  • The purpose of this study was to investigate the transport characteristics of passage cultured l1uman nasal epithelial cell monolayers grown on Transwell@ inserts using liquid-covered culture (LCC) method. The monolayer of passage 2 and 3 exhibited tight barrier (TEER>1,000 ohmxcm$^2$) in 2-3 days after seeding. In the morphological studies by actin staining and SEM/TEM, the existence of tight junction was clearly observed. (omitted)

Characterization of Odontoblasts in Supernumerary Tooth-derived Dental Pulp Stem Cells between Passages by Real-Time PCR (과잉치 치수유래 줄기세포의 Real-time PCR에 의한 계대간 상아질모세포 발현 특성)

  • The aim of this study is to compare the properties of odontoblast gene of early passage cells and late passage cells derived from impacted maxillary supernumerary teeth. Impacted supernumerary teeth with maxilla were extracted from 12 patients (8 males, 4 females) between 6 - 9 years old without medical history. Real-time polymerase chain reaction (PCR) was conducted to compare characterization of odontoblast cell in the 3rd and 10th passage, and between with bone inducing additive group and without additive group. Genes for odontoblasts characteristics are osteonectin (ONT), alkaline phosphatase (ALP), osteocalcin (OCN), dentin matrix protein 1 (DMP-1) and dentin sialophosphoprotein (DSPP). The level of gene expression was in a decreasing order of ONT, ALP, OCN, DMP-1 and DSPP in the 3rd passage, and in decreasing order of ONT, DMP-1, OCN, ALP, and DSPP in the 10th passage in the undifferentiation and differentiation group. The order of ONT, DMP-1, and OCN did not changed. ALP and DMP-1 were switched in order. ALP and DMP-1 may be used as important markers for differentiating between the 3rd passage and 10th passage cells. Considering that supernumerary tooth was extracted young age and the time required to cultured 10th passage was short, supernumerary tooth can be considered a useful donor site of dental pulp stem cells.

Using AHP to Analyze the Evaluation Factors Related to Wildlife Passage Management (AHP기법을 활용한 야생동물이동통로의 기능개선을 위한 평가항목 분석)

  • The rapid increase of wildlife passage installation since the late 2000s was aimed to reduce roadkill caused by habitat fragmentation and losses related to road construction, but wildlife-vehicle collisions are now still occurred even near the wildlife passage area. This is the reason that the effectiveness of wildlife passage have not been evaluated in combination with absence of monitoring data and management strategy of the wildlife passage. The AHP method are used, in this study, to identify the evaluation factors affecting the effectiveness of the present 367 wildlife passages in a mitigation measures to reduce road effects on wildlife species. Ten evaluation factors are derived from third levels in the AHP analysis. Priority setting to identify appropriate management strategies in first level is selected among four second levels on facility, environment, wildlife species and management tool. The AHP analysis suggested that neighboring environments are the most important factor at the second level, and passage structure, harmony with natural surroundings, wildlife occurrence and monitoring of the passage are also important factors at the third levels. In summary, effective measurements of wildlife passage management is based on managing the passage with neighboring topography and natural surrounding. This is useful to establish wildlife passage management strategy in order to reduce the negative effects of roads on wildlife species.

Effect of Fusion Method and Passage Culture of Hanwoo (Korean Cattle) Ear Skin and Fetal Fibroblasts on the Development of Nuclear Transfer Embryos (한우의 귀세포와 태아섬유아세포의 융합 방법과 Passage 배양이 복제수정란의 발달에 미치는 영향)

  • The study was conducted to evaluate the effects of culture period and fusion method on the development of somatic cell nuclear transfer (SCNT) embryos reconstituted with Korean bovine fetal fibroblast cells (KbFF) and Korean bovine adult ear skin fibroblast cells (KbESF). KbFF were isolated from a day 51 Korean cattle (Hanwoo) fetus, and KbESF were isolated from a 28 month old Hanwoo calf. The cells were cultured up to 15 weeks (passage 15) in vitro for SCNT. Chamber and electrode needles were used for comparing fusion of reconstituted eggs. The doubling times of KbFF and KbESF were 17.3 hr and 24.3 hr, respectively. The fusion and cleavage rates were significantly higher in needle group (76.1 and 81.2% respectively, P<0.05) than those in chamber group. However, the blastocyst development rate was not different between both groups. Fusion and cleavage rates of NT eggs reconstituted with KbESF did not affected by passage number, however, blastocyst rates were lower in passage $1{\sim}4$ group (21.3%) than passage $5{\sim}8$ (39.4%) and $13{\sim}15$ groups (40.4%, P<0.05). Whereas, fusion rate was lower in passage $1{\sim}4$ group (61.5%) than those of passage $5{\sim}8$(75.0%) and $13{\sim}15$ (76.8%) groups, but cleavage and blastocyst rates were similar regardless of passage number in the KbFF. The results suggest that fusion method can affect the development of SCNT embryos, whereas the long term culture up to 15 passages may not affect the development of SCNT embryos.

  • A study on swirl chamber for diesel engine is to realize lower fuel consumption and exhaust emission than the current marketing engines. Author formerly reported the performance characteristics of small IDI diesel engine with swirl chamber by changing the jet passage area and its angle, and the depth and shape of the piston top cavity. Following after the first report, in this paper, the characteristics of fuel consumption, soot emission, and exhaust gas temperature were examined and analyzed after dimension of jet passage area expanded to $70.1mm^2$ The results were that the optimum values of the jet passage area depending on the depth of the piston top cavity were different at each engine speeds and loads, and in accordance with application of engine running conditions they were able to be selected as optimum dimensions of each design parameters.

  • The navigation problem of robot is one of the main themes to deal with conficts or interferences between obstacles and the robot itself In this case, while the robot avoids obstacles on the space, the passage route should be determined efficiently. In order to solve problems above, we have come up with the distance field space medel (DFM) and then, under known environment, we have presented the distance field A algorithm for passage route path search. In this research, the method of performing the 3-dimensional passage route path search of robot under unknown environment is proposed. It is shown that the authors can build the distance search model the does not need space division by taking into account of sensor information to a distance field space model, and constructing this information as virtual obstacle information.

Evaluation of virulence reversion of an attenuated porcine epidemic diarrhea vaccine strain by serial passages in suckling piglets

  • Porcine epidemic diarrhea is an infectious intestinal disease caused by the porcine epidemic diarrhea virus (PEDV). Especially, when suckling piglets are infected, the mortality rate is close to 100%. PEDV is classified into G1 and G2 types based on genetic differences. The G2 type PEDV outbreak in the United States in 2013 was highly pathogenic and contagious, and it has spread worldwide and caused continuous economic losses. Most commercial vaccines used are G1 type vaccines, and existing vaccines do not fully protect piglets due to genetic differences. In this study, we evaluated the safety of the newly developed G2 type attenuated HSGP vaccine strain by inoculating it into piglets and testing whether the vaccine virus spreads to the non-vaccinated, negative pigs and whether the vaccine reverts to its virulence during serial passage experiments. Each experiment lasted for 7 days for each passage, and fecal viral titers, clinical symptoms, and weight gain were measured daily. After the experiment, necropsy was performed to measure intestinal virus titer and pathological evaluation. As a result of the first passage, no transmission of the vaccine virus to negative pigs co-housed with vaccinated pigs was observed. In addition, after four consecutive passage experiments, the clinical symptoms and small intestine lesions were gradually alleviated, and no virus was detected in the feces in the fourth passage experiment. Therefore, it was concluded that the vaccine was safe without virulence reversion in accordance with the guidelines of the current licensing authority. However, further studies are needed on the genetic changes and biological characteristics of the mutant virus that occur during successive passages of the attenuated vaccine since the replication and clinical symptoms of the virus increased until the third passage during successive passages of the vaccine virus. Based on this study, it was concluded that virulence reversion and safety evaluation of attenuated vaccines through serial passage in target animals can be useful to evaluate the safety of attenuated viruses.

  • Accidents between ships occur frequently at the traffic congestion area. Once a maritime accidents occur, it is likely to end up with critical damaged accidents. This paper develop a simulator for assessing quantitative risk based on statical maritime traffic data and realtime traffic distribution. Ship's passage risk assessment simulator consist of import of division of passage data, traffic distribution analysis and passage risk assessment analysis. Maritime traffic data of WANDO waterway apply to simulator for calculation of quantitative risk rate of waterway.
