  • Acquaintance is scanty on primaquine (PQ) efficacy and Plasmodium vivax recurrence in Udupi district, Karnataka, India. We assessed the efficacy of 14 days PQ regimen (0.25 mg/kg/day) to prevent P. vivax recurrence. Microscopically, aparasitemic adults (${\geq}18years$) after acute vivax malaria on day 28 were re-enrolled into 15 months' long follow-up study. A peripheral blood smear examination was performed with participants at every 1-2 month interval. A nested PCR test was performed to confirm the mono-infection with P. vivax. Of 114 participants, 28 (24.6%) recurred subsequently. The median (IQR) duration of the first recurrence was 3.1 (2.2-5.8) months which ranged from 1.2 to 15.1 months, including initial 28 days. Participants with history of vivax malaria had significantly higher risk of recurrence, with hazard ratio (HR) (95% CI) of 2.62 (1.24-5.54) (P=0.012). Severity of disease (11.4%, 13/114) was not associated (P=1.00) with recurrence. Of 28 recurrence cases, the nPCR proved that P. vivax mono-infection recurrence rate was at least 72.7% (16/22) at first recurrence. In Udupi district, PQ dose of 0.25 mg/kg/day over 14 days seems inadequate to prevent recurrence in substantial proportion of vivax malaria. Patients with a history of vivax malaria are at high risk of recurrences.

  • The present work was designed to understand the mechanism of superovuiation and the cause of early embryo loss and Implantation failure in the superovulating immature female rats which were elaborated by a pituitary gland transplantation. A pituitary gland obtained from the orchidectomized rats was transplanted under the right kidney capsule of 28 day old female rats (PGT group) on the starting day of experiment which was designated as Day 2. The grafted pituitary glands were removed at 6h (RPGT 6h group), 12h (RPGT 12hgroup) and 24h (RPGT 24h group) after the transplantation. Control rats were treated with 41U PMSG on Day 2 (PMSG group). The estrous cycle and the levels of plasma progesterone and estradiol-17$\beta$ were observed on Day 0, Day 5, respectively. The implantation sites, the weights of ovary and uterus, and the number of corpora lutea were examined in all group on Day 8. The resuft obtained were summarized as follows: 1. The percentages of the number of the rats in proestrus and estrus were 93.0%, 82.6%, 0%, 90.7% and 89.5% in PMSG, PGT, RPGT6h, RPGT12h and RPGT24h group, respectively. The synchronization of estrus cycle was dchieved in all groups. 2. The mating rates of each group were 80.2, 75.0, 0, 56.4, 57.8% in PMSG, PGT, RPGT6h, RPGT12h, RPGT24h group, respectively. 3. The numbers of copora lutea on Day 8 were 47.1 i 4.9, 18.1 $\pm$0.5, 14.1 i 0.3 and 8.9 $\pm$ 0.3 in PGT, RPGT24h, RPGTl2h and PMSG group, respectively. There were signIficantly difference between all groups (P<0.05). 4. The numbers of implantation sites (18.1 +- 4.0) in PGT group on Day 8 were higher than those of PMSG (8.5 $\pm$ 2.5), RPGT 12h (9.8 i 0.2) and RPGT 24h group (10.8 i 0.2) (P<0.05). 5. The ovarlan weights in PGT (95.2 $\pm$ 14.3mgIlOOg BW), RPGT 12h (51.7 $\pm$ 0.6mgIlOOg BW), and RPGT24h (57.9 $\pm$ 0.9mg/l00g 8W) groups were significantly higher than those of PMSG group (30.4 $\pm$7.4mg/l00g BW) (P<0.05). 6. The uterine weights in PMSG (672.4 $\pm$ 4.7mg/l00g 8W), and PGT (660.7 $\pm$7.8mg/l00g BW) groupswere greater than those of RPGT 12h (403.0 $\pm$ 1.lmg/lOOg BW) and RPGT 24h (490.1 $\pm$ 0.9mg/l00g BW) group (P<0.05). 7. The plasma progesterone levels in PGT groups (15 lng/ml) on Day 5 were higher than those of PMSG (83ng/ml), RPGT 12h (S7ng/ml), RPGT24h (8lng/ml) group (P<0.05). 8.

  • Background: Propofol (2.6-diisopropylphenol) is a widely used intravenous anesthetic agent for the induction and maintenance of anesthesia during surgeries and sedation for ICU patients. Propofol has a structural similarity to the endogenous antioxidant vitamin E and exhibits antioxidant activities.13) However, the mechanism of propofol on hypoxia/reoxygenation (H/R) injury has yet to be fully elucidated. We investigated how P-PostC influences the autophagy and cell death, a cellular damage occurring during the H/R injury. Methods: The groups were randomly divided into the following groups: Control: cells were incubated in normoxia (5% CO2, 21% O2, and 74% N2) without propofol treatment. H/R: cells were exposed to 24 h of hypoxia (5% CO2, 1% O2, and 94% N2) followed by 12 h of reoxygenation (5% CO2, 21% O2, and 74% N2). H/R + P-PostC: cells post-treated with propofol were exposed to 24 h of hypoxia followed by 12 h of reoxygenation. 3-MA + P-PostC: cells pretreated with 3-MA and post-treated propofol were exposed to 24 h of hypoxia followed by 12 h of reoxygenation Results: The results of our present study provides a new direction of research on mechanisms of propofol-mediated cytoprotection. There are three principal findings of these studies. First, the application of P-PostC at the onset of reoxygenation after hypoxia significantly increased COS-7 cell viability. Second, the cellular protective effect of P-PostC in H/R induced COS-7 cells was probably related to activation of intra-cellular autophagy. And third, the autophagy pathway inhibitor 3-MA blocked the protective effect of P-PostC on cell viability, suggesting a key role of autophagy in cellular protective effect of P-PostC. Conclusions: These data provided evidence that P-PostC reduced cell death in H/R model of COS-7 cells, which was in agreement with the protection by P-PostC demonstrated in isolated COS-7 cells exposed to H/R injury. Although the this study could not represent the protection by P-PostC in vivo, the data demonstrate another model in which endogenous mechanisms evoked by P-PostC protected the COS-7 cells exposed to H/R injury from cell death.

  • Conditions for artificial culture of Lemna Paucicostata and its nutritional values were examined in this study. Lemna P. was cultured using artificial wastewater and a bioreactor (total volume $2,630\;cm^3$, working volume $2,240\;cm^3$) was operated at conditions of 6,250 lux and $28^{\circ}C$. Water flow affected the growth of Lemna P.: growth rate was very high (more than $1.1\;d^{-1}$) at a condition of no-water movement, but it was very low (less than $0.15\;d^{-1}$) when water moved slowly. The growth of Lemna P. was higher in $16h\;d^{-1}$ light cycle than in Sand $24h\;d^{-1}$, and it was also severely affected by the initial $NH_4$-N levels of wastewater. The growth rate of Lemna P. was high in lower $NH_4$-N level, indicating that the growth rate is in inverse proportion to $NH_4$-N concentration in wastewater. However, the contents of crude protein (CP) of Lemna P. were proportional to the initial $NH_4$-N concentration. The CP contents of Lemna P. cultured at 2, 10, 50 and 100 $NH_4$-N mg $L^{-1}$ was 18, 24, 37, 43%, respectively, showing the Lemna P. cultured at 50 and $100\;mg\;L^{-1}$ had similar protein contents to linseed (CP 35%), cottonseed (CP 38%) and soybean (CP 45%). Fat, protein, fiber, NDF and ADF contents of Lemna P. harvested at conditions of $16h\;d^{-1}$ light cycle and less than $2\;mg\;L^{-1}$ of $NH_4$-N level was 2.8, 18, 27, 20, 41 and 65.7%, respectively. Since the growth rate of Lemna P. was very high (more than $1.1\;d^{-1}$) at those conditions, it was convinced that mass production of valuable protein and fiber sources are feasible. In particular, since the Lemna P. has unsaturated fatty acids found mainly in animal fat as well as beneficial fatty acids to health such as C18:ln9c, C18:2n6c, C20:5n3 and C22:2, the Lemna P. biomass would be a highly valuable alternative feed source to grains.

  • Ozone is a strong oxidant and potent disinfecting agent. In this study, volatile basic nitrogen, thiobarbituric acid reactive substances, acid value and pH all of which are quality indicators in meat products, were evaluated. The meat was treated with the ozonated water (0.2 ppm) for 0, 5, 10, 30, and 60 min and then stored at $5^{\circ}C$ for 24 d. The volatile basic nitrogen content of meat was 22.40 mg% after 9 d of storage and 23.24 mg% at 15 d of storage with ozonated water (0.2 ppm) treatment. During 24 d of storage, the pH, thiobarbituric acid reactive substances, and acid value were decreased when subjected to ozonated water treatment. These results suggested that the ozonated water treatment effectively improved the chemical properties and food safety.

  • The proportion of dry matter (DM) and mineral elements (Ca, Mg, P, Na, K, Zn) released from eight feeds (2 rice straws, RSI and RS2; 2 grasses, NB21 and guinea; 2 leguminous fodders, glyricidia and erythrina; jak leaves and rice bran) were studied using the nylon bag procedure. Bag incubations up to 10 days were performed in the rumen of cows fed on a ration consisting of 50% wheat straw and 50% hay. Both the type of feed and the incubation time in the rumen significantly influenced (p < 0.01) the proportion of minerals released. In legumes, jak leaves and rice bran about 80% of the potentially degradable DM fraction was solubilized within 24 h in the rumen, and with the grasses, rice straws and jak leaves a considerable proportion of DM was released between 48 and 240 h in the rumen. During the early hours of incubation (up to 24 h) there were distinct differences between and within the feed classes in their ability to release all mineral elements studied. In all test feeds, high proportions of Mg and K were released within 24 h. Some feeds showed a tendency to ad/absorb Ca (grasses, rice straws and rice bran), P (jak leaves, rice straws), Na (glyricidia and rice bran) and Zn (jak leaves) from water and rumen fluid, and this was partly related to the low initial concentration. In terms of absolute quantity of mineral released, legumes (erythrina is superior to glyricidia) are a good source of Ca, Mg, P and Zn, and jak leaves a good source of Ca and Na. Within grasses, guinea contains appreciable quantity of available Mg and P. Rice bran is rich in available Mg, P and Zn.
