Effect of McN-A-343, Clonidine and Cocaine on the Contractile Response of Rat and Rabbit Vas Deferens to Field Stimulation (McN-A-343, Clonidine 및 Cocaine의 백서(白鼠)와 가토정관(家兎精管)의 자극효과(刺戟效果)에 대(對)한 영향(影響))

  • 1) Effects of McN-A-343, clonidine and cocaine on the contractile response of the rat and rabbit vas deferens to field stimulation were investigated. The action mechanisms of these drugs have shown to be associated with endogenous norepinephrine(NE). 2) The contractile response to the stimulation of 5-10 Hz was markedly inhibited by the cumulative doses of McN-A-343 (0.003, 0.03, $0.3{\mu}g/ml$), clonidine (0.003, $0.3{\mu}g/ml$) and cocaine $(0.03\;{\mu}g/ml)$. The inhibition was antagonized by yohimbine and pjperoxan. 3) The inhibitory effect of McN-A-343 $(0.03{\mu}g/ml)$ and cocaine $(0.03{\mu}g/ml)$ was markedly enhanced by the same dose of cocaine and McN-A-343, respectively. This enhanced inhibition was also antagonized by yohimbine. 4) The contractile response to the stimulation of 0.01 Hz and 5-10 Hz was markedly potentiated by comparatively large doses of McN-A-343 $(30\;{\mu}g/ml)$ and $(3\;{\mu}g/ml)$. This potentiation was not observed in the presence of thymoxamine. The potentiation by McN-A-343 also did not appear in the presence of atropine. 5) The contractile response to the above stimulation was potentiated by muscarine and the potentiation was markedly attenuated in the presence of thymoxamine and atropine.

  • Objectives : BHH10 is traditional medicine herb used for enhancing body resistance against various diseases. The aim of this study was to identify BHH10 extract induces osteogenic activity in human osteoblast-like MC3T3-E1 cells. Methods : MC3T3-E1, pre-osteoblast cell line, were treated with BHH10 of various concentrations($0.1{\mu}g/mL$, $1{\mu}g/mL$, $10{\mu}g/mL$). And then, the effect of BHH10 on osteoblast differentiation was examined by alkaline phosphatase(ALP) activity, von Kossa staining and RT-PCR for osteoblast differentiation markers such as osteocalcin(OCN), osteopontin(OPN). Results : BHH10 had dose-dependent effect on the viability of osteoblastic cells, and dose-dependently increased alkaline phosphatase(ALP) activity. BHH10 markedly increased mRNA expression for OCN, OPN in MC3T3-E1 cells. Also, BHH10 significantly induced mineralization in the culture of MC3T3-E1 cells. Conclusions : In conclusion, these results propose that BHH10 can play an important role in osteoblastic bone formation, osteogenesis, and may possibly lead to the development of bone-forming drugs.

Effects of Malty Culture supplementation diets during lactation on sow and litter performance (포유돈 사료에 Malty Culture의 첨가가 모돈 및 자돈 생산성에 미치는 영향)

  • This experiment was conducted to investigate the effects of supplemented diets with Malty Culture(MC) during lactation on sow and litter performance. Eighteen lactating sows were randomly assigned to three dietary treatments(basal diet, basal diet+MC0.5%, basal+MCl.0%). Average daily feed intake of sow was not affected by dietary treatments with treatment means ranging from 4.49 to 4.58kg(p>0.62). All sows lost their backfat thickness during lactation with no difference among the treatments(p>0.25). Fecal moisture was similar for sows fed the control and MC treatments with 70% moisture content. The growth rate of piglets was not significant difference among the treatments ranging from 47 to 48kg. Day to estrus postweaning was not affected by supplementation of MC with treatment means ranging from 5 to 7 days. There was not significant difference in the milk production(i.e, 8.9kg/d for control, 9.18kg/d for MC0.5 and 9.20kg/d for MC1.0). Survivability of piglet was affected by feeding added MC during lactation. There was no significant difference in the digestibility of DM, but the digestibility of N tended to increase in sows fed the MC0.5 and MC1.0 supplemented diets compared to the basal diet(linear effect, p<0.10; quadratic effect. p<0.08). Digestibility of P tended to increase by the treatment of supplemented MC. However, there was no significant difference among the treatments. In conclusion, the results of experiment suggest that lactation diet of supplemented MC could be a beneficial feeding for digestibility in sow and litter performance.

Karyotype Analysis and Physical Mapping of rDNAs Using McFISH in Jeffersonia dubia Benth (깽깽이풀의 핵형분석과 McFISH를 이용한 rDNA의 물리지도 작성)

  • Karyotype analysis and chromosomal locailization of 45S and 5S rDNAs using McFISH (multi-color fluorescence in situ hybridization) were carried out in Jeffersonia dubia Benth., which is one of medicinal plants belonging to Berberidaceae. The somatic metaphase chromosome number was 2n=2x=12 and the size of chromosomes ranged $1.95{\sim}3.50\;{\mu}m$. The chromosome complement consisted of two pairs of metacentrics (chromosomes 1 and 3), two pairs of submetacentrics (chromosomes 2 and 4) and two pairs of subtelocentrics (chromosomes 5 and 6). In McFISH, one pair of 45S rDNA site was detected on the centromeric region of chromosome 2 and three pairs of 5S rDNA sites were detected on the short arm of chromosomes 4, 5 and 6, respectively.

Calculation of Jaws-only IMRT (JO-IMRT) dose distributions based on the AAPM TG-119 test cases using Monte Carlo simulation and Prowess Panther treatment planning system

  • The aim of this study is to calculate the JO-IMRT dose distributions based on the AAPM TG-119 using Monte Carlo (MC) simulation and Prowess Panther treatment planning system (TPS) (Panther, Prowess Inc., Chico, CA). JO-IMRT dose distributions of AAPM TG-119 were calculated by the TPS and were recalculated by MC simulation. The DVHs and 3D gamma index using global methods implemented in the PTW-VeriSoft with 3%/3 mm were used for evaluation. JO-IMRT dose distributions calculated by TPS and MC were matched the TG-119 goals. The gamma index passing rates with 3%/3 mm were 98.7% for multi-target, 96.0% for mock prostate, 95.4% for mock head-and-neck, and 96.6% for C-shape. The dose in the planning target volumes (PTV) for TPS was larger than that for the MC. The relative dose differences in D99 between TPS and MC for multi-target are 1.52%, 0.17% and 1.40%, for the center, superior and inferior, respectively. The differences in D95 are 0.16% for C-shape; and 0.06% for mock prostate. Mock head-and-neck difference is 0.40% in D99. In contrast, the organ curve for TPS tended to be smaller than MC values. JO-IMRT dose distributions for the AAPM TG-119 calculated by the TPS agreed well with the MC.

The Effect of IGF-1 on ALP Activity of MC3T3-E1 Cell (MC3T3-E1세포의 ALP activity에 대한 IGF-I의 영향)

  • Polypeptide growth factors belong to a class of potent biologic mediators which regulate cell differentiation, proliferation, migration and metabolism. IGF-I is polypeptides secreted by skeletal cells and is considered as regulators of bone formation. The purpose of this study is to evaluate the effects of IGF-I on bone nodule formation and alkaline phosphatase activity of MC3T3-E1 cells. MC3T3-E1 cells were seeded at $1{\times}10^4$ cells/well, $1{\times}10^5$ cells/well in alpha-modified Eagle medium containing 10% fetal bovine serum, 10 mM ${\beta}-glycerophosphate$ and $5O{\mu}g/ml$ of ascorbic acid. Before 48 hours of indicated time, medium were changed with serum free medium. After 24 hours, 0.1, 1, 10 ng/ml IGF-I were added to the cells and cultured for 3, 7, 14, 21, 28 days. And histochemical analysis was done and ALP activity was measured and was expressed as nmol/min/mg of protein. The bone nodule formation in MC3T3-E1 cells of IGF-I was seen at 21, 28 days, but there were no difference between control group and experimental groups. The ALP activity decreased when it is compare to control 2 group except for 1 ng/ml, 10 ng/ml IGF-I of 21-day-groups and 1 ng/ml IGF-I of 28-day-groups. Dose response effects of IGF-I of ALP activity in MC3T3-E1 cells were seen the highest ALP activity at 1ng/ml until 21days and the highest ALP activity at 10 ng/ml of 28 daygroups. The peak times were seen at 7-day group, 14-day group on control group and experimental group respectively, and 1 ng/ml group was the highest ALP activity, From the above results, IGF-I was not seen notable effect on bone nodule formation and decreased ALP activity of MC3T3-E1 cells but the use of IGF-I to mediate biological stimulation of MC3T3-E1 cells shows promise for future therapeutic application.

  • The demand for multi-functionality, high density, high performance, and miniaturization of IC devices has caused the technology paradigm shift for electronic packaging. So, thermal management of new packaged chips becomes a bottleneck for the performance of next generation devices. Among various thermal solutions such as heat sink, heat spreader, TIM, thermoelectric cooler, etc. on-chip liquid cooling module was investigated in this study. Micro-channel was fabricated on Si wafer using a deep reactive ion etching, and 3 different micro-channel designs (straight MC, serpentine MC, zigzag MC) were formed to evalute the effectiveness of liquid cooling. At the heating temperature of $200^{\circ}C$ and coolant flow rate of 150ml/min, straight MC showed the high temperature differential of ${\sim}44^{\circ}C$ after liquid cooling. The shape of liquid flowing through micro-channel was observed by fluorescence microscope, and the temperarue differential of liquid cooling module was measuremd by IR microscope.

An Analytical Approach for Design of Nth-band FIR Digital Filters with Equi-Ripple Passband

  • In FIR (Finite Impulse Response) filter applications, Nth-band FIR digital filters are known to be important due to their reduced computational requirements. The conventional methods for designing FIR filters use iterative approaches such as the well-known Parks-McClellan algorithm. The Parks-McClellan algorithm is also used to design Nth-band FIR digital filters after Mintzer's research. However, a disadvantage of the Parks-McClellan algorithm is that it needs a large amount of design time. This paper describes a direct design method for Nth-band FIR Filters using Chebyshev polynomials, which provides a reduced design time over indirect methods such as the Parks-McClellan algorithm. The response of the resulting filter is equi-ripple in passband. Our proposed method produces a passband response that is equi-ripple to within a minuscule error, comparable to that of Mintzer's design method which uses the Parks-McClellan algorithm.

Photocatalytic Degradation of Algae and its By-product using Rotating Photocatalytic Oxidation Disk Reactor

  • This study examined the special technique of photocatalytic degradation (RPODisk) for removal of taste and odor causing materials, algae, and algal toxin. The RPODisk was effective for removal of these troublesome contaminants. It outperformed the fixed media and the UV irradiation for geosmin removal. The RPODisk performance was comparable to the combination of the UV irradiation with TiO2. The RPODisk performance was affected by the rotating speed. The faster the speed was, the better the performance. The RPODisk was also effective for removal of algae and algal toxin. The algal activity reduced by 80% after 30 mins of the treatment. More toxic microcystin (MC)-LR was more difficult to remove than MC-RR. The times for 50% removal were 23.7 mins for MC-LR and 14.1 mins for MC-RR. Almost 100 mins of the contact time was required to completely remove MC-LR at the rotating speed of 260 rpm.

Analysis of Melanocortin receptor 1 (MC1R) gene differential test for beef species between Hanwoo and Holstein using polmerase chain reaction -restriction fragment length polymorphism (PCR-RFLP) (MC1R gene의 PCR-RFLP를 이용한 한우.젖소고기 감별)

  • The objective of this study was to differentiate the beef species between Hanwoo and Holstein from a total of 1,081 beef samples using PCR-RFLP of MC1R gene. When a PCR product of 403 bp specific band amplified from bovine MC1R gene sequence was digested with restriction enzyme MspA1I, Hanwoo type showed 2 bands, 220 bp and 183 bp size bands. Holstein type, however, showed three bands, 220 bp, 138 bp and 45 bp size band, respectively. The results of the differential test for beef species were as following; 7 samples (0.64%) were determined to Holstein type, of which 4 were submitted from administrative authorities, other 3 from self-collection planing, and none from civilian clients including school.
