Molecular Cloning of $\beta$-Galactosidase Gene from Neisseria lactamica 2118 into Escherichia coli MC 1061 (Neisseria lactamica 2118의 $\beta$-galactosidase 유전자의 대장균으로의 클로닝)

  • The gene coding for $\beta$-galactosidase of Neisseria lactamica 2118 was cloned into Escherichia coli MC 1061. The isolated 6.5 Kb EcoR I fragement and 7.2 Kb BamH I fragment of chromosomal DNA in Southern hybridization were ligated to a vector plasmid pBR322 and then transformed into Escherichia coli MC 1061 cells. Finally, I obtained three clones as $\beta$-galactosidase positive clone by colony hybridization and Southern hybridization($\beta$-galactosidase probe: lac Z gene of pMC1871). Three recombinant plasmids(pNL.13. 17 and 24) were found to contain the 7.2Kb BamH I fragment originated from Neisseria lactamica 2118 chromosomal DNA by Southern hybridization and pNL 24 was showed high homology to probe especially and also its physical map was constructed.

Soil Respiration in Pinus densiflora, Quercus variabilis and Platycarya strobilacea Stands in Jinju, Gyeongnam Province (경남 진주지역 소나무, 굴참나무 및 굴피나무림의 토양호흡에 관한 연구)

  • Soil respiration rate was measured from March to November 2003 using the KOH absorption method in Pinus densiflora, Quercus variabilis, Platycarya strobilacea stands in Jinju, Gyeongnam Province. Throughout the study period, average soil temperature and moisture content were 16.2$^{\circ}C$, 25.1% for P. densiflora stand, 17.1$^{\circ}C$, 24.3% for Q. variabilis stand, and 17.6$^{\circ}C$, 25.1% for P. strobilacea stand, respectively. The seasonal fluctuations of soil respiration rate increasing in summer and decreasing in winter, which there were strong positive correlations of soil respiration and soil temperature in all study stands. However, there were no significant correlations between soil moisture and soil respiration. Soil respiration rates throughout the study period ranged from 0.12 to 0.77 for P. densiflora stand, 0.23 to 1.37 for Q. valiabilis stand, and 0.30 to 1.47 g $CO_2\cdotm^{-2}\cdothr^{ -1}$ for P. strobilacea stand, respectively. Mean soil respiration rates in P. densiflora, Q. variabilis, P. strobilacea stands were 0.43, 0.80, and 0.90 g $CO_2\cdotm^{-2}\cdothr^{ -1}$, respectively. The Q$_{10}$ values were 2.38 for P. densiflora stand, 2.11 for Q. variabilis stand, and 2.07 for P. strobilacea stand. Annual total soil respiration was 24 for P. densiflora stand, 49.3 for Q. variabilis stand, and 55.3 t $CO_2\cdotha^{-1}\cdotyr^{ -1}$ for P. strobilacea stand, respectively.y.

INHIBITORY EFFECT OF LACTOCOCCUS LACTIS 1370 ON THE FORMATION OF DENTAL PLAQUE IN CHILDREN (소아에서 Lactococcus lactis 1370에 의한 치태형성 억제 효과)

  • This study was undertaken to evaluate the clinical effect of inhibiting plaque formation of Lactococcus lactis 1370, a acid producing bacterium residing in the mouth. 30 children were asked to use 10ml of control mouth-wash and mouthwash containing Lactococcus lactis 1370. The plaque index and plaque area rate at 24h and 48h after the use of the mouthwashes were measured. And the number of Lactococcus lactis 1370 was counted at 1h, 3h, and 6h in the mouth. The results are as follow. 1. The mean plaque index at 24h after the use of the control mouthwash and the mouthwash containing Lactococcus lactis 1370 were 2.43 and 2.06, respectively. The inhibiting rate of plaque formation was 15% (P<0.05). 2. The mean plaque index at 48h after the use of the control mouthwash and the mouthwash containing Lactococcus lactis 1370 were 2.95 and 2.17, respectively. The inhibiting rate of plaque formation was 26%, showing more decrease than at 24h(P<0.05). 3. The mean plaque area rate at 24h after the use of the control mouthwash and the mouthwash containing Lactococcus lactis 1370 were 21.2% and 15.6%, respectively. The inhibiting rate of plaque formation was 26% (P<0.05). 4. The mean plaque area rate at 48h after the use of the control mouthwash and the mouthwash containing Lactococcus lactis 1370 were 33.0% and 17.8%, respectively. The inhibiting rate of plaque formation was 46% (P<0.05). 5. The number of Lactococcus lactis 1370 in the mouth decreased significantly from mouthwashing to 3h, but increased slightly between 3h and 6h. As seen with the above results, we think that using the mouth wash with Lactococcus lactis 1370 would prevent the formation of plaque in the mouth and can be an effective method to prevent dental caries and periodontal disease.

Effect of Pre-Fermentation Condition on Quality Characteristics of Small Packaged Cabbage Kimchi (소포장 양배추김치의 품질특성에 미치는 전 발효조건의 영향)

  • This study was carried out to investigate commercialization of Kimchi made of cabbage (Brassica oleracea var. capitata L.) using pre-fermentation conditions. The pre-fermentation conditions were 0, 18, 24, and 28 h at $20^{\circ}C$, and then the samples were stored at $10^{\circ}C$ to assess changes in quality characteristics. A comparison of the quality characteristics during storage showed that PF24 (pre-fermented cabbage Kimchi during 24 h at $20^{\circ}C$) and PF28 (pre-fermented cabbage Kimchi during 28 h at $20^{\circ}C$) had pH 4.47 and pH 4.23 on the second day of storage, respectively. It was possible to shorten the fermentation time to less than that of PF0 (not pre-fermented cabbage Kimchi at $20^{\circ}C$), by approximately 3 days. Total acidity was 0.26 to 0.29% immediately after making the Kimchi. However, PF0, PF18 (pre-fermented cabbage Kimchi during 18 h at $20^{\circ}C$), PF24 and PF28 became well-fermented when they were stored for 8~14 days, 3~10 days or 2~3 days. The number of lactic acid bacteria increased with the passage of time in all treatment groups regardless of fermentation conditions. However, the longer pre-fermentation time became, the faster the number of lactic acid bacteria increased. Most samples showed similar results late in the storage period; 7.2~7.4 log CFU/mL. PF0 had the greatest volume change 2.1 times increase late in the storage period. The sensory evaluation showed significant differences for flavor, taste, and overall acceptability after a partial storage period. PF28 stored for 2~3 days showed excellent flavor, and PF24 and PF28 stored for 2~3 days showed the highest scores of 6.27 to 6.67. The PF24 and PF28 treated samples were appropriate for commercializing small packed cabbage Kimchi and for alleviating the expansion problem of the packing material. However, because mass commercial production requires a large number of samples to be used at once, the results should be assessed for industrial product development in the future.

Effect of pH on Hydrogen Fermentation of Food Waste with Livestock Wastewater (음식물쓰레기 수소발효 시 pH 영향 및 축산폐수와의 혼합 발효)

  • In the modern industrial society, huge amount of organic wastes have exceeded the society's self-cleaning capability, caused pollution of the whole environment, including water quality, soil, and the air, and become a big burden of waste treatment. Moreover, the emission of green house gases brought by the continual combustion of fossil fuels has facilitated the global warming. The simultaneous effect of initial and operational pH on $H_2$ yield was expressed using mathematical equation and optimized. The optimal initial and cultivation pH was 7.50 and 6.01, respectively. Addition of livestock wastewater to food waste substantially decreased the amount of alkali requirement and also improved the $H_2$ fermentation performance.

Taxonomic Status of Siberian Flying Squirrel from Korea (Pteromys volans aluco Thomas 1907)

  • Sequences of mitochondrial DNA (mtDNA) cytochrome b gene (1,140 bp) and control region (803 bp) of Siberian flying squirrels from Korea (Pteromys volans aluco) and Mt. Changbai of northeast China (P. v. arsenjevi) were obtained to reexamine the taxonomic status of the Korean subspecies. In the cytochrome b gene, six haplotypes of P. v. aluco formed a clade with six haplotypes of P. v. arsenjevi, and in control region, seven haplotypes of P. v. aluco formed a clade with six haplotypes of P. v. arsenjevi. Furthermore, six haplotypes of cytochrome b gene of P. v. aluco from this study formed a clade with four haplotypes of P. v. arsenjevi in far-east Russia obtained from GenBank. We also investigated the research papers previously published that reported the length of tail vertebrae of P. volans, and found that the length was not sufficiently large as to be a key character of P. v. aluco. This result is not consistent with morphological description for its haplotype. Therefore, we conclude that P. v. aluco from Korea might possibly be a synonym of P. v. arsenjevi from northeast China and nearby Russia.

Antigenotoxicity of Ginseng Petroleum Ether Extract and its Action Mechanism (인삼 지용성성분인 유전독성억제효과와 작용기전)

  • Panax ginseng C.A. Meyer has been extensively used in the traditional oriental medicine as a restorative, tonic and prophylatic agent. Petroleum ether extract of panax ginseng C.A. Meyer (GPE) and its several fractions (PI-P5) were tested for the evaluation of antigenotoxicity against N-methyl-N-nitrosourea (MNU) and benzo(a)pyrene [B(a)P]-induced micronucleated reticulocytes in mouse peripheral blood. GPE and P2 showed more significant anticlastogenicity than other fractions did. To elucidate the anticlastogenic action mechanism of GPE and P2 against B(a)P, the alteration of B(a)P metabolism was studied. GPE and P2 inhibited B(a)P metabolism in the presence of 8-9 mix and decreased B(a)P-DNA binding in calf thymus DNA with 8-9 mix. They also decreased [$^3H$] MNU induced DNA binding and methylation to 7-methyl guanine and $O^{6}-methyl$ guanine adducts in calf thymus DNA by RPLC analysis. These results suggest that the anticlastogenicity of GPE and P2 on the B(a)P or MNU-induced clastogenicity is due to decrease of DNA binding with B(a)P or MNU, the inhibition of metabolism with B(a)P and the inhibition of methylation in DNA. Therefore, GPE and P2 may be useful chemopreventive agents of alkylating agent like MNU and secondary carcinogen like B(a)P.

Aquatic Plants for Wastewater Treatment (수생식물을 이용한 수질정화에 관한 연구)

  • Water parsley(Oenanthe javanica(Blume) DC) was raised with varying population density(S) in the laboratory aquarium unit to determine the growth equation. The population density was measure after 7 days. The resultant growth curve was well fit to the equation 1/S = A+B (1/S0) with a high correlation coefficient ($R^2$ = 0.999). The maximum specific absorption rate was $9.011 \times 10^{-5}$ kg $NO_x-N/kg$ water parsley$\cdot$day and $1.31 \times 10^{-5}$ kg $PO_4-P/kg$ water parsley$\cdot$day when the average population density was $2.62 kg/m^2$. The relationship between population density and nutrient absorption rate, the absorption rate of $NO_x-N$ was 5.04~5.24 mg/l$\cdot$day when the population density was $7.51~10.0 $mg/m^2\cdot day$ and the absorption rate of $PO_4-P$ was 0.56~0.78 mg/l$\cdot$day when the population density was 5.02~10.0 $kg/m^2\cdot day$. Taking into account the nutrient absorption rate and growth rate, the population density between $7.0 kg/m^2\cdot day$ and $8.0 kg/m^2 \cdot day$ was selected. The removal rate of nutrient was investigated after 7 days culture. Removal rate of $NO_x-N$ was 95.6~99.95% with initial concentration of 35 mg $NO_x-N/l$, and the removal rate of $PO_4-P$ was also high, indicating 80.24~98.9% with initial concentration of 5.95 mg $PO_x-P/l$.

Studies on the structure and expression of penicillin G acylase gene I (Penicillin G acylase 유전자의 구조와 발현기작에 관한 연구 I)

  • The penicillin G acylase(pga) gene was cloned in the vector plasmid pKM $300(Ar^r,\;Tc^r,\;6.33kb)$ for the study of the structure and expression of the pga gene. This recombinant plasmid pPAKS-1 DNA(24.5 Kb) was cleaved into 2 fragments by restriction enzyme Eco R1.1fragment by BamH1, 4fragments by Hind III, and 2 fragments by Pst I. The pga gene was located on the Eco R1.Hind III-C fragement of pPAKS-1. The recombinant plasmids pPAKS-1 and pPAKS-2, in which the Hind III-B and Hind III-D fragments pPAKS-1 are deleted, are characterized. The results are summarized as follows : 1. Doubling times of bacterial strain bearing pPAKS-1 and pPAKS-2 are 90 and 60 minutes, respectively. 2. pPAKS-1 and pPAKS-2 are present at about 16-32 and 70 copies per cell, respectively, are 0.66 and 5.5 units, respectively, which represent 2-fold and 20-fold higher enzyme 4. pPAKS-1 is very unstable, but pPAKS-2 is stable.

Optimization of Batch Expression of Sesame Oil (참기름의 회분식 착유의 최적화)

  • In order to optimize the batch expression of sesame oil, recovery of expressed oil(REO) from roasted and unroasted sesame seeds were observed at different temperature, pressure, pressing duration and moisture content, and relatinship between REO and effects of expression factors were analysed. REO was high at 2.5~4.5% moisture content, 30~$50^{\circ}C$ and 30~50MPa, and decreased abruptly with increasing moisture content above 4.5%. The optimum temperature, pressure, pressing duration and moisture content were $40.1^{\circ}C$, 54.4MPa, 21.7min and 1.3% for unroasted seeds and $44.4^{\circ}C$, 37.8MPa, 14.4min and 2.52% for roasted seeds, respectively. REO in optimum condition was 84.6% in unroasted seed and 81.7% in roasted seed. From the statistic analysis between effects of expression factors and REO, importance of their effects was decreased in the order of moisture content, pressure, temperature and pressing duration. And also interaction effects were high in $pressure{\times}moisture$ content, $temperature{\times}moisture$ content and temperature pressure. The multiple regression equation between REO(Y) and temperature(T), pressure(P), moisture content(M), and pressing duration(D) were as follows ; $Y=18.20$ $35.66P$$24.52M-4.45P^{2}-1.20TM-4.02PM-6.62M^{2},\;r^{2}=0.89$, for unrosated sesame seed, $Y=117.93$$16.40P-58.61M-2.75P^{2}$$1.79TM-1.65PM$$7.16M^{2},\;r^{2}$$=0.91$ for roasted sesame seed.
