widely. However, there are not an available sequence type (ST) classification methods and previous studies for T. rubrum until now. Therefore, currently, molecular biological tools using their DNA sequences are used for genotype identification and classification. In the present study, in order to characterize the genetic diversity and the phylogenetic relation of T. rubrum clinical isolates, five different housekeeping genes, such as actin (ACT), calmodulin (CAL), RNA polymerase II (RPB2), superoxide dismutase 2 (SOD2), and ${\beta}$-tubulin (BT2) were analyzed using by multilocus sequence typing (MLST). Also, DNA sequence analysis was performed to examine the differences between the sequences of Trichophyton strains and the identified genetic variations sequence. As a result, most of the sequences were shown to have highly matched rates in their housekeeping genes. However, genetic variations were found on three different positions of ${\beta}$-tubulin gene and were shown to have changed from $C{\rightarrow}G$ (1766), $G{\rightarrow}T$ (1876), and $C{\rightarrow}A$ (1886). To confirm the association with T. rubrum inheritance, a phylogenetic tree analysis was performed. It was classified as four clusters, but there was little significant correlation. Even so, MLST analysis is believed to be helpful for determining the genetic variations of T. rubrum in cases where there is more large-scale data accumulation. In conclusion, the present study demonstrated the first MLST analysis of T. rubrum in Korea and explored the possibility that MLST could be a useful tool for studying the epidemiology and evolution of T. rubrum through further studies.

Efficient Modulation for the Last Symbol in OFDM Systems (OFDM 시스템의 마지막 심볼을 위한 효율적인 변조 방식)

  • OFDM modulation has been used for a transmission scheme in 4G LTE (Long Term Evolution) and Wi-Fi systems to mitigate the effects of frequency selective fading channels. An OFDM modulation is a block transmission scheme because an OFDM symbol consists of multiple subcarriers with narrow bandwidth. Therefore, all OFDM symbols in a frame should be filled out with data and padding bits. Depending on the amount of data, more padding bits than information bits can occupy the last OFDM symbol. Such inefficiency causes the loss of throughput. To overcome this problem, an efficiency padding method is proposed by using the property of DFT (Discrete Fourier Transform). In the proposed method, symbol duration of the last symbol is changed depending on the number used data subcarriers in the last symbol. With numerical evaluation, it is examined that throughput enhancement achieved by the proposed method can be about 20% depending a transmission scheme and data length.

RECYCLING PROCESS OF U3O8 POWDER IN MnO-Al2O3 DOPED LARGE GRAIN UO2 PELLETS

  • The effect of various process variables on the powder properties of recycled $U_3O_8$ from MnO-$Al_2O_3$ doped large grain $UO_2$ pellets and the effect of those recycled $U_3O_8$ powders on the sintered density and grain size of MnO-$Al_2O_3$ doped large grain $UO_2$ pellets have been investigated. The evolution of morphology, size, and BET surface area of the recycled $U_3O_8$ powders according to the respective variation of the thermo-mechanical treatment variables of oxidation temperature, powder milling, and sequential cyclic heat treatment of oxidation and then reduction was examined. The correlation between the BET surface area of recycled $U_3O_8$ powder and the sintered pellet properties of MnO-$Al_2O_3$ doped pellets showed that the pellet density and grain size of doped pellets were increased and then saturated by increasing the BET surface area of the recycled $U_3O_8$ powder. The density and grain size of the pellets were maximized when the BET surface area of the recycled $U_3O_8$ powder was in the vicinity of $3m^2/g$. Among the process variables applied in this study, the cyclic heat treatment followed by low temperature oxidation was a potential process combination to obtain the sinter-active $U_3O_8$ powder.

  • $Al_2O_3$ composites powders with 1∼11 wt% $TiO_2$ were prepared by wet method and sintered at 1350$^{\circ}C$, 1450$^{\circ}C$ for 2h. Mechanical properties and microstructural evolution were investigated in this study. $Al_2O_3$-3 wt% $TiO_2$ composite were high bulk density of 2.37 g/$cm^3$ and low apparent porosity of 6.3%. The composites containing of 3 wt% $TiO_2$ showed moderately high bending strength of 68.9 MPa and the young's modulus of 35.5 MPa. The composites with increasing $TiO_2$ contents exhibit reduced thermal expansion coefficient due to the formation of $Al_2TiO_5$ phase.

Impacts of wave and tidal forcing on 3D nearshore processes on natural beaches. Part II: Sediment transport

  • This is the second of two papers on the 3D numerical modeling of nearshore hydro- and morphodynamics. In Part I, the focus was on surf and swash zone hydrodynamics in the cross-shore and longshore directions. Here, we consider nearshore processes with an emphasis on the effects of oceanic forcing and beach characteristics on sediment transport in the cross- and longshore directions, as well as on foreshore bathymetry changes. The Delft3D and XBeach models were used with four turbulence closures (viz., ${\kappa}-{\varepsilon}$, ${\kappa}-L$, ATM and H-LES) to solve the 3D Navier-Stokes equations for incompressible flow as well as the beach morphology. The sediment transport module simulates both bed load and suspended load transport of non-cohesive sediments. Twenty sets of numerical experiments combining nine control parameters under a range of bed characteristics and incident wave and tidal conditions were simulated. For each case, the general morphological response in shore-normal and shore-parallel directions was presented. Numerical results showed that the ${\kappa}-{\varepsilon}$ and H-LES closure models yield similar results that are in better agreement with existing morphodynamic observations than the results of the other turbulence models. The simulations showed that wave forcing drives a sediment circulation pattern that results in bar and berm formation. However, together with wave forcing, tides modulate the predicted nearshore sediment dynamics. The combination of tides and wave action has a notable effect on longshore suspended sediment transport fluxes, relative to wave action alone. The model's ability to predict sediment transport under propagation of obliquely incident wave conditions underscores its potential for understanding the evolution of beach morphology at field scale. For example, the results of the model confirmed that the wave characteristics have a considerable effect on the cumulative erosion/deposition, cross-shore distribution of longshore sediment transport and transport rate across and along the beach face. In addition, for the same type of oceanic forcing, the beach morphology exhibits different erosive characteristics depending on grain size (e.g., foreshore profile evolution is erosive or accretive on fine or coarse sand beaches, respectively). Decreasing wave height increases the proportion of onshore to offshore fluxes, almost reaching a neutral net balance. The sediment movement increases with wave height, which is the dominant factor controlling the beach face shape.
