DEVELOPMENT OF NEW WHITENING AGENT. THE INHIBITORY EFFECTS OF LAGENARIA LEUCANTHA ON MELANOGENESIS AND DEPIGMENTATION EFFECT OF GOLD FISH

  • In this study, we demonstrated the whitening effect of Lagenaria leucantha through the melanin biosynthesis of S bikiniensis and inhibition of melanogenesis in cultured Bl6 melanocytes. And we confirmed the whitening effect of Lagenaria leucantha through the depigmentation of gold fish in vivo. The melanogenesis of B$_{16}$ melanocytes was founded to be activated dose and time dependently by the treatment of u- MSH. When the B$_{16}$ melanocytes was treated with 200nM of $\alpha$-MSH, the morphology of melanocytes was remarkably changed. The melanin content and the synthesis of tyrosinase were strikingly increased. Lagenaria ieucantha inhibited the melanin formation stimulated by $\alpha$-MSH without affection of cell viability. However, Lagenaria leucantha didn't inhibit tyrosinase activity and showed weak suppression on the synthesis of tyrosinase. These results suggest Lagenaria leucantha might inhibit melanin formation with tyrosinase independent manner. Lagenaria ieucantha also inhibition melanin biosynthesis with 18mm inhibition zone in S.bikiniensis. To evaluate the inhibitory activity of melanogenesis of Lagenaria leucantha in vivo, we examined its effect on depigmentation of gold fish. Lagenaria ieucantha remarkably reduced the size and density of melanophores in gold fish. These results suggest that Lagenaria ieucantha can be used as a whitening agent in cosmetics.ics.s.

  • Respectively the powder made of ZnO added $SnO_2$ was prepared by coprecipitation method and the thick film gas sensor was fabricated by screen-printing technique, The morphology and phase of the powder and film was investigated by SEM and XRD. The specific area of the particle was linearly increased with ZnO contents. Target gas was di(propylene glycol) methylether ($CH_3$($OC_3$$H_{6}$ )$_2$OH, DPGME), which is simulant gas of blister gas. The gas sensing characteristics for DPGME were examined with flow type measurement system and the concentrations of target gas were controlled from 500 ppb to 1500 ppb. ZnO (2 wt%) added $SnO_2$ showed maximum sensitivity to DPGME at $300^{\circ}C$.

Functional Anaylsis of sprD Gene Encoding Streptomyces griseus Protease D(SGPD) in Streptomyces griseus

  • The chromosomal sprD gene encoding Streptomyces griseus protease D (SGPD), a chymotrypsin-like protease, was disrupted in Streptomyces griseus by insertion of the neomysin-resistance gene. The production of chymotrypsin activity of sprD disruptant was not completely abolished, but delayed by 24 h, compared with that of wild-type strain. The aerial mycelial formation of sprD disruptant was retarded, and specifically the formation of spores was not observed in the central region of colonies. However, normal morphological development into spores was observed in the marginal region of colonies. In addition, the production of yellow pigment that might be dependent on A-factor was also decreased in the sprD disruptant, compared with that of the wild-type strain. Introduction of the sprD gene, which was placed on a high copy-numbered plasmid into S. griseus ${\Delta}sprD$, partially restored the ability of morphological development, and a significant level of sporulation was observed. When the overexpression vector for sprD, pWHM3-D, was introduced in S. griseus, there was no significant change in the chymotrypsin activity or colonial morphology, in contrast to Streptomyces lividans, indicating the presence of a tight regulation system for the overexpression of the sprD gene in S. griseus.

Effect of dietary energy levels on growth performance, blood parameter and intestinal morphology of Pekin ducks in low ambient temperature

  • This study was carried out to investigate the effects of dietary energy levels on growth performance, blood parameter, and intestinal morphology of Pekin ducks in low temperature. A total of 500, 21-d-old Pekin ducks (initial BW = 1,089 ± 5.21 g) were evenly assigned to five dietary treatments (2,950, 3,000, 3,050, 3,100, or 3,150 kcal AME/kg, calculated on an as-is basis) with four replicates (pens) for each treatment (25 ducks per pen). During the experiment, hens were provided with feed and water ad libitum. Overall, increasing dietary energy levels corresponded to an increase of final body weight and body weight gain (linear, p < 0.01). Feed intake decreased (linear, p < 0.01) and feed conversion ratio increased (linear, p < 0.01) with increasing levels of energy. There were no significant differences (p < 0.05) in the level of leukocytes between groups. However, heterophils decreased (quadratic, p < 0.05) and lymphocytes increased (linear, p < 0.01) as inclusion of dietary energy levels increased. The H/L ratio increased (linear, p < 0.01) with increasing dietary energy levels while serum corticosterone levels decreased at overall experimental periods. Triglycerides increased (linear and quadratic, p < 0.05) with increasing dietary energy levels. There were no significant changes in villus height or crypt depth of the jejunum at overall experimental. In conclusion, increasing concentrations of dietary energy levels up to 2,950-3,150 kcal/kg in diet. Additionally, 3,150 kcal/kg dietary energy had been revealed more beneficial and could be practiced as protective management for the Pekin ducks reared under low ambient temperature (8℃ to 10℃).

  • 한국과학기술정보연구원과 가톨릭대학교 의과대학 가톨릭응용해부연구소에서는 기초기술연구회 National agenda project 일환으로 척추 연구자들이 쉽게 사용할 수 있는 기초 자료를 구축하고 있다. 컴퓨터 시뮬레이션 연구에 활용할 수 있는 모델 제작을 위한 형상 정보를 제공하기 위해 60대, 70대 기증시신 20여 표본을 활용하여 고해상도 척추 (whole spine) CT (pixel dimension : 0.4xmm, thickness: 0.6mm)를 촬영하고 이를 3차원 모델링 소프트웨어(Mimics, Ver.14, Materialise, Belgium)를 사용하여 3차원 형상 모델(shell model, STL format)로 구축하고 있다. 또한 3차원 형상 모델을 활용하여 목, 등, 허리 척추의 주요 부위를 계측하여 수치화 하였다. 시신 기반 자료의 한계를 극복하기 위해 척추센터를 내원한 환자 중 고령자 호발 질환을 중심으로 선정하여 X-Ray, 3D CT, MRI, BMD 자료를 구축하여 보강하고 있다.

  • In oxide matrix-SiC(W) composites, instability and glassy phase formation due to oxidation at the high temperature and the diffusion of Si, respectively, cause brittle fracture and low reliability for ceramic materials. The mode of contribution in each mechanisms induced by matrix-whisker debonding, varies with the morphology of matrix-whisker interfaces. This work has described the dispersion behaviours and stabilization mechanisms in slip systems, and multiple toughening mechanisms by dint of two second phase different from each other when spherical ZrO2 and chemically stable Al2O3(W) is respectively added in Al2O3 matrix. To obtain complexshaped components, slip casted bodies were sintered at 1$600^{\circ}C$, 2hrs up to 98~99% R.D.. Multiple toughening mechanisms in comparison with theories reported until now will be discussed as a result of the phase analysis of ZrO2 by athermal behaviours and microstructural characterizations as well as measured mechanical properties.

Microwave Sol-Gel Preparation of NaLa(MoO4)2:Eu3+/Yb3+ Particles and Their Upconversion Photoluminescence Properties

  • $NaLa_{1-x}(MoO_4)_2:Eu^{3+}/Yb^3$ phosphors with doping concentrations of $Eu^{3+}$ and $Yb^{3+}$ ($x= Eu^{3+}+Yb^{3+}$, $Eu^{3+}=0.05$, 0.1, 0.2 and $Yb^{3+}= 0.2$, 0.45) were successfully synthesized by the microwave-modified sol-gel method, and the upconversion and spectroscopic properties were investigated. Well-crystallized particles showed a fine and homogeneous morphology with particle sizes of $2-5{\mu}m$. Under excitation at 980 nm, $NaLa_{0.5}(MoO_4)_2:Eu_{0.05}Yb_{0.45}$ particles exhibited a strong 525-nm emission band and a weak 550-nm emission band in the green region, and a very weak 665-nm emission band in the red region. The strong 525-nm emission in the green region corresponds to the $^7F_1{\rightarrow}^5D_1$ transition and the weak 550-nm emission in the green region corresponds to the $^7F_0{\rightarrow}^5D_2$ transition, while the very weak emission 665-nm band in the red region corresponds to the $^5D_0{\rightarrow}^7F_3$ transition. The Raman spectra of the doped particles indicated the domination of strong peaks at higher frequencies of 762, 890, 1358 and $1430cm^{-1}$ and weak peaks at lower frequencies of 323, 388 and $450cm^{-1}$ induced by the disorder of the $[MoO4]^{2-}$ groups with the incorporation of the $Eu^{3+}$ and $Yb^{3+}$ elements into the crystal lattice or by a new phase formation.

Physical and electrical properties of PLA-carbon composites

  • Polylactic acid or polylactide (PLA) is a biodegradable thermoplastic that can be produced from renewable material to create various components for industrial purposes. In 3D printing technology, PLA is used due to its good mechanical, electrical, printing properties, environmentally friendly and non-toxic properties. However, the physical properties and excellent electrical insulation properties of PLA have limited its application. In this study, with the carbon black (CB) as filler added into PLA, the lattice spacing and morphology were investigated by using X-ray diffraction (XRD) and scanning electron microscope (SEM), respectively. The physical properties of PLA-carbon composite were evaluated by using tensile test, shore D hardness test and density and voids measurement. Impedance test was conducted to investigate the electrical properties of PLA-Carbon composites. The results demonstrate that the inclusion of carbon black as filler enhances the physical properties of the PLA-carbon composites, including tensile properties, hardness, and density. The addition of carbon black also leads to improved electrical conductivity of the composites. Better enhancement toward the electrical properties of PLA-carbon composites is observed with 1wt% of carbon black in N774 grade. The N550 grade with 2wt% of carbon black shows better improvement in the physical properties of PLA-carbon composites, achieving 10.686 MPa in tensile testing, 43.330 in shore D hardness test, and a density of 1.200 g/cm3 in density measurement. The findings suggest that PLA-carbon composites have the potential for enhanced performance in various industrial applications, particularly in sectors requiring improved physical and electrical properties.

Effects of Feeding Solid-state Fermented Rapeseed Meal on Performance, Nutrient Digestibility, Intestinal Ecology and Intestinal Morphology of Broiler Chickens

  • This trial was conducted to determine the effects of feeding a diet containing solid-state fermented rapeseed meal on performance, nutrient digestibility, intestinal ecology and intestinal morphology of broiler chickens. A mixed liquid culture, containing approximately 5 log cfu/ml Lactobacillus fermentum, Enterococcus faecium, Saccharomyces cerevisae and Bacillus subtilis was prepared in a 1:1:1:1 ratio. A basal substrate (BS) containing 75% rapeseed, 24% wheat bran and 1% brown sugar was mixed with the liquid culture in a ratio of 10:3. Over the 30-day fermentation, isothiocyanates were reduced from 119.6 to 14.7 mmol/kg. A total of 168, day-old male Arbor Acres broiler chicks were assigned to one of three dietary treatments including a corn-soybean meal based control diet as well as two experimental diets in which the control diet was supplemented with 10% of the BS containing unfermented rapeseed meal or 10% of the BS containing rapeseed meal subjected to solid state fermentation. There were 8 pens per treatment and 7 birds per pen. From days 19-21 and days 40-42, uncontaminated excreta were collected from each pen for digestibility determinations. In addition, digesta from the colon and ceca were collected to determine the number of lactobacilli, enterobacteria and total aerobes. The middle sections of the duodenum, jejunum, and ileum were collected for intestinal morphology. Over the entire experimental period (d 1-42), the weight gain and feed conversion of birds fed fermented rapeseed meal were superior (p<0.05) to that of birds fed nonfermented rapeseed meal and did not differ from the soybean control. On day 42, birds fed fermented rapeseed meal had higher (p<0.05) total tract apparent digestibility coefficients for dry matter, energy, and calcium than birds fed non-fermented rapeseed meal. Colon and ceca digesta from broilers fed the fermented feed had higher (p<0.05) lactobacilli counts than birds fed the control and non-fermented rapeseed meal diets on day 21 and 42. Fermentation also improved (p<0.05) villus height and the villus height:crypt depth ratio in the ileum and jejunum on day 21 and 42. The results indicate that solid-state fermentation of rapeseed meal enhanced performance and improved the intestinal morphology of broilers and may allow greater quantities of rapeseed meal to be fed to broilers potentially reducing the cost of broiler production.

Dietary protease improves growth performance, nutrient digestibility, and intestinal morphology of weaned pigs

  • This study was conducted to evaluate effects of dietary protease (PR) on growth performance, nutrient digestibility, and intestinal morphology of weaned pigs. A total of 75 weaned pigs [7.06 ± 0.18 kg of average body weight (BW); 28 day old] were randomly allotted to 3 dietary treatments in a randomized complete block design (blocks = BW and sex): a diet based on corn and soybean meal to meet the requirement of crude protein (CP) as a positive control (PC; CP = 24.49%), a low protein diet as a negative control (NC; CP = 22.51%), and NC + 0.02% PR. The PR used in this study was a commercial product containing 75,000 protease units/g derived from Nocardiopsis prasina produced in Bacillus licheniformis. Pigs were fed the dietary treatments for 6 weeks and the diets containing 0.2% chromic oxide for the last week of this study. Blood, feces, ileal digesta, and ileum samples were collected from randomly selected two pigs in each pen on respective time points. Measurements were growth performance, apparent ileal digestibility (AID) and apparent total tract digestibility (ATTD) of dry matter (DM), CP, and energy, frequency of diarrhea, packed cell volume (PCV), and ileal morphology of weaned pigs. Pigs fed PC and PR had higher (p < 0.05) final BW, average daily gain (ADG), and gain to feed ratio (G:F) during overall experimental period than those fed NC. Pigs fed PC and PR had higher (p < 0.05) AID or ATTD of DM, CP, or energy than those fed NC. Moreover, pigs fed PR had higher ratio between villus height and crypt depth (p < 0.05) and number of goblet cells (p < 0.05) than those fed NC. Addition of PR decreased (p < 0.05) frequency of diarrhea for the first two weeks after weaning compared with PC and NC. In addition, pigs fed PR had lower (p < 0.05) PCV on d 14 after weaning than those fed PC and NC. In conclusion, addition of PR in nursery diets with a low protein level significantly improved growth performance, nutrient digestibility, and intestinal morphology of weaned pigs.
