Establishement of Antibody Selection by Ribosome Display (Ribosome Display를 이용한 항체선별 방법의 확립)

  • Background: Phage display is the most widely used technique among display methods to produce monoclonal antibody fragment with a specific binding activity. Having a large library for efficient antibody display/selection is quite laborious process to have more than $10^9$ members of transformants. To overcome these limitations, several in vitro selection approaches have been reported. Ribosome display that links phenotypes, proteins, directly to genotype, mRNA, is one of the in vitro display methods. Ribosome display can reach the size of scFv library up to $10^{14}$ molecules and it can be further diversified during PCR steps. To select the high affinity scFv from one pot library, we established ribosome display technique by modifying the previously reported eukaryotic translation system. Methods: To establish the antibody selection system by ribosome display, we used 3D8, anti-DNA antibody. A 3D8 scFv was synthesized in vitro by an in vitro transcription-translation system. The translated 3D8 scFv and the encoding 3D8 mRNA are connected to the ribosome. These scFv-ribosome-mRNA complexes were selected by binding to their specific antigens. The eluted mRNAs from the complexes are reverse transcribed and re-amplified by PCR. To apply this system, antibody library from immunized mouse with terminal protein (TP)-peptide of hepatitis B virus DNA polymerase TP domain was also used. This TP-peptide encompasses the 57~80 amino acid residues of TP. These mRNA/ribosome/scFv complexes by our system were panned three times against TP-peptide. The enrichment of antibody from library was determined by radioimmunoassay. Results: We specifically selected 3D8, anti-DNA antibody, against ssDNA as a model system. The selected 3D8 RNAs sequences from translation complexes were recovered by RT-PCR. By applying this model system, we enriched TP-peptide-specific scFv pools through three cycles of panning from immunized library. Conclusion: We show that our translating ribosome complexes are well maintained and we can enrich the TP-specific scFv pools. This system can be applied to select specific antibody from an antibody library.

A Time-multiplexed 3d Display Using Steered Exit Pupils

  • This paper presents the multi-user autostereoscopic 3D display system constructed and operated by the authors using the time-multiplexing approach. This prototype has three main advantages over the previous versions developed by the authors: its hardware was simplified as only one optical array is used to create viewing regions in space, a lenticular multiplexing screen is not necessary as images can be produced sequentially on a fast 120Hz LCD with full resolution, and the holographic projector was replaced with a high-frame-rate digital micromirror device (DMD) projector. The whole system in this prototype consists of four major parts: a 120Hz high-frame-rate DMD projector, a 49-element optical array, a 120Hz screen assembly, and a multi-user head tracker. The display images for the left/right eyes are produced alternatively on a 120Hz direct-view LCD and are synchronized with the output of the projector, which acts as a backlight of the LCD. The novel steering optics controlled by the multiuser head tracker system directs the projector output to regions referred to as exit pupils, which are located in the viewers’eyes. The display can be developed in the "hang-on-the-wall"form.

  • In this paper, a novel 2D/3D convertible display system based on integral imaging is proposed. Combining two liquid crystal display panels with integral imaging, it is possible to convert the display between two-dimensional mode and three-dimensional mode without mechanical movement. The proposed method is proven by preliminary experiments.

  • Various kinds of tile-based ultra-high resolution display devices have been developed by, for example, constructing display walls using many commodity LCD displays. To represent 3D applications like computer games on these devices, one has to develop 3D applications or develop particular APIs only for representing on these devices. If one can develop a distributed rendering system using legacy 3D APIs such as OpenGL and Direct3D by extending a remote rendering system, commercial computer games can be represented on such display devices without modifying their source codes. The purpose of the paper is to propose a new Dired3D-based distribute rendering system by extending a Direct3D-based remote rendering system and to show its feasibility technically by appling it to a sample Direct3D application and performing a few experimentations.

  • A reflection-type three-dimensional (3D) screen using retroreflector is proposed to improve the visibility of a projected 3D image while keeping its perspective. For the projection-type 2D display, the diffuser is used to represent the 2D scene, overcoming the limitation of the aperture of the projection lens set. If the diffuser is adopted for the projected 3D image, only 2D images sectioned and blurred should be displayed on the screen. The proposed screen can make the 3D image with the aperture limitation visible to be applied to the 3D image projection systems. The feasibility of the proposed screen is verified by experiments.

  • This paper describes research results that 2-D contents, which display in smart phone or tablet PC to be able to see the 3-D stereoscopic by use projection technique. For this research, we have construct four brown-glass into pyramid shape, project each of the four LCD monitors that output from the PC screen into of the four inverted pyramid-shaped mirror and display the 3-D image to the center of the mirror system. For the test, We use tablet PC and server PC(desktop PC) connected by wireless network, tablet PC select contents which is displayed in the server PC, and displayed selected contents into the 3-D image to the center of the mirror system. Through the test, we have showed that it is possible to display 3-D stereoscopic to the 2-D contents by use projection technique. Although, display image is depending on the observer's viewing angle.

The Study on the Fabrication and Characterization of Dielectric Materials of Front and Back Panel for PDP(Plasma Display Panel)

  • The glass compositions of $PbO-SiO_2-B_2O_3$ system and $P_2O_5-PbO-ZnO$ system for the transparent dielectric materials for front panel and $P_2O_5-ZnO-BaO$ and $SiO_2-ZnO-B_2O_3$ for the reflective dielectric materials for back panel of PDP(Plasma Display Panel) were investigated. As a transparent dielectric materials for front panel, $PbO-SiO_2-B_2O_3$ glass showed good dielectric properties, high transparency and proper thermal expansion matching to soda-lime glass substrate. And the reflective dielectric materials for back panel were prepared from parent glass of $SiO_2-ZnO-B_2O_3$ system and oxide filler. It was found that these glass-ceramics are useful materials for reflective dielectric layers, as those have a similar thermal expansion to soda-lime glass plate, high reflectance, low sintering temperature.

  • In this paper, we describe the development of the polarized-light stereoscopic monitoring system based on full HDTV. The system consists of the stereoscopic camera part, image processing device for the stereoscopic image recording and stereoscopic display. The developed stereoscopic imaging display system reduces eyestrain and viewer fatigue.

  • This paper introduces two systems for editing and evaluating 3D content. The systems, a non-linear editing system and an evaluation system, were developed to improve viewing safety and comfort for 3D content observers and provide better usability for the creators from the viewpoint of ergonomics.
