SAMs of 2-Aminoethanethiol Modified with 3,4-Dihydroxyphenylalanine for the Electrocatalytic Oxidation of NADH on Gold Electrodes

  • 2-Aminoethanethiol (aet) has been used to make self-assembled monolayer (SAMs) on gold electrodes, which are subsequently modified with 3,4-dihydroxy phenylalanine (dpa). Such modified electrodes having various types of Au/aet-dpa were employed in the electrocatalytic oxidation of NADH. The purpose of this study to characterize the responses of such modified electrodes in terms of the immobilization procedure, pH of the solution and applied potential. The reaction of the surface immobilized dpa with NADH was studied using the rotating disk electrode technique and a value of $2.2{\times}10^4M^{-1}s^{-1}$ was obtained for the second-order rate constant in 0.1 M Tris/$NO_3^-$buffer (pH=8.0). The hydration behavior of the films was characterized by quartz crystal microbalance. When used as a NADH sensor, the Au/aet-dpa electrode exhibited good sensitivity and an excellent correlation (r ${\geq}$ 0.99) for NADH concentration which extended to $3.8{\times}10^{-3}$ M.

복합해조류 발효추출물의 항산화, 미백 활성 (The Anti-Oxidant and Whitening Activities of Seaweeds Mixture Fermentation Extracts)

  • 조류 소재에 관한 연구는 꾸준히 진행되고 있다. 본 연구에서는 미역, 다시마, 불등가사리 등 해조류를 혼합한 발효추출물의 항산화, 미백, 보습의 활성 실험을 진행하였다. 또한 유산균으로는 김치유산균 Lactobacillus sakei균주를 이용하였다. 복합해조류 발효추출물의 생리활성 실험을 진행하였으며, 1,1-diphenyl-2-picrylhydrazyl(DPPH), 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS) 라디칼 소거능에서는 양성대조군 보다 높은 저해효능을 보였다. 미백효능 실험에서는 3,4-dihydroxy-L-phenylalanine (L-DOPA), mushroom tyrosinase 실험을 수행하였으며, 발효 전보다 발효 후의 미백활성이 높게 나타나는 것을 확인할 수 있었다. 한편, 해조류 소재의 유효성분인 fucose에 대한 HPLC분석을 수행하였으며, 용매조건에 대한 표준화 방법을 새로이 정립하였다. 이 연구는 복합해조류 추출물이 항산화, 미백 등 화장품 소재로서 가능성을 제안할 수 있다.

Specific urinary metabolites in canine mammary gland tumors

  • The identification of biomarkers that distinguish diseased from healthy individuals is of great interest in human and veterinary fields. In this research area, a metabolomic approach and its related statistical analyses can be useful for biomarker determination and allow non-invasive discrimination of healthy volunteers from breast cancer patients. In this study, we focused on the most common canine neoplasm, mammary gland tumor, and herein, we describe a simple method using ultra-high-performance liquid chromatography to determine the levels of tyrosine and its metabolites (epinephrine, 3,4-dihydroxy-L-phenylalanine, 3,4-dihydroxyphenylacetic acid, and vanillylmandelic acid), tryptophan and its metabolites (5-hydroxyindolacetic acid, indoxyl sulfate, serotonin, and kynurenic acid) in canine mammary cancer urine samples. Our results indicated significantly increased concentrations of three tryptophan metabolites, 5-hydroxyindolacetic acid (p < 0.001), serotonin, indoxyl sulfate (p < 0.01), and kynurenic acid (p < 0.05), and 2 tyrosine metabolites, 3,4-dihydroxy-L-phenylalanine (p < 0.001), and epinephrine (p < 0.05) in urine samples from the mammary gland tumor group compared to concentrations in urine samples from the healthy group. The results indicate that select urinary tyrosine and tryptophan metabolites may be useful as non-invasive diagnostic markers as well as in developing a therapeutic strategy for canine mammary gland tumors.

마늘의 alcohol 침지 중 휘발성 향기성분과 침출유리아미노산 함량 (Elution Profiles of Volatile Compounds and Free Amino Acids during Alcohol Soaking of Garlic(Allum sativum L.))

  • 마늘주의 산업화에 앞서 기초 자료로써 활용하고자, 마늘을 소주 등의 술에 침지하였을 때 마늘의 휘발성 기능성분과 유리아미노산이 술로 침출되는 정도 및 형태에 관하여 조사하고자, 생마늘을 20% 주정에 5주간 침지하고 매주 휘발성 향기성분과 유리아미노산들이 침출되는 양적 및 형태적 변화를 조사하였다. 마늘의 주요 유리 아미노산은 L-aspartic acid, L-glutamic acid, L-arginine, L-alanine, L- proline, L-asparagine 및 L-serine이었으며, 유리 아미노산들 모두 침지 기간의경과에 따라 침출량이 증가하였다. 또한, L-threonine, L-proline, L-valine 및 L-leucine 등의 중성아미노산들과 방향족아미노산인 tyrosine과 phenylalanine은 3주 후부터 80% 이상 침출된 반면에, 산성, 염기성 및 함황아미노산들은 5주 후에도 80% 미만이었다. 생마늘의 휘발성 향기성분은 diallyl trisulfide, diallyl disulfide, methyl allyl disulfide, 2-vinyl-4H-1,3-dithiin, 3-vinyl-3,4-dihydro-1,2-dithiin, 3,5-diethyl-1,24-trithiolane, isobutyl isothiocyanate 및 diallyl sulfide 등의 황화합물들이 주요 성분들이었다. 생마늘로부터 주정으로 침출된 휘발성 향기성분은 allyl alcohol, diallyl disulfide, 3,5-diethyl-1,2,4-trithiolane, diallyl trisulfide, 3,4-dimethoxyfuran의 5종이었으며, 침지기간의 경과에 따라 3,5-diethyl-1,2,4-trithiolane는 계속 침출량이 증가하였지만 그 외 4종은 감소하였다. 침 지기간 중 furfural, 5-methylfurfural, 5-hydroxymethylfurfural, dimethyl pyrazine, furfuryl alcohol, 3-hydroxy-2-bytanone과 2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one이 새로이 생성되었으며, 침지기간의 경과에 따라 침출주에 대부분 축적되는 양상을 보였다.

Hydroxylated Hydrocinnamides as Hypocholesterolemic Agents

  • Several hydroxylated cinnamic acid derivatives were prepared from the corresponding acids and amino acid residues, and their hypocholesterolemic activities were evaluated in high cholesterol-fed mice. The presence of the double bond in hydroxylated cinnamide derivatives decreases cholesterol-lowering activities and the number of free phenolic hydroxy groups affect greatly the activities. 3,4-Dihydroxy hydrocinnamides obtained from amino acid derivatives containing a hydrophobic side chain such as alanine, valine, phenylalanine, and isoleucine exhibited potent cholesterol-lowering activities.

Inhibitory Effects of Herbal Extracts on Dopa Oxidase Activity of Tyrosinase

  • Tyrosinase catalyzes the rate-limiting steps in melanin biosynthesis which is involved in skin-coloring and local hyperpigmentation of human beings, and unfavorable darkening of food products, Inhibitory effects on 3,4-dihydroxy-phenylalanine (dopa) oxidase activity of tyrosinase by 594 kinds of herbal extracts prepared from herbal medicines and wild plants in Korea were estimated. Two herbal extracts prepared from radicis cortex of Morus alba and rhizoma of Curcuma longa were selected as those exhibiting potent inhibitory effects on the enzyme activity. These herbal extracts were subjected to sequential fractionations with methylene chloride, ethyl acetate, n-butanol, and polar residue. The inhibitory effects on dopa oxidase activity of tyrosinase were shown in ethyl acetate fraction of Morus alba, and in methylene chloride fraction of Curcuma langa. The ethyl acetate fraction of Marus alba exhibited 50% of inhibition on dopa oxidase activity of tyrosinase at the concentration of 12 ${\mu}g/ml$, and methylene chloride fraction of Curcuma langa at 51 ${\mu}g/ml$.

AOT/이소옥탄/역미셀계에서의 Tyrosinase 반응 (Tyrosinase Reaction in AOT/Isooctane/Reverse Micelles)

  • 지용성 물질은 물에 용해되지 않아 수용액 효소반응계에서의 특정효과를 평가하기가 어렵다. 생체내에서 멜라닌을 합성하는 효소인 tyrosinase의 지용성 저해제를 탐색하고자 유기용매, 계면활성제 및 물로 구성된 광학적으로 투명한 RM을 도입하였다. Tyrosinase의 겉보기 반응이 물에서와 유사한 RM의 형성조건 중 유기용매는 isooctane, 계면활성제는 100 mM의 dioctyl sulfosuccinate(AOT)가 적절하였다. RM에서 물과 AOT의 몰 농도 비율이 15일 때 tyrosinase(103.5 units)는 기질인 3,4-dihydroxy-L-phenylalanine(0.18 mM)에 대하여 수용액에서와 유사한 거동을 나타내었다. 또한 tyrosinase 저해제인 cinnamic acid 존재 하에서도 tyrosinase 촉매 반응의 반응 생성물이 반응 시간에 비례하여 형성됨이 확인되어 지용성 물질도 RM에서는 tyrosinase 저해활성이 가능함을 시사하였다. 따라서 RM은 수용성인 효소와 기질 그리고 지용성인 저해제를 모두 하나의 상(phase)에 용해시킬 수 있어 지용성 물질의 tyrosinase 저해 효과를 분석할 수 있는 중요한 수단으로 활용할 수 있을 것으로 판단되었다.
