Vitamin C (ascorbic acid) is an essential component for collagen biosynthesis and also for the proper functioning of the cardiovascular system in humans. Unlike most of the animals, humans lack the ability to synthesize ascorbic acid on their own due to a mutation in the gene encoding the last enzyme of ascorbate biosynthesis. As a result, vitamin C must be obtained from dietary sources like plants. In this study, we have developed two different kinds of transgenic potato plants (Solanumtuberosum L. cv. Taedong Valley) overexpressing strawberry GalUR and mouse GLoase gene under the control of CaMV 35S promoter with increased ascorbic acid levels. Integration of the these genes in the plant genome was confirmed by PCR and Southern blotting. Ascorbic acid(AsA) levels in transgenic tubers were determined by high-performance liquid chromatography(HPLC). The over-expression of these genes resulted in 2-4 folds increase in AsA intransgenic potato and the levels of AsA were positively correlated with increased geneactivity. The transgenic lines with enhanced vitamin C content showed enhanced tolerance to abiotic stresses induced by methyl viologen(MV), NaCl or mannitol as compared to untransformed control plants. The leaf disc senescence assay showed better tolerance in transgenic lines by retaining higher chlorophyll as compared to the untransformed control plants. Present study demonstrated that the over-expression of these gene enhanced the level of AsA in potato tubers and these transgenics performed better under different abiotic stresses as compared to untransformed control. We have also investigated the mechanism of the abiotic stress tolerance upon enhancing the level of the ascorbate in transgenic potato. The transgenic potato plants overexpressing GalUR gene with enhanced accumulation of ascorbate were investigated to analyze the antioxidants activity of enzymes involved in the ascorbate-glutathione cycle and their tolerance mechanism against different abiotic stresses under invitro conditions. Transformed potato tubers subjected to various abiotic stresses induced by methyl viologen, sodium chloride and zinc chloride showed significant increase in the activities of superoxide dismutase(SOD, EC 1.15.1.1), catalase, enzymes of ascorbate-glutathione cycle enzymes such as ascorbate peroxidase(APX, EC 1.11.1.11), dehydroascorbate reductase(DHAR, EC 1.8.5.1), and glutathione reductase(GR, EC 1.8.1.7) as well as the levels of ascorbate, GSH and proline when compared to the untransformed tubers. The increased enzyme activities correlated with their mRNA transcript accumulation in the stressed transgenic tubers. Pronounced differences in redox status were also observed in stressed transgenic potato tubers that showed more tolerance to abiotic stresses when compared to untransformed tubers. From the present study, it is evident that improved to lerance against abiotic stresses in transgenic tubers is due to the increased activity of enzymes involved in the antioxidant system together with enhanced ascorbate accumulated in transformed tubers when compared to untransformed tubers. At moment we also investigating the role of enhanced reduced glutathione level for the maintenance of the methylglyoxal level as it is evident that methylglyoxal is a potent cytotoxic compound produced under the abiotic stress and the maintenance of the methylglyoxal level is important to survive the plant under stress conditions.
농작물 생산에 있어서 병원균 침입에 의한 피해를 줄이는 것은 아주 중요한 과제이다. 식물은 스스로도 생체 방어 신호 전달 기작을 가지고 있지만 그 피해를 줄이는데 있어서 한계가 있다. 본 연구는 콩과 식물에서 분리한 식물생체방어 신호전달 유전자인 SCaM-5를 토마토에 형질전환하여 형질전환 식물체를 작성하고 병 저항성에 관한 실험을 수행한 것이다. SCaM-5 유전자가 형질전환 된 토마토에서는 pathogen-related(PR-5) 유전자를 항상 발현시킴으로서 계속적으로 식물 생체방어 신호기작을 활성 시킨다는 사실을 확인하였다. 또한, SCaM-5를 형질전환 시킨 토마토는 식물에 막대한 피해를 주는 중요한 곰팡이 (P. capsici와 F. oxysporm)와 bacteria (Pst DC3000)에 대하여 병 저항성을 가진다는 것을 검증하였다.
본 연구에서 프로모터-리포터 분석방법을 통해 인슐린 유사 성장인자-1의 프로모터를 자극하는 천연물을 선별한 결과 사인 추출물이 가장 좋은 프로모터 자극효과를 나타냈으며, 무모생쥐에서 패쇄첩포 후 RT-PCR로 실험한 결과 IGF-1 mRNA를 $35\%$ 증가시키는 것으로 나타났다. 사인 추출물의 피부 주름개선 효과를 알아보기 위하여 인체 섬유아세포에서는 type-I collagen과 MMP-1 합성 변화를 관찰하였으며, 무모생쥐에서는 콜라젠의 증가와 진피 두께를 관찰하였다. 그 결과, 동위원소를 이용한 콜라젠 증가실험에서 type-I collagen은 $38\%$ 증가하였으며 무모생쥐에서 실시한 RT-PCR 결과에서는 mRNA가 $21\%$ 증가하는 것으로 관찰되었다. MMP-1 효소발현의 경우 ELISA 분석을 통해서 $63\%$의 높은 발현저해능을 확인하였고 Western blot에서도 발현이 저해되는 것을 확인하였다 추출물을 무모생쥐에 패쇄첩포 하였을 경우 대조군에 비해 진피 두께가 두꺼워지고 콜라젠 양도 증가되는 것으로 조직염색 관찰을 통해 확인하였다. 이상의 결과를 통해 사인추출물은 피부 주름개선에 좋은 효과를 나타내는 것으로 보이며, 여기에는 인슐린 유사성장 인자-I의 발현증가와 관련된 기전이 관여하는 것으로 판단된다.
SHI-RELATED SEQUENCE (SRS) 유전자는 zinc-binding RING finger motif를 갖고있는 식물 특이적 전사인자로서 식물 생장과 발달에서 중요한 역할을 하고 있다. 배추 생장점 부위로부터 분리된 Brassica rapa SHI-RELATED SEQUENCE (BrSRS) 유전자인 BrSRS7과 BrLRP1은 식물 생장과 발달에서 중요한 조절자이다. BrSRS7과 BrLRP1 유전자가 원예작물의 생장과 발달에 미치는 영향을 보고자 35S 프로모터에 유전자를 결합시켜 두 종류의 식물 형질전환 벡터를 제작한 후 아그로박테리움을 이용하여 페튜니아에 형질전환하였다. 형질전환체들은 전체적으로 식물체 크기가 감소되었고 잎은 작으면서 upward-curled된 특성을 나타내었고 줄기 사이의 길이가 줄었다. 페튜니아의 꽃 모양은 도입된 유전자에 따라 오각형, 별 모양, 둥근 모양으로 다르게 나타났다. 이러한 특성들은 $T_2$, $T_3$ 세대진전 후에도 안정적으로 나타났다. RT-PCR로 유전자 발현을 분석한 결과 BrSRS7과 BrLRP1 유전자는 정단 분열 조직에서 형태 형성에 관여하는 지베릴린과 옥신 관련 유전자인 PtAGL15, PtIAMT1 발현을 조절함으로서 페튜니아의 생장과 발달에 중요한 역할을 하는 것으로 추측된다. 따라서 본 연구결과로 BrSRS 계열 유전자는 왜성 특성, 식물 형태 변형을 갖는 화훼 품종을 개발하는데 유용하게 이용될 수 있을 것으로 사료된다.
본 연구는 경남 화훼연구소에서 개발 육성된 스프레이형 국화 18 품종들에 대한 재분화율을 각각 조사하고, 그 중에서 '무랑루즈'가 가장 우수한 재분화 능력이 있음을 확인하였다. 특히 BA($0.5mg{\cdot}L^{-1}$)와 NAA($1.0mg{\cdot}L^{-1}$)를 함유한 MS배지를 신초재분화 배지로 이용함으로써 스탠다드형 국화인 신마와 함께 스프레이형 국화인 '무랑루즈'의 잎 절편체부터 효율적으로 신초 형성을 유도하고 완전한 식물체로 재분화시킬 수 있었다. 이러한 신초재분화 조건에서 '무랑루즈'잎 절편에 아그로박테리움을 감염하고 10일 동안 함께 배양한 후에, 선발항생제 kanamycin 농도를 저농도($10mg{\cdot}L^{-1}$)에서 고농도($30mg{\cdot}L^{-1}$)로 단계적으로 선발과정을 강화함으로써 효율적인 국화 형질전환체 제작이 가능하였다. 조사한 kanamycin 저항성 식물체 모두에서 전이유전자 $AtSICKLE$가 존재함을 genomic PCR 방법으로 확인하였다. 비록, 국화에서 그 기능이 비효율적인 CaMV 35S 프로모터를 사용했기 때문에 RT-PCR로 확인한 형질전환체에서의 전이유전자 $AtSICKLE$의 발현율은 상대적으로 낮았으나, 그 발현이 상대적으로 높은 개체에선 잎의 상편생장에 의한 엽형의 변화가 나타났다. 이러한 연구의 결과는 모델 식물체에서 분리되고 연구된 많은 유전자들이 가지는 화훼작물의 분자육종학적 가치를 국화에서 대량으로 조사할 수 있는 기반을 제공 할 것으로 기대된다.
식물생명공학 연구에 있어서 모델 식물인 Arabidopsis thaliana (애기장대)와 아주 유사한 염생 식물인 salt cress (Thellungiella halophila 또는 Thellungiella parvula)는 고염 스트레스에 대하여 강한 내성을 가지고 있다. 본 연구에서는 고염에 저항성을 가지는 유용유전자를 선별하기 위하여, 200 mM NaCl을 처리한 T. halophila 또는 T. parvula 식물로부터 mRNA를 분리하여 cDNA library를 작성하였다. Full length cDNA library를 Agrobacteria-methods 형질전환 방법에 필요한 binary vector pool을 작성하고 Arabidopsis에 형질전환 시켰다. 형질전환되어진 Arabidopsis를 항생제 Basta 선별을 통하여 형질전환체 pool을 구축하였다(305,400 종자). 이와 같은 방법을 통해 구축 되어진 pool중에서 168,500 종자를 이용하여 고염 스트레스 조건하에서 종자 발아율과 뿌리 생장 촉진되는 현상이 나타나는 내염성 형질전환체를 1차 선별하였다(7,157 개체; 4.24%). 1차 선별된 형질전환체 중에서 1,551개체를 이용하여 2차 선별을 수행하여 내염성 형질전환체 165 개체를 확보하였다(10.6%). 선별된 형질전환체 중 대부분 개체는 고염 스트레스에 대응하여 종자 발아 및 뿌리 생장 모두 야생형보다 촉진된 표현형을 보였다. 그 중 몇 몇의 형질전환체는 야생형에 비하여 특이적인 기관 발달 현상이 나타났다. 예를 들면, 꽃과 줄기의 발달이 야생형과는 다른 표현형을 보이는 형질전환체가 선별되었다. 이렇게 스트레스에 내성을 가지는 형질전환체로부터 유전자 분리 방법을 통하여 해당 유용유전자를 확보할 수가 있다. 본 연구에서는 향후 halopyte 식물체를 이용하여 고염 뿐만 아니라 다양한 환경스트레스(건조, 냉해, 고열, 호르몬 등) 신호전달에 관여하는 유용유전자를 보다 쉽게 확보하는 방법을 제시함으로서, 환경재해극복에 관여하는 신호전달 기작을 보다 쉽게 이해할 수 있을 것으로 사료된다.
